Abstract:Thiopetheride antibiotics are a class of ribosomal peptides that are produced by microbial secondary metabolism, rich in sulfur, and highly modified amino acid residues. Thiopeptide antibiotics have a series of very important biological activities, including anti-infection, anti-tumor and immunosuppression, and their mechanism of action targeting ribosomes is different from those commonly used in clinical practice, which makes thiopeptide antibiotics have great development potential, but their poor water solubility and low bioavailability limit their clinical application. In order to improve the physicochemical properties of thiopeptide antibiotics, researchers tried to modified the structure of thiopeptide antibiotics by means of chemosemi-synthesis, combinatorial biosynthesis and precursors directed mutation biosynthesis. The complex structure of thiopeptide antibiotics provides many modification sites for its chemical semi-synthetic modification. In recent years, research on the chemical semi-synthetic modification of thiopeptide antibiotics has developed rapidly. In this paper, the research progress of thiopeptide antibiotic analogues obtained by chemical semi-synthetic modification in recent ten years is reviewed.

Abstract:A synthetic route for the preparation of 2-(benzylthio)-5-[1-(4-fluorobenzyl)-3-phenyl-1H-pyrazol-5-yl]-1,3,4-oxadiazoles derivatives from ethyl 3-phenyl-1H-pyrazole-5-carboxylate has been developed. The configurations of the intermediate 1,3,4-oxadiazole-2-thione and 3-bromobenzylthio substituted 1,3,4-oxadiazole were determined by single crystal X-ray diffraction analysis. The in vitro xanthine oxidase inhibitory activity of these compounds was evaluated.

Abstract:Benzothiazoles have played an important role in the field of biochemistry and medicinal chemistry due to their highly pharmaceutical and biological activity. The development of synthetic processes is undoubtedly one of the most significant problems facing researchers. In this review paper, we provided recent advances in the synthesis of benzothiazole compounds related to green chemistry from condensation of 2-aminobenzenethiol with aldehydes/ketones/acids/acyl chlorides and the cyclization of thioamide or carbon dioxide (CO2) as raw materials, and the future development trend and prospect of the synthesis of benzothiazoles were anticipated.

Abstract:The construction of N-aryl sulfonamides is usually an important reaction in organic synthesis because these compounds are privileged in the structure of many natural products and pharmaceutical molecules. Also, N-aryl sulfonamides are well-known due to their antibacterial, antitumor, and anti-HIV biological activities. Therefore, research on the laboratory synthesis of N-aryl sulfonamides is an important challenge in organic synthesis. In this review, we discussed on methods reported up to now for the synthesis of N-aryl sulfonamides with the goal of stimulating further progresses in this field. We believe that this review will open up a new road for the preparation of pharmaceutically and biologically active sulfonamides.

Abstract:Compounds possessing a thioamide function play a crucial role in organic synthesis, serving as key building blocks. They are also important in the pharmaceutical chemistry and drug design, owing to their ability to mimic the amide function in biomolecules while retaining or developing biological activity. From the synthetic viewpoint, several methods have been developed for preparing thioamides using sulfuration agents. The purpose of this review is to give an update of the last decade of contributions focusing on the formation of thioamides employing different sulfur sources. When appropriate, the cleanness and practicality of the new methods are highlighted.

Abstract:Thietanes are important aliphatic four-membered thiaheterocycles that are found in the pharmaceutical core and structural motifs of some biological compounds. They are also useful intermediates in organic synthesis. Various synthetic methods of thietanes have been developed, including inter- and intramolecular nucleophilic thioetherifications, photochemical [2 + 2] cycloadditions, ring expansions and contractions, nucleophilic cyclizations, and some miscellaneous methods. The recently developed methods provide some new strategies for the efficient preparation of thietanes and their derivatives. This review focuses on the synthetic methods to construct thietane backbones developed during 1966 to 2019.

Abstract:Aromatic azo compounds have -N=N- double bonds as well as a larger π electron conjugation system, which endows aromatic azo compounds with wide applications in the fields of functional materials. The properties of aromatic azo compounds are closely related to the substituents on their aromatic rings. However, traditional synthesis methods, such as the coupling of diazo salts, have a significant limitation with respect to the structural design of aromatic azo compounds. Therefore, many scientists have devoted their efforts to developing new synthetic methods. Moreover, recent advances in the synthesis of aromatic azo compounds have led to improvements in the design and preparation of light-response materials at the molecular level. This review summarizes the important synthetic progress of aromatic azo compounds in recent years, with an emphasis on the pioneering contribution of functional nanomaterials to the field.

Abstract:Pyridinium 1,4-zwitterionic thiolates, as a novel kind of sulfur-containing synthon, have been applied to the synthesis of 12aH-benzo[f]pyrido[1,2-d][1,4]thiazepines and benzo[b]thiophenes. Benzopyridothiazepines were produced through a 1,5-dipolar cycloaddition reaction from pyridinium 1,4-zwitterionic thiolates with arynes, whereas benzothiophenes as side products were generated via a [3 + 2] cascade cyclization reaction. The [5 + 2] reaction mode of pyridinium 1,4-zwitterionic thiolates is disclosed for the first time.

