Abstract:In this work, a new immobilized cellobiase enzyme system was used in the enzymatic hydrolysis of biomass for the generation of cellulosic ethanol. The application of the immobilized cellobiase enzyme enabled easy post-hydrolysis separation of the enzyme from the reaction mixture. The cellobiase enzyme was immobilized in porous solid silica host material via acid catalyzed non-surfactant templated sol–gel reaction of tetraethyl orthosilicate (TEOS). The pore size and porosity of the host material were controlled by varying the template (d-fructose) content. The immobilized cellobiase enzyme samples when used in the hydrolysis of pre-treated biomass yielded hydrolysis efficiency up to 74% (with FeCl3 pre-treated biomass) and 81% (with oxalic acid pre-treated biomass) when compared to hydrolysis with free cellobiase enzyme. The solid immobilized enzyme samples could be easily recycled from the reaction mixture after the hydrolysis reaction. When re-used in multiple batches, the immobilized samples retained their enzymatic activity in biomass hydrolysis. The re-usability of the immobilized cellobiase with retained activity would actively bring down the enzyme cost. Thus, our immobilized cellobiase enzyme opens up great potential in the enzymatic hydrolysis of biomass for generating cellulosic ethanol, as it leads to overall process cost reduction.

Abstract:Cellulosic material is the most abundant renewable carbon source in the world. Cellulose may be hydrolyzed using cellulase to produce glucose, which can be used for production of ethanol, organic acids, and other chemicals. Cellulase is a complex enzyme containing endoglucanase (EC 3.2.1.4), exoglucanase (EC 3.2.1.91) and cellobiase (EC 3.2.1.21). The hydrolysis of natural cellulose to glucose depends on the synergism of these three components. The mostly used cellulase produced by Trichoderma reesei has high activity of endoglucanase and exoglucanase, but the activity of cellobiase is relatively low. Therefore, improving the activity of cellobiase in cellulase reaction system is the key to enhance the sacchrification yield of cellulosic resources. Aspergillus niger LORRE 012 was a high productivity strain for cellobiase production. It was found that the spores of this strain were rich in cellobiase. In this work, the cellobiase was immobilized efficiently by simply entrapping the spores into calcium alginate gels instead of immobilizing the pure cellobiase proteins. The immobilized cellobiase was quite stable, and its half-life was 38 days under pH 4.8, 50 degrees C. The thermal stability of the immobilized cellobiase was improved, and it was stable below 70 degrees C. The suitable pH range of the immobilized cellobiase was pH 3.0 - 5.0, with the optimal pH value 4.8. The Km and Vmax value of the immobilized cellobiase were 6.01 mmol/L and 7.06 mmol/min x L, respectively. In repeated batch hydrolysis processes, 50 mL of substrate (10 g/L cellobiose) and 10 mL of immobilized beads containing cellobiase were added into a 150 ml flask. After reacting at pH 4.8, 50 degrees C for several hours, the hydrolysate was harvested for assay, and the immobilized beads were used for the next batch hydrolysis with the fresh substrate. This process was repeated, and the yield of enzymatic hydrolysis kept higher than 97% during 10 batches. The continuous hydrolysis process was carried out in a column reactor (inside diameter 2.8 cm, inside height 40 cm) packed with the immobilized beads. Using 10 g/L cellobiose as substrate, the hydrolysis yield reached 98% under 0.4 h (-1) dilution rate and pH 4.8, 50 degrees C. After corncob was treated by 1% dilute acid, the cellulosic residue (100 g/L) was used as substrate, and hydrolyzed by the cellulase (15 IFPU/g substrate) from Trichoderma reesei, at pH 4.8, 50 degrees C for 48 h. The concentration of reducing sugar in the hydrolysate was only 48.50 g/L (hydrolysis yield 69.5%). When the hydrolysate was further treated by the immobilized cellobiase, the cellobiose was hydrolyzed into glucose, and the feedback inhibition caused by the cellobiose accumulation disappeared sharply. By the synergism of immobilized cellobiase and the cellulase from T. reesei left in the hydrolysate, other oligosaccharides were mostly converted to monosaccharides. At 48 h, the reducing sugar concentration was increased to 58.78 g/L, the hydrolysis yield of the corncob residue was improved to 84.2%, and the ratio of the glucose in the total reducing sugar was increased from 53.6% to 89.5%. The reducing sugars converted from corncob could be used further in the fermentation of valuable industrial products. This research results were meaningful in the conversion and utilization of renewable biomass.
Extrusion process to enhance the pretreatment effect of ionic liquid for improving enzymatic hydrolysis of lignocellulosic biomass

