Abstract:Hydrogen sulfide (H2S) can undergo dual nucleophilic reaction, which is a wise and effective way to distinguish biothiols and H2S. A novel H2S fluorescence probe, 4-{2-[4-(2-disulfide pyridyl-benzoyloxy)-phenyl]-vinyl}-1-methyl-pyridinium[e]iodide (DSPBP), with two nucleophilic reaction sites has been developed. The spectra results showed that DSPBP could detect H2S in ratiometric and colorimetric signals and has excellent selectivity and sensitivity. The fluorescence ratiometric signals (F-520/F-450) displayed a prominent increase from 0.74 to 7.08, the fluorescence color turned to yellow form blue simultaneously. The linear range was 2-14 mu M and its detection limit was 25.7 nM. Moreover, the biocompatibility of DSPBP was fine and its toxicity was very low. It has been successfully used for imaging H2S in cells. (C) 2018 Elsevier B.V. All rights reserved.

Abstract:Hydrogen sulfide has emerged as an important biological messenger and much attention has been paid to the design of fluorescent probes for H2S to meet the requirement of accurate measurement of H2S. In this work, a new red emission fluorescent probe for H2S was developed based on the reduction reaction of azide with H2S to amine with the fluorophore of dicyanomethylenedihydrofuran because of its long excitation and emission wavelength. The probe has a high selectivity for H2S over competitive anions and sulfide-containing analytes. Finally, the probe was applied to sense H2S in living cells.

Abstract:We present a new two-photon fluorescent probe (T-HS) for the detection of H2S. With the addition of hydrogen sulfide, the absorption and fluorescence spectra of the probe show regular changes. The probe exhibited favorable properties, such as large turn-on fluorescence signal, good selectivity and low cytotoxicity. Moreover, the probe T-HS was successfully used for the fluorescence imaging of H2S in live cells and zebrafish.

Abstract:Fluorescent probes for hydrogen sulfide have received considerable attention because of the biological significance of H2S recognized recently. Two-photo microscopy offers advantages of increased penetration depth, localized excitation, and prolonged observation time. However, two-photon fluorescent probes for H2S are still rare. In this work, we introduced a dinitrophenyl ether group into the 4-position of 1,8-naphthalimide, which acts as the H2S reactive site, to efficiently yield compound NI–NHS as a two-photo fluorescent probe for H2S. The probe NI–NHS has a high selectivity for H2S over competitive anions and sulfide-containing analytes. This probe exhibits turn-on fluorescence detection of H2S in bovine serum and two-photon fluorescent imaging of H2S in living cells.

Abstract:A novel probe based on the fluorescence off-on strategy was prepared to optically detect hydrogen sulfide (H2S) via an excited state intramolecular proton transfer (ESIPT) mechanism. The probe shows high sensitivity and excellent selectivity to H2S. It also displays a large Stokes shift (similar to 140 nm) and a remarkable quantum yield enhancement (Phi = 0.412) after interaction with H2S. Moreover, the cellular imaging experiment demonstrated that it has potential utility for H2S sensing in biological sciences. (C) 2015 Elsevier B.V. All rights reserved.

Abstract:Since Hydrogen Sulfide (H2S) was recognized as a gas transmitter, its detection and quantification have become a hot research topic among chemists and biologists. In this area, fluorescent probes have shown great advantages: fast and strong response, low detection limit and easy manipulation. Here we developed a new fluorescent probe that detected H2S selectively among various bioactive and inorganic salts. This probe was based on the core structure of fluorescein and reacted with H2S through azide-reduction. Great linearity was achieved correlating fluorescence intensity and H2S concentrations in solution. The detection of H2S in cancer cells was also achieved.

