Abstract:Diblock and miktoarm star-shaped thermoresponsive copolymers composed of single-stranded DNA (ssDNA) and poly(N-isopropylacrylamide) were successfully synthesized with combination of atom transfer radical polymerization (ATRP) and click chemistry. This approach should be generalizable to other DNA-functionalized copolymers. Such copolymers self-assemble into spherical micelles with ssDNA corona in aqueous solution above the lower critical solution temperature. The micellar size can be tuned from the macromolecular architecture. These DNA-encoded micellar particles are able to encapsulate and release hydrophobic guest molecules upon changing temperature.

Abstract:Confined microenvironments in biomacromolecules arising from molecular crowding account for their well‐defined biofunctions and bioactivities. To mimick this, synthetic polymers to form confined structures or microenvironments are of key scientific value, which have received significant attention recently. To create synthetic confined microenvironments, molecular crowding effects and topological cooperative effects have been applied successfully, and the key is balance between self‐association of structural units and self‐repulsion from crowding‐induced steric hindrance. In this article, formation of confined microenvironments from stimuli‐responsive dendronized polymers carrying densely dendritic oligoethylene glycols (OEGs) moieties in their pendants is presented. These wormlike thick macromolecules exhibit characteristic thermoresponsive properties, which can provide constrained microenvironments to encapsulate effectively guest molecules including dyes, proteins, or nucleic acids to prevent their protonation or biodegradation. This efficient shielding effect can also mediate chemical reactions in aqueous phase, and even enhance chirality transferring efficiency. All of these can be switched off simply through the thermally‐induced dehydration and collapse of OEG dendrons due to the amphiphilicity of OEG chains. Furthermore, the switchable encapsulation and release of guests can be greatly enhanced when these dendronized polymers are used as major constituents for fabricating bulk hydrogels or nanogels, which provide a higher‐level confinement.

Abstract:Polymeric microgels form a classof promising materials for variousapplications based on their highly tunable swelling and cooperativeinteractions between the cross-linked polymer chains. Here, we reporton a convenient route for the fabrication of polymeric microgels through in situ cross-linking polymerization of thermally collapsedmacromonomers carrying 3-fold dendritic oligoethylene glycol (OEG)pendants, affording thermoresponsive dendronized microgels with intriguingmicroconfinement. This methodology has been proven to be versatileand can be used to prepare intelligent microgels through homopolymerizationof thermoresponsive dendronized macromonomers, or via copolymerization with either hydrophobic or hydrophilic, and dendronizedor nondendronized comonomers. The thermoresponsive microgels of uniformsizes undergo shrinking or swelling by changing the temperature aboveor below their phase transition temperatures and thus exhibit tunablemicroconfinement to encapsulated solvatochromic dye molecules. Thisoutstanding capacity for confinement by the microgels is comparedto linear copolymers with or without dendritic architectures and isproposed to originate from both dendritic crowding effects and cooperativeinteractions between polymer chains within the network. This remarkablefeature of microconfinement opens a new era for microgels to reversiblyencapsulate and protect guest molecules and provides a platform tomanipulate the functions of the encapsulated guests through externalstimuli.

Abstract:Thermoresponsive dendritic polymers combine the "smart" property of thermoresponsive polymers and the nano-sized, tunable non-linear architectures of dendritic polymers, which have been found applications from materials science to biology. Recent research progresses on thermoresponsive dendritic polymers including dendrimers, dendronized polymers and hyperbranched polymers are reviewed. In contrast to their linear counterparts, the thermoresponsive behavior of these dendritic polymers can be influenced by the dendritic architecture, which paved a way to fabricate molecule-based nano-devices for encapsulation and recognition. Their synthetic methodology, structural-related thermoresponsive behavior as well as their applications in nanomaterials, sensors and biomaterials are described in details.
Topological Effects of Dendronized Polymers on Their Thermoresponsiveness and Microconfinement

Abstract:Topological structures can provide dendritic polymers with featured properties through cooperative effects beyond their molecular structures. Herein, we report the synthesis of a series of oligoethylene glycol (OEG)-based dendronized polymers, which carry the first generation of 2-fold, 4-fold, or 6-fold dendritic OEG pendants with methoxyl or ethoxyl terminals. Depending on the OEG branching density, these polymers show different molecular thicknesses and exhibit characteristic thermoresponsiveness with phase transition temperatures in the range of 16.6-67.2 degrees C. Furthermore, the higher the OEG branching density, the higher the shielding to the interior parts from the dendritic OEGs cooperatively to exhibit "molecular envelope" effects. This molecular microconfinement formed from the crowded OEG dendrons was further investigated through covalently linked or physically mixed dye. Topological structures of the polymers are discovered to play a key role in encapsulation of the dye guest and can modulate its transition within the aggregates through the thermally induced phase transitions.

