Abstract:A rapid and sensitive magnetic particles (MPs)-based chemiluminescence enzyme immunoassay (MPs-CLEIA) for determination of luteinizing hormone (LH) in human serum was developed and validated.The method was developed based on a sandwiched immunoreaction format, in which MPs served as both the solid phase and separator. The horseradish peroxidase(HRP)-luminol-H(2)O(2) chemiluminescent system was chosen as the detection system.This method had a detection limit of 0.2 mIU/mL and a linear range of 0.5-200 mIU/mL. The assay showed little cross-reactivity, and the intra- and inter-assay coefficients of variation were less than 10%. The recoveries were from 101.6% to 104.1%. The method has been successfully applied to detection of LH in human serum and showed good correlation compared with radioimmunoassay (RIA).The MPs-CLEIA provided apparent advantages over RIA, and facilitated the development of high-throughput screening and automated operation systems in clinical diagnosis.
Tracing the history of nuclear releases: determination of 129I in tree rings.

Abstract:Concentrations of the long-lived radioisotope 1291 were measured in dated tree rings in order to determine whether the distribution of this isotope reflects the history of nuclear deposition. 129I concentrations and 129I/127I ratios were analyzed in tree rings and bark samples from four trees at West Valley, NY, and from one tree at Rochester, NY. West Valley was the site of short-lived nuclear fuel reprocessing activities (1966-1972), while Rochester, located 115 km to the northeast, provided a regional control site for the study. The selected trees reflect different modes of fluid and nutrient transport in trees, with three species of ring-porous trees (elm, oak, and locust), one semidiffuse (cherry), and one diffuse-porous tree (maple). The results show that 1291 levels in ring-porous trees, in which xylem or hydrologic tissue is localized in the outermost growth ring, are generally well correlated with the expected 1291 deposition pattern for the region. In contrast, tree rings of the more common semidiffuse to diffuse-porous wood, where xylem is disseminated throughout the trunk, show a less well developed 129I signal, probably due to the transport of iodine ions across annual rings. Iodine concentrations in the tree rings range from 0.04 to 2 mg/kg, 129I/127I ratios from 6 x 10(-10) to 3.8 x 10(-6). Tree bark and the outermost rings show significantly higher 129I concentrations than the wood of the trunk. The 129I/127I ratios for bark are very similar to values obtained for surface soil and water at the two localities, while inner rings have ratios similar to those in deeper layers of the soil, reflecting different pathways for 129I uptake and the differences in ambient 1291 levels between the atmosphere and deep soil. Although ring porous trees preserve the depositional pattern of nuclear releases, rings older than or close to the onset of the nuclear age have 129I/127I ratios significantly above the preanthropogenic level, suggesting that even in these trees some redistribution of 129I occurs throughout the trunk. Our results indicate that growth rings from ring-porous wood are useful in time-series analyses of regional 129I deposition, yielding reliable information on relative changes in 129I concentrations but requiring caution in the reconstruction of absolute ambient concentrations during any given time.
Utilization of RRA [radioreceptor Assay] in Bioactive LH Measurement for a Variety of Animal Species.

Abstract:Using the established RRA to measure the LH bioactive level of variety animal species, we detected the LH variations of several animals in different physiological conditions to confirm the veracity of RRA results. Preliminary application of LH/hCG RRA to measure serum LH level indicated that the rabbits were specific response to LRH-A_(3) stimulation and the RRA could specifically measure the LH change induced by LRH-A_(3). The LH/hCG RRA could be a convenient alternative to RIA and the Leydig cell bioassay for monitoring the LH profiles of sera and biological fluids in a variety of species.
Development and evaluation of an ELISA method for the determination of lipoprotein lipase mass concentration--comparison with a commercial, one-step enzyme immunoassay

