Abstract:Hydrogen sulfide (H2S) has recently been considered as a crucial gaseous transmitter occupying extensive roles in physiological and biochemical processes throughout the life of plant species. Furthermore, plenty of achievements have been announced regarding H2S working in combination with other signal molecules to mitigate environmental damage, such as nitric oxide (NO), abscisic acid (ABA), calcium ion (Ca2+), hydrogen peroxide (H2O2), salicylic acid (SA), ethylene (ETH), jasmonic acid (JA), proline (Pro), and melatonin (MT). This review summarizes the current knowledge within the mechanism of H2S and the above signal compounds in response to abiotic stresses in plants, including maintaining cellular redox homeostasis, exchanging metal ion transport, regulating stomatal aperture, and altering gene expression and enzyme activities. The potential relationship between H2S and other signal transmitters is also proposed and discussed.

Abstract:Hydrogen sulfide (H2S) nowadays is regarded as a novel gaseous signaling molecule in plants during environmental stress response. The spermidine (Spd), an important plant growth regulator, plays critical roles in regulating stress tolerance in plant species. Objectives of this study were to examine whether Spd could act as a H2S signaling activator to regulate stress-responsive transcription factors (TFs) and antioxidant defense and to further explore the possible crosstalk among Spd-induced hydrogen peroxide (H2O2), nitric oxide (NO), and H2S signaling in leaves of white clover. Results showed that dehydration or exogenous application of Spd could significantly increase L/D-cysteine desulfhydrate (L/DCD) activities and then cause a quick accumulation of H2S, followed by significant improvement of antioxidant enzyme (SOD, CAT, GPDX, APX, GR, DHAR, and MDHAR) activities and transcript levels of TFs (bZIP37, bZIP107, DREB2, DREB4, and WRKY108715) and genes encoding antioxidant enzymes. The inhibitors (AOA and NH2OH) of L/DCD activities could effectively attenuate dehydration- or Spd-induced these effects in leaves. Additionally, dehydration-induced increase in H2S accumulation, L/DCD and antioxidant enzyme activities, and genes expression were remarkably abolished by the application of Spd biosynthesis inhibitor (DCHA), whereas exogenous Spd significantly alleviate the inhibitory effect of DCHA on these parameters under dehydration condition. This indicated that H2S which was derived from L/DCD pathway might act as a downstream signaling of Spd, and Spd-activated H2S signaling was involved in dehydration-regulated TFs and antioxidant defense associated with dehydration tolerance. Further analyses found that Spd-induced H2O, production could not be attenuated by scavengers of NO and H2S, but the scavenger of H2O2 effectively inhibited Spd-induced increases in NO and H2S. The Spd-induced H2S signaling was also substantially inhibited by the application of the NO scavenger. The result implied that Spd-induced H2O2 could be an upstream signal molecule of NO and H2S, and the Spd-induced H2S might act as the downstream signaling of NO in leaves. These obtained results provide the basic evidence to understand Spd-regulated signal transduction in white clover.
Elucidating the downstream pathways triggered by H2S signaling in Arabidopsis thaliana under drought stress via transcriptome analysis

Abstract:Hydrogen sulfide (H 2 S) is a crucial signaling molecule in plants. Recent studies have shown that H 2 S plays an equally important role as nitric oxide (NO) and hydrogen peroxide (H 2 O 2 ) in plant signaling. Previous studies have demonstrated the involvement of H 2 S in regulating drought and other stressful environmental conditions, but the exact downstream molecular mechanisms activated by the H 2 S signaling molecule remain unclear. In this study, we conducted a comprehensive genome-wide transcriptomic analysis of both wild type (WT) and double mutant ( lcd/des1 ). Arabidopsis thaliana plants were exposed to 40% polyethylene glycol (PEG) to induce drought stress and 20 μM sodium hydrosulfide (NaHS). The resulting transcriptome data were analyzed for differentially significant genes and their statistical enrichments in the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways. The results indicated significant upregulation of genes related to photosynthesis, carbon fixation, plant secondary metabolite biosynthesis, inositol and phosphatidylinositol signaling pathways, and stress-responsive pathways in mutant plants under drought stress. Mutant plants with impaired H 2 S signaling mechanisms displayed greater susceptibility to drought stress compared to wild-type plants. In summary, all findings highlight the pivotal role of H 2 S signaling in stimulating other drought-responsive signaling pathways.