Abstract:Extrusion process using 1-ethyl-3-methylimidazolium acetate ([EMIM]Ac) in co-solvent, dimethyl sulfoxide (DMSO), was conducted to improve the pretreatment effect on the enzymatic saccharification of pussy willow (Salix gracilistyla Miq.). The use of a twin screw extruder capable of continuous processing increased the solid loading to 50% in [EMIM]Ac and its co-solvent DMSO. The water-soluble fraction was increased by increasing the [EMIM]Ac amount in co-solvent and the extrusion temperature but decreasing the screw rotating speed and solid loading. The surface morphologies of the extruded product showed remarkable disruption of the cell wall structures and fibrillation with increasing amounts of [EMIM]Ac in co-solvent. With increasing [EMIM]Ac amount in co-solvent and extrusion temperature and with decreasing screw rotating speed and solid loading, the crystalline structure of cellulose was more disrupted, resulting in the increase in saccharification yield. The highest yield of glucose and xylose was 99.0% and 99.5%, respectively, from the pretreated products using only [EMIM]Ac under solid loading of 15% at 160 °C extrusion temperature and 5 rpm screw rotating speed. The initial saccharification rate was faster in the pretreated product precipitated in alcohol and acetone than in water.

Abstract:Industrial waste corn cob residue (from xylose manufacturing) without pretreatment was hydrolyzed by cellulase and cellobiase. The cellulosic hydrolysate contained 52.4 g l −1 of glucose and was used as carbon source for lactic acid fermentation by cells of Lactobacillus delbrueckii ZU-S2 immobilized in calcium alginate gel beads. The final concentration of lactic acid and the yield of lactic acid from glucose were 48.7 g l −1 and 95.2%, respectively, which were comparative to the results of pure glucose fermentation. The immobilized cells were quite stable and reusable, and the average yield of lactic acid from glucose in the hydrolysate was 95.0% in 12 repeated batches of fermentation. The suitable dilution rate of continuous fermentation process was 0.13 h −1 , and the yield of lactic acid from glucose and the productivity were 92.4% and 5.746 g l −1 h −1 , respectively. The production of lactic acid by simultaneous saccharification and fermentation (SSF) process was carried out in a coupling bioreactor, the final concentration of lactic acid was 55.6 g l −1 , the conversion efficiency of lactic acid from cellulose was 91.3% and the productivity was 0.927 g l −1 h −1 . By using fed-batch technique in the SSF process, the final concentration of lactic acid and the productivity increased to 107.6 g l −1 and 1.345 g l −1 h −1 , respectively, while the dosage of cellulase per gram substrate decreased greatly. This research work should advance the bioconversion of renewable cellulosic resources and reduce environmental pollution.

Abstract:The practice to kill terminally ill patients on their own demand has resulted in the Netherlands in a decriminalisation of active euthanasia which thus has fundamentally changed the way to deal with dying patients. Sooner or later this development will extend to other European countries as well as to the USA. Involuntary euthanasia of severely handicapped newborn children or of demented persons is propagated by the practical ethics of P. Singer and other representatives of utilitarianistic philosophy. According to the standpoint of utilitarianism a human being should only have the right to live as long as he or she is a person, i. e. has rationality and self-consciousness. The next step toward the elimination of elderly people can easily be predicted. For economical reasons these persons may be withheld from life-saving medical treatment or may be supposed to commit suicide. A moral pressure is created to make a decision for suicide as soon as severe invalidity occurs. The consideration of such ideas shows that in today's debate on euthanasia the issue is no longer the right of a few severely and terminally ill human beings to their "own death". Instead, the right to live of a large group of handicapped and "socially useless" or "unproductive" persons is at stake. This is the danger of today's discussion of euthanasia.
Enzymatic hydrolysis of structurally upgraded lignocellulosic biomass with the aid of humic acid: a case study in a membrane integrated bioreactor