Abstract:We developed a non-competitive, enzyme-linked, immunosorbent assay (ELISA) for the quantitation of lipoprotein lipase (LPL) in human postheparin plasma using affinity-purified antihuman milk lipoprotein lipase antibodies produced in chicken eggs and a monoclonal antibody directed against human lipoprotein lipase. We compared our ELISA method with a commercially available sandwich-enzyme immunoassay (Markit-F LPL EIA Kit, Dainippon Pharmaceutical Co, Ltd. Osaka, Japan). The reference values for lipoprotein lipase catalytic activity concentration and mass concentration in healthy Finns were determined. Lipoprotein lipase activity concentration (mean +/- SD) was 297 +/- 112 U/l in women, and mass concentration as measured by the ELISA method was 1058 +/- 367 micrograms/l. The corresponding values for men were 247 +/- 97 U/l and 815 +/- 207 micrograms/l, respectively. Across the whole concentration range of the ELISA method, the control samples' intra- and inter-assay coefficients of variation (CV) were 5.1% and 6.5%, respectively. The correlation between the ELISA and EIA methods was good, r = +0.81. The importance of the correct standardisation of immunoassays is discussed.

Abstract:Abstract The performance of enzyme immunoassay (EIA) and radioimmunoassay (RIA) in the quantitative analysis of ecdysteroids was compared. The EIA was found to be at least equivalent to the RIA with respect to analytical range and sensitivity and to be more comfortable with respect to safety and time saving. When biological samples were analyzed by both assays a good correlation (r = 0.83) was found. Since the EIA has certain advantages over the RIA, we now recommend the use of the former assay for the quantification of ecdysteroids.

Abstract:BackgroundThe escalating prevalence of allergic diseases on a global scale, affecting approximately 20% of the world's population, has become a significant health concern. Clinical determination of total IgE in serum and plasma is used as an aid to the diagnosis of allergic diseases. The tIgE CLIA Microparticles Kit is a chemiluminescent microparticle immunoassay for the quantitative determination of total IgE concentrations in human serum. This kit, integrated with the AutoLumo® 2000 Plus Analyzer, offers an automated, cutting-edge platform for total IgE detection. This streamlined combination provides clinicians with a reliable tool for comprehensive and efficient allergic diseases diagnosis.MethodsThe performance of tIgE CLIA Microparticles Kit was evaluated on AutoLumo® A2000 Plus automated chemiluminescence analyzer and compared to a well-characterized platform (Elecsys IgE II on Cobas e601, Roche). This assay is based upon the two-step sandwich method. During detection, the patient sample and the anti-IgE coated microparticles solution were added sequentially. Unbound components were washed off after incubation. The formation of a complex occurred with the subsequent addition of a HRP conjugated anti-IgE antibody solution. The catalysis of chemiluminescent substrates by this complex generated a measurable light signal recorded as RLUs (Relative Light Units). A 2-point calibrator was used to analyze the tIgE antibody concentrations in the serum sample. The limit of blank (LoB), limit of detection (LoD), limit of quantitation (LoQ), accuracy, repeatability, linearity, specificity, on-board stability and real-time stability of the assay kit were evaluated. Serum samples (n=220) that had been previously well characterized were used for method comparisons.ResultsThe tIgE CLIA Microparticles assay demonstrated a measurement range of 5-2500 IU/mL, with LoB, LoD, and LoQ values of ≤3 IU/mL, ≤4 IU/mL, and ≤5 IU/mL, respectively. The accuracy of the assay kit was assessed by trueness and precision following CLSI EP15-A2 and EP5-A2 guidelines. The trueness was evaluated using WHO IRP 11/234 as the reference standard, the estimate %bias between the expected and measured mean value was ≤5%. The average %CV of the intra-run and inter-run precision were 5.00% and 3.24% respectively, with a linearity of R≥0.990. The specificity to tIgE is evident, with no cross-reactivity to IgA, IgG, and IgM (≤100 μg/mL), and minimal interference from rheumatoid factor (RF) and anti-nuclear antibodies (ANA) ( 0.990) and the bias was within 15% when compared to a reference kit according to the CLSI EP9-A2 guideline.ConclusionsAutolumo® Analyzers, in conjunction with the tIgE CLIA Microparticles Assay, presents a fully automated chemiluminescence immunoassay platform for accurate and cost-effective screening to aid allergic diseases diagnostics with excellent specificity, precision, and on-board stability.