Abstract:For a long time, hydrogen sulfide (H2S) has been considered as merely a toxic by product of cell metabolism, but nowadays is emerging as a novel gaseous signal molecule, which participates in seed germination, plant growth and development, as well as the acquisition of stress tolerance including cross-adaptation in plants. Cross-adaptation, widely existing in nature, is the phenomenon in which plants expose to a moderate stress can induce the resistance to other stresses. The mechanism of cross-adaptation is involved in a complex signal network consisting of many second messengers such as Ca2+, abscisic acid, hydrogen peroxide and nitric oxide, as well as their crosstalk. The cross-adaptation signaling is commonly triggered by moderate environmental stress or exogenous application of signal molecules or their donors, which in turn induces cross-adaptation by enhancing antioxidant system activity, accumulating osmolytes, synthesizing heat shock proteins, as well as maintaining ion and nutrient balance. In this review, based on the current knowledge on H2S and cross-adaptation in plant biology, H2S homeostasis in plant cells under normal growth conditions; H2S signaling triggered by abiotic stress; and H2S-induced cross-adaptation to heavy metal, salt, drought, cold, heat, and flooding stress were summarized, and concluded that H2S might be a candidate signal molecule in plant cross-adaptation. In addition, future research direction also has been proposed.

Abstract:Throughout their whole life cycle, higher plants are often exposed to diverse environmental stresses, such as drought, salinity, heavy metals and extreme temperatures. In response to such stress, plant cells initiate signalling transduction, resulting in downstream responses, such as specific gene transcription and protein expression. Accumulating evidence has revealed that hydrogen sulphide (H2 S) serves as a signalling molecule in plant acclimation to stressful conditions. More important, H2 S interacts with other signalling molecules and phytohormones, contributing to transcriptional regulation and post-translational modification. Overall, the H2 S-mediated signalling pathway and its interaction with other signals remains elusive. Here, we describe the role of the H2 S signalling network in regulating physiological and molecular processes under various abiotic stresses.

Abstract:Hydrogen sulfide (H2S), once recognized only as a poisonous gas, is now considered the third endogenous gaseous transmitter, along with nitric oxide (NO) and carbon monoxide (CO). Multiple lines of emerging evidence suggest that H2S plays positive roles in plant growth and development when at appropriate concentrations, including seed germination, root development, photosynthesis, stomatal movement, and organ abscission under both normal and stress conditions. H2S influences these processes by altering gene expression and enzyme activities, as well as regulating the contents of some secondary metabolites. In its regulatory roles, H2S always interacts with either plant hormones, other gasotransmitters, or ionic signals, such as abscisic acid (ABA), ethylene, auxin, CO, NO, and Ca2+. Remarkably, H2S also contributes to the post-translational modification of proteins to affect protein activities, structures, and sub-cellular localization. Here, we review the functions of H2S at different stages of plant development, focusing on the S-sulfhydration of proteins mediated by H2S and the crosstalk between H2S and other signaling molecules.

Abstract:Hydrogen sulfide (H2S) was initially recognized as a toxic gas and its biological functions in mammalian cells have been gradually discovered during the past decades. In the latest decade, numerous studies have revealed that H2S has versatile functions in plants as well. In this review, we summarize H2S-mediated sulfur metabolic pathways, as well as the progress in the recognition of its biological functions in plant growth and development, particularly its physiological functions in biotic and abiotic stress responses. Besides direct chemical reactions, nitric oxide (NO) and hydrogen peroxide (H2O2) have complex relationships with H2S in plant signaling, both of which mediate protein post-translational modification (PTM) to attack the cysteine residues. We also discuss recent progress in the research on the three types of PTMs and their biological functions in plants. Finally, we propose the relevant issues that need to be addressed in the future research.