Abstract:Taking apart in numerous physiological and pathological activities, hydrogen sulfide (H 2 S) has been selected as an excellent target spot for the early diagnosis of cancer. So far, there are many mature probes that apply single optical imaging to monitor endogenous H 2 S. Nevertheless, a single modality is not an ideal method to afford accurate diagnostic information in comprehensive biological organisms. Herein, we developed a dual-modal imaging probe BWS. This designed probe showed a specific response to H 2 S with both chemiluminescence and NIR fluorescence light-up. The concurrence of fluorescence and chemiluminescence signal provided "double insurances" for highly accurate monitoring of H 2 S. Satisfactorily, this dual-modal imaging probe performed precise visualization of endogenous H 2 S in living cells and in vivo . We envisaged that this chemiluminescence/fluorescence real-time dual-modality strategy for H 2 S detection will expand and optimize the multimodal imaging methods for efficient diagnosis and treatment of cancer.

Abstract:Hydrogen sulfide (H2S), as one of the critical gaseous signaling molecules, has important physiological functions in the human body, and abnormal levels of hydrogen sulfide are closely related to tumors, Parkinson’s disease, Alzheimer’s disease, and other diseases. In order to enable the detection of H2S in the physiological environment, herein, a new H2S fluorescence probe, named C-HS, based on a coumarin–chalcone fluorescence platform was developed. The fluorescence probe provides specific recognition of H2S within a wide pH detection range (5.5–8.5), a rapid recognition response (within 10 min) for H2S molecules, and a high selectivity for competing species. The probe C-HS possesses low cytotoxicity and is used to achieve the detection of exogenous/ endogenous H2S in living cells, indicating that the constructed probe C-HS has the ability to track changes in intracellular H2S levels. Therefore, probe C-HS could be a potential tool for the early diagnosis of H2S-related diseases.

Abstract:In comparison with other biological detection technologies, fluorescence bioimaging technology has become a powerful supporting tool for intracellular detection, and can provide attractive facilities for investigating physiological and pathological processes of interest with high spatial and temporal resolution, less invasiveness, and a rapid response. Due to the versatile roles of hydrogen sulfide (H2S) in cellular signal transduction and intracellular redox status regulation, fluorescent probes for the detection of this third signalling gasotransmitter have rapidly increased in number in recent years. These probes can offer powerful means to investigate the physiological actions of H2S in its native environments without disturbing its endogenous distribution. In this feature article, we address the synthesis and design strategies for the development of fluorescent probes for H2S based on the reaction type between H2S and the probes. Moreover, we also highlight fluorescent probes for other reactive sulfur species, such as sulfane sulfurs and SO2 derivatives.

Abstract:Reactive sulfur species (RSS), primarily containing glutathione (GSH), cysteine (Cys), hydrogen sulfide (H2S) and hydrogen polysulfides (H2Sn, n > 1), were crucial endogenous biological signal molecules. Among them, H2S and H2Sn played extremely momentous roles in many physiological and pathological processes. Limited by the vivo environmental diversity and complexity, it was highly desirable to rely on the single fluorescence probe platform to visualize H2Sn and H2S according different fluorescence signals. Herein, we reported a two-photon excited fluorescent probe MC-S-n that had the following merits: (I) ratiometric and colorimetric: reducing detection errors and conducting visual analysis; (II) two-photon microscope: better performance in vivo and vitro imaging; (III) two specific sulfide reaction sites: studying on the mutual transformation of H2Sn and H2S in biological systems. Furthermore, take advantages of low cytotoxicity (cell viability >= 85.99%), low limit of detection (21 nM), significant photostability, excellent pH stability, quick response behavior (175 s) and high penetration depth (110 mu m), MC-S-n can be employed to visualize endogenous and exogenous H2Sn and H2S in cells, liver tissues and zebrafish.

Abstract:As a signal transduction molecule in organism, H2S plays an important role in many physiological processes. In this paper, a novel H2S fluorescent probe SBD was designed based on 1,8-naphthalimide and dinitrobenzenesulfonyl group. In DMSO-PBS solution, probe SBD could recognize H2S and change the color of solution from yellow to pink, realizing the naked eye recognition for H2S. In addition, probe SBD displayed high fluorescence enhancement rate (45-fold), good selectivity, strong anti-interference ability and the detection limit was as low as 152 nM. Most importantly, probe SBD showed very low cytotoxicity and had been successfully applied to image exogenous H2S in living cells.