Abstract:Benzo[ b ]thiophene fused compounds with a unique active heterocyclic skeleton have wide applications in the fields of medicinal chemistry, organic synthesis, and organic functional materials, which resulted in rapid development of many efficient methods for the construction of benzo[ b ]thiophene-fused heterocycles in recent years. Among these methods, the domino reaction of benzo[ b ]thiophene derivatives is a practical and powerful synthetic route to access benzo[ b ]thiophene-fused heterocycles by virtue of the particularity of sulfur atom. This review summarizes the latest developments in the construction of benzo[ b ]thiophene-fused heterocycles by ring formation at the C2-C3-position of benzo[ b ]thiophene derivatives in the past decade. Additionally, this review is divided into four parts according to the four kinds of benzo[ b ]thiophene derivatives used, including thioaurone, thioisatin, substituted benzo[ b ]thiophene, and azadiene.

Abstract:Thiochromenes are versatile sulfur-containing heterocyclic compounds that have received considerable interest in drug discovery because of their ability to act as crucial building blocks in synthesizing bioactive compounds. In particular, these scaffolds have found utility in the design of anticancer, anti-HIV, antioxidant, and antimicrobial agents, among others. Despite their pharmacological potential, the synthesis of these scaffolds is less explored in contrast to its oxygen-containing counterpart. The review classifies synthetic processes into Michael addition, cycloaddition, ring-opening, coupling, cyclization, Diels-Alder reactions, and others. Reaction mechanisms, circumstances, and important instances are thoroughly discussed in each area. For instance, chiral catalysts and substrates like mercapto benzaldehyde and cinnamaldehyde are used in Michael addition processes to achieve excellent enantioselectivity. In cycloddition reactions, readily available substrates such as thioisatins and alkynes achieve regioselectivity and product production. Thiochromenes are also synthesized by ring-opening reactions with epoxides or aziridines. These reactions demonstrate the importance of catalysts and solvents in reaction control, particularly palladium catalysts for aryl halides and thiol coupling processes. The other major class discussed is the cyclization reactions with alkynyl thiols and alkynes under regulated temperature and pressure to efficiently synthesize thiochromene. The use of chiral substrates and catalysts, Diels-Alder processes increase yields and selectivity and enhance the variety of thiochromene compounds. This review emphasizes the versatility of thiochromenes in drug discovery and consolidates the existing literature on thiochromenes, scrutinizing the gaps and opportunities for synthesizing novel thiochromene-containing lead molecules.

Abstract:Immediately after the invention of 'Click Chemistry' in 2002, the regioselective 1,2,3- triazole scaffolds resulted from respective organic azides and terminal alkynes under Cu(I) catalysis have been well recognized as the functional heterocyclic core at the centre of modern organic chemistry, medicinal chemistry, and material sciences. This CuAAC reaction has several notable features including excellent regioselectivity, high-to-excellent yields, easy to execute, short reaction time, modular in nature, mild condition, readily available starting materials, etc. Moreover, the resulting regioselective triazoles can serve as amide bond isosteres, a privileged functional group in drug discovery and development. More than hundreds of reviews had been devoted to the 'Click Chemistry' in special reference to 1,4-disubstituted triazoles, while only little efforts were made for an opposite regioisomer i.e., 1,5-disubstituted triazole. Herein, we have presented various classical approaches for an expeditious synthesis of a wide range of biologically relevant 1,5- disubstituted 1,2,3-triazole analogues. The syntheses of such a class of diversly functionalized triazoles have emerged as a crucial investigation in the domain of chemistry and biology. This tutorial review covers the literature assessment on the development of various synthetic protocols for the functionalized 1,5-disubstituted triazoles reported during the last 12 years.

Abstract:NH-1,2,3-Triazoles are a kind of N-containing heterocylic compounds with important biological activities, which have found broad use in medicinal intermediates. Besides, NH-1,2,3-triazoles serve as valuable synthetic intermediates for the preparation of 2-substituded-1,2,3-triazoles through N-alkylation reaction. Because of their potent applications in both synthetic organic chemistry and medicinal chemistry, the progess in the synthesis of NH-1,2,3-triazoles, especially those reported in the past decade, is surveyed according to the research by our group and other groups. Some classical reaction mechanics are also reviewed in this paper.

Abstract:Thietanes are important structural motifs of some biological compounds and useful intermediates in organic synthesis. Various synthetic methods of thietanes have been developed recently with readily available saturated three‐membered heterocycles as starting materials or key intermediates. Both saturated three membered heterocycle‐2‐methyl halides and sulfonates and thiiranes are widely utilized materials. Both nucleophilic and electrophilic ring expansions of thiiranes are efficient methods for synthesis of thietanes. In this Focus Review, we have described various strategies for the synthesis of strained four‐membered thiaheterocyclic thietanes from readily available strained and saturated three‐membered heterocycles.

Abstract:In medicinal chemistry, one of the most significant heterocyclic compounds are quinazolines, possessing broad range of biological properties such as anti-bacterial, anti-fungal, anti-HIV, anti-cancer, anti-inflammatory, and analgesic potencies. Owing to its numerous potential applications, in the past two decades, there is an increase in the importance of designing novel quinazolines, exploring promising routes to synthesize quinazolines, investigating different properties of quinazolines, and seeking for potential applications of quinazolines. The present review article describes synthesis of quinazolines via eco-friendly, mild, atom-efficient, multi-component synthetic strategies reported in the literature. The discussion is divided into different parts as per the key methods involved in the formation of quinazoline skeletons, aiming to provide readers an effective methodology to a better understanding. Consideration has been taken to cover the most recent references. Expectedly, the review will be advantageous in future research for synthesizing quinazolines and developing more promising synthetic approaches.
