Abstract:Hydrogel materials are of great value in medical field due to their similarity to human soft tissue.With silk fibroin as raw materials,organic solvent induction method can be employed to shorten the gelling time of silk fibroin to several hours.Silk fibroin/collagen composite hydrogel prepared with carbodiimide (EDC) as activation cross-linking agent has a storage modulus of up to 10 kPa.In vivo experiments indicate that silk fibroin hydrogel created by enzymatic crosslinking has good biocompatibility and biodegradability.We first explain the gelling mechanism of silk fibroin hydrogels,and then systematically introduce the research progress on preparation and modification methods of various silk fibroin hydrogels.Besides,potential applications of silk fibroin hydrogels as novel biomaterial in clinical applications are analyzed.Basic theoretical and technological issues to be solved for clinical applications of silk fibroin hydrogels are also discussed.

As a versatile natural polymer, silk fibroin has been combined with various nanoparticles. The introduction of nanoparticles brings many optimized and new functionalities to silk fibroin, such as mechanical, biological, thermal, electrical, florescent and magnetical properties. This review focuses on the preparation methods and enhanced properties of silk fibroin-nanoparticle composites, especially their biomedical applications.

Abstract:Silk fibroin (SF) is a structural protein derived from natural silkworm silks. Materials fabricated based on SF usually inherit extraordinary physical and biological properties, including high mechanical strength, toughness, optical transparency, tailorable biodegradability, and biocompatibility. Therefore, SF has attracted interest in the development of sustainable biodevices, especially for emergent bio-electronic technologies. To expand the function of current silk devices, the SF characteristic sequence has been used to synthesize recombinant silk proteins that benefit from SF and other functional peptides, such as stimuli-responsive elastin peptides. In addition to genetic engineering methods, innovated chemistry modification approaches and improved material processing techniques have also been developed for fabricating advanced silk materials with tailored chemical features and nanostructures. Herein, this review summarizes various methods to synthesize functional silk-based materials from different perspectives. This review also highlights the recent advances in the applications of natural and recombinant silks in tissue regeneration, soft robotics, and biosensors, using B. mori SF and silk-elastin-like proteins (SELPs) as examples.

Abstract:Benefiting from the remarkable biological and physicochemical properties, silk fibroin hydrogels occupy a vital position in the biomedical field. We review the recent developments of silk fibroin‐derived hydrogels, outline their unique structures and important properties, and highlight their applications in various biomedical fields, including tissue engineering, drug delivery, and wearable sensors. Silk fibroin hydrogels occupy an essential position in the biomedical field due to their remarkable biological properties, excellent mechanical properties, flexible processing properties, as well as abundant sources and low cost. Herein, we introduce the unique structures and physicochemical characteristics of silk fibroin, including mechanical properties, biocompatibility, and biodegradability. Then, various preparation strategies of silk fibroin hydrogels are summarized, which can be divided into physical cross‐linking and chemical cross‐linking. Emphatically, the applications of silk fibroin hydrogel biomaterials in various biomedical fields, including tissue engineering, drug delivery, and wearable sensors, are systematically summarized. At last, the challenges and future prospects of silk fibroin hydrogels in biomedical applications are discussed.

Abstract:We present a silk biomaterial platform with highly tunable mechanical and degradation properties for engineering and regeneration of soft tissues such as, skin, adipose, and neural tissue, with elasticity properties in the kilopascal range. Lyophilized silk sponges were prepared under different process conditions and the effect of silk molecular weight, concentration and crystallinity on 3D scaffold formation, structural integrity, morphology, mechanical and degradation properties, and cell interactions in vitro and in vivo were studied. Tuning the molecular weight distribution (via degumming time) of silk allowed the formation of stable, highly porous, 3D scaffolds that held form with silk concentrations as low as 0.5% wt/v. Mechanical properties were a function of silk concentration and scaffold degradation was driven by beta-sheet content. Lyophilized silk sponges supported the adhesion of mesenchymal stem cells throughout 3D scaffolds, cell proliferation in vitro, and cell infiltration and scaffold remodeling when implanted subcutaneously in vivo.