Abstract:Convenient chemical modification of biomacromolecules to create novel biocompatible functional materials satisfies the current requirements of sustainable chemistry. Dendronization of chitosan with dendritic oligoethylene glycols (OEGs) paves a strategy for the preparation of functional dendronized chitosans (DCSs) with unprecedent thermoresponsive behavior, which inherit biological features from polysaccharides and the topological features from dendritic OEGs. In addition, densely packed dendritic OEG chains around the backbone provide efficient cooperative interactions and form an intriguing confined microenvironment based on the degradable biopolymers. In this perspective, we describe the principle for the preparation of the thermoresponsive DCSs, and focus on the molecular envelop effect from the hydrophobic microconfinement to the encapsulated guest molecules or moieties. Particular attention is put on their capacity to regulate behavior and the functions of the encapsulated guests through thermally-mediated dehydration and collapse of the densely packed dendritic OEGs. We believe that the methodology described here may provide prospects for the fabrication of functional materials from biomacromolecules, especially when used as environmentally friendly nanomaterials or in accurate diagnosis and therapy. Dendronization of chitosan with 3-fold dendritic oligoethylene glycols generates stimuli-responsive and biocompatible dendronized chitosans, which form microconfinement to protect guests or modulate reactions. This confinement can be greatly enhanced through the formation of hydrogels.

Abstract:Thermoresponsive biotinylated dendronized copolymers carrying dendritic oligoethylene glycol (OEG) pendants were prepared via free radical polymerization, and their protein recognitions based on biotin–avidin interaction investigated. Both first (PG1) and second generation (PG2) dendronized copolymers were designed to examine possible thickness effects on the interaction between biotin and avidin. Inherited from the outstanding thermoresponsive properties from OEG dendrons, these biotinylated cylindrical copolymers show characteristic thermoresponsive behavior which provides an envelope to capture avidin through switching temperatures above or below their phase transition temperatures (Tcps). Thus, the recognition of polymer-supported biotin with avidin was investigated with UV/vis spectroscopy and dynamic laser light scattering. In contrast to the case for PG1, the increased thickness for copolymer PG2 hinders partially and inhibits the recognition of biotin moieties with avidin either below or above its Tcp. This demonstrates the significant architecture effects from dendronized polymers on the biotin moieties to shift onto periphery of the collapsed aggregates, which should be a prerequisite for protein recognition. These kinds of novel thermoresponsive copolymers may pave a way for the interesting biological applications in areas such as reversible activity control of enzyme or proteins, and for controlled delivery of drugs or genes.

Abstract:Regulation of protein activity is important in their applications for biomedicine and therapeutics. Here, an approach for the regulation of protein bioactivity through molecular confinement provided by oligoethylene glycol (OEG)-based dendronized chitosan (DCS) hydrogels is reported. Structural effects on their thermoresponsiveness are investigated. The highly transparent hydrogels are formed from thermoresponsive DCSs through their thermal dehydration and exhibit an intriguing reversible sol-gel transition property when triggered at physiological temperatures. The thermo-gelling behavior and mechanical strength of these hydrogels are investigated, and possible effects from hydrophobicity of the OEG dendrons, grafting rates of the dendrons on the chitosan main chain, and solid content of polymers are examined. These DCS hydrogels are found to have lamellar morphologies and can provide characteristic hydrophobicity microenvironments formed through the crowded OEG dendrons, which show a higher level of confinement to guest proteins. This allows the DCS hydrogels remarkable activity protection capability to proteins. Furthermore, these DCS hydrogels inherit the degradability from chitosan, allowing protein release from these hydrogels through the controllable ways without impairing their activities.

Abstract:Thermoresponsive dendronized polymers constituted with a polyacylhydrazone backbone pendanted with oligoethylene glycol (OEG)-based dendrons were synthesized through polycondensation of dialdehydes and diacylhydrazines. First (G1) and second (G2) generation OEG dendrons were selected for comparison to examine the dendritic effects. Inherited from OEG dendrons, these polymers showed characteristic thermoresponsive behavior. By virtue of the dynamic property of the acylhydrazone linkage, formation of these polymers are concentration and pH-dependent. A dimer from monoaldehyde and monoacylhydrazine was prepared as a model to support the remarkable macromolecular effects for enhanced structural stability of polyacylhydrazones. Constitutional stability of these polyacylhydrazones were also testified by switching solutions to strong acidic or basic conditions. The structural vibration was simply testified by addition of a new monomer as a competitor to dendronized polyacylhydrazones, thus, resulting in polymers with tunable phase transition temperatures. Furthermore, G1 dendrons were found to provide efficient shielding for acylhydrazones from an affinity to metal ions, but this shielding effect diminished and exhibited specific interactions with Cu2+ when the dendron collapsed.

