Abstract:Total immunoreactive insulin (IRI) is conventionally determined by radioimmunoassays. IRI measurement in rats can be made more sensitive, accurate, and practical, as demonstrated by a new modified enzyme-linked immunosorbent assay (ELISA). It is characterized by indirect binding of an anti-insulin antibody by an antiglobulin antibody and uses the principle of competitive saturation. In this ELISA, IRI can be determined in a wide range of concentrations, corresponding to the standards. The standard curve ranges from 100 to 0.049 ng/ml IRI (1 ng/ml ∼ 23.4 μU/ml ∼ 172 pM rat insulin). The statistical analysis shows between- and within-assay coefficients of variation of ≤ 15%.

Abstract:Prolactin (PRL) is produced by the pituitary gland and plays a vital role in the production of milk after a baby is born. PRL levels are normally elevated in pregnant and nursing women, and high levels of PRL in the human body cause hyperprolactinemia, infertility, galactorrhea, infrequent or irregular periods, amenorrhea, breast pain, and loss of libido. Accordingly, herein, a novel label-free immunosensor using a bismuth sulfide/polypyrrole (Bi2S3/PPy)-modified screen-printed electrode (SPE) for the fast and facile detection of the peptide hormone PRL. Bi2S3 nanorods were synthesized via a facile hydrothermal technique, and PPy was prepared by chemical polymerization. Subsequently, the Bi2S3/PPy-modified SPE was prepared using 3-mercaptopropionic acid (MPA) and EDC/NHS. Owing to the cross-linking effect of EDC/NHS, antibody-PRL (anti-PRL) was firmly stabilized on the modified SPE surface. These layer-by-layer modifications enhanced the conducting properties, anti-PRL loading capacity, and sensitivity of the developed immunosensor. Under optimized conditions, the PRL immunosensor demonstrated a broad linear range of approximately 1–250 ng/mL, a low detection limit of approximately 0.130 ng/mL (3 × SD/b), good specificity, reproducibility, and stability. PRL was successfully evaluated in human and mouse serum samples, and the corresponding outcomes were compared with those of the electrochemical and ELISA methods.

Abstract:Abstract Objective To generate monoclonal antibodies against pre-proinsulin (PPI), and establish sandwich ELISA method to provide a basis for PPI detection in recombinant human insulin production. Methods The Balb /c mice were immunized with PPI, and the hybridomas secreting anti-PPI monoclonal antibodies were obtained by conventional cell fusion technique and ELISA screening.The antibody was purified using a Protein G gel column and identified for purity by SDS-PAGE. Pairing effect was found by the sandwich ELISA, and the specificity of the paired antibody was determined. A paired antibody with better specificity was selected to establish sandwich ELISA, and construct a quantitative curve, the accuracy and sensitivity of the method were evaluated. Results Six anti-PPI monoclonal antibodies were obtained, named P1, P2, P3, P4, P5 and P6, of which P5 had the highest titer. The sandwich ELISA method was established with P5 for plating and P2 for detection antibodie. The linear range of the quantitative curve of PPI by sandwich ELISA was 0. 645-82.5 pg/mL, the recovery was 89%–95%, and the limit of detection was 3.06 pg/mL. Conclusion Six monoclonal antibodies against PPI were generated and the sandwich ELISA method was established to detect PPI in process control and product release control for recombinant human insulin production.

