Abstract:An efficient protocol was developed to access 3-fluoro-2-hydroxy-2-substituted benzo[b]furans with Selectfluor™ as the fluorinating reagent in MeCN and water. By utilizing SOCl2/Py as the dehydrating agent, the compounds above were readily converted to 3-fluorinated, 2-substituted benzo[b]furans in high yields.

Abstract:A transition‐metal‐free protocol is proposed for the selective oxidation of flavones, providing various sulfoxide/sulfone‐flavonoids in excellent yields. This strategy features excellent selectivities, high yields and broad substrate scope. The combination of oxone (2KHSO5·KHSO4·K2SO4) as the sole oxidant and NaHCO3/Na2CO3 as the buffer play an important role in the process, in which the amounts of oxone and reaction temperatures are the key to the outstanding selectivities and high efficiency of the reaction. It should be noteworthy that a class of novel sulfone‐flavonols is also synthesized through triple oxidation. Moreover, the glycosylation of sulfone‐flavonols has been realized.

Abstract:Linkage chemistry and functional molecules derived from the stereogenic sulfur(VI) centre have important applications in organic synthesis, bioconjugation, drug discovery, agrochemicals and polymeric materials. However, existing approaches for the preparation of optically active S(VI)-centred compounds heavily rely on synthetic chiral S(IV) pools, and the reported linkers of S(VI) lack stereocontrol. A modular assembly method, involving sequential ligand exchange at the S(VI) centre with precise control of enantioselectivity, is appealing but remains elusive. Here we report an asymmetric three-dimensional sulfur(VI) fluoride exchange (3D-SuFEx) reaction based on thionyl tetrafluoride gas (SOF 4 ). A key step involves the chiral ligand-induced enantioselective defluorinative substitution of iminosulfur oxydifluorides using organolithium reagents. The resulting optically active sulfonimidoyl fluorides allow for further stereospecific fluoride-exchange by various nucleophiles, thereby establishing a modular platform for the asymmetric SuFEx ligation and the divergent synthesis of optically active S(VI) functional molecules.

Abstract:An efficient selective oxidation of sulfides to sulfoxides has been developed by means of AlCl3-promoted oxygen transfer from phenyliodine diacetate [PhI(OAc)(2)]. AlCl3 proved to be the optimal Lewis acid for the activation of PhI(OAc)(2). Various substituted sulfides were selectively transformed into the corresponding sulfoxides in good to excellent yields (<= 99%). The high efficiency, excellent functional-group compatibility, broad substrate scope, and mild conditions render the current transformation useful for the synthesis of sulfoxides.

Abstract:Fluorination of 2-aminopyridines and pyridin2(1H)-ones in the presence of Selectfluor, water, and chloroform under mild conditions has been realized. This method gives fluorinated pyridines in good to high yields with high regioselectivities. The electron-deficient pyridine system is activated by an amino or hydroxyl group at C2. The regioselectivity of the fluorination reaction is strongly dependent upon the substituent pattern in the 2-amino pyridine or pyridin-2(1H)-one. The transformation of the 3-fluoro-substituted pyridine derivative into fluorinated zolimidine was achieved as well.

Abstract:Selective oxidation of sulfides to sulfoxides was successfully performed by employing readily available Fe（NO3）3 9H2O as the active catalyst with oxygen as the oxidant in 2,2,2-trifluoroethanol（TFE） without the formation of sulfones.Nitrate anion could play a crucial role in promoting the reaction due to the oxidation capacity under acidic media.High yields of sulfoxides were exclusively obtained from the corresponding sulfides.Furthermore,both aromatic and aliphatic sulfides gave moderate to high yields of sulfoxides with this protocol.

Abstract:Selective S–C bond cleavage of disulfides presents a significant challenge due to the fact that S–S bonds are weaker than S–C bonds. In this study, we present a novel chloride-induced Selectfluor radical cation process for converting readily available symmetrical disulfides into unsymmetrical β-fluorodisulfides through selective S–C bond cleavage. Mechanistic investigations and DFT calculations suggest the involvement of a chlorinated disulfide radical, which subsequently reacts with alkenes to form β-fluorodisulfides via the atom transfer radical addition (ATRA) mechanism. Furthermore, this method exhibits broad functional group tolerance, enabling the synthesis of various target products in moderate to good yields.