Abstract:Substrate accessibility by an enzyme is the rate-limiting step in cellulose digestion. The newly schematized humic acid (HA)-assisted alkali pretreatment was done to delignify and upgrade the bagasse's characteristics for the pragmatic accessibility of the enzyme. The proposed pretreatment resulted in ~90–100% lignin recovery with distinctive properties. After delignification, the upgraded biomass harnessed as solid support to immobilize the enzyme, ultimately reducing the cost and step for designing physical support for immobilization. HA, an active ligand, with its enriched functional moieties such as carboxyl, carbonyl, hydroxyl, and amide group, bind prior to the substrate (i.e., delignified bagasse) that immobilizes enzyme and hydrolyses cellulose portion. The interaction mechanisms of pretreated bagasse with enzyme were demonstrated through molecular docking using Auto Dock software and UV (Ultraviolet) Spectrophotometric analysis. The pretreatment efficiency was analyzed as a case study in a large-scale reactor by subjecting pretreated bagasse to batch enzymatic hydrolysis with a low enzyme-loading rate of 14 FPU/g of cellulose. Cellulose conversion of 88% (i.e., 20.92 g/L of glucose) was achieved in 48 h, respectively. Subsequently, the flat-sheet cross-flow ultrafiltration (UF) membrane-based continuous recycling unit was operated at a flux of 22.5 L/m2hr toward the purification of glucose (i.e., 15.75 g/L of glucose). The performance of PES10-UF membrane module chaperoning with hydrolysis reactor for the designed hydrolysis scheme was examined in light of critical flux, pure water permeability, and irreversible fouling studies. The flow rate of 300 LPH (L/hr) and the transmembrane membrane pressure (TMP) of 1.5 bar showed high antifouling performance.

Abstract:This study aims to establish a natural cellulosic biomass pretreatment process using ionic liquid (IL) for efficient enzymatic hydrolysis and second generation bioethanol. The IL 1-Butyl-3-methylimidazolium Chloride/FeCl3 ([Bmim]Cl/FeCl3) was selected in view of its low temperature pretreatment ability and the potential of accelerating enzymatic hydrolysis, and it could be recyclable. The yield of reducing sugars from sugarcane residue pretreated with this IL at 80 oC for 1 h reached 46.8% after being enzymatically hydrolyzed for 24 h. Sugarcane residue regenerated were hydrolyzed more easily than that treated with water. The fermentability of the hydrolyzates, obtained after enzymatic saccharification of the regenerated sugarcane residue, was transformed into bioethanol using Candida shehatae. This microbe could absorb glucose and xylose efficiently, and the ethanol production was 0.38 g/g glucose within 30 h fermentation. In conclusion, the metal ionic liquid pretreatment in low temperature shows promise as pretreatment solvent for natural biomass.
Ethylenediamine Enhances Ionic Liquid Pretreatment Performance at High Solid Loading