Abstract:CM310 is a recombinant humanized monoclonal antibody targeting Interleukin (IL)-4 receptor alpha (IL-4R alpha). IL-4R alpha blockade prevents IL-4 and IL-13 from binding to their receptor, thereby inhibiting downstream signaling pathways that drive Type 2 helper T-cell (Th2) inflammation. CM310 holds potential for treating Th2-related inflammatory diseases, such as asthma, atopic dermatitis and chronic sinusitis with nasal polyposis. In this study, a direct enzyme-linked immunosorbent assay (ELISA) was developed to measure the concentrations of CM310 in rat serum. Seven calibration standards (ranging from 25 to 1600 ng/mL) and three quality controls (70, 500 and 1250 ng/mL) were defined. The limit of detection (LOD), lower limit of quantification (LLOQ) and upper limit of quantification (ULOQ) were 13, 25 and 1600 ng/mL, respectively. The method exhibited excellent precision and accuracy and successfully applied to in vitro serum stability and pharmacokinetic (PK) studies. In conclusion, we have developed and validated a highly sensitive and selective method for measuring CM310 in Sprague-Dawley rats. The development and validation ELISA method met the acceptable criteria, which suggested that these can be applied to quantify CM310, as well as in PK studies.

Abstract:A sandwich ELISA for rat apo E was developed. Blood was drawn from rats that had been administered with triamcinolone diacetate to increase apo E-rich HDL in serum. Apo E was purified from the d less than 1.225 g/ml lipoproteins, and antiserum was raised in rabbits. Diluted samples and standards were added into the wells of polystyrene microtiter plates precoated with immunoaffinity-purified IgG and incubated for 90 min. After washing, immunoaffinity-purified Fab'-horseradish peroxidase conjugate was added to each well and incubated for 90 min. The bound enzyme was assayed by a colorimetric method. Samples and standards were pretreated with 6 M guanidine. HCl to maximize the antigenic response of apo E. The sensitivity lies around 1 pg apo E, and the working range was 0.1 to 1.0 ng. All assay procedures were completed within 4-5 hr. The mean intra- and interassay coefficients of variation were 1.8 and 4.1%, respectively. Serum apo E concentrations were 21.2 +/- 2.1 and 61.3 +/- 17.0 mg/dl (mean +/- SD) for young (8-12 weeks old, n = 9) and old (36-40 weeks old, n = 16) rats, respectively. As determined by gel filtrations, most of the apo E in fasted rat serum was associated with larger HDL particles (or HDL1) and a small portion of apo E was present in a free form.

Abstract:Abstract In the rat two major molecular variants of prolactin are recorded i.e. 23,000 M(r) and glycosylated 26,000 M(r). In order to further characterize the glycosylated 26,000 rat prolactin molecular variant, rat pituitary cell lysates were digested with several glycoen-zymes and the digestion products submitted to sodium dodecyl sulphate polyacrylamide gel electrophoresis and subsequent immunoblotting. The results were as follows: treatment with 1) neuraminidase, specific for sialic acid, yielded an M(r) decrease of the glycosidic variant from 26,000 to 24,500, 23,800, 23,000 and 22,000; 2) endo-alpha-N-acetylgalactosaminidase, which releases the disaccharide Gal (beta 1-3) GalNac from O-glycans, split 26,000 rat prolactin into a doublet of M(r) 26,000 to 25,500; and 3) mixed exoglycosidases from Turbo cornutus caused a gradual M(r) shift from 26,000 to 23,000. Affinity chromatography on wheat germ agglutinin Sepharose 6MB and soybean agglutinin agarose of rat pituitary homogenates and competitive inhibition tests showed that glycosylated rat prolactin has distinct affinity for these lectins. From the experimental data it is proposed that glycosylated rat prolactin is O-linked through threonine by the disaccharide Gal (beta 1-3) GalNac and possesses at least GalNac, and/or Gal and sialyl residues.

Abstract:Prolactin is a 23 kDa protein synthesised and released by lactotrophs located in the anterior pituitary gland. The main function of prolactin is the development of the mammary glands and the production and maintenance of milk secretion during pregnancy and lactation. Prolactin is present in 3 forms in the circulation monomeric, dimeric and macroprolactin which is one of the causes of variability between assay manufacturers. Although most assays have been standardised using the WHO's 3rd international standard there is still considerable variability between commercially available assays and as such laboratories need to establish their own reference intervals. Clinicians use reference intervals to distinguish between healthy and diseased individuals. Direct methods to establish reference intervals are expensive and time consuming and are impractical for most routine clinical laboratories. Indirect methods using the laboratories own data are more commonly used but also have drawbacks. Determination of reference intervals for prolactin is particularly difficult due to various physiological causes for variation in prolactin levels including age, sex and menopause. The presence of macroprolactin, its detection and methods for reporting its presence adds to reference interval determination complexity and harmonisation of these methods may improve clinical care.

