Abstract:Resistance to anoikis allows cancer cells to survive during systemic circulation; however, the mechanism underlying anoikis resistance remains unclear. Abstract:Objectives:As a zinc-dependent proteinase family member, A Disintegrin and Metalloproteinase 9 (ADAM9) has the metalloproteinase domain that performs proteolytic function to liberate or shed factors, such as bFGF, VEGF, EGF, TGF-α and IGF, from cell surface and make them more accessible to tumor cells. It was shown in previous studies that the upregulation of ADAM9 in breast cancer is associated with metastasis.In this study, we examined the effect of ADAM9 on the cell proliferation, migration and invasion and elucidated the molecular mechanism in TNBC. We hypothesize that ADAM9 sheds Tumor Necrosis Factor alpha (TNF-α), which activates TNF receptor and then triggers PI3K-dependent NF-kappa B pathway, affecting metastatic phenotypes of triple negative breast cancer (TNBC).Materials and Methods:Correlation of the levels of ADAM9 expression and survival time in triple negative breast cancer patients was analysed by the Kaplan-Meier (KM) plotter integrative data analysis tool. The TNBC subjects were divided into two groups of high and low ADAM9 expression levels with respect to the median. We also compared the protein expression levels of ADAM9 in a highly aggressive, invasive and poorly differentiated TNBC cell line, MDA-MB-231, and a non-tumorigenic epithelial cell line, MCF-10A.ADAM9 was knocked down in MDA-MB-231 using siRNA. In comparison to negative and blank controls, we examined the cell proliferation, migration and invasion, and the phosphorylation levels of AKT subject to ADAM9 knockdown. We also profiled the mRNA expression levels of ADAM9 and NF-kappa B in breast cancer patients.Results:As shown in Figure 1(a), the survival curves of these two groups were plotted and compared using Kaplan-Meier survival analysis, indicating that lower ADAM9 expression is linked to better survival rate (p=0.0038). Figure 1(b) illustrates significantly higher ADAM9 protein level in MDA-MB-231, compared with MCF-10A (p=0.0019).It is shown in Figure 2 and Figure 3(a) that the cell proliferation, migration and invasion, and the phosphorylation of AKT are significantly suppressed by ADAM9 knockdown in the aggressive TNBC cell line. Figure 3(b) shows significant association between ADAM9 and NF-kappa B in the mRNA expression level (p<0.001).Conclusions:The results clearly showed the activation of PI3K-dependent NF-kappa B pathway triggered by ADAM9. Abstract:PURPOSE:To investigate the influence of p75NTR ICD on anoikis resistance ability of tumor cells based on generation of an anoikis cell model and discuss the activation of related signaling pathways.METHODS:Immunohistochemistry was used to detect p75NTR ICD expression in lymph node metastatic and non-lymph node metastatic,primary specimens of oral squamous cell carcinoma (OSCC).Westem blot was used to detect the expression of p75NTR ICD in a pair of low-and high-metastatic potential OSCC cell lines.Poly-HEMA was used to pre-coat the cell culture dish in order to generate anoikis cell model.Over-expression of p75NTR ICD was conducted in tumor cells with plasmid transfection.Flow cytometry was used to detect apoptosis level of tumor cells after suspension culture.Laser scanning confocal microscopy analysis and Western blot were performed to detect the activation of related signal pathways of tumor cells in anoikis cell model.SPSS 22.0 software package was used for statistical analysis.RESULTS:The positive staining of p75NTR ICD in lymph node metastatic OSCC cells was located in the cytoplasm,while on the cytomembrane in nonlymph node metastatic OSCC.The expression of p75NTR ICD was much higher in HN-12,which was more metastatic as well,compared with HN-4.Larger sphere was formed in HN-12 anoikis cell model than in HN-4,lower apoptosis level of HN-12 in anoikis cell model was observed than in HN-4.NF-κB signaling was activated in HN-12 anoikis cell model,while not in HN-4.CONCLUSIONS:Cellular localization of p75NTR ICD in tumor cells was related to cervical lymph node metastasis.Over expression of p75NTR ICD in tumor cells shows higher anoikis resistance;p75NTR ICD promotes anoikis resistance through activating NF-κB signaling.

