Abstract:Limonoids , a class of abundant natural tetracyclic triterpenoids, present diverse biological activity and provide a versatile platform amenable by chemical modifications for clinical use. Among all of the limonoids isolated from natural sources, obacunone, nomilin , and limonin are the primary hub of limonoid-based chemical modification research. To date, more than 800 limonoids analogs have been synthesized, some of which possess promising biological activities. This review not only discusses the synthesis of limonoid derivatives as promising therapeutic candidates and details the pharmacological studies of their underlying mechanisms from 2002 to 2022, but also proposes a preliminary limonoid synthetic structure-activity relationship (SAR) and provides future direction of limonoid derivatization research.

Abstract:Covering: July 2010 to December 2021 Limonoids, a kind of natural tetranortriterpenoids with diverse skeletons and valuable insecticidal and medicinal bioactivities, are the characteristic metabolites of most plants of the Meliaceae family. The chemistry and bioactivities of meliaceous limonoids are a continuing hot area of natural products research; to date, about 2700 meliaceous limonoids have been identified. In particular, more than 1600, including thirty kinds of novel rearranged skeletons, have been isolated and identified in the past decade due to their wide distribution and abundant content in Meliaceae plants and active biosynthetic pathways. In addition to the discovery of new structures, many positive medicinal bioactivities of meliaceous limonoids have been investigated, and extensive achievements regarding the chemical and biological synthesis have been made. This review summarizes the recent research progress in the discovery of new structures, medicinal and agricultural bioactivities, and chem/biosynthesis of limonoids from the plants of the Meliaceae family during the past decade, with an emphasis on the discovery of limonoids with novel skeletons, the medicinal bioactivities and mechanisms, and chemical synthesis. The structures, origins, and bioactivities of other new limonoids were provided as ESI. Studies published from July 2010 to December 2021 are reviewed, and 482 references are cited.

Abstract:Limonoids are a group of secondary metabolisms containing furan ring and highly oxidized tetracyclic triterpene.It has many significant biological activities,such as anticancer,antiviral,antioxidant,antimicrobial,insect antifeedant,antiinflammatory and analgesic atherosclerosis.Limonoids are mainly found in Rutaceae and Meliaceae families,which are particularly abundant in the citrus seeds.The research on the species,structure and biosynthetic pathways of limonoids in citrus seeds and the research progress on the bioactivity of limonin and its analogues in recent years are discussed.Its application in food industry,agriculture,medicine and animal husbandry are also reviewed to provide references for the application of limonin and its analogues in citrus seeds.

Abstract:Limonin is a natural tetracyclic triterpenoid compound, which widely exists in Euodia rutaecarpa (Juss.) Benth., Phellodendron chinense Schneid., and Coptis chinensis Franch. Its extensive pharmacological effects have attracted considerable attention in recent years. However, there is no systematic review focusing on the pharmacology, toxicity, and pharmacokinetics of limonin. Therefore, this review aimed to provide the latest information on the pharmacology, toxicity, and pharmacokinetics of limonin, exploring the therapeutic potential of this compound and looking for ways to improve efficacy and bioavailability. Limonin has a wide spectrum of pharmacological effects, including anti-cancer, anti-inflammatory and analgesic, anti-bacterial and anti-virus, anti-oxidation, liver protection properties. However, limonin has also been shown to lead to hepatotoxicity, renal toxicity, and genetic damage. Moreover, limonin also has complex impacts on hepatic metabolic enzyme. Pharmacokinetic studies have demonstrated that limonin has poor bioavailability, and the reduction, hydrolysis, and methylation are the main metabolic pathways of limonin. We also found that the position and group of the substituents of limonin are key in affecting pharmacological activity and bioavailability. However, some issues still exist, such as the mechanism of antioxidant activity of limonin not being clear. In addition, there are few studies on the toxicity mechanism of limonin, and the effects of limonin concentration on pharmacological effects and toxicity are not clear, and no researchers have reported any ways in which to reduce the toxicity of limonin. Therefore, future research directions include the mechanism of antioxidant activity of limonin, how the concentration of limonin affects pharmacological effects and toxicity, finding ways to reduce the toxicity of limonin, and structural modification of limonin—one of the key methods necessary to enhance pharmacological activity and bioavailability.