Abstract:Traditionally, hydrogen sulfide (H2S) has been thought to be a cytotoxin. Nowadays, H2S is emerging to be a multifunctional signaling molecule, involved in the overall process of life cycle of plants from seed germination to maturation and senescence, and response and adaptation of plants to environmental stress. Among adaptive mechanisms, cross-stress tolerance is one of the most important strategies for plants’ response to environmental stress, involved in stress perception, signaling transduction and integration, gene expression, and physiological and biochemical responses. In these processes, signaling transduction and integration is a key step in the formation of cross-stress tolerance, referring to an H2S-centered signaling network. In this chapter, the current advance of understanding on H2S in plants signaling and cross-stress tolerance to heat, cold, salt, drought, hypoxia/anoxia, and heavy metal stress is discussed. The subjective is to motivate the study of cross-stress tolerance mechanisms and establish the foundation of acquiring transgenic plants with cross-stress tolerance.
Elucidating the intricacies of the H2S signaling pathway in gasotransmitters: Highlighting the regulation of plant thiocyanate detoxification pathways

Abstract:In recent decades, there has been increasing interest in elucidating the role of sulfur-containing compounds in plant metabolism, particularly emphasizing their function as signaling molecules. Among these, thiocyanate (SCN-), a compound imbued with sulfur and nitrogen, has emerged as a significant environmental contaminant frequently detected in irrigation water. This compound is known for its potential to adversely impact plant growth and agricultural yield. Although adopting exogenous SCN- as a nitrogen source in plant cells has been the subject of thorough investigation, the fate of sulfur resulting from the assimilation of exogenous SCN- has not been fully explored. There is burgeoning curiosity in probing the fate of SCN- within plant systems, especially considering the possible generation of the gaseous signaling molecule, hydrogen sulfide (H2S) during the metabolism of SCN-. Notably, the endogenous synthesis of H2S occurs predominantly within chloroplasts, the cytosol, and mitochondria. In contrast, the production of H2S following the assimilation of exogenous SCN- is explicitly confined to chloroplasts and mitochondria. This phenomenon indicates complex interplay and communication among various subcellular organelles, influencing signal transduction and other vital physiological processes. This review, augmented by a small-scale experimental study, endeavors to provide insights into the functional characteristics of H2S signaling in plants subjected to SCN--stress. Furthermore, a comparative analysis of the occurrence and trajectory of endogenous H2S and H2S derived from SCN--assimilation within plant organisms was performed, providing a focused lens for a comprehensive examination of the multifaceted roles of H2S in rice plants. By delving into these dimensions, our objective is to enhance the understanding of the regulatory mechanisms employed by the gasotransmitter H2S in plant adaptations and responses to SCN--stress, yielding invaluable insights into strategies for plant resilience and adaptive capabilities.

Abstract:For the past 300 years, hydrogen sulfide (H 2 S) has been considered a toxic gas. Nowadays, it has been found to be a novel signaling molecule in plants involved in the regulation of cellular metabolism, seed germination, plant growth, development, and response to environmental stresses, including high temperature (HT) and low temperature (LT). As a signaling molecule, H 2 S can be actively synthesized and degraded in the cytosol, chloroplasts, and mitochondria of plant cells by enzymatic and non-enzymatic pathways to maintain homeostasis. To date, plant receptors for H 2 S have not been found. It usually exerts physiological functions through the persulfidation of target proteins. In the past 10 years, H 2 S signaling in plants has gained much attention. Therefore, in this review, based on that same attention, H 2 S homeostasis, protein persulfidation, and the signaling role of H 2 S in plant response to HT and LT stress were summarized. Also, the common mechanisms of H 2 S-induced HT and LT tolerance in plants were updated. These mechanisms involve restoration of biomembrane integrity, synthesis of stress proteins, enhancement of the antioxidant system and methylglyoxal (MG) detoxification system, improvement of the water homeostasis system, and reestablishment of Ca 2+ homeostasis and acid-base balance. These updates lay the foundation for further understanding the physiological functions of H 2 S and acquiring temperature-stress-resistant crops to develop sustainable food and agriculture.

