Abstract:Modulation of chiroptics, chiral phenomena of tonic switches. One of the most effective methods for applications, including chirality-specific biosensing and pho- the optical properties, is, pivotal in a variety of advanced :achieving this control is assembly of the optical moieties into chiral nanostructures. Lipopeptide self-assemblies have been extensively employed as soft templates to organize composites into low-dimensional superstructures due to their rigidity and ease of functionalization. Therefore, an appealing approach is to provide chiroptical control by using lipopeptide self assemblies as templates to assemble chromophores. Herein, two lipopeptidic molecules, namely, C-14-FFK and C-14-FK, composed of phenylalanine and lysine residues conjugated to a myristic acid chain, were custom-designed. Spectroscopic and microscopic characterizations indicated that C-14-FFK self-assembled to wide, slightly left-handed nanoribbons, while C-14-FK formed narrow, intensely right-handed nanofibers. The different chirality was derived from the distinct self-assembly driving forces, especially the molecular bending dimensions. These superstructures' presented an ideal capability to serve as soft templates to assemble porphyrin (ZnTPyP) through noncovalent electrostatic attractive interactions, or assemble the phenolic groups through covalent conjugation to peptide backbones. The distinct exciton coupling of the chromophores allowed their achiral optics to become chiral, showing negative Cotton effect when templated by nanoribbons and positive Cotton effect with nanofibers as templates. Following replacement of the lipopeptides with their D-type enantiomers, the handedness of the superstructures and the associated chiroptics were reversed and presented "mirror" symmetric CD signals to their L-type counterparts. These findings may pave the way to the formation of morphologically and chioptically controllable nanomaterials.

Abstract:Chiral self-assembly of peptides has attracted great interest owing to their promising applications in biomedicine, chemistry, and materials science. However, compared with the rich knowledge about their chiral self-assembly at the molecular or nanoscale, the formation of long-range-ordered hierarchical helical arrays (HHAs) from simple peptides remains a formidable challenge. Herein, we report the self-templated assembly of an amyloid-like dipeptide into long-range-ordered HHAs by their spontaneous fibrillization and hierarchical helical assembly within a confined film. The chiral interactions between the peptide and diamines result in geometry frustration and the phase transition of self-assembling peptide films from achiral spherulite structures into chiral HHAs. By changing the chirality and enantioselective interactions, we can control the phase behavior, handedness, and chiroptics of the self-assembled HHAs precisely. Moreover, the redox activity of the HHAs allows the in situ decoration of nanoparticles with high catalytic activity. These results provide insights into the chiral self-assembly of peptides and the fabrication of highly ordered materials with complex architectures and promising applications in chiroptics and catalysis.

Abstract:Chirality is one of the most important structural features in biological systems. Peptides, as a class of small biomolecules, can self-assemble into various chiral supramolecular polymers with unprecedented levels of diversity and complexity, which have practical ramifications in various fields from catalysis to sensing, optics, and biomedicine. In this review, we will focus on the chiral transfer during peptide self-assembly from the molecular scale to the nano-, micro- and mesoscale, and highlight the vital roles of chirality in determining the structures and functions of the self-assembled supramolecular polymeric materials. By rational control of the chiral self-assembly of peptide monomers, supramolecular polymers with sophisticated hierarchical structures and enhanced chemical and biological functions can readily be achieved. These can provide inspiration for researchers to design highly efficient asymmetric catalysts, templates fabricating materials with tunable chiroptics, or to synthesize biomaterials with chirality-encoded properties for biomedical applications.

Electrostatic and Aromatic Interaction-Directed Supramolecular Self-Assembly of a Designed Fmoc-Tripeptide into Helical Nanoribbons

