Don) Makino (Orchidaceae). Their structures were elucidated on the basis of extensive spectroscopic analyses. All the compounds obtained were tested in vitro for cytotoxic activities against colon (HCT-116), cervix (Hela), and breast (MDA-MB-231) cancer cell lines. They all showed moderate or weak cytotoxicities to the above cancer cell lines.

Abstract:Five new benzylphenanthrenes, cremaphenanthrenes L-P (1-5) were isolated from the EtOAc-soluble extract of the tubers of Cremastra appendiculata. Their structures were elucidated based on the spectroscopic data and chemical methods. Compounds 1-5 were tested in vitro for their cytotoxic effects against colon (HCT-116), cervix (Hela), and breast (MCF-7 and MDA-MB-231) cancer cell lines. Compound 1 showed moderate cytotoxic activities against HCT-116, MCF-7, and MDA-MB-231 cancer cell lines with IC50 values ranging from 15.84 to 24.18 mu M and weak cytotoxicity to Hela cell line with IC50 value of 68.81 mu M. Abstract:Eleven previously unknown compounds and 23 known compounds, including 20 phenanthrene or 9,10-dihydrophenanthrene derivatives, five bibenzyls, seven malate or tartrate benzyl ester glucosides, adenosine and gastrodin were isolated from tubers of Cremastra appendiculata. Among the obtained compounds, two are the first isolated dimers with one phenanthrene or bibenzyl unit connected to C-3 of 2,3,4,5-tetrahydro-phenanthro[2,1-b]furan moiety. In addition, 33 of these compounds were evaluated in vitro for their cytotoxic activity against two cancer cell lines. Among the compounds examined, one compound showed moderate cytotoxic activity, while five showed weak cytotoxic activity against the A549 cell line.

Abstract:Nineteen new phenanthrene-based tylophorine analogues with various functional groups on the piperidine moiety were designed, synthesized, and evaluated for in vitro anticancer activity against four human tumor cell lines. Analogues 15 and 21 showed approximately 2-fold enhanced inhibitory activity as compared with our prior lead compound (PBT-1). Analogues 23 and 24 with S- and R-configured substituents, respectively, at the piperidine 3'-position exhibited comparable cytotoxicity to that of PBT-1. Furthermore, mechanistic studies to investigate the effects of the new compounds on Akt protein in lung cancer cells and the NF-kB signaling pathway suggested that the compounds may exert their inhibitory activity on tumor cells through inhibition of activation of both Akt and NF-kB signaling pathway.

Abstract:Six new anthracyclines (1-6) were obtained by the investigation of the solid fermented extract of a marine-derived Streptomyces sp. NB-A13. The structures were elucidated on the basis of spectroscopic analysis, including 1D and 2D NMR, HRESIMS, UV and IR. The absolute configurations of compounds 1-5 were established by calculated electronic circular dichroism (ECD) using time dependent density functional theory. Among the isolates, four compounds (2, 3, 5 and 6) showed moderate cytotoxic activity against PC3 cells (prostate cancer cells) with IC50 values of 7.0 +/- 0.3 mu M, 0.92 +/- 0.05 mu M, 2.1 +/- 0.7 mu M and 9.8 +/- 0.7 mu M, respectively.

Abstract:Phytochemical investigation on the nonpolar extracts of P. acidus leaves afforded three new cleistanthane diterpenoids, named phyllanacidins A–C (1–3), in addition to three known ones (4–6). Among them, compounds 2 and 3 represent the first examples of 17-nor cleistanthane diterpenoids. Their structures including absolute configurations were elucidated on the basis of extensive spectroscopic data and single crystal X-ray diffraction analysis. All the isolated cleistanthane diterpenoids were evaluated for their cytotoxic activities against five human cancer cell lines. Compounds 4–6 exhibited moderate cytotoxicity with IC50 values ranging from 16.11 to 30.82 μM.