Abstract:Higher biomass loading (≥20% w/w) in pretreatment unit operations for lignocellulosic biorefineries will lead to a better economic feasibility and higher green efficiency. However, the industrial development behind high biomass loading in ionic liquid pretreatment has been hindered by unsatisfactory lignin fractionation and fermentable sugar release. Here, this study demonstrates that the addition of ethylenediamine (EDA) effectively improved glucose and xylose release in pretreatment using a 1-ethyl-3-methylimidazolium acetate ([Emim][OAc])-based system at corn stover loadings as high as 36% w/w under facile conditions. It was discovered that synergistic pretreatment with a 10% v/v EDA ratio in [Emim][OAc] was optimal for maximizing fermentable sugar yields, showing glucose yields of 94% and xylose yields of 48% based on untreated biomass; meanwhile, xylan loss was avoided due to low EDA loading. Additionally, the synergistic effect between [Emim][OAc] and EDA further reduced cellulose crystallinity when compared to the single pretreatment with [Emim][OAc] or EDA. In synergistic pretreatment, it was revealed that EDA played a crucial role in the cleavage of lignin–carbohydrate complex linkages, lignin relocalization, and removal at high solid loading, which provided "easy and simple" pathways for [Emim][OAc] to attack crystalline domains of native cellulose. Also, there were more native β-O-4 structures preserved in treated lignin after this synergistic pretreatment, offering a potential to make downstream lignin valorization even easier and more efficient. In addition, to achieve excellent performance, synergistic pretreatment could diminish requirements for pretreatment severity relative to single [Emim][OAc] pretreatment, which reduced glucan loss as well as energy consumption in the pretreatment process. Selective Separation and Recovery of the Lignin from Lignocellulosic Biomass Using Ionic Liquid

Abstract:Lignocellulosic biomass represents a potentially key and sustainable source for transformation into biofuels and other commodity chemicals. However, efficient means of separating and depolymerising the hard components are required. The use of ionic liquids (ILs) in biomass pretreatment has attracted significant attention as promising green solvents for its effectiveness at decreasing biomass recalcitrance. In this study, an efficient pretreatment method using the low-cost IL 1-butyl-3-methylimidazolium acesulfamate [BMIM]Ace and organic solvents for the extraction of lignin and recovery of carbohydrate-enriched materials (CEMs) from Eucalyptus. The lignin was recovered from the IL by precipitation with acetone, allowing the IL to be recycled. The results showed a synergy effect with exceeding 89.6% of the CEMs recovery and 14.1% of the lignin obtained. It was found that the presence of some organic solvents have affected the extraction efficiency, probably due to the difference of dielectric constants of the organic solvents. The regenerated ionic liquid showed good retention of structure and properties. The purity of the lignin fractions were characterized by high-performance anion exchange chromatography (HPAEC). Moreover, the lignin fractions were characterized with Fourier transform infrared (FT-IR). The recovered CEMs can be serving as a valuable source for enzymatic saccharification. These results indicates a potential approach in further optimization of the choice of low-cost ILs and organic solvents systems for the efficient utilization of the lignocellulosic biomass, and all constituents can be fully recovered and further processed.
Dissolution pretreatment of cellulose by using levulinic acid-based protic ionic liquids towards enhanced enzymatic hydrolysis

Abstract:In this study, an economically competitive and sustainable levulinic acid-based protic ionic liquids were identified to be good solvents for the dissolution pretreatment of cellulose towards enhanced enzymatic hydrolysis. The influences of protic ionic liquids species, dissolution pretreatment time, and pretreatment temperature on the physico-chemical structures of cellulose were systematically investigated by various analytical techniques. The findings indicate that the pretreatment efficiency was correlated to the basicity of the organic bases, and the presence of ketone group in the levulinate anion with particular hydrogen bonding forming ability via keto–enol tautomerism. The DBN derived protic ionic liquids exhibited best performance at 100 °C in one hour, as evidenced by a 94% glucose yield. This solvent system was also suitable for the dissolution pretreatment of corn stover-based lignocellulosic biomass for sugars production, although a higher temperature and longer pretreatment time was required. Furthermore, the solvent system could be recycled and reused.