Abstract:The fluorohydroxylation of allenyl phosphine oxides with Selectfluor in commercial MeCN without prior treatment or a mixed solvent of anhydrous MeCN (refluxed over CaH2 and distilled before use) and 7.0 equiv of H2O or MeNO2/H2O=10/1 afforded 2-fluoro-3-hydroxy-1(E)-alkenyl diphenyl phosphine oxides in moderate yields with very high regio- and stereoselectivities. The E-stereoselectivity is believed to be controlled by the phosphine oxide functionality. In the reaction of 3-(4-methoxyphenyl)-1,2-propadienyl diphenyl phosphine oxide, further fluorination on the electron-rich phenyl ring was also observed.

Abstract:The efficient and selective photocatalytic conversion of chemicals with visible light and naturally abundant resources has long been desired, but this requires finely designed sensitizers that are capable of converting light into chemical energy for the required energy/electron transfer, bond formation and scission, etc. Inspired by flavin (FL) based enzymes that are capable of initiating many redox reactions in biological systems in visible light, FL and naphthalimide chromophores were fused together as a heavy-atom-free triplet photosensitizer (NI-FL). It is expected that the extended conjugation within NI-FL may benefit absorption in the visible light range, promoting the intersystem crossing to triplet excited states for efficient chemoselective conversions. The absorption and emission maximum of NI-FL are redshifted by ∼40 nm and the absorption is more than doubled (1.53 × 104 M-1 cm-1 at 484 nm) with respect to FL (7.5 × 103 M-1 cm-1 at 439 nm), and a long-lived triplet excited state of intramolecular electron transfer nature was captured (τT = 45.3 μs). The performances of FL and NI-FL in the photooxidation of sulfides in air were also examined. Apart from nearly quantitative selectivity for sulfoxide, NI-FL demonstrates a 0.5-5 fold enhanced performance with respect to FL and the conversion proceeds through radical intermediates formed by electron transfer at excited states with substrates. Mechanistic investigation satisfactorily explained the observed photophysical properties and the dominant role of radical intermediates in the NI-FL catalyzed photooxidation. The findings may help to understand photooxidation sensitized by FL derivatives and benefit the design of novel efficient photosensitizers.

Abstract:A sulfur redox process has been developed between sulfinate and thiosulfate, which efficiently affords diverse unsymmetrical disulfides and provides a new method to modify pharmaceuticals and natural products without requiring an extra oxidant or reductant. Gram-scale investigation further demonstrates the practicality and application potential of this process. Isolated key intermediates and a series of control experiments afford an unusual process, which reveals the mechanism of comproportionation and the transition-metal-free sulfur redox process.

Abstract:The development of efficient approaches to access sulfonyl fluorides is of great significance because of the widespread applications of these structural motifs in many areas, among which the emerging sulfur(vi) fluoride exchange (SuFEx) click chemistry is the most prominent. Here, we report the first three-component aminofluorosulfonylation of unactivated olefins by merging photoredox-catalyzed proton-coupled electron transfer (PCET) activation with radical relay processes. Various aliphatic sulfonyl fluorides featuring a privileged 5-membered heterocyclic core have been efficiently afforded under mild conditions with good functional group tolerance. The synthetic potential of the sulfonyl fluoride products has been examined by diverse transformations including SuFEx reactions and transition metal-catalyzed cross-coupling reactions. Mechanistic studies demonstrate that amidyl radicals, alkyl radicals and sulfonyl radicals are involved in this difunctionalization transformation.

Abstract:A systematic investigation has been conducted into the mechanism for the persulfate-promoted benzylic mono-and difluorination with Selectfluor by using density functional theory (DFT). In the monofluorination, the role of SO4 center dot- (from the substrate-assisted cleavage of S2O82- in the initiation phase) is just to trigger fluorination by producing the initial benzyl radical. The cationic N-radical from the fluorine transfer between the benzyl radical and Selectfluor can abstract hydrogen from the substrate, generating the benzyl radical to complete the reaction cycle. The difluorination contains the monofluorination and the second fluorine transfer. The benzylic C-H activation in the monofluorination product can be achieved by cationic N-radical or SO4 center dot-. Whether in monofluorination or in difluorination, the rate-determining step is the generation of SO4 center dot-. (C) 2017 Elsevier B.V. All rights reserved.

Abstract:A metal‐free direct trifluoromethylthiolation of sulfonium ylides with an electrophilic trifluoromethylthiolating reagent has been established, in which sulfonium salt or α‐bromoacetic ester is employed as sulfonium ylide precursors. This trifluoromethylthiolation enables the straightforward construction of SCF3‐substituted sulfonium ylides from a wide range of substrates, including ketones, esters, and even PEGylated substrates. Moreover, the application of this approach in large‐scale preparation and the fluorescence and fluorine‐19 magnetic resonance imaging capabilities of the product are also explored.
