Studies on the Activities of Hainan Dry Noni Fruit Hydro-Alcoholic Extracts and Their Novel Compounds Identification Using Uplc-Esi-Ms/Ms Spectrometry

Abstract:Morinda citrifolia Linn (noni) is an important common plant distributed widely in the Pacific Asian region. Its fruits and leaves are food sources and used as a treatment in folk medicine. The objective of the study is to investigate the activities of the dry noni fruit hydro-alcoholic extracts and identify the novel compounds in these hydro-alcoholic extracts using UPLC-ESI-MS/MS spectrometry. The extraction of both NFE (Noni Fruit Ethanol) and NFM (Noni Fruit Methanol) were obtained with recovery rates of 72.2 % and 99.4 %, respectively. The antimicrobial effect of the crude extracts was subjected to disc diffusion test screening against SA Staphylococcus aureus (SA) and, Candida albicans (CA) , with MIC of NFE 35.34 mg/mL, 47.80 mg/mL and NFM 117.40 mg/mL, 108.01 mg/mL respectively. Further on, cell viability assay showed that IC 50 values of extract NFE and NFM on human UMUC-3 bladder carcinogenic cells were 865.1 µg/mL and 789.1 µg/mL respectively for 72 h incubation, with less effect on SVHUC-1 Human uroepithelium normal cell line. A UPLC-ESI-MS/MS analysis of both extracts identified six novel compounds, Cyanidin 3-(2G-xylosylrutinoside), Inulobiose, Clausarinol, Pectachol, 4,7-Megastigmadien-9-ol, and D-1-[(3-Carboxypropyl) amino]-1-deoxyfructose. It is speculated that they may have potential antimicrobial and/or anti-cancer activities, which need further investigation in the future. These findings form a basis on screening of folk medicinal plant extracts with contributing properties to be beneficial as a food and health source.
