Abstract:(S)-6-Nitroindoline-2-carboxylic acid, a substructure occurring in numerous biologically active natural products, was synthesized with moderate yield (53%) and high enantiomeric excess (> 99.5%) starting from the nitration of L-phenylalanine, which is a commercially available chiral pool compound, followed by successively bromination and intramolecular cyclization. The route was carried out in gram quantities and it is suitable for industrial application due to its convenient reaction conditions and low cost.

Abstract:(S)-Indoline-2-carboxylic acid was synthesized by use of a nitro amination approach with l-phenylalanine as chiral pool. The first step of the synthesis was nitration of l-phenylalanine, with urea nitrate (UN)/H2SO4 as nitrating reagent, to give 2,4-dinitro-l-phenylalanine in 75.7 % yield in one-pot synthesis and 69.1 % yield by step-wise nitration. Intramolecular nitro amination of 2,4-dinitro-l-phenylalanine gave (S)-6-nitro-indoline-2-carboxylic acid in 65.7 % yield and more than 99.5 % enantiomeric excess (ee). The title compound, (S)-indoline-2-carboxylic acid, was obtained in 85.9 % yield and high ee by one-pot transformation of (S)-6-nitroindoline-2-carboxylic acid. The total synthesis consisted of three operations and gave the title compound in 42 % yield and more than 99.5 % ee.

Abstract:Highly enantioselective synthesis of 1,3-bis(hydroxymethyl)-2-oxindoles was established through chiral Nd-III-N,N'-dioxide in up to 93% yield and >99% cc under mild conditions. The key features of this approach include using cheap and readily available formalin as a hydroxymethylation Cl unit and unprotected 3-substituted-2-oxindoles as nucleophiles directly. The initial C-aldol product from 3-substituted-2-oxindole was converted to the corresponding 1,3-bis(hydroxymethyl)-2-oxindole derivative immediately under the reaction conditions. Moreover, the catalyst was water-tolerant, and ligand and Nd(OTf)(3) could be easily recovered and reused with no loss in catalytic activity and enantioselectivity. The synthetic utility of this methodology was accomplished with the synthesis of key intermediates of physostigmine and coerulescine.

Abstract:A general synthetic route of S-(-)-2-Carboxyindoline was proposed which started from o-nitrotoluene and diethyl oxalate, via condensation, nitro-group reduction, heterocyclic reduction and hydrolysis, 2-carboxyindoline was achieved with total yield of 40.6%. Especially, using 10% Pd-C as catalysts, the heterocycle is reduced easily with hydrogen at 160 ℃ and 6.2 MPa, the catalysts also can be reclaimed. Through steps of chlorination, esterification, fractional crystallization, and hydrolysis, the S-(-)-2-carboxyindoline was resoluted with L-carnitine oxalate as the chiral resolution reagent. The titled product can be achieved with 32% yield and ??α 52809 = ?27.5?? (c =1.0, DMSO).

Abstract:Indole-2-carboxylic acid and indoline-2-carboxylic acid are common building block in natural products with bioactivities and anti-hypertension pharmaceuticals. The syntheses and transformation from each other were reviewed. Indoline-2-carboxylic acid can be got from the reduction of indole-2-carboxylic acid and the S enantiomer can be achieved from the resolution of the racemic mixture with chemical reagents or enzymes. The chiral pool synthesis, emerged recently, will play an important role in the future. Indole-2-carboxylic acid can be formed from Fischer way or substituted anilines and benzaldehydes.

Abstract:Chiral sulfide is an important fine chemical product. The intermediate product L-2-chlorosuccinic acid was prepared from L-aspartate via diazotization and chlorination. Thus,(+)-2R,2'S-thiodisuccinic acid was prepared with the total yield of 40.1%. Its structure was characterized by 1H-NMR,[α20D] =+27.3 (c=1.0,DMSO) and the optical purity was 99.3%. A novel synthesis of chiral sulfide was introduced,in which natural amino acid was applied as chiral pool.

Abstract:A procedure for synthesis of 2-methoxyphenothiazine (1) has been developed, starting from resorcinol and aniline by condensation, following methylation and cyclization. p-Toluenesulfonic acid and polysubstituted aromatics were employed as the catalyst of condensation and the solvent of cyclization, respectively, to improve the yield. The use of parallel experiments, statistical experimental design, and multivariate modeling made the total yield of the procedure as high as 74.2%. J. Heterocyclic Chem., (2012).

Abstract:Chiral N-alkylated indoles are privileged structural motifs found in many natural products and pharmaceuticals and a search for effective methods with which to prepare chiral N-alkylated indoles is ongoing. Efficient syntheses of enantioenriched N-alkylated indoles has been developed but not with standing these advances, the development of a facile method to prepare chiral N-alkylated indoles from simple and available starting materials under mild conditions is still needed. In this paper, we report a nitrogen replacement process driven by dearomatization that directly incorporates natural a-amino acids into 2-alkynylanilines leading to the formation of a variety of multi-functional chiral N-alkylated indoles.

