Abstract:In mixtures of water and dimethyl sulfoxide, 4 '-(n-dodecyloxy)-1,1 '-biphenyl-4-carbonyl Ala-Ala dipeptides can self-assemble into tubular structures that are formed by coiled nanoribbons. The twist and stacking handedness of biphenylene groups were studied using circular dichroism and confirmed by theoretical chemical calculations. The handedness of the coiled nanoribbons and the stacking handedness of biphenylene groups are controlled by the chirality of alanine at the C-terminus, whereas the twist handedness of biphenylene groups is determined by the chirality of alanine at the N-terminus. H-1 NMR spectra indicated that the hydrogen bond formed by the N-H group of alanine at the N-terminus plays an important role in the formation of organic self-assemblies. On the basis of small-angle X-ray scattering characterization, a dimer structure was proposed to form through the terminal COOH groups.

Abstract:Supramolecular peptide assemblies have been widely used for the development of biomedical, catalytical, and optical materials with chiral nanostructures in view of the intrinsic chirality of peptides. However, the assembly pathway and chiral transformation behavior of various peptides remain largely elusive especially for the transient assemblies under out-of-equilibrium conditions. Herein, the N-fluorenylmethoxycarbonyl-protected phenylalanine-tyrosine dipeptide (Fmoc-FY) was used as a peptide assembly platform, which showed that the assembly proceeds multistep evolution. The original spheres caused by liquid-liquid phase separation (LLPS) can nucleate and elongate into the formation of right-handed helices which were metastable and easily converted into microribbons. Interestingly, a bipyridine derivative can be introduced to effectively control the assembly pathway and induce the formation of thermodynamically stable right-handed or left-handed helices at different stoichiometric ratios. In addition, the chiral assembly can also be regulated by ultrasound or enzyme catalysis. This minimalistic system not only broadens the nucleation-elongation mechanisms of protein aggregates but also promotes the controllable design and development of chiral biomaterials.

Abstract:Modulation of chiroptics, chiral phenomena of tonic switches. One of the most effective methods for applications, including chirality-specific biosensing and pho- the optical properties, is, pivotal in a variety of advanced :achieving this control is assembly of the optical moieties into chiral nanostructures. Lipopeptide self-assemblies have been extensively employed as soft templates to organize composites into low-dimensional superstructures due to their rigidity and ease of functionalization. Therefore, an appealing approach is to provide chiroptical control by using lipopeptide self assemblies as templates to assemble chromophores. Herein, two lipopeptidic molecules, namely, C-14-FFK and C-14-FK, composed of phenylalanine and lysine residues conjugated to a myristic acid chain, were custom-designed. Spectroscopic and microscopic characterizations indicated that C-14-FFK self-assembled to wide, slightly left-handed nanoribbons, while C-14-FK formed narrow, intensely right-handed nanofibers. The different chirality was derived from the distinct self-assembly driving forces, especially the molecular bending dimensions. These superstructures' presented an ideal capability to serve as soft templates to assemble porphyrin (ZnTPyP) through noncovalent electrostatic attractive interactions, or assemble the phenolic groups through covalent conjugation to peptide backbones. The distinct exciton coupling of the chromophores allowed their achiral optics to become chiral, showing negative Cotton effect when templated by nanoribbons and positive Cotton effect with nanofibers as templates. Following replacement of the lipopeptides with their D-type enantiomers, the handedness of the superstructures and the associated chiroptics were reversed and presented "mirror" symmetric CD signals to their L-type counterparts. These findings may pave the way to the formation of morphologically and chioptically controllable nanomaterials.

Abstract:Eight dipeptides, composed of phenylalanine and phenylglycine, that are able to self-assemble into twisted nanoribbons in deionized water are synthesized. The handedness of the nanoribbons is controlled by the chirality of the phenylalanine and the steric hindrance owing to the phenyl group of the phenylglycine. When the phenylalanine is at the C-terminal, pi-pi stacking by the phenyl groups, hydrogen bonding by the NH group of the phenylalanine, and hydrophobic associations of the alkyl chains control the stacking of the molecules. When phenylglycine is at the C-terminal, the chiral pi-pi stacking by the phenyl groups of the phenylalanines is suppressed. The hydrogen bonds formed by the NH groups of the phenylalanines had a greater contribution on forming organic self-assemblies than those formed by the NH groups of the phenylglycines.

Abstract:Four Phe-Ala lipodipeptides with different stereochemical structures are observed to self-assemble into twisted nanoribbons in water. The handedness of the twisted nanoribbons is controlled by the chirality of the phenylalanine near the alkyl chain, while the stacking handedness of the phenyl and carbonyl groups is determined by the alanine at the C-terminal. The homochiral and heterochiral lipodipeptides self-assemble into parallel and antiparaller beta-sheet structures, respectively. The H-1 NMR, FTIR, X-ray diffraction, and circular dichroism characterizations indicate that these phenomena are mainly driven by the interaction between neighboring phenyl groups and H-bonding among the amide groups.

