Abstract:The simple and one-pot method for the synthesis of polysubstituted tetrahydroquinolines from readily available anilines and aldehydes under reagent-free conditions has been developed. The scope of the transformation has been demonstrated. This method has been successfully applied to the rapid formal synthesis of (±)-martinellic acid and martinelline.

Abstract:An efficient method for the synthesis of substituted quinolines is described. By FeCl3-promoted C–C bond formation, 2,3-disubstituted and 2,3,4-trisubstituted quinolines were successfully synthesized from the reaction between imines and electron-deficient alkynes in good yields. The reaction can also be performed in a sequential three-component manner by using aromatic amines and aromatic aldehydes instead of the corresponding imines.

Abstract:We have developed an efficient, convenient new method for the synthesis of tetrahydroquinoline derivatives via PhSiH3‐mediated dearomatization of quinoline quaternary ammonium salts in moderate to good yields for the first time. This process has successfully constructed various substituted tetrahydroquinoline/isoquinoline derivatives, and has the advantages of mild reaction conditions and simple operation. Dearomatization of quinoline/isoquinoline quaternary ammonium salts to synthesize valuable tetrahydroquinolines was firstly reported. Under this method, we used phenylsilane as a reducing agent to prepare a series of tetrahydroquinoline and tetrahydroisoquinoline derivatives conveniently and quickly in moderate to good yields. This method showed good functional group tolerance, wide substrate range, mild reaction conditions and simple operation.

Abstract:A highly efficient method for the synthesis of 1,2,3,4-tetrahydroquinoline derivatives via a molecular iodine catalyzed domino reaction of anilines with cyclic enol ethers, such as 2,3-dihydrofuran and 3,4-dihydro-2H-pyran, is described. The reaction may proceed through an aza-Diels–Alder process between an in situ generated 2-azadiene and another equivalent of cyclic enol.

Abstract:Utilizing simple precursors for the exquisite construction of molecules with diversity and complexity is urgent. Herein, a novel A3-X type reaction for functionalized 3-haloquinolines has been developed by PhI (OAc)2-mediated four-component tandem addition/cyclization/oxidation/aromatization process with simple aromatic amine, aldehyde, alkyne and halide salt. Broad functional group tolerance, very mild condition, easy for further transformation illustrate the concision, high efficiency and practicability of this A3-X type reaction.

Abstract:A general and transition-metal-free method for the construction of the carbazol-4-amine motif via a vinylogous Michael addition/cyclization/isomerization/elimination reaction of 3-nitroindoles with alkylidene malononitriles has been developed. This novel methodology allows the facile synthesis of a series of di- and trisubstituted carbazol-4-amine derivatives in moderate to good yields. A gram-scale experiment was successfully performed, highlighting the practicability of this method. Moreover, this strategy is also applicable to 3-nitrobenzothiophene, affording the corresponding dibenzo[ b, d]thiophen-1-amine derivatives in moderate yields.

Abstract:Herein, we describe a general and selective deprotonation functionalization reaction of tetrahydroquinolines at the 4–position using organolithiums and phosphoramide ligands. In addition to the development of a direct deprotonation alkylation reaction with primary and secondary alkyl halides, a Negishi cross–coupling protocol was realized to afford products with a range of aromatic halides. These methods were applied to the late-stage installation of tetrahydroquinolines into a variety of substrates including pharmaceuticals as well as natural product analogues. The use of thorough mechanistic investigations revealed the aggregation state of the newly formed tetrahydroquinoline anion to be a separated ion pair, which proved critical to optimizing the reaction conditions.

Abstract:A wide range of isoquinoline ring-modified Quinap oxides with different steric and electronic variations have been constructed through a palladium-catalyzed imidoylative cyclization of arylethenyl isocyanides with 2-diphenylphosphinyl-1-naphthyl bromides. The Pd-catalysis plays dual roles in the formation of both axial C–C bond and isoquinoline ring. This de novo synthetic strategy features good functional group tolerance, high yields and easy scale-up, providing Quinap derivatives with substitution patterns that could not be obtained using coupling reactions. Chiral ligands 7 and 12 can be readily prepared by transformation of the resulting Quinap oxide to their BINOL esters, and have been proven to be superb chiral ligands for the copper-catalyzed enantioselective A3-coupling and alkynylation of quinoline reactions. In general, the enantioselectivies obtained using ligands 7 and 12 are excellent, and the ee values are higher than those using Quinap as ligand, even three times higher in some cases. Mechanism studies revealed that a monomeric copper(I) complex bearing a single chiral ligand was formed and served as the catalytically active species.

Abstract:The Winterfeldt oxidation (NaOH, DMF, air, rt) of substituted 1,2,3,4-tetrahydro-gamma-carbolines has been developed, which provides a convenient and efficient method for the synthesis of the corresponding dihydropyrrolo[3,2-b]quinolones in moderate to excellent yields (38-94%). The generality and substrate scope of this reaction are explored and a possible mechanism is proposed. The results imply that electron-withdrawing groups on N-2 of tetrahydro-gamma-carbolines and N-5-H are necessary. The synthesis of 5 or 7-substituted pyrrolo[3,2-b]quinolones in near quantitative yields was also achieved through deprotection and aromatization of N-1-Boc-dihydropyrrolo[3,2-b]quinolones.

Abstract:Atom- and step-economy in IBX assisted diversity-oriented synthesis is achieved with a versatile AQ auxiliary α-amino acid analogs offering rapid access to polycyclic spiro-quinolines featuring a quaternary stereocenter in 20%–91% yields under mild conditions via 7,8-dearomatization of quinolines. Free of a preinstalled activation group is highlight of this intramolecular oxidation spiroannulation tandem reaction. This type of N-heterospirocycles, traditionally difficult to access, may open the door to a potentially interest scaffold for synthetic and medicinal chemistry.

Abstract:A method was proposed for the regioselective synthesis of new 2,3-disubstituted quinolines based on the reaction of anilines with 4-chloro-1,1-diethoxybutane and N -(4,4-diethoxybutyl)tosylamide. Structure of the resulting compounds was proven by NMR spectroscopy techniques ( 1 H, 13 C, 15 N, COSY, HSQC 1 H– 13 C, HMBC 1 H– 13 C, HMBC 1 H– 15 N, NOESY), the composition was confirmed by elemental analysis data. The proposed method is distinguished by the possibility of obtaining a wide range of substituted quinolines in one step and the absence of the need to use expensive metal catalysts.

Abstract:A new and metal-free three-component method for the synthesis of 2,4-disubstituted quinolines via the reactions of anilines, α-keto acids and alkyl lactates is reported. The reactions proceed in the presence of p-toluene sulfonic acid (p-TSA) and tert-butyl peroxybenzoate (TBPB) to provide diverse quinoline products via the construction of new CC double, C-C single and CN double bonds without producing any organic mass-based side product. Notably, the anti-inflammatory activity of the quinolines has been investigated by measuring their ability to inhibit NO release by lipopolysaccharide (LPS) induced RAW264.7 cells, leading to the identification of 4i, 4t and 4x as potent anti-inflammatory compounds in vitro.

Abstract:A one-pot procedure has been developed to prepare the derivatives of 2-arylimidazo(2, 1-a)-isoquinolines (2a similar to 2b), 2-arylimidazo(1, 2-a) quinolines (4a similar to 4b) and 2-arylimidazo(1, 2-a) phthalazines (6a similar to 6c) in moderate yields (51% similar to 62%) by the reaction of isoquinolinium salts, quinolinium salts or phdialazinium salts with ammonium acetate in the presence of tetrakispyridine cobalt(II) dichromate (TPCD).