Abstract:The use of salbutamol (SAL), a beta-adrenergic receptor agonist (beta-agonist) that could enhance the lean meat-to-fat ratio in livestock, has been prohibited as an additive in animal feeds for livestock production in many countries due to its harmful effect to the consumers. Herein, we present a sensitive and group-specific ELISA based on monoclonal antibodies (mAb) for the determination of SAL in swine meat and liver samples. The SAL derivative, containing the carboxylic group, was prepared by directly reacting SAL with succinic anhydride and then the derivative was linked to carrier proteins. Mice immunized with SAL-bovine serum albumin (BSA) conjugate were utilized for mAb generation. A hybridoma clone (1C3) specifically secreting mAb against SAL with higher sensitivity was successfully obtained. Under optimal conditions, the IC50 and LOD values of the mA beta-based ELISA for SAL were calculated to be 0.47 ng mL(-1) and 0.049 ng mL(-1), respectively, indicating high sensitivity of the assay. The mAb displayed high cross-reactivity (CR) with four beta-agonists, i.e., clenbuterol (55%), terbutaline (44%), brombuterol (33%) and cimbuterol (134%) because these compounds contain the same tert-butyl group on the nitrogen atom as does SAL. The CR of the mAb with other beta-agonists, including cimaterol, clorprenaline, isoprenaline, neophryn, ractopamine and phenylethylamine A, was less than 2.2% due to relatively large structural differences compared to that of SAL. Swine meat and liver samples were spiked with different SAL content and detected by ELISA. The recovery rates were found in the range of 76.4-107.6% and 71.8-106.8%, respectively, with coefficients of variation less than 20%. Spiked samples were also analyzed by HPLC. A good correlation between both methods was obtained. These results demonstrated that the proposed ELISA is a feasible quantitative/screening method for SAL analysis in swine meat and liver samples. In addition, the recovery results of the beta-agonists that displayed the high CR values with mAb in an additional spiking experiment revealed that the ELISA could also be an alternative method for multiple beta-agonist detection.

Abstract:BACKGROUND: All beta-agonists are banned as feed additives for growth promotion in animals due to toxic effects on humans after consuming the beta- agonist contaminated meats. Phenylethanolamine A (PA) is a newly emerged beta-agonist. Thus there is a need to develop highly sensitive and specific analytical methods for the detection of PA in food samples. In this study, the monoclonal antibody(mAb) against PA was produced by hybridoma technology and used for the development of enzyme-linked immunosorbent assay (ELISA).RESULTS: The IC50 values and limits of detection (LODs) of the ELISA using homogeneous combination of coating antigen/antibody for PA were 0.16 ng mL(-1) and 0.011 ng mL(-1), respectively. The cross-reactive (CR) values of the assay with 14 structurally related beta-agonists were lower than 0.59%. Swine liver and meat samples were spiked with PA at different content and analysed by ELISA. Acceptable recovery rates of 91.40-105.51% and intra-assay coefficients of variation of 1.56-9.92% (n = 3) were obtained. The ELISA for seven spiked samples was confirmed by LC-MS/MS with a high correlation coefficient of 0.9881.CONCLUSION: The proposed mAb-based ELISA was highly sensitive and specific for PA and could be used as a quantitative/screening method for PA analysis in food samples. Abstract:Using the established RRA to measure the LH bioactive level of variety animal species, we detected the LH variations of several animals in different physiological conditions to confirm the veracity of RRA results. Preliminary application of LH/hCG RRA to measure serum LH level indicated that the rabbits were specific response to LRH-A_(3) stimulation and the RRA could specifically measure the LH change induced by LRH-A_(3). The LH/hCG RRA could be a convenient alternative to RIA and the Leydig cell bioassay for monitoring the LH profiles of sera and biological fluids in a variety of species.