Solvent-based Delignification and Decrystallization of Wheat Straw for Efficient Enzymatic Hydrolysis of Cellulose and Ethanol Production with Low Cellulase Loadings

Abstract:Several solvents and ionic liquids, namely formic acid (Formiline process), concentrated phosphoric acid (CPA), N-methylmorpholine-N-oxide (NMMO) and 1-allyl-3-methylimidazolium chloride ([AMIM]Cl), were used to pretreat wheat straw in order to increase cellulose digestibility for ethanol production. When being directly used to pretreat the raw wheat straw under corresponding optimized conditions, the improvement of cellulose hydrolyzability followed the order of CPA > Formiline > NMMO > [AMIM]Cl. However, when Formiline pretreated (delignified) substrates were further post-treated by the above cellulose solvents, the cellulose digestibility was significantly improved particularly with low cellulase loadings. Cellulose solvent post-treatment resulted in deconstruction of hydrogen-bond networking, alteration of cellulose polymorphs, decrease in crystallinity, depolymerization of cellulose chain with dramatic reduction in particle size, thus greatly increasing cellulose accessible surface area. CPA post-treatment showed the best efficacy. Semi-simultaneous scarification and fermentation (sSSF) of a CPA post-treated substrate obtained an ethanol concentration of 41.6 g L−1 with 91.2% of yield at a relatively low cellulase loading (5 FPU per g solid) within 24 h incubation. A biorefining process was proposed based on Formiline pretreatment coupled with CPA post-treatment to achieve a co-production of ethanol, furfural and high-purity lignin, which greatly increases the potential revenue of the process.
A zwitterionic liquid vs an ionic liquid analogue upon biomass pretreatment ability

Abstract:One-pot ethanol production, which involves successive biomass pretreatments, enzymatic hydrolysis, and fermentation in a single vessel, is essential for practical bioethanol production. To achieve this, low-toxicity pretreatment solvents such as a zwitterionic liquid, OE2imC3C, is necessary. Although the cellulose-dissolving ability of OE2imC3C has been studied, its biomass pretreatment ability has not been thoroughly investigated. Herein, we investigated the biomass pretreatment ability of OE2imC3C at various temperatures and concentrations. The mildest conditions for successful pretreatment were 75 wt.% OE2imC3C at 90 °C. On the other hand, OE2imC3C exhibited a lower pretreatment ability than [C2mim]OAc, which is the best-known cellulose-dissolving ionic liquid. The main reason for the low pretreatment ability of OE2imC3C was its high viscosity. Reducing their viscosity is a key factor for designing zwitterionic liquids with high pretreatment ability.

Abstract:The inhibition effect of impurities in the raw lignocellulosic biomass greatly impedes its direct acid-catalyzed alcoholysis process to produce downstream chemicals. Previous literatures widely investigated the inhibition problems of lignin content and cellulose crystallite in the raw biomass for the production of alkyl levulinate (AL), while the effect of inorganic matters on this process only received cursory attention. In this contribution, the vital importance of ash removal in raw material for efficient catalytic alcoholysis of cellulose fraction been highlighted. Moreover, it has been found that the facile pretreatment of Napier grass stem greatly facilitates the acid-catalyzed alcoholysis of its cellulose fraction towards ethyl levulinate (EL), a versatile commodity chemical. EL yield as high as 72.1 % can be accomplished from the NaOH-pretreated Napier grass stem in ethanol/γ-valerolactone co-solvent medium over Al(OTf) 3 at 170 °C for 150 min, which is almost ten times that from the pristine one in ethanol medium. It is also interesting to note that a facile and green water prewashing treatment can efficiently remove the inorganic component of the raw biomass, which thus largely promotes the conversion of Napier grass stem to EL. The activation energies of cellulose degradation and glucopyranoside alcoholysis after pretreatment decreased to 62.4 kJ/mol and 68.6 kJ/mol, respectively. This work presents a highly-reliable strategy for valorizing grass lignocellulosic feedstock to produce bio-based ester compounds.