Abstract:Hydrogen sulfide (H2S) serves as an important gaseous signaling molecule that is involved in intra- and intercellular signal transduction in plant-environment interactions. In plants, H2S is formed in sulfate/cysteine reduction pathways. The activation of endogenous H2S and its exogenous application has been found to be highly effective in ameliorating a wide variety of stress conditions in plants. The H2S interferes with the cellular redox regulatory network and prevents the degradation of proteins from oxidative stress via post-translational modifications (PTMs). H2S-mediated persulfidation allows the rapid response of proteins in signaling networks to environmental stimuli. In addition, regulatory crosstalk of H2S with other gaseous signals and plant growth regulators enable the activation of multiple signaling cascades that drive cellular adaptation. In this review, we summarize and discuss the current understanding of the molecular mechanisms of H2S-induced cellular adjustments and the interactions between H2S and various signaling pathways in plants, emphasizing the recent progress in our understanding of the effects of H2S on the PTMs of proteins. We also discuss future directions that would advance our understanding of H2S interactions to ultimately mitigate the impacts of environmental stresses in the plants.

Abstract:Hydrogen sulfide (H2S) is considered the third gas signal molecule in recent years. A large number of studies have shown that H2S not only played an important role in animals but also participated in the regulation of plant growth and development and responses to various environmental stresses. Waterlogging, as a kind of abiotic stress, poses a serious threat to land-based waterlogging-sensitive plants, and which H2S plays an indispensable role in response to. In this review, we summarized that H2S improves resistance to waterlogging stress by affecting lateral root development, photosynthetic efficiency, and cell fates. Here, we reviewed the roles of H2S in plant resistance to waterlogging stress, focusing on the mechanism of its promotion to gained hypoxia tolerance. Finally, we raised relevant issues that needed to be addressed.

Abstract:Hydrogen sulfide (H2S) is an important gaseous molecule in various plant developmental processes and plant stress responses. In this study, the transgenic Arabidopsis thaliana plants with modulated expressions of two cysteine desulfhydrases, and exogenous H2S donor (sodium hydrosulfide, NaHS) and H2S scavenger (hypotaurine, HT) pre-treated plants were used to dissect the involvement of H2S in plant stress responses. The cysteine desulfhydrases overexpressing plants and NaHS pre-treated plants exhibited higher endogenous H2S level and improved abiotic stress tolerance and biotic stress resistance, while cysteine desulfhydrases knockdown plants and HT pre-treated plants displayed lower endogenous H2S level and decreased stress resistance. Moreover, H2S upregulated the transcripts of multiple abiotic and biotic stress-related genes, and inhibited reactive oxygen species (ROS) accumulation. Interestingly, MIR393-mediated auxin signaling including MIR393a/b and their target genes (TIR1, AFB1, AFB2, and AFB3) was transcriptionally regulated by H2S, and was related with H2S-induced antibacterial resistance. Moreover, H2S regulated 50 carbon metabolites including amino acids, organic acids, sugars, sugar alcohols, and aromatic amines. Taken together, these results indicated that cysteine desulfhydrase and H2S conferred abiotic stress tolerance and biotic stress resistance, via affecting the stress-related gene expressions, ROS metabolism, metabolic homeostasis, and MIR393-targeted auxin receptors.