Abstract:A dual-signal amplification strategy for electrochemiluminescence (ECL) immunosensor was designed based on L-cysteine capped CdSe quantum dots (QDs) and gold nanoparticles (AuNPs) conjugated horseradish peroxidase (HRP) for highly sensitive detection of ractopamine (RAC). The electrochemical detection was based on the HRP catalyzed o-phenylenediamine (OPD) to consume the self-produced H2O2, which has been extensively used as a co-reactant of QDs. The enzymatic reaction rate was proportional to the amount of HRP bound to the electrode. In the presence of RAC standard solution, the immobilized RAC coating antigen competed with RAC solution for the Ab-AuNPs-HRP complexes. With the increase of the RAC concentration, the amount of immobilized HRP decreased, which led to the increase of ECL intensity. Under optimized conditions, ECL intensity changed linearly with the logarithm of RAC concentration in the range of 0.005-500 ng mL(-1) with the detection limit of 0.0017 ng mL(-1) (S/N = 3). The ECL immunosensor possesses high sensitivity, satisfied reproducibility and selectivity, and may provide a feasible route for practical application of RAC detection. A Neutralizing Prolactin Receptor Antibody Whose In Vivo Application Mimics the Phenotype of Female Prolactin Receptor-Deficient Mice

Abstract:The prolactin receptor (PRLR) has been implicated in a variety of physiological processes (lactation, reproduction) and diseases (breast cancer, autoimmune diseases). Prolactin synthesis in the pituitary and extrapituitary sites is regulated by different promoters. Dopamine receptor agonists such as bromocriptine can only interfere with pituitary prolactin synthesis and thus do not induce a complete blockade of PRLR signaling. Here we describe the identification of a human monoclonal antibody 005-C04 that blocks PRLR-mediated signaling at nanomolar concentrations in vitro. In contrast to a negative control antibody, the neutralizing PRLR antibody 005-C04 inhibits signal transducer and activator of transcription 5 phosphorylation in T47D cells and proliferation of BaF3 cells stably expressing murine or human PRLRs in a dose-dependent manner. In vivo application of this new function-blocking PRLR antibody reflects the phenotype of PRLR-deficient mice. After antibody administration female mice become infertile in a reversible manner. In lactating dams, the antibody induces mammary gland involution and negatively interferes with lactation capacity as evidenced by reduced milk protein expression in mammary glands and impaired litter weight gain. Antibody-mediated blockade of the PRLR in vivo stimulates hair regrowth in female mice. Compared with peptide-derived PRLR antagonists, the PRLR antibody 005-C04 exhibits several advantages such as higher potency, noncompetitive inhibition of PRLR signaling, and a longer half-life, which allows its use as a tool compound also in long-term in vivo studies. Therefore, we suggest that this antibody will help to further our understanding of the role of auto- and paracrine PRLR signaling in health and disease.
Prolactin Receptor Expression in the Gastrointestinal Tract: Characterization of the Prolactin Receptor of Gastric Mucosa.