Abstract:Silk fibroin is a natural polymer that has various material forms and wide applications. Hydrogel is one of the most attractive silk materials because of its hydrophilicity, biocompatibility, and flexibility. However, its applications are still quite limited because they have a complicated preparation process and/or low mechanical strength. Herein, a simple way to prepare tough silk fibroin hydrogels via a solvent-exchange method is introduced. The degummed silk fiber was directly dissolved in a calcium chloride/formic acid solution and then water was used to replace the solvent. The silk fibroin hydrogel that was obtained using this facile method exhibited even better mechanical properties than most silk fibroin hydrogels that have been reported in the literature. Also, the silk fibroin hydrogel maintained biocompatibility that was as good as that prepared via other methods. Finally, the possibility of using this regenerated silk fibroin hydrogel as a multi-functional platform (such as a catalyst carrier, photothermal agent, and underwater adhesive) has been discussed. Therefore, such a natural, sustainable, robust, and good biocompatible silk fibroin hydrogel that is prepared by an improved method may have great potential for further applications.

Abstract:Silk protein is a promising natural material applied in various fields, but the application of silk protein-based hydrogel is quite limited because of its long gelation time and poor mechanical properties. Here, we present a facile way to prepare strong silk protein hydrogels simply by adding surfactant into silk fibroin aqueous solution and incubating at 60 °C. The resulting silk protein hydrogels demonstrate fairly good mechanical properties; for example, the silk protein hydrogel made by adding sodium dodecyl sulfate (SDS) has the compressive and tensile moduli of 3.0 and 3.3 MPa, respectively, which are close to some tissues in the body, such as cartilages, tendons, and ligaments. The effect of different types of surfactant on the formation of strong silk protein hydrogel, and the possible reason for the improvement of the mechanical properties of the hydrogel are also discussed. Preparation and Characterization of Natural Silk Fibroin Hydrogel for Protein Drug Delivery

Abstract:In recent years, hydrogels have been widely used as drug carriers, especially in the area of protein delivery. The natural silk fibroin produced from cocoons of the Bombyx mori silkworm possesses excellent biocompatibility, significant bioactivity, and biodegradability. Therefore, silk fibroin-based hydrogels are arousing widespread interest in biomedical research. In this study, a process for extracting natural silk fibroin from raw silk textile yarns was established, and three aqueous solutions of silk fibroin with different molecular weight distributions were successfully prepared by controlling the degumming time. Silk fibroin was dispersed in the aqueous solution as "spherical" aggregate particles, and the smaller particles continuously accumulated into large particles. Finally, a silk fibroin hydrogel network was formed. A rheological analysis showed that as the concentration of the silk fibroin hydrogel increased its storage modulus increased significantly. The degradation behavior of silk fibroin hydrogel in different media verified its excellent stability, and the prepared silk fibroin hydrogel had good biocompatibility and an excellent drug-loading capacity. After the protein model drug BSA was loaded, the cumulative drug release within 12 h reached 80%. We hope that these investigations will promote the potential utilities of silk fibroin hydrogels in clinical medicine.

Abstract:Hydrogels are an attractive class of materials that possess similar structural and functional characteristics to wet biological tissues and demonstrate a diversity of applications in biomedical engineering. Silk fibroin (SF) is a unique natural polymer due to its fibrous protein nature, versatile formats, biocompatibility, tunable biodegradation and is thus a good hydrogel candidate for bio-applications. Compared to synthetic polymer hydrogels, poor mechanical performance is still a fatal drawback that hinders the application of SF hydrogels as structural materials. Researchers have attempted to develop strategies to construct silk fibroin-based high-strength hydrogels (SF-HSHs). Herein, we firstly provide an overview of the approaches of processing SF-HSHs with a focus on the physical/non-covalent crosslinking mechanisms. The examples of SF-HSHs are discussed in detail under four categories, including physical-crosslinked, dual-crosslinked, double network and composite hydrogels respectively. A brief section follows to elucidate on the gelation mechanisms of SF-HSHs before a description of the utility of SF-HSHs in biomedicine and devices is presented. Finally, the potential challenges and future development of SF-HSHs are briefly discussed. This review aims to enhance our understanding of the structure-mechanical property-function relationships of soft materials made from natural polymers and guide future research of silk fibroin-based hydrogels for biomedical applications.