Abstract:Hydrogen sulfide (H(2)S) plays a crucial role in human and animal physiology. Its ubiquity and versatile properties have recently caught the attention of plant physiologists and biochemists. Two cysteine desulfhydrases (CDes), L-cysteine desulfhydrase and D-cysteine desulfhydrase, were identified as being mainly responsible for the degradation of cysteine in order to generate H(2)S. This study investigated the expression regulation of these genes and their relationship to drought tolerance in Arabidopsis. First, the expression pattern of CDes in Arabidopsis was investigated. The expression levels of CDes gradually increased in an age-dependent manner. The expression of CDes was significantly higher in stems and cauline leaves than in roots, rosette leaves and flowers. Second, the protective effect of H(2)S against drought was evaluated. The expression pattern of CDes was similar to the drought associated genes induced by dehydration, and H(2)S fumigation was found to stimulate further the expression of drought associated genes. Drought also significantly induced increased H(2)S production, a process that was reversed by re-watering. In addition, seedlings after treatment with NaHS (a H(2)S donor) showed a higher survival rate and displayed a significant reduction in the size of the stomatal aperture compared to the control. These findings provide evidence that H(2)S, as a gasotransmitter, improves drought resistance in Arabidopsis.

Abstract:Based on the research conducted so far, hydrogen sulphide (H2S) plays a crucial role in the development and stress resilience of plants. H2S, which acts as a signalling molecule, responds to different stresses such as heavy metals, drought, and salinity, and it regulates various aspects of plant growth and development including seed germination, root development, stomatal movement, flowering, and fruit ripening. Additionally, H2S is involved in mediating legume‐Rhizobium symbiosis signalling. It modulates plant responses to external environmental stimuli by interacting with other signalling molecules like phytohormones, nitric oxide, and reactive oxygen species. Furthermore, H2S exerts these regulations since it can modify protein functions through a reversible thiol‐based oxidative posttranslational modification called persulfidation, particularly in stress response and developmental processes. As a result, H2S is recognised as an important emerging signalling molecule with multiple roles in plants. Research in this field holds promise for engineering stress tolerance in crops and may lead to potential biotechnological applications in agriculture and environmental management.

Does a DVD improve compliance with home exercise programs for people who have sustained a traumatic hand injury? Results of a feasibility study

Abstract:Abstract Purpose: This feasibility study sought to determine if compliance and understanding of a home exercise program following a traumatic hand injury is improved when patients are provided with a DVD and a brochure when compared to using brochures only. Method: Patients who presented with a traumatic hand injury and commenced on a hand therapy protocol were randomly assigned into two groups. The control group received brochures while the experimental group were provided with exercise instructions on DVD as well as brochures. Compliance was measured through the use of exercise diaries, clinic attendance, a checklist to measure correctness and understanding of exercises and a follow-up survey. Results: No significant improvement was found in the mean exercise compliance score (p = 0.344) between the intervention and control groups. From the survey results almost half of all participants reported that pain interfered in their ability to perform their home exercises and a third acknowledged that time limited their ability to perform their exercises. Conclusion: Findings demonstrate the multidimensional nature of compliance. The provision of DVD technology, while not shown to cause a statistically significant change in overall compliance, did help improve understanding of exercises; as such DVDs could be utilised as part of a program that facilitates the patient--therapist relationship. Implications for Rehabilitation Limited time and level of pain are highlighted as reasons for non compliance with exercise and treatment programs. The use of DVDs can improve understanding and execution of exercises and can be part of a treatment program that facilitates increased patient therapist contact for rural and remote clients.