Abstract:ADAM10 is a metalloproteinase that regulates invasiveness in many tumors. Here, we found that ADAM10 expression correlates with the invasiveness of pituitary adenomas and contributes to invasion by cleaving L1 and CD44. In high-grade pituitary adenoma patients, ADAM10 expression levels were found to be elevated compared with low-grade pituitary adenomas. In a phorbol 12-myristate 13-acetate (PMA)-stimulated pituitary adenoma cell line, AtT-20 cells, we found that the cleavage of L1 was correspondingly enhanced with the increased interaction between Src and Shc. Increases in PMA-induced L1 cleavage and the phosphorylation of residue 418 of Src (418Src) were promoted by overexpression of ADAM10. Inversely, knockdown of Adam10 suppressed PMA-induced L1 cleavage and the phosphorylation of Src, which was blocked by the Src inhibitor PP2 and the MEK inhibitor PD98059. On the other hand, calcium flux activation in AtT-20 cells resulted in increased CD44 cleavage, with reduction of the interaction between calmodulin and ADAM10. The induction of enhanced CD44 cleavage by calcium flux activation was inhibited by knockdown of Adam10. In addition, Adam10 knockdown repressed AtT-20 cell migration, which was reversed by CD44EXT (CD44 ectodomain cleavage). Abstract:Tumor necrosis factor-alpha (TNFalpha), a potent proinflammatory cytokine, is released from cells by proteolytic cleavage of a membrane-anchored precursor. The TNF-alpha converting enzyme ( TACE; a disintegrin and metalloprotease17; ADAM17) is known to have a key role in the ectodomain shedding of TNFalpha in several cell types. However, because purified ADAMs 9, 10, and 19 can also cleave a peptide corresponding to the TNFalpha cleavage site in vitro, these enzymes are considered to be candidate TNFalpha sheddases as well. In this study we used cells lacking ADAMs 9, 10, 17 ( TACE), or 19 to address the relative contribution of these ADAMs to TNFalpha shedding in cell-based assays. Our results corroborate that ADAM17, but not ADAM9, - 10, or - 19, is critical for phorbol ester- and pervanadate-stimulated release of TNFalpha in mouse embryonic fibroblasts. However, overexpression of ADAM19 increased the constitutive release of TNFalpha, whereas overexpression of ADAM9 or ADAM10 did not. This suggests that ADAM19 may contribute to TNFalpha shedding, especially in cells or tissues where it is highly expressed. Furthermore, we used mutagenesis of TNFalpha to explore which domains are important for its stimulated processing by ADAM17. We found that the cleavage site of TNFalpha is necessary and sufficient for cleavage by ADAM17. In addition, the ectodomain of TNFalpha makes an unexpected contribution to the selective cleavage of TNFalpha by ADAM17: it prevents one or more other enzymes from cleaving TNFalpha following PMA stimulation. Thus, selective stimulated processing of TNFalpha by ADAM17 in cells depends on the presence of an appropriate cleavage site as well as the inhibitory role of the TNF ectodomain toward other enzymes that can process this site.

Abstract:Ectodomain shedding mediated by tumor necrosis factor-alpha (TNF-alpha)-converting enzyme [TACE; also known as ADAM17 (a disintegrin and metalloproteinase 17)] provides an important switch in regulating cell proliferation, inflammation, and cancer progression. TACE-mediated ectodomain cleavage is activated by signaling of the mitogen-activated protein kinases (MAPKs) p38 and ERK (extracellular signal-regulated kinase). Here, we found that under basal conditions, TACE was predominantly present as dimers at the cell surface, which required its cytoplasmic domain and enabled efficient association with tissue inhibitor of metalloproteinase-3 (TIMP3) and silencing of TACE activity. Upon activation of the ERK or p38 MAPK pathway, the balance shifted from TACE dimers to monomers, and this shift was associated with increased cell surface presentation of TACE and decreased TIMP3 association, which relieved the inhibition of TACE by TIMP3 and increased TACE-mediated proteolysis of transforming growth factor-alpha. Thus, cell signaling altered the dimer-monomer equilibrium and inhibitor association to promote activation of TACE-mediated ectodomain shedding, a regulatory mechanism that may extend to other ADAM proteases.
Direct Activation of TACE-Mediated Ectodomain Shedding by P38 MAP Kinase Regulates EGF Receptor-Dependent Cell Proliferation

Abstract:Inflammatory stimuli activate ectodomain shedding of TNF-alpha, L-selectin, and other transmembrane proteins. We show that p38 MAP kinase, which is activated in response to inflammatory or stress signals, directly activates TACE, a membrane-associated metalloprotease that is also known as ADAM17 and effects shedding in response to growth factors and Erk MAP kinase activation. p38alpha MAP kinase interacts with the cytoplasmic domain of TACE and phosphorylates it on Thr(735), which is required for TACE-mediated ectodomain shedding. Activation of TACE by p38 MAP kinase results in the release of TGF-alpha family ligands, which activate EGF receptor signaling, leading to enhanced cell proliferation. Conversely, depletion of p38alpha MAP kinase activity suppresses EGF receptor signaling and downstream Erk MAP kinase signaling, as well as autocrine EGF receptor-dependent proliferation. Autocrine EGF receptor activation through TACE-mediated ectodomain shedding intimately links inflammation and cancer progression and may play a role in stress and conditions that relate to p38 MAP kinase activation.