Abstract:Abstract Encouraged by the in vitro potent inhibitory activity on butyrylcholinesterase (BChE) of 95% ethanol extract of Cremastra appendiculata (D. Don) Makino tubers, a further phytochemical investigation on C. appendiculata tubers was conducted, which led to the isolation of a pair of new biphenanthrene atropisomers, namely cremaphenanthrene F-G (1–2). Their structures were elucidated on the basis of extensive spectroscopic analyses and chemical method. It is the first time that biphenanthrene atropisomers have been isolated from the plant kingdom. Compound 1 showed potent BChE inhibitory effect with IC50 value of 14.62 ± 2.15 μM. Compound 2 exhibited weak BChE inhibitory effect with IC50 value of 79.56 ± 0.78 μM. Meanwhile, 1 and 2 were found to be inactive for acetylcholinesterase (AChE) inhibition. These findings suggested that compound 1 was a promising selective BChE inhibitor for AD prevention and treatment. Graphical Abstract

Abstract:Three new dihydrophenanthrenes, retusiusine D (1), retusiusine E (2), retusiusine F (3), and a new phenanthrene retusiusine G (4), together with two known dihydrophenanthrenes 4,7-dihydroxy-2,3-methylenedioxy-9,10-dihydrophenanthrene (5) and epemeranthol-A (6) were isolated from the tubers of Bulbophyllum retusiusculum. Their structures were established on the basis of extensive spectroscopic analyses. Compounds 1 and 2 exhibited potent cytotoxic activities against SMMC-7721 and weak cytotoxic activities against HL-60. Compound 4 showed moderate cytotoxic activity against SMMC-7721 and MCF-7.

Abstract:Five undescribed abietane diterpenoids, along with eight known analogs, were isolated from Phlegmariurus carinatus. Their structures were unambiguously elucidated by extensive analysis of spectroscopic data and comparison between the literature. The absolute configuration of phlecarinatone C was determined by evaluating ECD spectra. Four undescribed abietane diterpenoids and eight known analogs were tested for their neuroprotective and cytotoxic activities, separately. Teuvincenone C showed potential neuroprotective effect against Hemin-induced HT22 cell damage. Importantly, phlecarinatone C showed pronounced cytotoxic effect against U251 cells in vitro assays. The biological evaluation revealed that phlecarinatone C could inhibit proliferation, migration, and invasion in a concentration-dependent manner of U251 cells. Meanwhile, phlecarinatone C effectively reversed epithelial-to-mesenchymal transition (EMT) and promoted U251 cells apoptosis via a mitochondrial apoptotic pathway. Taken together, phlecarinatone C might be a valuable candidate for anti-metastatic agents against glioblastoma treatment.

Abstract:Four pairs of novel enantiomeric phenanthrenes, named (+)/(−)-blephenanthropyran A [(+)/(−)- 1 ], (+)/(−)-blephenanthropyran B [(+)/(−)- 2 ], (+)/(−)-blephenanthrofuran A [(+)/(−)- 3 ], and (+)/(−)-blephenanthrofuran B [(+)/(−)- 4 ], were isolated from the tubers of Bletilla striata (Thunb.) Reichb.f. Their structures and absolute configurations were elucidated by extensive spectroscopic analysis and electronic circular dichroism calculations. Compounds (+)/(−)- 1 and (+)/(−)- 2 are 9,10,11,12-tetrahydrophenanthropyrones, while (+)/(−)- 3 and (+)/(−)- 4 are 9,10,11,12-tetrahydrophenanthrofurans. Since B. striata is known as "an excellent medicine for lung diseases" in traditional Chinese medicine, the effects of the isolates in A549 lung cancer cells were investigated. The results showed that compound (+)- 3 significantly inhibited the viability of A549 cells. Furthermore, (+)- 3 could promote the apoptosis of A549 cells by upregulating the protein expression of caspase3, cleaved caspase3, cleaved PARP, and Bax. Thus, compound (+)- 3 is an effective proliferation inhibitor and apoptosis inducer in lung cancer cells.

Abstract:Chemical exploration of Artemisia myriantha Wall. ex Bess (Asteraceae) aerial parts afforded one new seco-guaianolide (1) and one new germacranolide (2). Structure elucidation of the new compounds was unambiguously performed based on HRESIMS, 1D and 2D NMR spectroscopic analyses. The two new compounds exhibited in vitro strong cytotoxic activities against human cancer cell lines HCT-8 with IC50 values of 0.76 and 0.28 μM, respectively.