Abstract:Thermoresponsive dendronized polymers, displaying remarkable phase behavior, are currently being studied for their potential exploitation as polymeric sensors and biomaterials. Understanding the conformational transitions occurring at the LCST is essential for improved design and translation of these polymers. The combination of NMR and molecular dynamics simulations opens a unique window onto the thermal behavior, showing that the peripheries of the dendrons, while driving the thermal properties, largely retain mobility above the critical temperature. The cores of the dendrons and the polymeric main chain are highly rigid below the thermal transition and increasingly so above the LCST. Both the experimental and computational studies reveal stretching of the interior segments of the dendrons with associated changes in spatial arrangements of the structural units. Furthermore, diffusion-ordered NMR and DLS below and above the LCST show a further hierarchy of dynamics within different size aggregates. The combination of the detailed experimental study and molecular dynamics simulations provides a detailed understanding of thermoresponsive behavior of these dendronized polymers.
Impact of Cyclization and Dendronization on Multi-Tunable Thermoresponsive Behaviors of Polyacrylamide Copolymers

Abstract:Topological constraints can play a crucial role in the thermoresponsive behaviors of architectural polymers. To reveal phase transitions under confined conditions, two pairs of linear and cyclic polyacrylamide-type thermoresponsive heterofunctional-dendron-bearing polymers (THDPs) with pH-responsive dimethylamino groups, namely, PNADA and PNADD, are designed. Thermoresponsive behaviors can be efficiently regulated by topology, composition, solvent isotopes and pH. Despite the cyclization-induced increase in the LCST-type cloud point (T-c,T-l) of PNADA solutions, cyclic PNADD solutions only present a medium T-c,T-l as compared with the linear analogue with dual LCSTs at pH 7.5 and 10 due to a more confined microenvironment. A solvent switch can alter the type and temperature of phase transitions, in which linear PNADD shows unusual dual LCST and UCST behavior in D2O. The incorporation of bulky dendrons into cyclic architecture can endow subunits with remarkably confined mobility and a distinct response upon heating, during which morphological transitions among spheres, vesicles, nanoribbons and lamellae occur. Owing to their abundant compositions and functions, topologically confined THDPs can hold great promise in the on-demand regulation of phase transitions and nanostructures.
Thermoresponsive Double Network Cryogels from Dendronized Copolymers Showing Tunable Encapsulation and Release of Proteins.

Abstract:A series of double-network cryogels (DNCs) were prepared from oligoethylene glycol (OEG)-based dendronized copolymers as the first network and polyvinyl alcohol (PVA) as the second network. The dendronized copolymers were composed of dendritic OEG units, as well as triethoxysilyl, amine or carboxyl groups. The dendritic OEGs ensured characteristic thermoresponsiveness and triethoxysilyl groups were for crosslinking, while amino and carboxyl groups were designed to provide charged species for specifically interacting with proteins. PVA behaves as a mutual support which ensures the stability and mechanical properties of the cryogels. These DNCs exhibited good thermoresponsiveness and biocompatibility. Upon increasing the temperature above their cloud points (Tcps), the cryogels became dehydrated with enhanced hydrophobicity, and thus their porous structures shrank. This property was utilized to mediate encapsulation and release of proteins. The loading and release efficiencies of different proteins were found to be dominated by hydrophilic to hydrophobic transitions of the first network, and at the same time, by the properties of proteins. At room temperature, the cryogel could efficiently capture model proteins, while above Tcps, more than 80% of lysozyme could be released in 24 h with enzyme activity remaining unchanged. These series of smart cryogels based on thermoresponsive dendronized copolymers may provide promising applications for the reversible capture of enzymes or proteins.
Confinement Effects On The Kinetics And Thermodynamics Of Protein Dimerization