Abstract:Supramolecular self-assembly offers an efficient pathway for creating macroscopically chiral structures in biology and materials science. Here, a new peptide consisting of an N-(9-fluorenylmethoxycarbonyl) headgroup connected to an aromatic phenylalanine-tryptophan dipeptide and terminated with zwitterionic lysine (Fmoc-FWK) and its cationic form (Fmoc-FWK-NH2) were designed for self-assembly into chiral structures. It was found that the Fmoc-FWK peptide self-assembled into left-handed helical nanoribbons at pH 11.2-11.8, whereas it formed nanofibers at pH 5 and 12 and large flat ribbons composed of many nanofibers in the pH range of 6-11. However, only nanofibers were observed in the cases of Fmoc-FWK-NH2 at different values. A series of structural characterizations based on CD, FTIR, UV-vis and fluorescence spectroscopy reveal that the electrostatic and aromatic interactions and the associated hydrogen bonding direct the self-assembly into various structures. The enhanced π-π stacking and hydrogen bonding were found in the helical nanoribbons. This difference in intermolecular interactions should be derived from the ionization of carboxyl and amino groups from lysine residues at different pH values. Furthermore, we performed molecular dynamics simulations to gain insight into the assembly mechanisms. The results imply that a relatively rigid molecular conformation and the strong intramolecular aromatic interaction between Trp and Fmoc groups favor chiral self-assembly. This study is the first attempt to design a Fmoc-tripeptide for the fabrication of helical structures with macroscopic chirality, which provides a successful example and allows us to create new peptide-based chiral assembly systems.

Abstract:Self‐assembly of peptides into highly ordered chiral nanostructures has great potential applications in optics, sensing, biomedicine, and templated synthesis. However, the detailed mechanism for the chiral amplification and spontaneous ordering during the self‐assembly of peptides remains largely unexplored. Here, it is shown that by topology control of the co‐assembly of a simple dipeptide and a chiral diamine within a confined film, the supramolecular structures can be controlled precisely from hierarchically organized helical bundles to microcrystals and then parallel aligned nanohelices. The peptide and diamines can co‐assemble into aligned microcrystals when the molar ratio (χ = Cdiamine/Cpeptide) of the two components is 0.5. However, by changing the χ value, excess peptides or diamines create topological defects during the self‐assembly process. This leads to chiral amplification and the formation of long‐range‐ordered nanohelical arrays with controlled phase behaviors. Moreover, the formation of right‐ or left‐handed nanohelices can be further inversed by the topological control, without the need to change the chirality of the peptides. These findings provide new insights into the chiral self‐assembly of amyloid‐related peptides and offer a facile strategy to fabricate highly ordered functional materials.

Abstract:Beyond the common supramolecular helical polymers in solutions, controlling single-crystal helical self-assembly with precisely defined chirality and architectures has been challenging. Here, we report that simply merging static homochiral amino acids with dynamic chiral disulfides can produce a class of building blocks featuring supramolecular helical single-crystal self-assembly with unusual stereodivergency. Analysis of 20 single-crystal structures of 1,2-dithiolanes gives an atom-precision understanding of the chirality transfer from the molecular to supramolecular level, featuring homochiral and heterochiral helical supramolecular self-assembly in the solid state. The underlying structure-assembly relationship reveals that the synergistic interplay of intermolecular H-bonds and the 1,2-dithiolane ring with adaptive chirality plays a key role in determining the assembly pathway, also involving the effects of residue groups, substituents, molecular stacking, and solvents. The confinement effect in the solid state can stabilize the dynamic stereochemistry of disulfide bonds and selectively result in specific conformers that can minimize the energy of global supramolecular systems. We envision that these results represent a starting point to use dynamic chiral disulfide as a functional entity in supramolecular chemistry and may inspire a new class of supramolecular helical polymers with dynamic functions.

Abstract:The self-assembly of peptides into ordered nanostructures is important for understanding both peptide molecular interactions and nanotechnological applications. However, because of the complexity and various self-assembling pathways of peptide molecules, design of self-assembling helical peptides with high controllability and tunability is challenging. We report a new self-assembling mode that uses in-tether chiral center-induced helical peptides as a platform for tunable peptide self-assembly with good controllability. It was found that self-assembling behavior was governed by in-tether substitutional groups, where chirality determined the formation of helical structures and aromaticity provided the driving force for self-assembly. Both factors were essential for peptide self-assembly to occur. Experiments and theoretical calculations indicate long-range crystal-like packing in the self-assembly, which was stabilized by a synergy of interpeptide π-π and π-sulfur interactions and hydrogen bond networks. In addition, the self-assembled peptide nanomaterials were demonstrated to be promising candidate materials for applications in biocompatible electrochemical supercapacitors.
Reversible Manipulation of Supramolecular Chirality using Host-Guest Dynamics between β-Cyclodextrin and Alkyl Amines.