Production and Characterization of Anti-Estrone Monoclonal Antibody

Abstract:Objective Determination of estrone (E-1) levels has a significant meaning in evaluating physiological effect and diagnosing some diseases. In order to detect free E-1 in biological fluids, a monoclonal antibody specific for E-1 was prepared after the complete antigen of E-1 was synthesized. The purified monoclonal antibody was fully characterized for later inummoassay. Methods 3-O-carboxymethyl ether derivative of E-1 was synthesized and in turn coupled to bovine serum albumin (BSA) to form complete antigen E-1-BSA. A monoclonal antibody (McAb) specific for E-1 was produced both in vitro and in vivo by a hybridoma anti-E-1. Anti-E-1 was prepared by fusion of SP2/0 murine myeloma cells with spleen cells isolated from immunized BALB/c mouse. The McAb was characterized by enzyme-linked immunosorbent assay (ELISA), SDS-PAGE and Western-blotting. The specificity of the immunoassay was investigated by determining the cross-reactions of E-1 analogs when free E-1 was detected by competitive indirect enzyme-linked immunosorbent assay (CI-ELISA). Results Analysis revealed that anti-E, McAb (E-1-McAb) was of the lgG1 type, the molecular weight of E-1-McAb was 164 000 daltons. The affinity constant of E-1-McAb with coated complete antigen was 8.2x10(8)L/mol. The linear range for free E-1 determined by CI-ELISA was 10pg/mLsimilar to10ng/mL. The detection limit was 21.4 pg/mL (defined as twice the standard deviation of the blank). Conclusion The CI-ELISA developed with E-1-McAb was both sensitive and specific. The prepared E1-McAb can be used in some immunoassays.

Abstract:Abstract Background The detection of specific IgE is a key method for diagnosing allergic diseases. An accurate, user friendly and cost-effective way to clinically diagnosing allergies is beneficial for the patients. Our detection kit together with the Autolumo A2000 Plus chemiluminescence fully automated platform provide a new and more effective (40 minutes) way for detection of specific IgE with simple operation, high sensitivity and specificity. Methods The Animal Dander Allergen Mix Assay kit detects specific IgE against cat dander (E1), horse dander (E3), cow dander (E4) and dog dander (E5). Performance evaluation of the animal dander mix, E1, E3, E4 and E5 assay kits for the detection of specific IgE in serum was performed on our automated chemiluminescence platform (Autolumo A2000 Plus) and compared to a well-characterized platform (ImmunoCAP, Phadia). During detection, magnetic particle suspension, antigen solution, patient sample, and enzyme conjugate were added sequentially. A streptavidin-biotinylated antigen-sIgE antibody-anti-IgE-HRP conjugate complex is formed, wherein the amount of the anti-IgE-HRP conjugate is positively correlated to the amount of sIgE antibodies in the patient sample. After the substrate is added, the HRP catalyze the chemical reaction to generate a recordable signal in the form of light. The intensities of the light were obtained by using a chemiluminescence quantitative analyzer, and the standard curve was generated according to the 7-point calibration, and then the sIgE antibody concentrations in the serum sample were analyzed. The overall agreement, method comparison, precision, linearity, and stability of our mixed and single assay kits were evaluated using serum samples (n > 140) that had been characterized with Phadia. Results The overall agreement for the animal dander mix assay is 91.84%, with the average intra-run and inter-run precision to be 1.97% and 4.21% respectively. The limit of blank (LoB) and limit of detection (LoD) for the animal dander mix assay is 0.019 kU/L and 0.045 kU/L respectively. The sensitivity for our single E1, E3, E4, and E5 assays are 97%, 85.6%, 70.8%, and 80.2%, respectively. All single assays have a specificity >90%, total precision <5% CV, and a linearity with R2 > 0.999. The LoB, LoD and recommended LoQ for all single assays are 0.019 kU/L, 0.037 kU/L, and 0.1 kU/L respectively. The average signal drifts after 10 days incubation in 37C is less than 10% indicating the assays are stable. Conclusion Autolumo A2000 Plus is a fully automated chemiluminescence Immunoassay platform that provides accurate and cost-effective screening for allergy diagnostics with excellent overall agreement, precision, and stability.