Abstract:ABSTRACT There is evidence that prolactin (PRL) influences gastrointestinal function. However, the sites at which prolactin exerts these effects are not known. A monoclonal antibody was therefore generated against the rabbit mammary gland prolactin receptor (MAb 218) and used to study the distribution of the prolactin receptor in the rabbit gastrointestinal tract (GIT) by immunohistochemistry. MAb 218 is an IgG 1 κprecipitating antibody which precipitates major affinity cross-linked mammary gland prolactin receptor subunits of molecular masses 45 and 80 kDa. It has an affinity of 0·8 × 109 mol/l for the prolactin receptor and does not react with GH or insulin receptors in precipitation assays. MAb 218 immunoreactivity was observed in classical prolactin target cells such as mammary gland epithelium, and this immunoreactivity was abolished by preincubation of MAb 218 with purified prolactin receptor but not by preincubation with purified GH receptor. In the GIT, the most intense immunoreactivity was associated with the oesophageal epithelium, chief (zymogenic) cells of the fundic mucosa, pancreatic islets of Langerhans and surface epithelial cells of the duodenum and jejunum. Other specific elements of the GIT were immunoreactive at lower levels or were immunonegative. No immunoreactivity was observed in these locations with a control monoclonal antibody (MAb 50·8) of identical isotype to 218. To support the immunohistochemical findings, rabbit gastric mucosal membranes were used to show the presence of lactogen-specific binding. Scatchard analysis of 125I-labelled human GH binding to the gastric mucosal membranes with rat prolactin as displacing ligand yielded an affinity constant (Ka) of 1·0 ± 0·2 × 109 mol/l with a capacity of 3·5 ± 0·4 fmol/mg protein. Affinity cross-linking and sodium dodecyl sulphate-polyacrylamide gel electrophoresis of the gastric receptor revealed lactogenic hormone-binding subunits of molecular masses 43, 68 and 83 kDa. The 68 kDa subunit was not seen in rabbit mammary gland or ovarian tissue, and may be unique to gastric mucosa. In conclusion, we have demonstrated the presence of a high affinity lactogenic receptor in specific epithelial cell subpopulations of the GIT. The Development of an Indirect Competitive Immunomagnetic-Proximity Ligation Assay for Small-Molecule Detection

Abstract:The development of an indirect competitive immunomagnetic-proximity ligation assay (ICIPLA), which is a novel method for detecting small molecules, is described in this report. Free small molecules in samples can be detected using a proximity ligation assay (PLA); the detection is based on the proximity effect caused by a high concentration of small molecule-BSA conjugates bound to streptavidin magnetic beads. As an indirect format competitive immunoassay, the ICIPLA method has the advantage in that the quantity of monoclonal antibody (mAb) used for small-molecule detection is 8-fold lower than that required for the competitive immunomagnetic-proximity ligation assay (CIPLA) described in our previous work. Small molecules can be detected using a single monoclonal antibody, and the PLA method can be used to amplify high-performance signals. In this work, the small molecular compound ractopamine (RAC) was selected as a target for ICIPLA. The limit of detection (LOD) was 0.01 ng ml(-1), and the method exhibited a broad dynamic range of up to six orders of magnitude. We also employed the ICIPLA method to detect RAC in serum, urine, and muscle extracts; the results indicated that the LOD and dynamic range were not altered. The cross-reactivity studies showed that the cross-reactivity values for all RAC analogs were below 0.01%. These results suggest that ICIPLA is a sensitive, specific and practical method for small-molecule detection. This is the first report of the improved PLA technology for small-molecule detection by indirect competitive formats in the biological samples.

Abstract:Complete antigens for β-adrenergic agonist ractopamine were synthesized using bovine serum albumin(BSA) and ovalbumin(OVA) as carrier proteins. The WHB dark-eye rabbits were used as antibody generation animals. B- lymphocytes from immuned rabbits and 240E myeloma cells were fused to generate hybridoma cells. And four clones with continuous growth and the ability to produce highly specific antibodies were obtained. Supernatant from clone A3-2-6 was used for the development of an immunoassay. The selected antibody was specific for ractopamine with an IC50 of 4.09 ngmL and its linear range of the inhibition rate vs log ractopamine competition curve generated in PBST was 0.1~100.0 ngmL. β-adrenergic agonists which had similar structures to ractopamine such as clenbuterol hydrochloride, isoxsuprine, epinephrine, norepinephrine, isoprenaline, salbutamol and dopamine showed no cross-reactivity. In conclusion, a highly specific rabbit monoclonal antibody(RabMab) to ractopamine hydrochloride is developed, and it has wide linear range.