Abstract:In the cell, protein complexes form by relying on specific interactions between their monomers. Excluded volume effects due to molecular crowding would lead to correlations between molecules even without specific interactions. What is the interplay of these effects in the crowded cellular environment? We study dimerization of a model homodimer when the mondimers are free and when they are tethered to each other. We consider a structured environment: Two monomers first diffuse into a cavity of size L and then fold and bind within the cavity. The folding and binding are simulated by using molecular dynamics based on a simplified topology based model. The confinement in the cell is described by an effective molecular concentration C similar to L-3. A two-state coupled folding and binding behavior is found. We show the maximal rate of dimerization occurred at an effective molecular concentration C-op similar or equal to 1 mM, which is a relevant cellular concentration. In contrast, for tethered chains the rate keeps at a plateau when C < C-op but then decreases sharply when C > C-op. For both the free and tethered cases, the simulated variation of the rate of dimerization and thermodynamic stability with effective molecular concentration agrees well with experimental observations. In addition, a theoretical argument for the effects of confinement on dimerization is also made.
Bifunctional and Bioreducible Dendrimer Bearing a Fluoroalkyl Tail for Efficient Protein Delivery Both In Vitro and In Vivo

Abstract:Rational design of stimuli-responsive polymers for cytosolic protein delivery with robust efficiency is of great importance but remains a challenging task. Here, we reported a bioreducible and amphiphilic dendrimer bearing a fluoroalkyl tail for this purpose. The fluorolipid was conjugated to the focal point of a cysteamine-cored polyamidoamine dendrimer via disulfide bond, while phenylboronic acid moieties were decorated on dendrimer surface for efficient protein binding. The yielding polymer showed high protein binding capability and complex stability and could efficiently release the cargo proteins in a glutathione-responsive manner. The designed polymer was effective in the delivery of various membrane-impermeable proteins into living cells with reserved bioactivities. In addition, the polymer efficiently delivered a toxin protein saporin into 4T1 breast cancer cells and inhibited the tumor growth in vivo after intravenous injection. Remarkable Structure Effects on Thermoresponsive Properties of Dendritic Macromolecules

Abstract:Amphiphilic dendritic macromonomers and their corresponding dendronized polymers were synthesized, and their thermoresponsive properties investigated. These dendritic macromolecules are constructed with a second generation lysine-based dendron as the interior (hydrophobic part) and oligoethylene glycol (OEG) linear chains or dendrons (hydrophilic part) covered in their periphery. By changing from OEG linear chains to OEG dendrons, these dendritic macromolecules carry on their periphery different density of OEG moieties, to investigate the structural effects on their thermoresponsiveness. Topology of dendritic macromolecules changes through polymerization, and the fan-shaped dendritic macromonomers transfer into the corresponding cylindrically shaped dendronized polymers. Furthermore, the amphiphilic characteristic of these dendritic entities can be switched due to the thermally-induced dehydration of OEG moieties. It was found that topology, OEG density in the periphery and switchable amphiphilicity of these dendritic macromolecules show significantly effects on their thermoresponsiveness.
Impact of Confinement within a Hydrogel Mesh on Protein Thermodynamic Stability and Aggregation Kinetics

Abstract:Though protein stability and aggregation have been well characterized in dilute solutions, the influence of a confining environment that exists (e.g., in intercellular and tissue spaces and therapeutic formulations) on the protein structure is largely unknown. Herein, the effects of confinement on stability and aggregation were explored for proteins of different sizes, stability, and hydrophobicity when encapsulated in hydrophilic poly(ethylene glycol) hydrogels. Denaturation curves show linear correlations between confinement size (mesh size) and thermodynamic stability, i.e., unfolding free energy and surface area accessible for solvation (represented by m-value). Two clusters of protein types are identifiable from these correlations; the clusters are defined by differences in protein stability, surface area, and aggregation propensity. Proteins with higher stability, larger surface area, and lower aggregation propensity (e.g., lysozyme and myoglobin) are less affected by the confinement imposed by mesh size than proteins with lower stability, smaller surface area, and higher aggregation propensity (e.g., growth hormone and aldehyde dehydrogenase). According to aggregation kinetics measured by thioflavin T fluorescence, confinement in smaller mesh sizes resulted in slower aggregation rates than that in larger mesh sizes. Compared to that in buffer solution, the confinement of a hydrophobic protein (e.g., human insulin) in the hydrogels accelerates protein aggregation. Conversely, the confinement of a hydrophilic protein (e.g., human amylin) in the hydrogels decelerates or prevents aggregation, with the rates of aggregation inversely proportional to mesh size. These findings provide new insights into protein conformational stability in confined microenvironments relevant to various cellular, tissue, and therapeutics scenarios.

