Abstract:Resistance to anoikis allows cancer cells to survive during systemic circulation; however, the mechanism underlying anoikis resistance remains unclear. Abstract:Objectives:As a zinc-dependent proteinase family member, A Disintegrin and Metalloproteinase 9 (ADAM9) has the metalloproteinase domain that performs proteolytic function to liberate or shed factors, such as bFGF, VEGF, EGF, TGF-α and IGF, from cell surface and make them more accessible to tumor cells. It was shown in previous studies that the upregulation of ADAM9 in breast cancer is associated with metastasis.In this study, we examined the effect of ADAM9 on the cell proliferation, migration and invasion and elucidated the molecular mechanism in TNBC. We hypothesize that ADAM9 sheds Tumor Necrosis Factor alpha (TNF-α), which activates TNF receptor and then triggers PI3K-dependent NF-kappa B pathway, affecting metastatic phenotypes of triple negative breast cancer (TNBC).Materials and Methods:Correlation of the levels of ADAM9 expression and survival time in triple negative breast cancer patients was analysed by the Kaplan-Meier (KM) plotter integrative data analysis tool. The TNBC subjects were divided into two groups of high and low ADAM9 expression levels with respect to the median. We also compared the protein expression levels of ADAM9 in a highly aggressive, invasive and poorly differentiated TNBC cell line, MDA-MB-231, and a non-tumorigenic epithelial cell line, MCF-10A.ADAM9 was knocked down in MDA-MB-231 using siRNA. In comparison to negative and blank controls, we examined the cell proliferation, migration and invasion, and the phosphorylation levels of AKT subject to ADAM9 knockdown. We also profiled the mRNA expression levels of ADAM9 and NF-kappa B in breast cancer patients.Results:As shown in Figure 1(a), the survival curves of these two groups were plotted and compared using Kaplan-Meier survival analysis, indicating that lower ADAM9 expression is linked to better survival rate (p=0.0038). Figure 1(b) illustrates significantly higher ADAM9 protein level in MDA-MB-231, compared with MCF-10A (p=0.0019).It is shown in Figure 2 and Figure 3(a) that the cell proliferation, migration and invasion, and the phosphorylation of AKT are significantly suppressed by ADAM9 knockdown in the aggressive TNBC cell line. Figure 3(b) shows significant association between ADAM9 and NF-kappa B in the mRNA expression level (p<0.001).Conclusions:The results clearly showed the activation of PI3K-dependent NF-kappa B pathway triggered by ADAM9. Abstract:PURPOSE:To investigate the influence of p75NTR ICD on anoikis resistance ability of tumor cells based on generation of an anoikis cell model and discuss the activation of related signaling pathways.METHODS:Immunohistochemistry was used to detect p75NTR ICD expression in lymph node metastatic and non-lymph node metastatic,primary specimens of oral squamous cell carcinoma (OSCC).Westem blot was used to detect the expression of p75NTR ICD in a pair of low-and high-metastatic potential OSCC cell lines.Poly-HEMA was used to pre-coat the cell culture dish in order to generate anoikis cell model.Over-expression of p75NTR ICD was conducted in tumor cells with plasmid transfection.Flow cytometry was used to detect apoptosis level of tumor cells after suspension culture.Laser scanning confocal microscopy analysis and Western blot were performed to detect the activation of related signal pathways of tumor cells in anoikis cell model.SPSS 22.0 software package was used for statistical analysis.RESULTS:The positive staining of p75NTR ICD in lymph node metastatic OSCC cells was located in the cytoplasm,while on the cytomembrane in nonlymph node metastatic OSCC.The expression of p75NTR ICD was much higher in HN-12,which was more metastatic as well,compared with HN-4.Larger sphere was formed in HN-12 anoikis cell model than in HN-4,lower apoptosis level of HN-12 in anoikis cell model was observed than in HN-4.NF-κB signaling was activated in HN-12 anoikis cell model,while not in HN-4.CONCLUSIONS:Cellular localization of p75NTR ICD in tumor cells was related to cervical lymph node metastasis.Over expression of p75NTR ICD in tumor cells shows higher anoikis resistance;p75NTR ICD promotes anoikis resistance through activating NF-κB signaling.

Abstract:Inflammatory stimuli activate ectodomain shedding of TNF-alpha, L-selectin, and other transmembrane proteins. We show that p38 MAP kinase, which is activated in response to inflammatory or stress signals, directly activates TACE, a membrane-associated metalloprotease that is also known as ADAM17 and effects shedding in response to growth factors and Erk MAP kinase activation. p38alpha MAP kinase interacts with the cytoplasmic domain of TACE and phosphorylates it on Thr(735), which is required for TACE-mediated ectodomain shedding. Activation of TACE by p38 MAP kinase results in the release of TGF-alpha family ligands, which activate EGF receptor signaling, leading to enhanced cell proliferation. Conversely, depletion of p38alpha MAP kinase activity suppresses EGF receptor signaling and downstream Erk MAP kinase signaling, as well as autocrine EGF receptor-dependent proliferation. Autocrine EGF receptor activation through TACE-mediated ectodomain shedding intimately links inflammation and cancer progression and may play a role in stress and conditions that relate to p38 MAP kinase activation.