Abstract:Silk proteins are natural macromolecules with unique properties.Due to excellent physiochemical properties and abundant sources,they are playing an increasingly important role in the field of biomaterials.More and more biomedical studies were conducted by using silk proteins as biomaterials,and significant progress has been achieved.This article reviews the different kinds of silk proteins used for biomaterials and processing techniques for transforming natural silk proteins into various biomedical scaffolds,and summarizes the latest advances of international groups focusing on silk-based material research.In addition,the development history of silk-based biomaterials and their application status in tissue engineering are elaborated.Furthermore,the prospects and trends of silk-based scaffold biomaterials are described.It is anticipated to provide good references for increasing the added value of silk industry and extending the application of silkbased biomaterials in clinical application,wishing that the silk fiber with thousands years of history expand their application space and scope continuously.
Progress in Hydrogels Based on Regenerated Silk Fibroin

Abstract:Regenerated silk fibroin (RSF) is a fibrillar protein obtained from Bombyx mori silkworm silk. Recently, RSF-based hydrogels have attracted extensive research interest, which has yielded an improved understanding in the gelling mechanisms, physical and mechanical properties as well as the applications in regenerative medicine and tissue engineering. Mechanistically, RSF hydrogel networks can be formed via two pathways, chemical cross-linking including enzymatic cross-linking, and physical cross-linking through conformational transition, during which thermodynamically more stable beta-sheet structures are induced. An advantage of RSF hydrogels is that the physical and mechanical properties of hydrogels can be further controlled by the beta-sheet content in the conformation on top of concentration and cross-linking density. RSF can be mixed with other polymers and/or inorganic components to prepare composite hydrogels for a wider spectrum of mechanical properties. Furthermore, we summarize most recent applications of RSF hydrogels in biomaterials, including controlled drug release, muscle, tendon and bone tissue engineering.

Abstract:As a natural biomaterial, silk fibroin (SF) holds great potential in biomedical applications with its broad availability, good biocompatibility, high mechanical strength, ease of fabrication, and controlled degradation. With emerging fabrication methods, nanoand microspheres made from SF have brought about unique opportunities in drug delivery, cell culture, and tissue engineering. For these applications, the size and distribution of silk fibroin particles (SFPs) are critical and require precise control during fabrication. Herein, we review common and emerging SFPs fabrication methods and their biomedical applications, and also the challenges and opportunities for SFPs in the near future. Lay Summary: The application of silk in textile has an extraordinarily long history and new biomedical applications emerged owing to the good biocompatibility and versatile fabrication options of its major protein component, silk fibroin. With the development of nanotechnology and microfabrication, silk fibroin has been fabricated into nano- or microspheres with precisely controlled shape and distribution. In this review, we summarize common and emerging silk fibroin particle fabrication methods and their biomedical applications, and also discuss their challenges and opportunities in the nearest future.

Abstract:Silk sericin, a natural protein extracted from silkworm cocoons, has been extensively studied and utilized in the biomedical field because of its superior biological activities and controllable chemical-physical properties. Sericin is biocompatible and naturally cell adhesive, enabling cell attachment, proliferation, and differentiation in sericin-based materials. Moreover, its abundant functional groups from variable amino acids composition allow sericin to be chemically modified and cross-linked to form versatile constructs serving as alternative matrixes for biomedical applications. Recently, sericin has been constructed into various types of biomaterials for tissue engineering and regenerative medicine, including various bulk constructions (films, hydrogels, scaffolds, conduits, and devices) and micro-nano formulations. In this review, we systemically summarize the properties of silk sericin, introduce its different forms, and demonstrate their newly-developed as well as potential biomedical applications.

Its unique chemical structure results in an uncommon combination of macroscopically useful properties, yielding a strong, fine and flexible material which, in addition, presents good biodegradability and better biocompatibility. Therefore, silk fibroin in various formats, appears as an ideal candidate for supporting biomedical applications. In this review, we will focus on the hydrogels obtained from silk fibroin or in combination with it, paying special attention to the synthesis procedures, characterization methodologies and biomedical applications. Tissue engineering and drug-delivery systems are, undoubtedly, the two main areas where silk fibroin hydrogels find their place.

