Abstract:This paper describes a simple chiral primary amine-catalyzed highly efficient and practical protocol for the synthesis of both Wieland-Miescher ketone and Hajos-Parrish ketone as well as their analogues. The reaction can be conducted in gram scale with 1% mol catalyst loading producing high enantioselectivity (up to 96% ee) and excellent yields (up to 98%). This procedure represents one of the most efficient methods for the synthesis of these versatile chiral building blocks.

Abstract:Due to their importance in natural products, organic synthesis, life science, and pharmaceuticals, reliable methods for the synthesis of chiral amines are urgently required. Here in this contribution, the dual chiral catalytic method has been established for the enantioselective reaction of terminal alkynes, aldehydes, and amines affording optically active propargylic amines with a decent enantioselectivity and a broad substrate scope. These propargylic amines have been demonstrated as platform molecules for the syntheses of different types of chiral amines. By applying this protocol, the asymmetric total syntheses of two linear monoamine natural products, (S)-5 and (S)-11, which were isolated from marine cyanobacteria Microcoleus lyngbyaceus, have been realized. The envisioned chiral ion pair, which has boosted the enantioselectivity greatly, and other key intermediates have been successfully characterized by MS studies.

Abstract:Enantioselective α-functionalizations of carbonyl compounds are fundamental transformations for the asymmetric synthesis of organic compounds. One of the more recent developments along this line is in aminocatalysis, which leads to the direct α-functionalization of simple aldehydes and ketones. However, most of the advances have been achieved with linear aldehydes and ketones as substrates. Effective aminocatalysis with α-branched carbonyls, particularly α-branched ketones, has remained elusive. The primary difficulty arises from the space-demanding α-substituent, which impedes iminium/enamine formation. In 2005, synthetic organic chemists revived catalysis using primary amines, which brought new attention to these challenges, because of the conformational flexibility of primary amines. On the basis of early biomimetic studies by Hine, in 2007 we developed the bioinspired chiral primary amine catalysts featuring primary-tertiary diamines. This type of catalyst involves enamine/iminium catalysis, and we could apply this chemistry to all of the major types of ketones and aldehydes. In this Account, we present research from our laboratory that significantly expands aminocatalysis to include α-branched ketones such as β-ketocarbonyls and α-substituted vinyl ketones. Our primary amine catalysis methodology, when used alone or in conjunction with metal catalysts, provides convenient access to both enantiopure α-tertiary and quaternary ketones, structures that are not available via other approaches. Our mechanistic studies showed that acidic additives play the critical role in facilitating catalytic turnover, most likely by shuttling protons during the enamine/iminium tautomerizations. These additives are also critical to induce the desired stereochemistry via ammonium N-H hydrogen bonding. Proton transfer by shuttling is also stereoselective, resulting in enantioselective enamine protonation as observed in the reactions of α-substituted vinyl ketones. In addition, we have carried out density functional theory studies that help to delineate the origins of the stereoselectivity in these reactions.

Abstract:Herein, a visible-light-induced chiral N-heterocyclic carbene (NHC) catalyzed asymmetric amination of ketenes has been developed. This strategy provides a convenient and facile synthetic protocol for the efficient construction of α-chiral amides under mild reaction conditions with moderate to excellent yields and stereoselectivities (up to 95% yields and 97:3 e.r.). Furthermore, chiral 2-phenyl-1-porpylamine derivative and (R)-ibuprofen could be obtained after simple transformations of chiral products.

A Concise Asymmetric Route to Chiral Α-Aminoxy Acids

Abstract:An efficient three-step method has been developed for enantioselective synthesis of chiral alpha-aminoxy acids from aldehydes. In this method, chiral propargylic alcohols are obtained by asymmetric addition of terminal alkynes to aldehydes. The hydroxyl group then converted to aminoxy group via Mitsunobu reaction and oxidative cleavage of the internal carbon-carbon triple bond produce the carboxylic acid group. This represents a convenient approach to the general asymmetric synthesis of alpha-aminoxy acids with the advantages of substantial overall yields (37-73%) and high enantioselectivities (81-99% ee).

Abstract:Carbonyls and amines are yin and yang in organocatalysis as they mutually activate and transform each other. These intrinsically reacting partners tend to condense with each other, thus depleting their individual activity when used together as cocatalysts. Though widely established in many prominent catalytic strategies, aminocatalysis and carbonyl catalysis do not coexist well, and, as such, a cooperative amine/carbonyl dual catalysis remains essentially unknown. Here we report a cooperative primary amine and ketone dual catalytic approach for the asymmetric α-hydroxylation of β-ketocarbonyls with H2O2. Besides participating in the typical enamine catalytic cycle, the chiral primary amine catalyst was found to work cooperatively with a ketone catalyst to activate H2O2via an oxaziridine intermediate derived from an in-situ-generated ketimine. Ultimately, this enamine–oxaziridine coupling facilitated the highly controlled α-hydroxylation of several β-ketocarbonyls in excellent yield and enantioselectivity. Notably, late-stage hydroxylation for peptidyl amide or chiral esters can also be achieved with high stereoselectivity.
