Abstract:Morinda citrifolia L., commonly known as Noni, has a longstanding history in traditional medicine for treating various diseases. Recently, there has been an increased focus on exploring Noni extracts and phytoconstituents, particularly for their effectiveness against cancers such as lung, esophageal, liver, and breast cancer, and their potential in cancer chemoprevention. This study aims to provide a comprehensive review of in vitro and in vivo studies assessing Noni's impact on cancer, alongside an exploration of its bioactive compounds. A systematic review was conducted, encompassing a wide range of scientific databases to gather pertinent literature. This review focused on in vitro and in vivo studies, as well as clinical trials that explore the effects of Noni fruit and its phytoconstituents—including anthraquinones, flavonoids, sugar derivatives, and neolignans—on cancer. The search was meticulously structured around specific keywords and criteria to ensure a thorough analysis. The compiled studies highlight Noni's multifaceted role in cancer therapy, showcasing its various bioactive components and their modes of action. This includes mechanisms such as apoptosis induction, cell cycle arrest, antiangiogenesis, and immune system modulation, demonstrating significant anticancer and chemopreventive potential. The findings reinforce Noni's potential as a safe and effective option in cancer prevention and treatment. This review underscores the need for further research into Noni's anticancer properties, with the hope of stimulating additional studies and clinical trials to validate and expand upon these promising findings.

[Antitumor effect research progress of shikonin and its derivatives]

Abstract:Shikonin, the main active ingredient of Lithospermum, and its derivatives have been proved to have antitumor effects, and the anti-tumor mechanisms involve multiple targets. Based on recent literatures, this review focuses on the antitumor effects and its mechanisms. More emphases are given on the aspects of induction of apoptosis, induction of necrosis, acting on matrix metalloproteinase, acting on the protein tyrosine kinase and antiangiogenesis. The current status and problems of shikonin derivatives in antitumor effects are simply summarized and lookout for the development of antitumor drugs with shikonin as leading compounds.

Abstract:Single-polarization single-mode (SPSM) fiber can efficiently eliminate polarization mode coupling, polarization mode dispersion, and polarization-dependent loss. Up to now, most single-polarization fibers have been designed based on form birefringence, which would result in a non-Gaussian field distribution and a small effective mode field area. In this paper, a novel structure of SPSM photonic crystal fibers based on the resonant coupling phenomena is proposed and analyzed by using a full-vector finite-element method with a second-order transparent boundary condition. From the numerical results it is confirmed that this fiber has a near-Gaussian mode field within the wavelength range from 1.46 to 2.2 μm, where only one polarized mode exists effectively, and the mode field area is about 79 μm(2) at the wavelength of 1.55 μm, matching that of the conventional single-mode fiber.

Abstract:Extraction and characterization of antioxidative compositions from the extracts of fermented Xisha Noni (Morinda citrifolia L.) juice were studied. The antioxidative constituents of 184.6 g freeze-dried extracts of naturally fermented Xisha Noni juice were isolated successfully by petroleum ether, EtOAc and n-BuOH solvents, and the antioxidative effects were measured according to scavenging activity against hydroxyl generated in Fenton reaction system and superoxide anion radicals in pyrogallol autoxidation system. The EtOAc extract exhibited most significantly higher (P < 0.01) antioxidative activity than mannitol or vitamin C, while the petroleum ether and n-BuOH extracts showed lower activities compared to mannitol. Three antioxidant phenolic compounds, isoscopoletin, aesculetin and 3, 3′, 4′, 5, 7-pentahydroxyflavone (quercetin) were isolated from the EtOAc extract by several chromatography techniques for the first time. The results suggest that several compounds, in particular, the phenolic compounds, contribute separately or synergistically to the antioxidative activity of fermented Noni fruit juice.

Abstract:Noni (Morinda citrifolia L.) is a tropical crop with strong antibacterial, antioxidant and other abilities, and its fruit has a strong potential for adjuvant treatment of diseases. This study aimed to explore the dynamic change of endophytic bacteria in Noni fruit at different stages and the correlation between the antagonistic and antioxidant activity of the Bacillus which was screened and the change of the host's growth stage. In this study, though the high-throughput sequencing technology (HTS), 106 endophytic bacteria species were found in A, B, C, D, E and F stages of Noni fruit, among which the dominant group were Pantoea (0.3%–20.9%), and Candidatus_Uzinura (2.3%–35.2%) etc. The endophytic bacteria were isolated by culture-dependent method. Through their antagonistic experiments on Staphylococcus aureus and Escherichia coli, the results of 16S polyphasic taxonomic identification showed that the 34 antagonistic strains belonged to Bacillus. Five species of these Bacillus were identified by gyrA polyphase taxonomy, including Bacillus subtilis (76% of all Bacillus), Bacillus licheniformis (9%), Bacillus amyloliquefaciens (6%), Bacillus velezensis (6%) and Bacillus mojavensi (3%), and the RAPD showed these Bacillus are no signs of stable passage. In C, D, E and F stages, the average total antioxidant activity of Bacillus endophytic antagonists against Noni was 7.812 U/mL, 8.144 U/mL, 7.817 U/mL and 7.144 U/mL, which was much higher than that of Noni fruit, and antioxidant activity of Noni juice and Bacillus bacterial liquid vary with host's growth period showed the same trend, both rose slowly at first, and reached the highest in period E, then declined slightly in period F, it showed that the antagonistic Bacillus of Noni had synergistic function with Noni fruit. This study clarified the relationship of function between Noni fruit and endophytic bacteria, and laid a foundation for future study on the dynamic change of endophytic flora succession and efficacy.

