Abstract:Beauveriolides, including the main beauveriolide I {systematic name: (3 R ,6 S ,9 S ,13 S )-9-benzyl-13-[(2 S )-hexan-2-yl]-6-methyl-3-(2-methylpropyl)-1-oxa-4,7,10-triazacyclotridecane-2,5,8,11-tetrone, C 27 H 41 N 3 O 5 }, are a series of cyclodepsipeptides that have shown promising results in the treatment of Alzheimer's disease and in the prevention of foam cell formation in atherosclerosis. Their crystal structure studies have been difficult due to their tiny crystal size and fibre-like morphology, until now. Recent developments in 3D electron diffraction methodology have made it possible to accurately study the crystal structures of submicron crystals by overcoming the problems of beam sensitivity and dynamical scattering. In this study, the absolute structure of beauveriolide I was determined by 3D electron diffraction. The cyclodepsipeptide crystallizes in the space group I 2 with lattice parameters a = 40.2744 (4), b = 5.0976 (5), c = 27.698 (4) Å and β = 105.729 (6)°. After dynamical refinement, its absolute structure was determined by comparing the R factors and calculating the z-scores of the two possible enantiomorphs of beauveriolide I.

Abstract:Chemical investigation of the EtOAc extract of an insect-associated fungus Letendraea sp. 5XNZ4-2 cultured in Potato Dextrose Broth (1/2 PDB) medium lead to the isolation of three new polyketides, named letendronol D (1), phomopsiketones H-I (2-3). The structures of new compounds were elucidated by the analysis of HRESIMS and NMR spectroscopic data, and the absolute configurations were determined by modified Mosher’s method, ECD calculation and single-crystal X-ray diffraction. Cytotoxicity and antibacterial activities of 1 were assayed and regrettably 1 didn’t display any cytotoxicity and antibacterial activity. 3 was the first phomopsiketone derivative obtaining the lactone.

Abstract:Total synthesis of cyclodecapeptide antibiotics from Bacillus laterosporus, laterocidin and its analogues. was accomplished for the first time by solid-phase peptide synthesis followed by traceless on-resin cyclization of the linear precursors with protection of alpha-carboxyl group oil the Asp residue by Dmab as a temporary blocking group for on-resin head-to-tail cyclization, in which the carboxyl group of side chain of Asp was linked to Rink resin. Single alanine substitution or Asn substitution (Asp(10)-> Asn(10)) demonstrated improvements in antibacterial activity of note, D-Phe(2) and Pro(4) play an important role in on-resin head-to-tail cyclization and exerting antibacterial activity of Laterocidin. Abstract:Two novel 4-deacetyl and 2,4-deacyl 1-deoxybaccatin VI derivatives were prepared from 1-deoxybaccatin VI. Their structures were elucidated using extensive spectroscopic techniques (IR, 1HNMR, DEPT, COSY, HMQC and MS). In addition, the conformations for these compounds were obtained by X-ray diffraction methods. Comparisons to 1-deoxybaccatin VI and paclitaxel identify relative differences principally in the cyclohexenyl ring (A ring). Conformational analysis showed that removal of 4-acetyl or 2,4-diacyl moiety has a decisive influence on the conformation of the A ring.

Abstract:Two novel spiroketals, named as letenketals A and B, had been isolated from the cultural broth of marine fungal Letendraea sp. Two compounds were stereoisomers and contained the new carbon skeleton including benzodihydropyran ring and one tetrahydrofuran ring, which connected as spiro-fused ring system through one ketal carbon. Their relative and absolute configurations were determined by the 2D-NMR analysis together with single-crystal X-ray diffraction. The antibacterial and cytotoxic activities of letenketals were tested.

Abstract:In order to purify enough material for establishing the absolute stereochemistry of the new antifungal metabolite 3,6,8-trihydroxy-3-[3,5-dimethyl-2-oxo-3(E)-heptenyl]-2,3-diliydronaphthalen-1(4H)-one produced by Keissleriella sp., a marine filamentous fungus (strain number: YS 4108) [1], a repeated growth and fractionation of the fungal culture was performed to give instead a new antimicrobial metabolite, keisslone (1), the structure of which was elucidated on the basis of spectral analyses including homo- and hetero-nuclear correlation NMR experiments (HMQC, COSY, NOESY and HMBC). The absolute configuration of metabolite 1 was determined mainly by its CD data and NOESY spectrum. The compound 1 was shown to be inhibitory against the human pathogenic fungi Candida albicans, Trichophyton rubrum and Aspergillus niger with MICs of 50, 70, 40 mug/mL, respectively.

