Purification of an ionic liquid stable cellulase from Aspergillus aculeatus PN14 with potential for biomass refining

Abstract:Ionic liquids (ILs), the low temperature organic solvents may have application potential for a variety of industries including IL-based lignocellulosic biomass (LB) conversion to energy, fuels, and chemicals. Following IL-pretreatment, the LB polysaccharides are enzymatically saccharified to sugars. However, vulnerability of enzymes to traces of IL in pretreated biomass may compromise the overall process; therefore, IL-stable enzymes are desired for such applications. Previously, an IL-stable cellulase from an IL-tolerant fungal isolate Aspergillus aculeatus PN14 (from a chemically polluted niche) was successfully used for saccharification of IL-pretreated Parthenium hysterophorus biomass for biofuel-ethanol production. Rare studies are available on cellulases from A. aculeatus species. In the current study, cellulase from A. aculeatus PN14 was purified (threefold, specific activity 69.9 U/mg) to homogeneity using ammonium sulfate precipitation followed by diethyl-aminoethyl-sephadex chromatography. Native-PAGE and zymogram analysis established in-gel activity, while SDS-PAGE ascertained the estimated molecular weight (29 kDa) of cellulase. Though the temperature and pH optima of cellulase were 50 °C, and pH 8.0, respectively, but the enzyme exhibited substantial activity and stability (88–96%) over elevated temperatures (60–80 °C, > 80%), and high acidic and alkaline pH (pH 3–11, 60–85%). Activity was stimulated by Co 2+ , Mn 2+ , Mg 2+ , Zn 2+ and Ca 2+ , and inhibited by K + , Cu 2+ , Hg 2+ , ethylenediaminetetraacetic acid (EDTA), cetyltrimethylammonium bromide (CTAB), and sodium dodecyl sulfate (SDS). Double reciprocal plot analysis showed that cellulase had K m and V max of 3.91 mg/ml and 19.45 μmol/min/mg, respectively. A. aculeatus PN14 cellulase being equipped with desirable characteristics may have multitude potential for industrial applications.