Abstract:Back ground: Indian Noni (Morinda citrifolia L.) is a large shrub or small evergreen tree, also known as Indian mulberry. It is a medicinal plant that has been used as conventional medicine and food additives for a long time. As a drug, plant bioactivity is attributed to its secondary metabolites. M.citrifolia has very rich nutrients and consists of over 100 varieties of chemical compounds which have been isolated so far from the plant. In disorders such as skin ailments, respiratory infections, gastritis, menstrual, abortion, diabetes, and venereal diseases, it also shows beneficial effects. Materials and Methods: The relation of noni juice or its extract (fruit, leaves or root) to anticancer and/or immunostimulant properties was investigated in this study. A Medline quest was performed along with cross-referencing using the main search terms 'Morinda citrifolia and cancer'. Results: According to our search results for scientific studyM. citrifolia is used in literature for more than 40 forms of ailments. Crude extract is recorded from various plant and fruit juice components. There are records of amino acids, anthraquinones, fatty acids, flavonoids, iridoids, lignans, polysaccharides, sterols, carbohydrates, terpenoids, etc. in the crude extract of different sections of plant and fruit juice that are therapeutically beneficial for a wide range of pathological conditions. Conclusion: Literature indicates that Nonii pharmacologically active and used in various cancer types (Colon, breast, esophageal, colorectal Cancers, cardiovascular disorders, diabetes, hypertension, arthritis). This plant's high potential classifies the compounds as the noteworthy medication of the future, and clinical trials are needed to demonstrate their effect on life-threatening diseases such as cancer. Further analysis warrants the isolation of the active components. These properties can be substantiated by the preclinical or/and clinical investigations.

Abstract:Flavonoids, including flavones, flavonols, anthocyanins, flavanones, chalcones, flavan, proanthocyanidins, isoflavonoids and biflavonoids, etc, are natural components in our diet and plants. Several beneficial properties have been attributed to these compounds, including antioxidant, anti-inflammatory and anticarcinogenic effects, etc. Flavonoid preparations are marketed as herbal medicines or dietary supplements for a variety of alleged nontoxic therapeutic effects. However, they have yet to pass controlled clinical trials for efficacy, and their potential for toxicity is an understudied field of research. This review summarizes the current studies on the toxicity induced by flavonoids and gives some advices for ingesting flavonoids.

Abstract:Nowadays, most antiviral drugs are plant-based due to their low toxicity and high resistance. Morinda citrifolia (Noni) is such a perennial shrub a popular plant based medicine due to its wide therapeutic applications. The phytochemicals present in its fruit as well as its leaf have many antiviral properties and can enhance the human immune system. Herein, we report the identification of bioactive components present in the Morinda citrifolia (Noni) ethanolic leaf extract by GC-MS analysis and their biological interaction with human parallel-stranded 7-mer g-quadruplex DNA of MCF-7 using Biovia Discovery Studio software. In vitro analysis by MTT assay of the same extract reported a living cells of 59.74% at 100 μg/ml. The GC-MS analysis of ethanolic leaf extract revealed the presence of 27 bioactive components, and out of these, six major components were interacted with cancer DNA. The molecular docking interaction identified the most active bio component Phytol, with a binding energy of -27.0796 Kcal/mol. this value is much better than the commercially available doxorubicin. This novel study will provide insight into the development of bioactive components from Morinda citrifolia leaf extract against human breast cancer with minimal side effects.

Abstract:: As a medicinal fungus, Inonotus obliquus (IO) has been widely used in the treatment of cancer and digestive system diseases. Despite the progress that has been made in the studies of IO and its active compounds, their applications in the treatment of other important clinical diseases, such as cardiovascular diseases, which are major global issues with limited treatment strategies, are seldom reported. This review summarizes the separation and purification methods of chemical components of IO, the advances in their applications, and research progress on the pharmacological effects and related mechanisms of IO in disease prevention. This review will help researchers and clinicians to further understand the pharmacological functions and mechanisms of IO and its active components, which may extend their medical applications in the prevention and treatment of other diseases in addition to tumors and digestive system diseases in the near future.