Abstract:In the past two decades, substantial advances have been made on the asymmetric alkyne functionalization by the activation of inert alkynes. However, these asymmetric transformations have so far been mostly limited to transition metal catalysis, and chiral Bronsted acid-catalyzed examples are rarely explored. Here, we report a chiral Bronsted acid-catalyzed dearomatization reaction of phenol- and indole-tethered homopropargyl amines, allowing the practical and atom-economical synthesis of a diverse array of valuable fused polycyclic enones and indolines bearing a chiral quaternary carbon stereocenter and two contiguous stereogenic centers in moderate to good yields with excellent diastereoselectivities and generally excellent enantioselectivities (up to >99% enantiomeric excess). This protocol demonstrates Bronsted acid-catalyzed asymmetric dearomatizations via vinylidene-quinone methides.

Abstract:A chiral SPINOL derived phosphoric acid-catalyzed asymmetric N-alkylation reaction of indoles with cyclic α-diaryl-substituted N-acyl imines, which are generated in situ from 3-aryl 3-hydroxyisoindolinones, has been demonstrated. The transformation proceeds smoothly with a broad range of indoles and isoindolinone alcohols. A variety of indole derived N-alkylated tetrasubstituted chiral aminals were afforded in moderate to good yields (50-79%) with excellent enantioselectivities (up to 98% ee).

Abstract:An efficient method for the synthesis of five-membered chiral propargylic amines from 2-aryl-3H-indol-3-one and alkynylsilanes has been developed. The reaction proceeded under the catalytic system of PtCl(4), oxazoline-based ligand L11, Zn(CF(3)COO)(2), and AcOH in DCE at 95 °C via in situ desilylation of TMS-alkynes. This methodology also highlights a new protocol for the in situ desilylation of alkynylsilanes. The reaction showed a broad substrate scope with good yields and enantioselectivity.

Abstract:The Mannich reaction between chiral N-phosphonyl imines and a Ni(II)-complexed alanine Schiff base (Ala-Ni) is reported. With a chiral phosphonyl auxiliary, a single isomer of alpha-methyl-alpha,beta-diamino acid derivative containing vicinal chiral centers, including a chiral quaternary carbon center, can be obtained simply by washing the crude mixture with cosolvents. The absolute stereochemistry of the enantiomerically pure product has been unambiguously determined by X-ray crystallographic analysis.

Abstract:An efficient total synthesis of (2S,2'S,3aR,3'aR,8aR,8a R)-6,6'-dichloro-3a,3'a-dihydroxy-1,1',2,2', 3,3a,3',3'a,8,8a,8',8'a-dodecahydro-5,5'-bipyrrol[2,3-b]indole-2,2'-dica rboxylic acid, the central amino acid component of chloptosin (1), is described. Starting from m-chloronitrobenzene, this C-2-symmetrical amino acid was synthesized by using a 14-step route, in which a Zinin benzidine rearrangement, intramolecular Heck reaction, and selenocyclization and oxidative deselenation served as key steps. The absolute stereochemistry of the target was confirmed by X-ray single-crystal studies on a key intermediate (17).
Diastereoselective synthesis of chiral 3-substituted isoindolinones via rhodium(III)-catalyzed oxidative C-H olefination/annulation

Abstract:A new method for the direct and stereoselective synthesis of 3-substituted isoindolinones via Rh(iii)-catalyzed chiral N-sulfinyl amide directed asymmetric [4 + 1] annulation of benzamides with acrylic esters has been developed. The reaction proceeded through an oxidative C-H olefination and a subsequent cyclization by intramolecular aza-Michael addition, producing a series of diastereoisomeric chiral isoindolinones (20 examples) in generally good yields with a dr value up to 5.5 : 1. The absolute configurations of the newly formed C-stereocenters in the major and minor diastereomers of the catalysis product have been determined by X-ray crystal diffraction analysis to be S and R, respectively. The separation of the major diastereoisomers from the catalysis products and subsequent removal of the N-sulfinyl chiral auxiliary afforded enantiomerically pure (S)-isoindolinones. The application of the obtained (S)-isoindolinones in the synthesis of several biologically active isoindolinones such as (S)-PD172938, (S)-pazinaclone and (S)-pagoclone is presented.

Abstract:The first catalytic enantioselective dearomatization/rearomatization of 2-nitroindoles triggered by the Michael addition of 3-monosubstituted oxindoles was established. A wide range of 3-indolyl-3′-alkyloxindoles (up to 99% yield, 97% ee) and 3-indolyl-3′-aryloxindoles (up to 95% yield, 99% ee) were obtained by using an organocatalyst. This method provides an unprecedented strategy to access structurally diverse 3,3′-disubstituted oxindoles bearing a sterically congested triaryl-containing all-carbon quaternary stereocenter.