Abstract:Eleven triterpenoids were isolated from the fruits of Phellodendron chinense Schneid, and their structures were determined by spectroscopic analysis. The results show that four new tirucallane-type triterpenoids 1, 2, 5, and 6 and seven known compounds 3, 4, 7, 8, 9, 10, and 11 were isolated. Structurally, compound 6 was uncommon; it has a chlorine atom instead of a methyl group at the C-20 position. The cytotoxicities of the compounds was evaluated against the in vitro proliferation of four human tumor cell lines HEL, K562, MDA, and PC3 using adriamycin as the positive control. Compound 1 showed a similar cytotoxicity as the positive control; compounds 3 and 10 showed moderate cytotoxicities compared to the control (P<0.05). This indicates that these compounds have great potential for the development of new antitumor drugs.

Abstract:series of 2-phenoxy-N-phenylacetamides were designed and synthesized with a view to develop novel anticancer agents. All compounds were prepared with varied substitutions in the phenyl ring of aniline and evaluated for their anticancer activity. The majority of them displayed a moderate degree of cytotoxicity against several human cancer cell lines. Six compounds displayed potent cell growth inhibitory activity with IC 50 values below 20 μM. Two compounds ( TC-5 and TC-14 ) exhibited the most pronounced antiproliferative activity equivalent to or higher than that of positive control. The structure – activity relationship of these anticancer agents has been reviewed. The results indicate that incorporation of halogen into the benzene ring is beneficial for anticancer activity.

The in vitro cytotoxic activities of compounds 1 and 2 were studied against breast carcinoma MDA-MB-231, hepatocellular carcinoma HepG2 and lung carcinoma A549 cell lines. And compounds 1 and 2 exhibited anti-proliferative effects against these least one of the three types of cell lines with IC50 ranging from 12.57 ± 2.34 to 31.22 ± 1.36 μM.Graphical

Abstract:Two new dinor-eudesmane sesquiterpenoids, named multistalin A (1), and multistalin B (2), together with three sesquiterpene glycosides (3–5), and a norlabdane-type diterpene (6) were isolated from the root extract of Chloranthus multistachys Pei. Their structures were elucidated on the basis of spectroscopic analysis including 1D, 2D NMR techniques and HR-ESI-MS. In addition, the cytotoxicity activities of the isolated compounds against selected cancer cells (Hela and A-549) were evaluated by MTT assay.

Abstract:Six new cadinane-type sesquiterpenoids, named Chimnitensin A-F (1–6) were isolated from the leaves of Chimonanthus nitens Oliv. Their structures were elucidated by comprehensive spectroscopic analyses and comparison with structurally related known analogues. In vitro MTT assay showed that all six compounds had cytotoxicity against two selected human breast cancer cell lines (MDA-MB-468 and MDA-MB-231), which indicate their potential of developing into anticancer drugs.

Abstract:A new series of 3‐substituted 4‐(4‐methyloxy phenyl)‐1H‐pyrrole derivatives were synthesized and biologically evaluated for potential anticancer activity. Fifteen targeted compounds showed high selectivity toward normal cells and cancer cells: that is, all targeted compounds had no obvious cytotoxicity toward normal human cells (HUVEC and NIH/3T3), but some compounds exhibited broad‐spectrum proliferation inhibitory activity against the screened cancer cell lines. Among these pyrrole derivatives, compounds 3b and 3o showed potent anticancer activity against the MG‐63 cell line, with IC50 values of 14.9 and 12.7 μM, respectively. Other pyrrole derivatives also showed promising proliferation inhibitory activity, including compound 3d against A375 (IC50 = 18.6 μM), compound 3f and 3j against MGC80‐3 (IC50 = 19.9 μM), and compound 3o against MGC80‐3 (IC50 = 11.9 μM). Because the developed pyrrole derivatives showed strong anticancer activity and high selectivity, this new series of pyrrole derivatives could be considered as promising lead compounds for further development of potent and safe anticancer agents.