Abstract:Noni juice is a globally popular health beverage originating in the tropics. Traditional Tahitian healers believe the noni plant to be useful for a wide range of maladies, and noni juice consumers throughout the world have similar perceptions. Nevertheless, human clinical trials are necessary for a precise understanding of what the health benefits of noni juice are. A review of published human intervention studies suggests that noni juice may provide protection against tobacco smoke-induced DNA damage, blood lipid and homocysteine elevation as well as systemic inflammation. Human intervention studies also indicate that noni juice may improve joint health, increase physical endurance, increase immune activity, inhibit glycation of proteins, aid weight management, help maintain bone health in women, help maintain normal blood pressure, and improve gum health. Further, these studies point to notable antioxidant activity in noni juice, more so than other fruit juices which served as trial placebos. It is this antioxidant effect and its interaction with the immune system and inflammation pathways that may account for many of the observed health benefits of noni juice. However, the existing evidence does have some limitations as far as its general application to noni juice products; all the peer-reviewed human interventions studies to date have involved only one source of French Polynesian noni juice. Geographical factors and variations in processing methods are known to produce commercial noni juice products with divergent phytochemical and nutrient compositions. Therefore, other sources of noni products may have different toxicological and pharmacological profiles.

Abstract:The dynamic changes in phenolic composition and antioxidant activity, and the potential effect on foam cell formation and cholesterol efflux during noni (Morinda citrifolia Linn.) fruit juice fermentation were investigated in this study. The composition of phenolic compounds was significantly different at various fermentation times. Rutin, quercetin, and isoquercitrin were the major phenolics in fermented noni fruit juice based on a quantitative analysis of representative phenolics. The contents of caffeic acid, 2,4-dihydroxybenzoic acid, p-coumaric acid, rutin, and quercetin tended to increase, while those of isoquercitrin decreased during the fermentation process. Fermented noni juice extracts showed high antioxidant activities against 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid), hydroxyl radical scavenging activity, and ferric reducing antioxidant power. Notably, the highest antioxidant activity was observed after 28 days of fermentation. Furthermore, the treatment of fermented noni juice extracts was shown to reduce foam cell formation, intracellular cholesterol level, and the cholesterol esterification ratio. A correlation analysis indicated a strong positive relationship between the phenolic composition, antioxidant activity, and the ratio of cholesterol ester and total cholesterol. This study may provide a theoretical basis for the quality improvement and standardized production of fermented noni fruit juice, thus promoting the development of the noni food industry.

Abstract:Flavonoids derived from citrus plants are favored by phytomedicinal researchers due to their wide range of biological activities, and relevant studies have been sustained for 67 years (since the first paper published in 1955). In terms of a scientometric and critical review, the scientometrics of related papers, chemical structures, and pharmacological action of citrus flavonoids were comprehensively summarized. The modern pharmacological effects of citrus flavonoids are primarily focused on their anticancer activities (such as breast cancer, gastric cancer, lung cancer, and liver cancer), neuroprotective effects (such as anti-Alzheimer's disease, Parkinson's disease), and metabolic diseases. Furthermore, the therapeutic mechanism of cancers (including inducing apoptosis, inhibiting cell proliferation, and inhibiting cancer metastasis), neuroprotective effects (including antioxidant and anti-inflammatory), and metabolic diseases (such as non-alcoholic fatty liver disease, type 2 diabetes mellitus) were summarized and discussed. We anticipate that this review could provide an essential reference for anti-cancer and neuroprotective research of citrus flavonoids and provide researchers with a comprehensive understanding of citrus flavonoids.

Abstract:Background: Hyperuricemia is generally defined as the high level of serum uric acid and is well known as an important risk factor for the development of various medical disorders. However, the medicinal treatment of hyperuricemia is frequently associated with multiple side-effects. Methods: The therapeutic effect of noni (Morinda citrifolia L.) fruit juice on hyperuricemia and the underlying molecular mechanisms were investigated in mouse model of hyperuricemia induced by potassium oxonate using biochemical and high-throughput RNA sequencing analyses. Results: The levels of serum uric acid (UA) and xanthine oxidase (XOD) in mice treated with noni fruit juice were significantly decreased, suggesting that the noni fruit juice could alleviate hyperuricemia by inhibiting the XOD activity and reducing the level of serum UA. The contents of both serum creatinine and blood urine nitrogen of the noni fruit juice group were significantly lower than those of the model group, suggesting that noni fruit juice promoted the excretion of UA without causing deleterious effect on the renal functions in mice. The differentially expressed microRNAs involved in the pathogenesis of hyperuricemia in mice were identified by RNA sequencing with their target genes further annotated based on both Gene Ontology and Kyoto Encyclopedia of Genes and Genomes databases to explore the metabolic pathways and molecular mechanisms underlying the therapeutic effect on hyperuricemia by noni fruit juice. Conclusion: Our study provided strong experimental evidence to support the further investigations of the potential application of noni fruit juice in the treatment of hyperuricemia.