Abstract:Highly substituted pyridine scaffolds are found in many biologically active natural products and therapeutics. Accordingly, numerous complementary de novo approaches to obtain differentially substituted pyridines have been disclosed. This article delineates the evolution of the synthetic strategies designed to assemble the demanding tetrasubstituted pyridine core present in the limonoid alkaloids isolated from Xylocarpus granatum, including xylogranatopyridine B, granatumine A and related congeners. The most efficient and convergent construction of the core framework present in xylogranatopyridine B involved a Liebeskind pyridine synthesis and late-stage benzylic oxidation. By contrast, the synthesis of the bislactone limonoid alkaloids, such as granatumine A which exhibited moderate PTP1B-inhibitory activities, necessitated the development of a novel pyran-to-pyridine conversion. In addition, NMR calculations suggested structural misassignment of several limonoid alkaloids, and predicted their C3-epimers as the correct structures, which was further validated unequivocally through chemical synthesis. While preliminary results of the pNPP assays showed that these bislactone limonoid alkaloids were only weakly inhibitory against PTP1B, C3-deoxy-xylogranatin F, an unnatural synthetic analog, was demonstrated to be more potent than the other congeners.

Abstract:Background: Limonoids represent an important class of natural products which possess a broad range of biological activities. Albeit their enormous potentials as therapeutic candidates, they usually suffer from low bioavailability, poor aqueous solubility and relatively weak biological activities which result in significant challenges in the clinic applications. Therefore, the exploration and development of novel limonin derivatives with improved drug-like properties through the structural modifications recently have attracted great attention in the biological and medicinal chemistry field. Methods: Based on the structural modifications of C17-furan ring in limonin, a series of limonin derivatives was designed, synthesized and screened for their anti-inflammatory and analgesic activities in vivo. Results and Conclusion: Preliminary pharmacological studies revealed that most tested compounds exhibited more potent anti-inflammatory and analgesic efficacies than lead molecule limonin. Especially, for compound 3f, it exhibited a stronger anti-inflammatory effect than that of naproxen and comparable analgesic potency with aspirin. In the formalin test, 3f showed an obviously attenuated phase-II pain response which indicated that it may produce an anti-inflammatory effect in the periphery. Furthermore, the significantly low hERG inhibition (IC50 >100 μM) and high LD50 value of target molecule 3f further demonstrated it as a promising analgesic/anti-inflammatory candidate with excellent drug-like profiles.

Abstract:The lamellarins, a group of natural pyrrole alkaloids with interesting anticancer activities, were firstly isolated from a prosobranch mollus Lamellaria sp.. Due to their multidrug resistance (MDR) reversal in some cancer cell lines, total synthesis and structure modifications of lamellarins to provide drug candidates for the treatment of cancer have become one of the most promising project in the area of novel anticancer drug discovery. In this article, the latest results and progresses regarding the synthesis and modification of lamellarins are reviewed.

Abstract:The increasingly serious resistance of Tetranychus cinnabarinus Boisduval to a wide range of insecticides/acaricides poses a major challenge to their control. The citrus processing industry generates a huge quantity of various wastes that contain many limonoids. To effectively utilize these byproducts and discover more potent green acaricidal molecules as sustainable alternatives for traditional resistant pesticides, various limonoid-type derivatives (halogenated/seven-membered lactam derivatives of obacunone and halogenated/oxime esters/oxime ethers/seven-membered lactam derivatives of limonin) were synthesized based on a diversity-oriented synthetic strategy. The key steric configurations of 10 derivatives were further confirmed by X-ray crystallography. Compound 9m, which displayed greater than 9.7-fold potent acaricidal activity of limonin, was of preeminence. In addition, some interesting structure-activity relationships were observed. Moreover, a biocompatible O/W nanoemulsion delivery system was used to prepare the limonin-based agronanoacaricide, which exhibited pronounced control efficiency against T. cinnabarinus Boisduval in the greenhouse. This systematic investigation will provide valuable information and guidance for future value-added applications of novel eco-friendly natural product-based nanopesticides.

Abstract:Limonene and its derivatives have great market potential with diverse applications in food, pharmaceuticals, cosmetics, etc. Commercial production of limonene and its derivatives through extraction from plants suffers from the unstable market supply, while chemical synthesis of these compounds is hindered by high energy consumption and pollutant emission. Microbial biosynthesis provides a promising alternative approach for the sustainable supply of limonene and its derivatives. However, low efficiency and specificity of the biosynthetic enzymes and pathways in heterologous hosts make it still challenging for the commercialization of microbial limonene production. On the other hand, the limonene toxicity heavily reduces cellular fitness, which poses a serious challenge for improving limonene titer. Here, we critically review the recent progresses in engineering microbes for limonene biosynthesis and derivation with the emphasis on enzyme characterization and pathway optimization. In particular, we introduce the current trends in microbial limonene decoration for the bio-synthesis of bio-active molecules such as alpha-terpineol and perillyl alcohol. We also discuss the feasible strategies for relieving limonene toxicity and enhancing the robustness of microbial cell factories.