Abstract:A high-throughput and simple chemiluminescence (CL) enzyme immunoassay (CLEIA) for the determination of progesterone (P) in human serum was developed, with the highly sensitive 4-methoxy-4-(3-phosphatephenyl)-spiro-(1,2-dioxetane-3,2'-adamantane) (AMPPD)-alkaline phosphatase (ALP) system as the CL detection system. The results showed that the indirect immobilization of rabbit anti-progesterone polyclonal antibody (RAPA) through secondary antibody exhibited apparent advantages over direct coating in terms of antibody saving and improvement of the coating stability and uniformity. The direct analysis of P in human serum without extraction was realized by using 8-anilino-1-naphthalenesulphonic acid (ANS) to displace P from its binding proteins. The effect of several relevant parameters of the immunoreaction were examined and optimized. Compared with some commercial progesterone kits, the presented CLEIA has higher sensitivity with detection limitation as low as 0.06 ng/mL. The recoveries were 95.9-101%. The coefficient of variation was <8.4% and 9.9% for intra- and inter-assay precision, respectively. This method has been successfully applied to the evaluation of P in human serum. Abstract:Measuring low amounts of anti-erythropoietin antibodies (anti-EPO Abs) is important to evaluate the therapeutic safety of recombinant human erythropoietin (rhEPO). In this work, a simple, sensitive and high-throughput chemiluminescent (CL) imaging assay was developed for the detection of anti-EPO Abs in human sera. The influence of several physicochemical parameters, such as coating conditions, incubation time, detergent concentration and exposure time, were investigated. A calibration curve was established and the range of quantitative detection was 0.12-13.91 ng/mL. The limit of detection (LOD, 30;) for the CL-imaging assay was 0.033 ng/mL. Compared to conventional colorimetric enzyme-linked immunosorbent assay (ELISA), the LOD of the CL-imaging assay is 50-fold lower. The recoveries of anti-EPO Abs in the fortified serum were in the range 87.1-116.9% using the present method, which highlighted the validity of the CL-imaging assay system to accurately determine the anti-EPO Abs in serum samples. CL-imaging assay was used to evaluate the presence of anti-EPO Abs in serum samples obtained from chronic renal failure (CRF) patients treated with rhEPO. Abstract:Thermally sensitive hydrogel-antigen conjugates were synthesized with two different methods. Both kinds of conjugates can be used for the determination of free estrone (E-1) in serum in a temperature controlled phase-separation immunoassay. Antigen (bovine serum albumin (BSA)-E-1) was activated. by acrylic acid N-hydroxysuccinimide ester (NAS), and then copolymerized with N-isopropylacrylamide (NIPA) in the presence of ammonium persulphate (as an initiator) and N,N,N',N'-tetramethylene diamine(TEMED, as an accelerator). The copolymer prepared was called A. Another copolymer was prepared by copolymerization of NIPA and NAS with azoisobutyronitrile (AIBN) as an initiator. This copolymer reacted with BSA-E-1 to form copolymer-BSA-E-1 conjugate named B-Some properties of A and B were investigated and compared. A (or B) immunologically reacted with fluoresceinated anti-E-1 monoclonal antibody (McAb*) competing with free E-1. Thus a competitive temperature controlled phase-separation inummoassay was established for the determination of E-1. The calibration range (for B) was 10similar to700 mug/L. The detection limit was 1 mug/L (n=3). The results demonstrated that the immunoassay of E-1 based on B was more sensitive and efficient than that based on A.
Different Biotinylation Strategies for Competitive Immunoassay of Estradiol

Abstract:Study on biotinylation strategies for competitive immunoassay of estradiol (E2) was carried out. Two types of competitive enzyme immunoassay (EIA) with Biotin-Avidin amplification system were established and optimized.The E2-Biotin conjugate was used as a tracer in one assay, and biotinylated antibody was used as a tracer in the other. In both of EIAs, horseradish peroxidase-labelled Avidin (Avidin-HRP) was used with a spectrometric determination of enzyme activity. The precision, sensitivity and specificity were measured and compared. The results showed that although both were satisfactory in specificity, the EIA with hapten-Biotin showed to be superior to the EIA with biotinylated antibody in sensitivity and precision. The limit of detection of serum E2 was 8 and 50 pg/mL with E2-Biotin and biotinylated antibody as tracer, respectively.