Abstract:Polymer-mediated intracellular protein delivery systems are important for the development of protein-based biotechnologies and therapeutics. However, intracellular delivery of cargo proteins in the presence of serum remains challenging due to competitive binding of serum proteins with the polymers. Here, we reported a dendrimer engineered with a high density of 4-diethylaminophenyl groups on the surface to address this issue. The dendrimer showed a pH-responsive phase-transition behavior and could assemble with cargo proteins into stable nanoparticles in serum solutions. It efficiently delivered cargo proteins into living cells, and exhibited a pH-responsive disassembly behavior after cell internalization. As a result, various cargo proteins were delivered into the cytosol of living cells with maintained bioactivity. Crowding for Confinement: Reversible Isomerization of First-Generation Donor-Acceptor Stenhouse Adduct Derivatives in Water Modulated by Thermoresponsive Dendritic Macromolecules

Abstract:Mimicking nature, the reversible isomerization of hydrophobic dyes in aqueous solutions is appealing for bio-applications. Here, we report on the reversible isomerization of first-generation solvatochromic donor-acceptor Stenhouse adducts (DASAs) in water within dendritic matrices, realized either through the dendronization of DASAs or the incorporation of DASA pendants into dendronized copolymers. These dendritic macromolecules contain three-fold dendritic oligoethylene glycols (OEGs), which afford the macromolecules water-solubility and unprecedented thermoresponsive behavior. The thermoresponsive behavior of both dendronized DASAs and dendronized copolymers is dominated by the peripherals of dendritic OEGs. However, the hydrophilicity of the acceptor from DASA moieties also play a role in mediating their thermal phase transitions, and more importantly, tailor the hydrophobic interactions between dendritic OEGs and DASA moieties. Intriguingly, dendritic topologies contribute confinement to encapsulate the DASA moieties through crowding effects, and cooperative interactions from the crowded dendritic OEGs modulate the DASA moieties with different isomerization in aqueous media. The thermally induced collapse of dendritic OEGs, accompanied by the aggregation of dendritic macromolecules, leads to the formation of hydrophobic domains, which exert enhanced crowding effects to efficiently encapsulate the DASA moieties. Compared to the low molar mass of dendronized DASAs, thermally collapsed dendronized copolymers can efficiently retard the hydration of DASA pendants through cooperation between neighboring dendritic OEGs and afford the DASA pendants with better confined microenvironments to mediate their isomerization recovery by up to 90% from a cyclic charged (hydrophilic) state into a noncharged (hydrophobic) linear state in water. This dendritic confinement exhibits excellent fatigue resistance after several cycles of alternating photo-irradiation and thermal annealing at elevated temperatures.

Effects of Confinement on Protein Folding and Protein Stability

Abstract:In a cell, proteins exist in crowded environments; these environments influence their stability and dynamics. Similarly, for an enzyme molecule encapsulated in an inorganic cavity as in biosensors or biocatalysts, confinement and even surface effects play important roles in its stability and dynamics. Using a minimalist model (two-dimensional HP lattice model), we have carried out Monte Carlo simulations to study confinement effects on protein stability. We have calculated heat capacity as a function of temperature using the histogram method and results obtained show that confinement tends to stabilize the folded conformations, consistent with experimental results (some reported here) and previous theoretical analyses. Furthermore, for a protein molecule tethered to a solid surface the stabilization effect can be even greater. We have also investigated the effects of confinement on the kinetics of the refolding and unfolding processes as functions of temperature and box size. As expected, unfolding time increases as box size decreases, however, confinement affects folding times in a more complicated way. Our theoretical results agree with our experimentally observed trends that thermal stability of horseradish peroxidase and acid phosphatase, encapsulated in mesoporous silica, increases as the pore size of the silica matrix decreases.
Dynamic control of protein folding pathway with a polymer of tunable hydrophobicity.

Abstract:While the knowledge of protein folding in a dilute solution is now well-advanced, little is known of the influence of surrounding conditions on the folding kinetics, in particular when the protein is in a dynamically responsive environment. Here we report a new procedure to control the pathways of protein folding by using a thermally responsive polymer that varies its hydrophobicity concomitant with the protein structural changes. The advantages of folding in a dynamic environment have been demonstrated first by Langevin dynamics simulations on the basis of coarse-grained models for both the protein and polymer and then by experiments for lysozyme refolding in the presence of poly(N-isopropylacrylamide-co-N-tert-butylacrylamide), a thermal responsive polymer that varies its hydrophobicity in response to temperature. The simulation suggests that decreasing the polymer hydrophobicity during the folding process may result in an optimized free-energy landscape that enhances both the folding yield and kinetics. The experiments affirm that an optimal folding condition can be identified when structural transitions of the protein collaborate with the polymer hydrophobicity tuned by variation of temperature.