Abstract:Citrus is the largest fruit in the world, and a large number of citrus processing by-products are produced every year. These by-products are rich in limonoids, which have many physiological functions such as anti-tumor, anti-inflammatory and anti-virus, and have been used in food, medicine and agriculture. It is of great theoretical and practical value to develop efficient extraction, separation and purification technology of limonoids, and to develop rapid analysis and detection methods of limonoids. In this article, the structural characteristics and content of limonoids in citrus are introduced, and the extraction, separation, purification and detection techniques of limonoids are emphatically summarized. It is found that emerging technologies such as ultrasonic-assisted and supercritical fluid extraction has better extraction efficiency, and the combination of macroporous resin adsorption, high-speed countercurrent chromatography and preparative high-performance liquid chromatography has better separation effect. In terms of detection technology, liquid chromatography is the most widely used analytical method for limonoids in citrus, which can accurately detect the types and contents of limonoids in citrus. However, efficient, green and rapid pretreatment technology and convenient and high-throughput detection technology need to be developed. This paper provides a reference for the in-depth research and comprehensive development and utilization of limonoids in citrus, and provides a basis for promoting the utilization of citrus by-product resources.

The field of natural product synthesis continuously evolves through the development of methodologies to improve stereoselectivity, yield, scalability, substrate scope, late-stage functionalization, and/or enable novel reactions. One of the more interesting and unique techniques to emerge in the last thirty years is the use of chemoenzymatic reactions in the synthesis of natural products. This review highlights some of the recent examples and progress in the chemoenzymatic synthesis of natural products from 2019–2022.

Abstract:With the implementation of pesticide reduction, plant-derived pesticides have received more and more attention because of their low toxicity and low residue. Limonene is a natural monocyclic terpene widely present in citrus essential oils, which has diverse biological activities, and has potential and application prospect in agricultural disease, pest and weed control. The research progress of limonene and its essential oil in the agricultural fields of insecticidal, acaricidal, phytocidal and antifungal activity in recent years, and the mechanism of its biological activities were briefly reviewed and summarized in this paper. Meanwhile, the registration of limonene formulations in China and the research progress and application of limonene nano-formulations against agricultural diseases and pests were introduced, and the future development trend and research direction of this field were prospected, which will provide the scientific basis for the application of limonene in pesticide reduction and agricultural disease, pest and weed control.

Abstract:Flavonoids are a well-known family of natural polyphenols because of their prevalent properties in the physiological and medicinal field. In addition to a plethora of natural flavonoids, the construction of flavonoid skeletons still situates at the convergence point in the medicinal chemistry. Not surprisingly, amplification in the organic synthetic protocols showcases an expected avenue for accessing to abundant flavonoid scaffolds with special pharmacological activities. Hence, it is necessary to address the recent progresses in the synthesis of flavonoids by using organic strategies, and some typical protocols on the construction of flavonoids are thereby collected from recent publications (from 2020). The synthetic strategies presented herein are mainly cataloged as the cyclization of 4-chromanone, the glycosylation on the flavonoid scaffold, and the application of flavonoids in the pharmacological researches, aiming at providing with a current picture for depicting the recent progress on the synthesis of flavonoids. Therefore, it is expected to be a reference for the further exploration on the designing of synthetic routines for flavonoids.

Abstract:The limonoids have attracted significant attention from the synthetic community owing to their striking structural complexity and medicinal potential. Recent efforts notwithstanding, synthetic access to many intact or ring D-seco limonoids still remains elusive. Here, we report the first de novo synthesis of gedunin, a ring D-seco limonoid with HSP90 inhibitory activity, that proceeds in 13 steps. Two enabling features in our strategy are the application of modern catalytic transformations to set the key quaternary centers in the carbocyclic core and the use of biocatalytic oxidation at C3 to establish a chemical handle to access the A-ring enone motif. The strategy presented herein may provide an entry point to a wider range of oxidized limonoids.

Abstract:A novel series of water-soluble derivatives of limonin were synthesized by introducing various tertiary amines onto the C (7)-position of limonin. Ten target compounds were characterized and screened for their anti-inflammatory and analgesic activity in vivo. Compound 3c exhibited the strongest analgesic and anti-inflammatory activity among the limonin and its derivatives tested; its analgesic activity is more potent than that of aspirin and its anti-inflammatory activity is stronger than that of naproxen.