Abstract:Host-guest interactions are widely employed in constructing responsive materials, although less is known to manipulate the chiral property of materials using such host-guest dynamics. With the supramolecular self-assembly based on beta-cyclodextrin (beta-CD) and alkyl amines (CH3(CH2)(n-1)NH2), we report that faster host-guest dynamics induces a dipole located above the cavity of beta-CD, whereas slower dynamics create in-cavity dipole. These two scenarios correspond to negative and positive chiral signals, respectively. Considering that a larger fraction of amines facilitates faster exchange between the threaded and unthreaded amines, the chiral signal for the right-handed helical ribbons can be manipulated simply by alternatively increasing the fraction of amines and beta-CD. Excitingly, enzyme responsive supramolecular chirality is obtained as a result of shifting the molar ratio by enzyme triggered hydrolysis of beta-CD. We expect that this strategy may open up an area of rationally designed chiral supramolecular materials based on host-guest chemistry.

Abstract:Most natural biomolecules may exist in either of two enantiomeric forms. Although in nature, amino acid biopolymers are characterized by l-type homochirality, incorporation of d-amino acids in the design of self-assembling peptide motifs has been shown to significantly alter enzyme stability, conformation, self-assembly behavior, cytotoxicity, and even therapeutic activity. However, while functional metabolite assemblies are ubiquitous throughout nature and play numerous important roles including physiological, structural, or catalytic functions, the effect of chirality on the self-assembly nature and function of single amino acids is not yet explored. Herein, we investigated the self-assembly mechanism of amyloid-like structure formation by two aromatic amino acids, phenylalanine (Phe) and tryptophan (Trp), both previously found as extremely important for the nucleation and self-assembly of aggregation-prone peptide regions into functional structures. Employing d-enantiomers, we demonstrate the critical role that amino acid chirality plays in their self-assembly process. The kinetics and morphology of pure enantiomers is completely altered upon their coassembly, allowing to fabricate different nanostructures that are mechanically more robust. Using diverse experimental techniques, we reveal the different molecular arrangement and self-assembly mechanism of the dl-racemic mixtures that resulted in the formation of advanced supramolecular materials. This study provides a simple yet sophisticated engineering model for the fabrication of attractive materials with bionanotechnological applications.

Abstract:Self-assembly is one of the most important issues of fabricating materials with precise chiral nanostructures. Herein, we constructed a chiral assembly system from amphiphiles containing hydrophobic/hydrophilic chiral coils bonded to hexabiphenyl, exhibiting controllable enantioselectivity over various aggregation behaviors. The chiral coils aroused various steric hindrances affecting intrinsic stacking tendency and compactness, leading to different aggregating behaviors, as concluded from the self-assembly investigation. The strong π-π stacking interaction between the long hexabiphenyl groups gave rise to a relatively compact arrangement in the aqueous solution, whereas the methyl side groups on the coil segments raised steric hindrance at the rigid-flexible interface, resulting in loose stacking and formation of nanostructures with a larger curvature. Compared with the achiral molecule 1 that formed micron-sized large sheets, molecules 2-4 containing chiral coils aggregated into nanodishes, which looked exactly like mosquito-repellent incense, to overcome surface tension. The helical structures effectively amplified chirality and exhibited strong circular dichroism (CD) signals, which indicate enantioselectivity. In addition, the relatively loose packing behavior permitted their co-assembly with a dye and aided efficient energy transfer, providing a foundation for the chiral application of supramolecules. Thus, by introducing a simple methyl side group in amphiphilic molecules, asymmetric synthesis and energy transfer efficiency can be realized.

Abstract:Changing the chirality of the terminal charged residues of an amphiphilic hexapeptide AcI4K2‐NH2 gives rise to distinct nanostructures and supramolecular handedness. The chiral alterations of the two C‐terminal lysine residues cause the formation of diverse single β‐strand conformations, and the final self‐assembled nanostructures and handedness are determined by the twisting direction and degree of single β‐strands. Peptide assemblies are promising nanomaterials, with their properties and technological applications being highly hinged on their supramolecular architectures. Here, how changing the chirality of the terminal charged residues of an amphiphilic hexapeptide sequence Ac‐I4K2‐NH2 gives rise to distinct nanostructures and supramolecular handedness is reported. Microscopic imaging and neutron scattering measurements show thin nanofibrils, thick nanofibrils, and wide nanotubes self‐assembled from four stereoisomers. Spectroscopic and solid‐state nuclear magnetic resonance (NMR) analyses reveal that these isomeric peptides adopt similar anti‐parallel β‐sheet secondary structures. Further theoretical calculations demonstrate that the chiral alterations of the two C‐terminal lysine residues cause the formation of diverse single β‐strand conformations, and the final self‐assembled nanostructures and handedness are determined by the twisting direction and degree of single β‐strands. This work not only lays a useful foundation for the fabrication of diverse peptide nanostructures by manipulating the chirality of specific residues but also provides a framework for predicting the supramolecular structures and handedness of peptide assemblies from single molecule conformations.

