Abstract:α-Hydroxy amides are a class of important intermediates in organic synthesis. It plays an important role in the field of science and medicine. However, the preparation methods of α-hydroxy amides are relatively few, and they are usually enzyme-or Lewis acid-catalyzed reactions. These types of methods are not ideal in operation, yield and/or stereoselectivity. We found a new way to synthesize α-hydroxy amides by transforming amino acids into α-hydroxyl acids, and then by amination of the α-hydroxy acids.

Abstract:Multicomponent reactions are excellent tools for rapid generation of small molecules with broad chemical diversity and molecular complexity. Herein, a novel one-pot multicomponent synthesis of beta-amino amides from aldehydes, anilines, carboxylic acids and ynamides has been successfully developed. This process is practical and efficient to unravel synthetic utility and scalability. Moreover, an isotope labeling reaction was conducted to elucidate a plausible reaction mechanism.

Abstract:A multicomponent reaction of N-indole carboxylic acids, aldehydes, amines, and C2 building blocks can be transformed to structurally diverse β-indole carboxamide amino amides. In this multicomponent reaction, the ynamides and triazenyl alkynes act as the C2 building block, and this protocol features readily available starting materials, high atom economy, and mild reaction conditions. Besides, the acyl triazene group in the product can be easily transformed to differential groups to expand the structural diversity.

Abstract:A novel annulation reaction of amidines with sulfur ylides and nitrobenzenes has been developed, affording various novel 4-hydroxy-5-phenylaminoimidazoles in moderate to excellent yields. The 4-hydroxy-5-phenylaminoimidazoles have been further converted into α-ketoamide and imidazol-4-imine derivatives. Moreover, a plausible mechanism for this multicomponent reaction is proposed.

Abstract:Acrylamidines were synthesized via the copper-catalyzed three-component reaction of propargyl acetates, sulfonyl azides and amines, which are readily accessible materials. The synthesized N,N'- bis(aryl) amidines could be converted into 2-styrylbenzoimidazoles by iodobenzene-catalyzed oxidative C-H amination using m-CPBA as a terminal oxidant.

Abstract:The three-component reaction of N-substituted hydroxylamines, aldehydes, and phenylacetylene catalyzed by CuCl/2,2'-bipyridine in the presence of NaOAc under neat conditions afforded the corresponding beta-lactams in good to excellent yields. Aromatic, heteroaromatic, and aliphatic aldehydes are tolerated in this reaction. The electronic effects of the aldehydes were studied for the reaction with N-methylhydroxylamine and N-benzylhydroxylamine. For N-methylhydroxylamine, electron-rich aldehydes provided higher yields than electron-deficient aldehydes, whereas for N-benzylhydroxylamine, no significant electronic effect was observed for the aldehydes.

Abstract:We present herein a new mode of three-component reactions between isocyanoacetates, amines and 3-formylchromones. Both experimental and DFT studies revealed that this Ag-catalyzed unusual transformation is initiated by a facile aza-Michael addition instead of the conventional imine condensation. This catalytic method enables an efficient synthesis of polysubstituted pyrroles.

Abstract:A multicomponent carbonyl alkylative amination reaction is described. A variety of <i>N</i>-arylamines, aldehydes, and hydrocarbons have been examined as reaction substrates using tetrabutylammonium decatungstate as photocatalyst, providing the corresponding α-branched tertiary and secondary amines in good to moderate yields. Abstract:An asymmetric three‐component reaction of α‐H diazoacetate with sulfonamide and imine has been developed under the co‐catalyzation of Rh2(OAc)4 and a chiral phosphoric acid (CPA), which proceed through Mannich‐type interception of transient ammonium ylide. The reaction offers a convenient access to α, β‐diamine acid derivatives with two adjacent chiral centers in 59%‐74% yields with 91%‐99% ee and 92:8 to > 95:5 dr. Moreover, the generated products could be easily converted to free chiral diamines and other useful chiral motifs.

Abstract:β - Amino acids are structural motifs widely found in therapeutic natural products, novel biomimetic polymers and peptidomimetics. As a convergent method, the synthesis of stereoenriched β-amino amides through the asymmetric Mannich reaction requires specialized amide substrates or a metal catalyst for enolate formation. By a redesign of the Ugi reaction, a conceptually different solution to prepare chiral β-amino amides was established using ambiphilic ynamides as two-carbon synthons. The modulation of ynamides or oxygen nucleophiles concisely furnished three classes of β-amino amides with generally good efficiency as well as excellent chemo- and stereo-control. The utility is verified in the preparation of over 100 desired products that bear one or two contiguous carbon stereocentres, including those that directly incorporate drug molecules. This advance also provides a synthetic shortcut to other valuable structures. The amino amides could be elaborated into β-amino acids, anti -vicinal diamines, γ-amino alcohols and β-lactams or undergo transamidation with amino acids and amine-containing pharmaceuticals.