Abstract:A competitive immunoassay for estradiol (E2) based on secondary antibody format was established. The donkey anti-rabbit IgG was used as the secondary antibody to coat micro-plates, and the horseradish peroxidase (HRP)–luminol–H2O2 chemiluminescent system with high sensitivity was chosen as the detection system. The addition of sodium trichloroacetate (CCl3COONa) in the enzyme buffer as a replaceable packing material can realize directly analysis of E2 in human serum without extraction, which improved reproducibility and resolution of the assay. Additionally, the method showed specific recognition of estrogen, without cross-reaction for the major steroids (estrone (E1), estriol (E3), dihydrotestosterone (DHT), androstenedione, testosterone (T)) commonly found in human serum. The chemiluminescence immunoassay with secondary antibody can be applied to detect E2 with good precision at concentrations as low as 1.48pgmL−1. The proposed method has been successfully applied to the determination of E2 in 97 human sera and showed a good correlation compared with the commercially radioimmunoassay (RIA) kit with a correlative coefficient of 0.9881. This method has exhibited great potential in the fabrication of diagnostic kit and can be used in the clinical analysis of E2 in human serum.

Abstract:To date there have been few published immunoassays for the important iron regulator hepcidin. This study describes a novel competitive radioimmunoassay (RIA) for the bioactive hepcidin peptide. A rabbit anti-hepcidin polyclonal antibody was produced using synthetic hepcidin radiolabelled with 125I to produce a competitive RIA. Normal patient (n=47) samples were collected and assayed for hepcidin to determine a reference range. Other patient groups collected were ulcerative colitis (UC; n=40), iron deficiency anaemia (IDA; n=15), chronic kidney disease not requiring dialysis (CKD; n=45) and chronic kidney disease requiring dialysis (HCKD; n=94). Detection limit of the assay was determined as 0.6 ng/mL. Intra-assay precision was 5 ng/mL (7.2%) and 50 ng/mL (5.8%), interassay precision was 5 ng/mL (7.6%) and 50 ng/mL (6.7%). Analytical recovery was 98% (5 ng/mL), 94% (10 ng/mL) and 97% (50 ng/mL). The assay was linear up to 200 ng/mL. No demonstrable cross-reactivity with human insulin, glucagon I, angiotensinogen I, beta-defensin 1-4, alpha-defensin-1 and plectasin was observed. There was significant correlation (r=0.96, P < or = 0.0001) between the hepcidin RIA and an established hepcidin SELDI-TOF-MS method. Analysis of the normal human samples gave a reference range of 1.1-55 ng/mL for hepcidin. Further statistical evaluation revealed a significant difference between male and female hepcidin levels. There was significant correlation between hepcidin and ferritin in the control group (r=0.6, P < or = 0.0001). There was also a significant difference between the normal and disease groups (P < or = 0.0001). Healthy volunteers (n=10) showed a diurnal increase in plasma hepcidin at 4.00 pm compared to 8.00 am. A robust and optimised immunoassay for bioactive hepcidin has been produced and the patient sample results obtained further validates the important role of hepcidin in iron regulation, and will allow further investigation of this important peptide and its role in iron homeostasis.