Abstract:This review mainly introduces the types of silk hydrogels, their processing methods, and applications. There are various methods for hydrogel preparation, and many new processes are being developed for various applications. Silk hydrogels can be used in cartilage tissue engineering, drug release materials, 3D scaffolds for cells, and artificial skin, among other applications because of their porous structure and high porosity and the large surface area for growth, migration, adhesion and proliferation of cells that the hydrogels provide. All of these advantages have made silk hydrogels increasingly attractive. In addition, silk hydrogels have wide prospects for application in the field of biomedical materials.

Fabrication and characterization of silk microfiber-reinforced methacrylated gelatin hydrogel with turnable properties

Abstract:Despite considerable research effort, the natural hydrogels presently available for tissue engineering suffer from several major drawbacks, one of the significant issue is their poor mechanical strength which are unable to satisfy some mechanical requirements for successful outcomes. Herein, to mimic the composition and structure of the natural extracellular matrix, the micron-sized silk fibers obtained by alkaline hydrolysis were used as a reinforcement phase in a GelMA hydrogel, resulting in a material with significantly greater stiffness than pure GelMA hydrogel alone. In addition, the hydrogel demonstrated tunable compressive strength, swelling capacity, and degradation properties based on the silk fiber length. Experiments with cells indicated that MC3T3-E1 pre-osteoblasts quickly adhered to and proliferated on the surface of the composite hydrogels, as revealed by FDA/PI staining and CCK-8 assays. In addition, various cellular responses, including cell adhesion, changes in cellular morphology and cell proliferation behavior, occurred on the composite hydrogel and varied with fiber length. Overall, this study introduces a series of fiber-reinforced, tunable composite hydrogels that could be useful for various tissue engineering applications.

Abstract:Silk fibroin (SF) is an excellent protein-based biomaterial produced by the degumming and purification of silk from cocoons of the Bombyx mori through alkali or enzymatic treatments. SF exhibits excellent biological properties, such as mechanical properties, biocompatibility, biodegradability, bioabsorbability, low immunogenicity, and tunability, making it a versatile material widely applied in biological fields, particularly in tissue engineering. In tissue engineering, SF is often fabricated into hydrogel form, with the advantages of added materials. SF hydrogels have mostly been studied for their use in tissue regeneration by enhancing cell activity at the tissue defect site or counteracting tissue-damage-related factors. This review focuses on SF hydrogels, firstly summarizing the fabrication and properties of SF and SF hydrogels and then detailing the regenerative effects of SF hydrogels as scaffolds in cartilage, bone, skin, cornea, teeth, and eardrum in recent years.

Abstract:Silk fibroin (SF) from Bombyx mori is a natural polymer with exceptional biocompatibility, low immunogenicity, and ease of processability. SF-based hydrogels have been identified as one of the most attractive candidate scaffolds for tissue engineering and can be fabricated through various physical or chemical crosslinking approaches. However, conventional SF hydrogels may suffer from several major drawbacks, such as structural inhomogeneity, poor mechanical properties or utilization of cytotoxic reagents. Herein, a dually crosslinked SF-based composite hydrogel with enhanced strength and elasticity was fabricated by inducing the formation of uniform and small β-sheet structures by sonication in a restricted enzymatic precrosslinked network. The composite hydrogel not only demonstrated concentration-dependent stiffness variation but also exhibited time-dependent changes in toughness behavior. Moreover, subsequent experimental results revealed that the hydrogels exhibit other advantages, including high water retention capacity and long-term stability under physiological conditions. Finally, a three-dimensional (3 D) construct of the cell-laden hydrogel was fabricated, confirming that the composite hydrogel could provide a biocompatible microenvironment with dynamically changing mechanical properties. The combination of physical and enzymatic crosslinking strategies contributes to a biocompatible composite hydrogel with unique mechanical properties that holds great potential for use in tissue engineering and regenerative medicine.