Firstly, production of H2S from different natural and artificial sources were discussed with its transformation from sulfur (S) to sulfate (SO42−) and then to sulfite (SO32−). The importance of different kinds of transporters that helps to take SO42− from the soil solution was presented. Mainly, these transporters are SULTRs (H+/SO42− cotransporters) and multigene family encodes them. Furthermore, these SULTRs have LAST (Low affinity transport proteins), HAST (High affinity transport proteins), vacuole transporters, and plastid transporters. Since it is well known that there is strong relationship between SO42− and synthesis of hydrogen sulfide or dihydrogen sulfide or sulfane in plant cells. Thus, cysteine (Cys) metabolism through which H2S could be generated in plant cell with the role of different enzymes has been presented. Furthermore, H2S in interaction with other molecules could help to mitigate biotic and abiotic stress. Based on this review work, it can be concluded that H2S has potential to induce cross-adaptation to biotic and abiotic stress; thus, it is recommended that it should be considered in future studies to answer the questions like what are the receptors of H2S in plant cell, where in plants the physiological concentration of H2S is high in response to multiple stress and how it induces cross-adaptation by interaction with other signal molecules.

Abstract:Hydrogen sulfide (H2S) is regarded as a “New Warrior” for managing plant stress. It also plays an important role in plant growth and development. The regulation of root system architecture (RSA) by H2S has been widely recognized. Plants are dependent on the RSA to meet their water and nutritional requirements. They are also partially dependent on the RSA for adapting to environment change. Therefore, a good understanding of how H2S affects the RSA could lead to improvements in both crop function and resistance to environmental change. In this review, we summarized the regulating effects of H2S on the RSA in terms of primary root growth, lateral and adventitious root formation, root hair development, and the formation of nodules. We also discussed the genes involved in the regulation of the RSA by H2S, and the relationships with other signal pathways. In addition, we discussed how H2S regulates root growth in response to abiotic stress. This review could provide a comprehensive understanding of the role of H2S in roots during development and under abiotic stress.

Abstract:Over the past decade, a plethora of research has illuminated the multifaceted roles of hydrogen sulfide (H 2 S) in plant physiology. This gaseous molecule, endowed with signaling properties, plays a pivotal role in mitigating metal-induced oxidative stress and strengthening the plant's ability to withstand harsh environmental conditions. It fulfils several functions in regulating plant development while ameliorating the adverse impacts of environmental stressors. The intricate connections among nitric oxide (NO), hydrogen peroxide (H 2 O 2 ), and hydrogen sulfide in plant signaling, along with their involvement in direct chemical processes, are contributory in facilitating post-translational modifications (PTMs) of proteins that target cysteine residues. Therefore, the present review offers a comprehensive overview of sulfur metabolic pathways regulated by hydrogen sulfide, alongside the advancements in understanding its biological activities in plant growth and development. Specifically, it centres on the physiological roles of H 2 S in responding to environmental stressors to explore the crucial significance of different exogenously administered hydrogen sulfide donors in mitigating the toxicity associated with heavy metals (HMs). These donors are of utmost importance in facilitating the plant development, stabilization of physiological and biochemical processes, and augmentation of anti-oxidative metabolic pathways. Furthermore, the review delves into the interaction between different growth regulators and endogenous hydrogen sulfide and their contributions to mitigating metal-induced phytotoxicity.

Abstract:Hydrogen sulfide regulates essential plant processes, including adaptation responses to stress situations, and the best characterized mechanism of action of sulfide consists of the posttranslational modification of persulfidation. In this study, we reveal the first persulfidation proteome described in rice including 3443 different persulfidated proteins that participate in a broad range of biological processes and metabolic pathways. In addition, comparative proteomics revealed specific proteins involved in sulfide signaling during drought responses. Several proteins involved in the maintenance of cellular redox homeostasis, the TCA cycle and energy-related pathways, and ion transmembrane transport and cellular water homeostasis, highlighting the aquaporin family, showed the highest differential levels of persulfidation. We revealed that water transport activity is regulated by sulfide which correlates to an increasing level of persulfidation of aquaporins. Our findings emphasize the impact of persulfidation on total ATP levels, fatty acid composition, ROS levels, antioxidant enzymatic activities, and relative water content. Interestingly, the persulfidation role on aquaporin transport activity as an adaptation response in rice differs from the current knowledge in Arabidopsis, which emphasizes the distinct role of sulfide improving rice tolerance to drought.