Abstract:Pretreatment of lignocellulosic materials is a prerequisite to facilitate the disruption of the natural recalcitrance of their carbohydrate–lignin shield and to allow enzymes to easily access the crystalline cellulose surfaces. Recently, pretreatment of ionic liquids (ILs) has been widely studied as a promising pretreatment technique; however, it is too expensive to be commercialized. In this study, an efficient acid-catalyzed aqueous IL pretreatment process was developed to optimize the total sugar conversion of pretreated biomass and to reduce IL usage. The experimental results demonstrated that the total sugar conversion was raised to 92.7 % with the synergistic effects of IL (1,3-dimethylimidazolium dimethylphosphate, [MMIM]DMP) and dilute hydrochloric acid (HCl) under pretreatment conditions of 110 °C for 2 h, compared to the conversion of only 27.3 % obtained with untreated corn stover. Moreover, the addition of the inorganic acids, especially HCl, to the IL pretreatment was found to not only significantly destroy the crystalline structure of cellulose in corn stover, promoting the conversion of cellulose and hemicellulose to monomeric sugars, but also provide an opportunity to reduce the usage of expensive IL solvents.

Development of a novel ultrasound-assisted alkali pretreatment strategy for the production of bioethanol and xylanases from chili post harvest residue

Abstract:A novel ultrasound-assisted alkali pretreatment strategy was developed which could effectively remove lignin and hemicelluloses and improve the sugar yield from chili post harvest residue. Operational parameters that affect the pretreatment efficiency were studied and optimized. Inhibitor analysis of the hydrolyzate revealed that major fermentation inhibitors like furfural, 5-hydroxymethyl furfural as well as organic acids like citric acid, succinic acid and propionic acid were absent. Hence fermentation can be carried out without detoxification of the hydrolyzate. Changes in structural properties of the biomass were studied in relation to the pretreatment process using Scanning Electron Microscopy (SEM) and the changes in chemical composition were also monitored. The biomass pretreated with the optimized novel method could yield 0.428g/g of reducing sugars upon enzymatic hydrolysis. The hydrolyzate obtained by this novel pretreatment strategy was found to be suitable for bioethanol and xylanase production.

Abstract:Lignocellulosic biomass (LB) is a promising sugar feedstock for biofuels and other high-value chemical commodities. The recalcitrance of LB, however, impedes carbohydrate accessibility and its conversion into commercially significant products. Two important factors for the overall economization of biofuel production is LB pretreatment to liberate fermentable sugars followed by conversion into ethanol. Sustainable biofuel production must overcome issues such as minimizing water and energy usage, reducing chemical usage and process intensification. Amongst available pretreatment methods, microorganism-mediated pretreatments are the safest, green, and sustainable. Native biodelignifying agents such as Phanerochaete chrysosporium, Pycnoporous cinnabarinus, Ceriporiopsis subvermispora and Cyathus stercoreus can remove lignin, making the remaining substrates amenable for saccharification. The development of a robust, integrated bioprocessing (IBP) approach for economic ethanol production would incorporate all essential steps including pretreatment, cellulase production, enzyme hydrolysis and fermentation of the released sugars into ethanol. IBP represents an inexpensive, environmentally friendly, low energy and low capital approach for second-generation ethanol production. This paper reviews the advancements in microbial-assisted pretreatment for the delignification of lignocellulosic substrates, system metabolic engineering for biorefineries and highlights the possibilities of process integration for sustainable and economic ethanol production.

Abstract:Abstract Cellulolytic enzymes can readily access the cellulosic component of lignocellulosic biomass after the removal of lignin during biomass pretreatment. The enzymatic hydrolysis of cellulose is necessary for generating monomeric sugars, which are then fermented into ethanol. In our study, a combination of a deep eutectic (DE) mixture (of 2-aminoethanol and tetra- n -butyl ammonium bromide) and a cyclic ether (tetrahydrofuran) was used for selective delignification of rice straw (RS) under mild conditions (100 °C). Pretreatment with DE-THF solvent system caused ~ 46% delignification whereas cellulose (~ 91%) and hemicellulose (~ 67%) recoveries remained higher. The new solvent system could be reused upto 10 subsequent cycles with the same effectivity. Interestingly, the DE-THF pretreated cellulose showed remarkable enzymatic hydrolysability, despite an increase in its crystallinity to 72.3%. Contrary to conventional pretreatments, we report for the first time that the enzymatic hydrolysis of pretreated cellulose is enhanced by the removal of lignin during DE-THF pretreatment, notwithstanding an increase in its crystallinity. The current study paves way for the development of newer strategies for biomass depolymerization with DES based solvents.

