Abstract:We have developed an organocatalyzed asymmetric three‐component reaction of thiazol‐4‐ones, acrolein and nitroolefins, which provides an efficient approach to access optically active spiro thiazol‐4‐ones. Under the catalysis of a bifunctional squaramide derived from L‐tert‐leucine, the reactions of a wide range of thiazol‐4‐ones, acrolein and nitroolefins took place smoothly to generate the corresponding spirothiazolone derivatives bearing four contiguous stereogenic centers in good yields with high levels of enantioselectivity. The title chiral spirothiazolones were obtained as a pair of separable epimers, which are formed through a double Michael addition followed by a Henry process.

Abstract:A new strategy for the construction of optically active 5 '-CF3 spiro[pyrrolidin-3,2 '-oxindole] was described. A series of unprecedented 1,3-dipoles were obtained by condensation of CF3CH2NH2 with isatins. The 1,3-dipolar cycloaddition reactions of these ketimines with enals gave the products bearing four contiguous stereogenic centers in excellent yields, diastereoselectivities and enantioselectivities.

Abstract:The first organocatalytic asymmetric synthesis of a spirooxindole skeleton incorporated with a cyclobutane moiety has been successfully developed on the basis of H-bond-directing dienamine activation. Structurally complex spirocyclobutyl oxindoles, which possess four contiguous stereocenters, including one spiro quaternary center, were obtained in good yields (up to 83%) with excellent β,γ-regioselectivity (>19:1) and stereocontrol (up to >19:1 dr and 97% ee).

Abstract:Asymmetric cascade Michael/cyclization reaction between 2-substituted thiazol-4-ones and 2-(2-oxoindolin-3-ylidene) malononitriles was investigated using a series of chiral bifunctional hydrogen-bonding organocatalysts. Good yields (up to 91%) and excellent enantioselectivities (up to 98% ee) were achieved by using a (1R, 2R)-1,2-diphenylethane-1,2-diamine derived thiourea catalyst. This method provides an elegant synthetic route to access novel thiazole-fused spirooxindoles with potential bioactivity.

Abstract:A catalytic asymmetric conjugate addition/Schmidt-type rearrangement of vinyl azides and (E)-alkenyloxindoles was realized. It afforded a variety of optically active 3,2′-pyrrolinyl spirooxindoles with high yields (up to 98%), and excellent diastereo- and enantioselectivities (up to 98% ee, >19 : 1 dr), even at the gram-scale in the presence of a chiral N,N′-dioxide–nickel(ii) complex. In addition, a possible catalytic cycle and transition state model were proposed to rationalize the stereoselectivity.