Abstract:Lovastatin and its derivatives are well-known as a class of lipid-lowering drugs in the medicinal chemistry of natural products, and they typically feature a rigid decalin core structure with almost no other carbon rings. Herein, aculeatones A ( 1 ) and B ( 2 ), two highly modified lovastatin derivatives with an unreported 6/6/3-tricyclic carbon skeleton, in addition to four undescribed lovastatin derivatives aculeatones C–F ( 3–6 ), were isolated from the fungus Aspergillus aculeatus . Their structures with absolute configurations were characterized by comprehensive spectroscopic, quantum chemical calculation, electronic circular dichroism (ECD), modified Mosher's method, and single-crystal X-ray diffraction studies. The plausible biosynthetic pathway for 1 and 2 was proposed, and the biomimetic formation of 1 was achieved starting from 3 . Biologically, compounds 3 , 5 , and 6 significantly inhibited the accumulation of lipid in an oleic acid-treated HepG2 cell model.

Abstract:Azalomycins F5a (1), F4a (2) and F3a (3) are three 36-membered polyhydroxyl macrolides with remarkable antifungal activity. By comparing the NMR spectral data of 1 and its degradation products with those of several universal NMR databases and identical fragments of known compounds, the relative configuration of 1 was assigned for the first time except that the stereochemistry of stereoclusters C6–C11 to C14–C36 was undefined. Moreover, the relative configurations of 2 and 3 were respectively established by comparing their NMR spectral data with those of 1.

Abstract:Synthesis and crystal structures of 1-deoxybaccatin VI derivatives, and the influence of C(9), C(10), and C(13) substituents of the tetracyclic moiety on molecular conformations were investigated. Comparison of conformations of 1-deoxybaccatin VI derivatives reveals subtle conformational variations in the tetracyclic ring system due to deacetylation at C(10) and C(13), and 9,10-acetonide. Additionally, C(2)-benzoyl group exhibits an unexpected conformational flexibility in 1-deoxybaccatin VI derivatives, which may be relevant to the bioactivity of taxanes.

Abstract:Comprehensive Summary Chemical investigation of Beauveria felina , a marine‐derived entomopathogenic fungus, obtained two new destruxin hexadepsipeptides (1 and 2) containing an L ‐Ala 5 residue instead of the usual N ‐Me‐ L ‐Ala 5 . Interestingly, the presence of the L ‐Ala 5 residue enabled two slowly‐exchanging solution conformers with cis / trans (ILE 3 )‐( N ‐Me‐Val 4 ) amide bond for 1 and 2 with 1.5 : 1 and 1 : 1 cis / trans ratios, respectively, while only the conformer with cis amide bond was found in the solid‐state. The solution and solid‐state conformers were studied by NMR, X‐ray diffraction and solution and solid‐state ECD measurements, as well as by TDDFT‐ECD calculations. Structural (conformational) parameter‐activity correlations suggested that the extended destruxin geometry with the preferred cis (ILE 3 )‐( N ‐Me‐Val 4 ) bond probably resulted in improved bioactivity and reduced toxicity for the destruxin class. This was the first report on the interconversion phenomenon of cis / trans amide bonds of destruxin class, which could give insights in the development of bioactive destruxin agents with lower toxicity.