Abstract:Four dipeptides with alkyl chains derived from L- and D-valines were synthesized, the handedness of their self-assemblies were controlled by the valine chirality at the terminals. The stacking of the carbonyl groups plays an important in the formation of chiral organic self-assemblies. Chiral 4,4'-biphenylene bridged polybissilsesquioxane nanotubes were prepared using the self-assemblies of these dipeptides as the templates. The chirality of the polysilsesquioxane nanotubes was mainly controlled by the valines at the terminals of the dipeptides, which was transferred from the valines at the terminals through electrostatic interaction. The valines near the alkyl chains could also affect the polysilsesquioxane chirality through hydrogen bonding.

Abstract:Host-guest interactions are widely employed in constructing responsive materials, although less is known to manipulate the chiral property of materials using such host-guest dynamics. With the supramolecular self-assembly based on beta-cyclodextrin (beta-CD) and alkyl amines (CH3(CH2)(n-1)NH2), we report that faster host-guest dynamics induces a dipole located above the cavity of beta-CD, whereas slower dynamics create in-cavity dipole. These two scenarios correspond to negative and positive chiral signals, respectively. Considering that a larger fraction of amines facilitates faster exchange between the threaded and unthreaded amines, the chiral signal for the right-handed helical ribbons can be manipulated simply by alternatively increasing the fraction of amines and beta-CD. Excitingly, enzyme responsive supramolecular chirality is obtained as a result of shifting the molar ratio by enzyme triggered hydrolysis of beta-CD. We expect that this strategy may open up an area of rationally designed chiral supramolecular materials based on host-guest chemistry.

Abstract:The handedness of the self-assemblies of several series of homogeneous lipodipeptide sodium salts is controlled by the chirality of the amino acid at the C-terminus. This phenomenon is named the "C-terminal determination'' rule. Herein, four heterogeneous C17H35CO-Ala-Phe sodium salts, designed and synthesized by changing the chirality of the amino acids, were able to self-assemble into twisted nanoribbons in deionized water. The handedness of the nanoribbons was controlled by the chirality of the alanine, which does not follow the "C-terminal determination'' rule. This phenomenon might be because the homochiral and heterochiral lipodipeptide sodium salt molecules packed into different structures. The X-ray diffraction and circular dichroism characterizations indicated that the molecules self-assembled into an interdigitated bilayer structure and there were no strong pi-pi stackings between neighboring phenyl groups.

Abstract:A series of C13H27CO-Gly-Ala-Ala tripeptides with different chiralities are synthesized. They can form physical gels in toluene and 1,4-dioxane. They self-assemble into right- or left-handed twisted nanoribbons in a mixture of methanol and water. It is found that the handedness of the nanoribbons is controlled by the chirality of the alanine residue at the center. Circular dichroism characterization also revealed that the peptide segment packing handedness was dominated by the chirality of the central alanine residue. The formation of self-assemblies was studied using H-1 NMR and particle size distribution. Upon decreasing the temperature, the micelle forms a fine aggregate and then grows into twisted nanoribbons. It was also found from H-1 NMR characterization that H-bonds formed by the central alanine residue played an important role in the formation of the self-assemblies. The X-ray diffraction patterns showed that the tripeptides packed into lamellar structures.

Abstract:We report interesting hierarchical self‐assembled architectures from a designed amphiphilic peptide. The bisignate cotton effects in circular dichroism spectra show typical peptide aggregation‐induced. The observation of peptide assembly structures from initial particles and fibrils to ribbon structures is supported by microscopy (atomic force microscopy and transmission electron microscopy). The visualization of individual peptide at the single molecular level offered insights of the intermolecular interactions responsible for the formation of aggregates, which is investigated by scanning tunneling microscopy. The orientation of intermolecular bonds between carboxylic and amine group and the hydrophobic interactions between alanine residues could be the dominant driving force for the assembly chirality at near‐neutral pH. The single molecular and aggregate level evidence in this manuscript will shed light on the understanding of hierarchical chiral self‐assembly pathway and the underlying mechanism. Abstract:A series of lipotripeptide stereoisomers based on alanine were synthesized, and their self-assembling behaviors were studied by means of circular dichroism spectra, ATR-IR, temperature-dependent 1H NMR, and X-ray diffraction patterns. In the mixed solvent of hexafluoroisopropanol/H2O (1/9, v/v), eight lipotripeptides were able to self-assembled into nanoflakes or nanoribbons driven by the hydrophobic association of alkyl chains, intermolecular hydrogen bonding among carboxyl groups at C-terminal and amide groups of alanine moieties in the peptide segment. It was found that the stacking chirality of carbonyl groups was determined by the chirality of alanine residue at C-terminal (i.e., "C-terminal determination" rule). Moreover, our research also highlighted the intermolecular hydrogen bonding on amide groups of each alanine residue, terminal carboxyl as well as the molecular packing structures can be subtly manipulated by changing the stereochemical sequence of peptide segment.