Recombinant human erythropoietin (rhEPO) was immobilized on the solid phase to capture anti-rhEPO Abs specifically. After the immunoreactions with Au-labeled goat-anti-rabbit IgG, a diluted HNO3 (2%) was used to dissociate Au nanoparticles which was then introduced to the ICP-MS for measurements. Under the optimized conditions, the calibration graph for anti-EPO Abs was linear in the range of 35.6–500ngmL−1 with a detection limit of 10.7ngmL−1 (3σ, n=9). The relative standard deviation (R.S.D.) for three replicate measurements of 30.9ngmL−1 of anti-EPO Abs was 8.43%. The recoveries of anti-EPO Abs in sera at the spiking level of 50, 100, 150, 200 and 400ngmL−1 were 99.2%, 101.5%, 95.0%, 94.0% and 102.9%, respectively. For the real sample analysis, 26 samples from healthy people and 53 samples from patients with rhEPO treatments were studied. One sample from patients showed significantly higher anti-EPO Abs from other samples, indicating a possibility of immune response of this patient.

Abstract:Pituitary pars intermedia dysfunction (PPID) is a neurodegenerative disease of senior horses. Loss of dopaminergic inhibition of the melanotropes of the pars intermedia leads to increased concentrations of pro-opiomelanocortin (POMC)-derived peptides. Diagnosis is challenging due to pre-analytical variables, such as sample storage, handling, and time to analysis. Our objective was to develop an ELISA for ACTH measurement, which could ultimately form the basis for a stall-side equine ACTH test. We selected 2 ACTH-specific monoclonal antibodies, CBL57 and EPR20361-248, based on the recognition of separate epitopes, strong and rapid color change, and minimal background interference, including no cross-reactivity with themselves, each other, and the test reagents. CBL57 was chosen as the detection antibody (or secondary antibody). EPR20361-248, functionalized on superparamagnetic iron oxide beads, was chosen as the capture antibody (or primary antibody) to bind ACTH in plasma. The incorporation of magnetic beads marks the initial stage in establishing a platform that could potentially be utilized in the field, similar to other stall-side tests. The concentrations of antibodies, magnetic beads, and incubation durations were optimized. Our immunoassay detected unglycosylated rat recombinant ACTH. Further studies are ongoing to optimize and validate our assay using equine plasma and serum samples.
Development and performance evaluation of chemiluminescent immunoassay reagent for serum HER2 protein

Abstract:Objective To develop an automated, economical, quantitative, easy to operate and easily obtained sample detection of human epidermal growth factor receptor-2 (HER2). Methods Chemiluminescence immunoassay was used in vitro to quantitatively detect the content of HER2 protein in serum. One HER2 monoclonal antibody (primary antibody) was coupled to the carboxyl magnetic beads,and another HER2 monoclonal antibody (secondary antibody) was labeled to acridinium ester, to assemble a double antibody sandwich method detection reagent. Results The detection range of HER2 protein in this research was from 0 to 500 ng /ml,and precision was CV lower than 5％. The limited of detection and quantification were 0. 075 and 0. 093 ng/ml, respectively. Linearity range is from 2. 135 to 780. 427 ng/ml. The antiinterference test of HAMA positive serum test showed no interference to this kit. The accelerated stability experiment showed that the reagents could be stably stored for 1 years. Conclusions Compared with Her2 protein chemiluminescence reagents sold on the market, the detection in this study has an excellent expression in various analytical performance,which establish a HER2 protein chemiluminescence detection prototype successfully.
Physiological zinc-binding proteins of medium molecular weight in the rat gut

Abstract:1. Gel filtration on Sephadex G 75 was used to separate the medium-molecular-weight zinc-binding proteins from the soluble fractions from the duodenal and jejuno-ileal segments of the rat gut at 30 min after the intragastric administration of a tracer dose of 65Zn. These proteins were resolved by ion-exchange chromatography on DEAE cellulose. 2. In both the duodenum and jejuno-ileal segment an appreciable fraction of the total soluble Zn was bound in a protein fraction that resembled metallothionein [MT] in its behaviour on gel filtration. These fractions, however, were not homogeneous, but contained several medium-molecular-weight Zn-binding proteins. In the duodenum, but not in the jejuno-ileal segment, two ofthese proteins appeared to be the isometallothioneins, ZnMT-I and ZnMT-11. 3. These results suggest a possible role for MT in the binding of newly-absorbed Zn in the duodenal mucosal cells. They also show that gel filtration alone is insufficient for the identification of MT in the intestine.