Abstract:Nerve growth factor (NGF) and its high-affinity receptor TRKA are overexpressed in epithelial ovarian cancer (EOC) displaying a crucial role in the disease progression. Otherwise, NGF interacts with its low-affinity receptor P75, activating pro-apoptotic pathways. In neurons, P75 could be cleaved by metalloproteinases (α and γ-secretases), leading to a decrease in P75 signaling. Therefore, this study aimed to evaluate whether the shedding of P75 occurs in EOC cells and whether NGF/TRKA could promote the cleavage of the P75 receptor. The immunodetection of the α-secretase, ADAM17, TRKA, P75, and P75 fragments was assessed by immunohisto/cytochemistry and Western blot in biopsies and ovarian cell lines. The TRKA and secretases' inhibition was performed using specific inhibitors. The results show that P75 immunodetection decreased during EOC progression and was negatively correlated with the presence of TRKA in EOC biopsies. NGF/TRKA increases ADAM17 levels and the fragments of P75 in ovarian cells. This effect is abolished when cells are previously treated with ADAM17, γ-secretase, and TRKA inhibitors. These results indicate that NGF/TRKA promotes the shedding of P75, involving the activation of secretases such as ADAM17. Since ADAM17 has been proposed as a screening marker for early detection of EOC, our results contribute to understanding better the role of ADAM17 and NGF/TRKA in EOC pathogenesis, which includes the NGF/TRKA-mediated cleavage of P75.
An Essential Role of Metalloprotease-Disintegrin ADAM12 in Triple-Negative Breast Cancer

Abstract:In the absence of HER2 overexpression, triple-negative breast cancers (TNBCs) rely on signaling by epidermal growth factor receptor (EGFR/ErbB1/HER1) to convey growth signals and stimulate cell proliferation. Soluble EGF-like ligands are derived from their transmembrane precursors by ADAM proteases, but the identity of the ADAM that is primarily responsible for ligand release and activation of EGFR in TNBCs is not clear. Using publicly available gene expression data for patients with lymph node-negative breast tumors who did not receive systemic treatment, we show that ADAM12L is the only ADAM with an expression level significantly associated with decreased distant metastasis-free survival times. Similar effect was not observed for patients with ER-negative non-TNBCs. There was a positive correlation between ADAM12L and HB-EGF and EGFR in TNBCs, but not in ER-negative non-TNBCs. We further demonstrate that ectopic expression of ADAM12L increased EGFR phosphorylation in a mouse intraductal xenograft model of early breast cancer. Finally, we detect strong correlation between the level of anti-ADAM12L and anti-phospho-EGFR immunostaining in human breast tumors using tissue microarrays. These studies suggest that ADAM12L is the primary protease responsible for the activation of EGFR in early stage, lymph node-negative TNBCs. Thus, our results may provide novel insight into the biology of TNBC.

Abstract:The membrane-anchored metalloproteinase tumor necrosis factor-alpha-converting enzyme (TACE/a disintegrin and metalloproteinase [ADAM] 17) is key in proteolytic ectodomain shedding of several membrane-bound growth factors, cytokines and receptors. The expression and activity of ADAM17 increases under some pathological conditions including stroke, and promotes neural progenitor cell migration and contributes to stroke-induced neurogenesis. Hypoxia initiates cellular invasive processes that occur under both physiological and pathological conditions such as invasion and metastasis of some tumors. In the present study, we sought to elucidate whether ADAM17 contributes to brain tumor invasion. To this end, we examined the role of ADAM17 in the invasiveness of two different brain tumor cell lines, 9L rat gliosarcoma and U87 human glioma, under normoxic and hypoxic conditions. Additionally, we tested the effects of ADAM17 suppression on in vitro tumor cell invasion by means of ADAM17 proteolytic inhibitors and specific small interfering RNA. We found that tumor cells upregulated ADAM17 expression under hypoxia, and that ADAM17 activity correlated with increased tumor cell invasion. Conversely, suppression of ADAM17 proteolysis decreased invasiveness induced by hypoxia in 9L and U87 cells. Furthermore, the contribution of ADAM17 to tumor invasion was independent of matrix metalloproteinase (MMP)-2 and MMP-9 activity. ADAM17 was also found to activate the epidermal growth factor/phosphoinositide-3 kinase/serine/threonine kinase signal transduction pathway. Our data suggest that hypoxia-induced ADAM17 contributes to glioma cell invasiveness through activation of the EGFR signal pathway.