Abstract:A simple and reproducible enzyme-linked immunosorbent assay (ELISA) was developed to determine the concentration of bee venom in rat plasma. The intra- and inter-assay coefficients of variation for the ELISA were less then 3% between 0.1 and 1000 ng mL(-1) venom, and the sensitivity of the detection was 0.1 ng mL-1. Total recovery of the bee venom added to rat plasma was determined. Using this ELISA, serum levels of bee venom were easily determined. The rats were administered a single intravenous injection or oral dose of bee venom (1 mg kg(-1) of body weight). The bioavailability of the bee venom under the two administrations was compared using pharmacokinetic parameters. Results showed that intravenous administration of bee venom produced high plasma concentrations with a short half-life. The area under the curve for oral administration was 10 times lower than for intravenous administration. This loss of bee venom may be due to the degradation that occurs in the enzymatic and acidic environment of the gastrointestinal tract.
Confusion in the interpretation of prolactin levels caused by inappropriately low reference intervals

Abstract:Objective. Serum prolactin measurements are important in the differential diagnosis of pituitary masses and subfertility. We observed discrepancies in serum prolactin levels in several patients measured with different immunoassays. Despite differences in assay results, the reference intervals derived by the manufacturers were similar. In this study we aimed to investigate prolactin assay differences and to re-establish RIs for different prolactin immunoassays. Methods. For the assay comparison, serum samples were collected from men and women visiting our the Amsterdam UMC hospital. Prolactin levels were measured using the AtellicaTM IM analyzer (Siemens Healthineers) and the Cobas (Roche Diagnostics) immunoassay. Reference intervals for prolactin were re-established for men, premenopausal women and postmenopausal women for both the Atellica and Cobas immunoassay. Results. Prolactin levels measured using the Cobas immunoassay were 1.75 times higher than those measured using the Atellica immunoassay. The re-established reference intervals for Atellica and Cobas confirmed these findings and were <0.32 U/L; <0.55 U/L for men; <0.64 U/L; <0.86 U/L for premenopausal women and <0.31 U/L; <0.59 U/L for postmenopausal women, respectively for Atellica and Cobas assays. The re-established RIs of the Atellica assay matched the current and manufacturer RIs whereas those for Cobas differed substantially. Conclusions. Prolactin levels are assay dependent and the re-established reference intervals are different for the Atellica and Cobas assays. We recommend that laboratory specialists and manufacturers periodically review prolactin assay RIs as incorrect reference intervals can lead to misinterpretation of prolactin levels and unnecessary referrals and further laboratory testing, as we have experienced.

Abstract:Pituitary pars intermedia dysfunction (PPID) is a neurodegenerative disease of senior horses. Loss of dopaminergic inhibition of the melanotropes of the pars intermedia leads to increased concentrations of pro-opiomelanocortin (POMC)-derived peptides. Diagnosis is challenging due to pre-analytical variables, such as sample storage, handling, and time to analysis. Our objective was to develop an ELISA for ACTH measurement, which could ultimately form the basis for a stall-side equine ACTH test. We selected 2 ACTH-specific monoclonal antibodies, CBL57 and EPR20361-248, based on the recognition of separate epitopes, strong and rapid color change, and minimal background interference, including no cross-reactivity with themselves, each other, and the test reagents. CBL57 was chosen as the detection antibody (or secondary antibody). EPR20361-248, functionalized on superparamagnetic iron oxide beads, was chosen as the capture antibody (or primary antibody) to bind ACTH in plasma. The incorporation of magnetic beads marks the initial stage in establishing a platform that could potentially be utilized in the field, similar to other stall-side tests. The concentrations of antibodies, magnetic beads, and incubation durations were optimized. Our immunoassay detected unglycosylated rat recombinant ACTH. Further studies are ongoing to optimize and validate our assay using equine plasma and serum samples.