[Gait impairment in the oldest old].

Abstract:To evaluate senile gait patterns in octogenarians and nonagenarians, we provided a standardized questionnaire on gait disabilities to 153 elderly subjects over 88 years of age. Subjects represented a relatively healthy subgroup of non-institutionalized residents who participated in a gerontological survey of all inhabitants of the city of Leiden who were 85 years or older. Of the 142 subjects who responded to this questionnaire, 87 persons (61%) claimed distinct diseases as a cause of gait impairment. Of the remaining 55 persons, 42 received a standardized gait assessment. Gait was classified as completely normal in 25 persons (18% of all responders), whereas in three other persons gait could not reliably be classified as either normal or abnormal. A wide spectrum of clear gait abnormalities-mainly with ataxic features-was encountered in the remaining 14 persons (10%). It is concluded that some elderly subjects have a mainly ataxic gait disturbance which seems unrelated to the presence of distinct diseases. Although additional investigations might still reveal underlying pathology in these subjects, their gait impairment may represent the "idiopathic senile gait disorder'. In addition, a relatively high number of very old community residents have a completely normal gait.
Rhamnolipid enhanced beta-glucosidase from Paenibacillus sp. LLZ1 for in situ lignocellulose saccharification in ionic liquids

Abstract:Low β-glucosidase activity and stability are challenging limitations for the in situ saccharification of lignocellulose pretreated by ionic liquids. The addition of 0.1% (v/v) rhamnolipid, an ionic biosurfactant, was found to enhance the activity of β-glucosidase from Paenibacillus LLZ1 by 39% through a synergistic effect with 5% [Emim]OAc. The concentration of [Emim]OAc was further optimized to 2.5% (v/v) obtaining as high as 48% PsBGL activity increase. Fluorescence and near-UV circular dichroism spectroscopy analyses revealed looser protein structure and more hydrophilic microenvironment in the system of rhamnolipid and [Emim]OAc. Though ILs showed deactivation effect on cellulases, rhamnolipid was speculated to alleviate protein denaturation significantly. The reaction system was further applied in the hydrolysis of cellobiose and bagasse cellulose. The conversion was increased by 33% for cellobiose and by 62% for bagasse cellulose, demonstrating the application potential of the reaction system. The improvement was attributed to both reduced cellobiose inhibition and effective substrate solubilizing, and the latter was monitored by scanning electron microscopy to show the morphological change of substrates directly. This study proved the effect of surfactant in in situ enzymatic saccharification of lignocellulose and provided a practical enzymatic hydrolysis system.

Abstract:Biomass in the form of agricultural and forest waste is among the most anticipated materials for sustainable processes and economical bio-based products. Cellulose is widely utilized, but it is not always in huge relative amounts; therefore, biomass is first enriched by removing unwanted constituents through pretreatment. The most efficient among the primary generations of ILs used for biomass valorization were based on aromatic cations. Despite their efficacies and extended tunability, these ILs are costly and their greenness is questionable. Aliphatic ILs are cheaper and environmentally more benign, thus, more desirable for biomass processing. In this study, a protic IL based on an aliphatic amine N,N-dimethylbutylamine (DMBA) and sulfuric acid has been employed for the cellulose enrichment of sugar cane bagasse and corn stover. The results showed high post-treatment cellulose content in corn stover (49%) and bagasse (39%). It exhibited fairly high lignin removal, i.e., 83% and 75% and glucose yields 82% and 91% from bagasse and corn stover, respectively. The extent of delignification, cellulosic content, and glucose yields are evident of the high efficiency of the IL.