Abstract:Chiral self-assembly of peptides has attracted great interest owing to their promising applications in biomedicine, chemistry, and materials science. However, compared with the rich knowledge about their chiral self-assembly at the molecular or nanoscale, the formation of long-range-ordered hierarchical helical arrays (HHAs) from simple peptides remains a formidable challenge. Herein, we report the self-templated assembly of an amyloid-like dipeptide into long-range-ordered HHAs by their spontaneous fibrillization and hierarchical helical assembly within a confined film. The chiral interactions between the peptide and diamines result in geometry frustration and the phase transition of self-assembling peptide films from achiral spherulite structures into chiral HHAs. By changing the chirality and enantioselective interactions, we can control the phase behavior, handedness, and chiroptics of the self-assembled HHAs precisely. Moreover, the redox activity of the HHAs allows the in situ decoration of nanoparticles with high catalytic activity. These results provide insights into the chiral self-assembly of peptides and the fabrication of highly ordered materials with complex architectures and promising applications in chiroptics and catalysis.

Chiral inversion induced by aromatic interactions in short peptide assembly

Abstract:Although hydrophobic interactions provide the main driving force for initial peptide aggregation, their role in regulating suprastructure handedness of higher-order architectures remains largely unknown. We here interrogate the effects of hydrophobic amino acids on handedness at various assembly stages of peptide amphiphiles. Our studies reveal that relative to aliphatic side chains, aromatic side chains set the twisting directions of single β-strands due to their strong steric repulsion to the backbone, and upon packing into multi-stranded β-sheets, the side-chain aromatic interactions between strands form the aromatic ladders with a directional preference. This ordering not only leads to parallel β-sheet arrangements but also induces the chiral flipping over of single β-strands within a β-sheet. In contrast, the lack of orientational hydrophobic interactions in the assembly of aliphatic peptides implies no chiral inversion upon packing into β-sheets. This study opens an avenue to harness peptide aggregates with targeted handedness via aromatic side-chain interactions.

Abstract:Self-assembly of chiral nanostructures is of considerable interest, since the ability to control the chirality of these structures has direct ramifications in biology and materials science. A new approach to design chiral nanostructures from self-assembly of N-(9-fluorenylmethoxycarbonyl)-protected phenylalanine-tryptophanlysine tripeptides is reported. The terminal charges can induce helical twisting of the assembled beta-sheets, enabling the formation of well-defined chiral nanostructures. The degree and direction of twisting in the beta-sheets can be precisely tailored through in situ pH and temperature modulations. This enables the assembly of reconfigurable chiral nanomaterials with easily adjustable size and handedness. These results offer new insight into the mechanism of helical twist formation, which may enable the precise assembly of highly dynamical materials with potential applications in biomedicine, chiroptics, and chiral sensing.
Solvent-Induced Handedness Inversion of Dipeptide Sodium Salts Derived from Alanine

Abstract:The relationship between amino acid sequences and their resulting nanostructure has been well studied, but that between amino acid chirality and nanostructure handedness has not. Four dipeptide sodium salts with long alkyl chains derived from L- and D-alanines were synthesized. The behavior of their self-assembly into physical gels in water and THF was studied using field-emission scanning electron microscopy, circular dichroism (CD), FT-IR spectroscopy, (1)H NMR spectroscopy, and X-ray diffraction. The dipeptide salts organized into twisted nanoribbons, whose handedness was controlled by the terminal alanine chirality. The handedness of nanoribbons formed in water was opposite to that of those formed in THF. The dipeptide salts self-assembled into similar interdigitated bilayer structures in water and THF, but CD spectra of the gels indicated that the stacking of carbonyl groups was opposite. The formation of this handedness inversion is proposed to arise from the difference in interlayer distance and chiral stacking of carbonyl groups near the C-terminals.

Abstract:Self-assembly of peptides and amyloid fibrils offers an appealing approach for creating chiral nanostructures, which has promising applications in the fields of biology and materials science. Although numerous self-assembled chiral materials have been designed, the precise control of their twisting tendency and their handedness is still a challenge. Herein, we report the self-assembly of chiral nanostructures with precisely tailored architectures by changing the amino acid sequences of the peptides. We designed a series of self-assembling tripeptides bearing different ʟ-amino acid sequences. The peptide with ʟ-Phe-ʟ-Phe sequence preferred to self-assemble into left-handed nanohelices, while with ʟ-Phe-ʟ-Trp, right-handed nanohelices would be formed. Moreover, the diameter of the self-assembled nanohelices could be tailored by changing the terminal amino acids (His, Arg, Ser, Glu and Asp). Circular dichroism (CD) and molecular dynamics simulations (MDs) revealed that both of the right- and left-handed nanohelices formed by the tripeptides showed negative cotton effects in the peptide adsorption region, but exhibited nearly opposite CD cotton effects in the aromatic regions. These results indicated that the handedness of the self-assembled helical nanofibers was not only determined by the chirality of the peptide backbone, but also is closely related to the aromatic stacking, hydrogen bonding and steric interactions induced by the side chains. The findings deepen our understanding on the chiral self-assembly of peptide and offer opportunities for the creation of highly functional chiral nanomaterials.