Abstract:The tunability nature of supramolecular chirality is subtly employed where various small amounts of achiral diaminopyridines are used to copolymerize with m‐phenylenediamine based on a copolymerization‐mediated strategy to modulate the macroscopic helicity as well as the chiroptical activity of the resulting robust covalent polymers in an enantiopure template molecular system. Designing artificial systems to mimic chiral superstructures and exploring how achiral molecules precisely modulate their chirality is crucial to understand the origin of homochirality in nature. Herein, the tunability nature of supramolecular chirality is subtly applied to fabricate robust chiral covalent polymers with controlled macroscopic helicity. When using conventional supramolecular transcription methods to prepare chiral poly(m‐phenylenediamine), simply introducing various small amounts of achiral diaminopyridines (DAPs) to copolymerize with m‐phenylenediamine (MPD) enables the resulting chiral copolymers with adjustable helical configurations and chiroptical activities in an enantiopure template system. The strong hydrogen bonds between DAPs and the template molecules ((S)/(R)‐PhgC16) proved to be the driving force for guiding the chiral polymerization mode of MPD. Impressively, the exquisite self‐sorting phenomenon is observed by tuning the stoichiometric ratio of DAP and (R)‐PhgC16, where the as‐prepared copolymers with opposite helicity can exist simultaneously to form a double network structure and their chiroptical activity can be well regulated. In addition, based on such a copolymerization strategy, a conceptual library of helical functional copolymers is built with editable polymer types including conducting polymers and designable amine‐aldehyde condensation‐based polymers. This copolymerization‐mediated synthetic strategy may facilitate the development of chiral polymers with controlled morphologies and dynamic compositions.

Abstract:The regulation of natural helical nanostructures is principally supported and actuated by hydrogen bonds (H-bonds) formed from hydrogen-bonding groups (peptide bonds and base pairs) to realize biological activities and specific biofunctional transformations. However, studying the role of H-bonding patterns on the handedness of supramolecular assemblies is still challenging, since supramolecular assemblies will be disassembled or destabilized with slightly varying H-bonding groups for most supramolecules. To circumvent this issue, herein, two types of self-assembled C2-symmetric phenylalanine derivatives differed by a single H-bonding group (ester or amide) are systematically designed for deciphering the role of H-bonding pattern on the chirality of supramolecular assemblies and their related biostability. Opposite handedness nanofibrous structures with tailorable diameter and helical pitch are achieved with the transition from ester to amide groups in the gelators. Experimental and theoretical evidence suggests that helical orientation of ester-containing gelators ascribes to intermolecular H-bonds. In contrast, the helical direction for the amide-counterparts is mainly due to intra- and intermolecular H-bonds. Moreover, these H-bonding groups greatly influence their stability, as revealed by in vitro and in vivo degradation experiments and the left-handed assemblies are more stable than the right-handed ones. Thus, the study offers a feasible model to have valuable insight into understanding the role of H-bonding patterns in biological folding.

Abstract:In the context of sustainable development, research regarding chirality has aroused enormous attention. Concurrently, chiral self-assembly is one of the most important subjects in supramolecular research, which can broaden the applications of chiral materials. This study focuses on the morphology control of amphiphilic rod-coil molecules composed of the rigid hexaphenyl unit and flexible oligoethylene and butoxy groups containing lateral methyl groups, carried out using an enantioseparation application. The methyl side chain being located on different blocks influences the driving force through steric hindrance, which determines the direction and degree of tilted packing during the π-π stacking of the self-assembly process. Interestingly, the amphiphilic rod-coil molecules aggregated into long helical nano-fibers, which further hierarchically aggregated into nano-sheets or nano-tubes upon increasing the concentration of the THF/H<sub>2</sub>O solution. In particular, the hierarchical-chiral assembly effectively amplified the chirality and was validated by the strong Cotton signals; playing a vital role in the enantioselective nucleophilic substitution reaction. These results provide new insights into the applications of chiral self-assemblies and soft chiral materials.