Abstract:Resistance to anoikis is a characteristic of malignant cells with increased tumorigenesis and metastasis. Altered FAK activity has been strongly implicated in the development, growth, progression, and metastasis of human cancers, but the mechanism of FAK in regulating anoikis is unknown. In this study, the resistance anoikis role of FAK and its downstream mediators was evaluated in the human lung cancer cell line A549. It has been shown that down regulation of FAK stimulates the apoptosis of cells and the down-regulation of p-ERK, p-PI3K, p-Src, and p-p38. Furthermore, in detached A549 cells, increased FAK phosphorylations (Tyr397, Tyr861, Tyr925) were detected in a time-dependent manner, and the specific inhibitors of MEK1, PI3K, and Src (PD98059, LY294002, and PP2) partly abolished the resistance to the anoikis characteristic of cancer cells. Altogether, our data suggested that Src is involved in the progress of detachment-induced FAK activation in lung tumor cells. PI3K/AKT, MAPK-ERK, and perhaps MAPK-p38 but not MAPK-JNK, appear to be the key downstream effectors of FAK in mediating cell survival. The increased FAK activity upon cell detachment may contribute to the metastasis potential of malignant tumors.

Abstract:BackgroundNDRG1 plays important roles in tumor growth and metastasis of colorectal cancer (CRC). The relation between NDRG1 and metastatic colorectal cancer (mCRC) has not been identified and the mechanism of NDRG1 involving in mCRC needs to be elucidated.MethodsCorrelations between NDRG1 and clinicopathological characteristics and prognosis of 164 patients with mCRC were evaluated. Sensitivity of NDRG1-knockdown colon cancer cell to irinotecan (CPT-11) was determined by MTT assay. Blocking of NF-B signaling by p65 siRNA interference was carried out to explore the mechanism of NDRG1 involving in epithelial-mesenchymal transition (EMT)-regulated invasion and metastasis of CRC.ResultsNDRG1 expression was significantly negatively correlated with differentiation (P=0.008) and lymph node metastasis (P=0.016) of mCRC. NDRG1 was a favorable prognostic factor of mCRC, although might be responsible for CPT-11 resistance in vitro. Knockdown of NDRG1 promoted EMT of CRC cells via NF-B signaling. Depletion of NDRG1 increased phosphorylation level of NF-B. E-cadherin expression was increased and Vimentin expression was reduced in the p65-siRNA treated group, compared with the control group (P<0.001).ConclusionsNDRG1 appears to prevent EMT-induced metastasis by attenuating NF-B signaling in mCRC. Abstract:<div>AbstractAnoikis resistance is a prerequisite for circulating tumor cells to survive. However, the mechanism underlying anoikis resistance is poorly understood. In the current study, the effect of TNF receptor–associated factor 6 (TRAF6)-induced NF-kB activation on anoikis susceptibility in tumor cells was evaluated. Differential TRAF6-binding proteins in anoikis-sensitive versus anoikis-resistant tumor cells were screened by LC/MS-MS analysis. The effects of TRAF6-binding proteins on the stability of TRAF6, the activation of NF-kB signaling and anoikis susceptibility in tumor cells were detected. We found that the loss of TRAF6 expression is an important molecular event linked to anoikis. X-linked inhibitor of apoptosis protein (XIAP), an E3 ligase, can bind, ubiquitinate, and degrade TRAF6 and may lead to inactivation of NF-κB signaling and anoikis sensitivity. High expression of prohibitin 1 (PHB1) competes with XIAP for binding to TRAF6 and confers anoikis resistance to tumor cells. PHB1 and TRAF6 knockdown eliminated tumor cells from the circulation in vivo. Significant correlations between elevated PHB1 and TRAF6 expression and distant metastasis were observed in patients with oral cancer. Collectively, we elucidated a novel mechanism governing anoikis. Our data also indicated that TRAF6 and PHB1 are potential therapeutic targets for tumor cells disseminating in the circulation.Implications:Our data implicate that PHB1 competes with XIAP for binding to TRAF6 and confers anoikis resistance to tumor cells.</div>

Abstract:In many cancer types, integrin-mediated signaling regulates proliferation, survival and invasion of tumorigenic cells. However, it is still unclear how integrins crosstalk with oncogenes to regulate tumorigenesis and metastasis. Here we show that oncogenic K-RasV12 upregulates α6-integrin expression in Madin–Darby canine kidney (MDCK) cells via activation of the mitogen-activated protein kinase/extracellular signal-regulated kinase (ERK)/Fos-related antigen 1-signaling cascade. Activated α6-integrins promoted metastatic capacity and anoikis resistance, and led to perturbed growth of MDCK cysts. Transcriptomic analysis of K-RasV12-transformed MDCK cells also revealed robust downregulation of αV-class integrins. Re-expression of αV-integrin in K-RasV12-transformed MDCK cells synergistically upregulated the expression of Zinc finger E-box-binding homeobox 1 and Twist-related protein 1 and triggered epithelial-mesenchymal transition leading to induced cell motility and invasion. These results delineate the signaling cascades connecting oncogenic K-RasV12 with α6- and αV-integrin functions to modulate cancer cell survival and tumorigenesis, and reveal new possible strategies to target highly oncogenic K-RasV12 mutants.