Abstract:The first total synthesis of the cyclic depsipeptide antibiotic LL-A03411-beta1 (LL) is described. The configuration of the -methyltryptophan (-MeTrp) residue was established by preparing all four stereoisomers of Fmoc--MeTrp which were used for the synthesis of LL via Fmoc solid phase peptide synthesis. The most active of the four peptides was the one containing (2R,3R)--MeTrp. The activity of LL was strongly inhibited by cardiolipin (CL), but not other common phos-pholipids found in bacterial cell membranes suggesting that LL interacts with CL in the lipid membrane

Abstract:Aspernigrin A was reisolated as a secondary metabolite of the Cynodon dactylon-associated endophytic fungus Cladosporium herbarum IFB-E002 coproducing rubrofusarin B, fonsecinone A, 7-hydroxy-4-methoxy-5-methylcoumarin, orlandin, kotanin, and 3beta,5alpha,6beta-trihydroxyergosta-7,22-diene. The structure of aspernigrin A, previously elucidated to be 4-benzyl-6-oxo-1,6-dihydropyridine-3-carboxamide (1), was revised as 6-benzyl-4-oxo-1,4-dihydropyridine-3-carboxamide (2) on the basis of its additional NMR spectroscopic data and the X-ray crystallographic analysis.

Abstract:In order to study the molecular structure of Lentinan and its structural character after sulfation modification,the Lentinan with 0.34 million dolton average molecular weight was obtained through the processes involve water extraction,ethanol precipitation,sevage method deprotein and Sephadex G-200 gel column chromatography from the sporophore of Lentinus edodes. Lentinans were sulfated by the means of Wolfrom,sulfuric acid and triethylamine-sulfur trioxide,respectively,and the Lentinan sulfates were determined by Infrared Spectrometry. The results showed that Wolfrom method which has higher proportion of sulfate substitution and yield is a ideal way. The 13C-NMR data showed that the sulfation were located on the carbon-6 in sulfated derivative of lentinan

Abstract:Comprehensive SummaryA pair of alkaloid enantiomers possessing a novel 1‐oxaspiro[4.4]non‐3‐ene‐2,7‐dione skeleton, trichodermotin A (1), was obtained from the fungus Trichoderma asperellum. Spectroscopic data, X‐ray diffraction, and ECD calculations were used to establish its structure and absolute configuration. (−)‐1 showed significant α‐glucosidase inhibitory activity (IC50 = 10.1 μmol/L vs. 60.1 μmol/L of positive control). A plausible biosynthetic pathway originating from L‐β‐phenylalanine was proposed, and a facile total synthesis was further accomplished. The key reaction of our synthetic strategy was a domino aza‐Michael/lactonization in one pot, leading to the pivotal 4‐amino‐oxaspiro[4.4]octane scaffold.

Abstract:The metabolites of the predacious fungus Dactylaria lutea ROUTIEN include the anthraquinone macrosporin (2) and three hydroxylated 1,2,3,4-tetrahydro derivatives of this anthraquinone, altersolanol A (5), altersolanol B (4) and dactylariol (6). The structure and relative configuration of dactylariol are established from spectroscopic studies, and its absolute configuration is proposed as 1R, 2R, 3R by virtue of its co-occurrence with altersolanol B. Dactylarin, suggested by other authors to have the structure (1), is shown to be identical with altersolanol B (4).

Abstract:In this research, twenty aromatic and branched aliphatic polyketides, including seven previously undescribed butenolide derivatives, piterriones A–G and one known analogue, along with twelve known altenusin derivatives, were isolated from the isopod-associated fungus Pidoplitchkoviella terricola. Their structures were elucidated by analysis of NMR (1D and 2D) and mass spectrometry data, and their absolute configurations were determined by Mosher's method, microscale derivatization, and comparison of their specific rotations and ECD spectra. Dihydroaltenuene B exhibited mushroom tyrosinase inhibitory activity with an IC50 value of 38.33 ± 1.59 μM, which was comparable to that of the positive control, kojic acid (IC50 = 39.72 ± 1.34 μM). A molecular-docking study disclosed the hydrogen bonding interactions between the 3-OH and 4′-OH of dihydroaltenuene B and the His244, Met280 and Gly281 residues of tyrosinase.

Abstract:Chemical investigation of the endophytic fungus Talaromyces wortmannii, isolated from Aloe vera, yielded two new cyclic peptides, talaromins A and B. Their structures were established on the basis of extensive NMR spectroscopic and mass spectrometric analysis. Both cyclopeptides contain ring systems comprised of six α-amino acid residues connected to a β-amino acid. The absolute configurations of the α-amino acids were determined by Marfey’s method.