Abstract:Electron donor-acceptor (EDA) complexes of triphenylene (TP) based side-chain discotic liquid crystalline polymers doped with chiral acceptors exhibit a remarkable supramolecular polymer effect upon reaching a critical doping amount, complying with the discrete columnar stack (DCS) based modular assembly mechanism. Furthermore, selective left-or right-handed homochirality determined by the enantiomeric nature of chiral dopants has been achieved, with a helical polymer backbone surrounded by chiral supramolecular columns through modular assembly between the chiral dopant and DCS subunits, revealing a strong circular dichroism (CD) activity reminiscent of the fascinating hierarchically assembled superstructure of tobacco mosaic virus (TMV). Such polymeric EDA stoichiometric complexation and generation of single-handedness provides inspiring insight into and guidance for constructing various chiral functional materials via non-covalent interactions in a facile way, and also enhances in-depth understanding of the origin of biological helical structures and chiral basis of life.

Abstract:Supramolecular chirality is involved not only in biological events such as gene communication, replication, and enzyme catalysis but also in artificial self-assembly systems and aggregated materials. The precise control of supramolecular chirality, and especially supramolecular chirality inversion (SMCI), would deepen the understanding of chiral transfer and regulation in both living systems and artificial self-assembly systems, providing efficient ways to construct advanced chiral materials with an optimum assembly pathway necessary for various functions. In this review, the fundamental principles of SMCI are comprehensively summarized, with a focus on the helical assemblies having opposite handedness or chiroptical properties of the compositions. Thereafter, various SMCI strategies that have been developed for chiral nanostructures and assembled materials are systematically reviewed, and the promising applications of SMCI, including chiroptical switches, chiral recognition, enantiomeric separation, asymmetric catalysis, chiral optoelectronic materials, chiral spin filters, and biomedical uses, are highlighted accordingly. Finally, the scientific challenges and future perspectives for assembling materials with SMCI are also discussed.

Enhanced Chirality Transfer in Self‐Assembled Nanocomposites Powered by A Trace Amount of Chiral Dimeric Molecules

Abstract:Chiral-selective self-assembly has markedly advanced the development of chiral materials. While the Sergeant and Soldiers principle allows for chirality amplification, it necessitates precise shape-matching between chiral and achiral molecules, leading to a low chirality transfer efficiency-where one chiral molecule influences the chirality of a limited number of achiral molecules. Here, we show that this efficiency can be markedly enhanced by introducing chiral dimeric molecules. In this work, a single chiral molecule can control the chirality of up to 200 achiral molecules and even direct the assembly of inorganic nanoparticles into chiral nanocomposites through a sequential chirality transfer process. Moreover, this approach exhibits remarkable robustness, operating effectively without necessitating a precise match between chiral and achiral molecules. Consequently, using the same chiral molecules at an exceptionally low molar fraction (0.5 mol %) allows for the chiral-selective assembly of achiral molecules over a broad spectrum, regardless of their packing habits, thus facilitating the creation of otherwise inaccessible chiral materials with modulated chiroptical properties. Last but not least, even a trace amount of chiral molecules can enhance the elastic modulus of the self-assembled nanocomposites by a factor of eight.

Solvent-Modulated Chiral Self-Assembly: Selective Formation of Helical Nanotubes, Nanotwists, and Energy Transfer

Abstract:As the medium for self-assembly processes, solvents strongly influence the supramolecular assemblies via specific solute-solvent interactions, which may result in effective modulation of properties, self-assembled nanostructures, and functions through varying the solvent. Here, two kinds of pyridine-cyanostilbene functionalized chiral amphiphiles (l/d-PyPhG and l-PyG) were designed, and their self-assembly behaviors in different solvents were investigated. It was found that both amphiphiles formed gels in dimethyl sulfoxide (DMSO) and self-assembled into right-handed nanotwists, while they formed suspensions in ethanol consisting of left-handed nanotubes. Although the molecular chirality in the compounds remained unchanged in the two solvents, the nanoassemblies showed opposite handedness at the nanoscale together with opposite circular dichroism (CD) and circularly polarized luminescence (CPL) signals. Furthermore, when the amphiphiles were co-assembled with an achiral dye, it was found that efficient energy transfer took place in the systems composed of nanotubes rather than those composed of nanotwists. Therefore, by assembling molecules with the same molecular chirality in different solvents, a selective formation of helical nanotubes or nanotwists and the regulation of handedness as well as energy transfer efficiency were achieved.
