Abstract:We demonstrate the capability of a molecular tracer based laser induced fluorescence photobleaching anemometer for measuring fluid velocity profile in microfluidics. To validate the feasibility and accuracy of this measurement system, the velocity profiles of the cylindrical and rectangular microchannels are measured, respectively. We compare our experimental results with theoretical prediction. The theoretical prediction shows a good consistency with practical measurement results. The spatial resolutions are analyzed and validated. Finally, the error source of the presented system is analyzed, which makes it possible to compensate some error quantitatively and guides the application of this system.

Abstract:In this work, an integrated optofluidic phase-shifting element, along with an image processing algorithm, was proposed to enable quantitative analysis for microfluidic applications using a digital holographic microscopic configuration. The advantages of the proposed system lie in its compactness, that the phase-shifting element can be simultaneously fabricated with other microfluidic functional components on a single chip, its robustness to resist environmental vibrations as no mechanical movement is involved for phase-shifting, and its capability of capturing transient events of fluid dynamics for quantitative measurement. The performance of the proposed system was validated with quantitatively label-free monitoring of concentration gradients under various conditions in microchannels.

Low Noisy Dual Wavelength Digital Holography and Application in the Ultra-precision Machining Surface Measurement

Abstract:Through the subtraction of two digital holographic wrapped phases for different wavelengths, it yields a beat phase image corresponding to an equivalent wavelength to remove the phase wrapping. By comparing the beat phase image with any recoding wavelength, the position and multiple of the single wavelength phase jump are confirmed. Hence, the wrapped phase in single wavelength phase is wrapped and the phase noise is kept fixed. Simulation and experimental results demonstrate the measure error induced by the phase noise can be reduced to 2Λ/λm. An optical microstructure element surface is measured by the proposed digital holography with dual wavelength of 650 nm and 632.8 nm. The measured optical element is produced with Fast Tool Servo (FTS). The unwrapped phase image of machining mark on the surface of microstructure element is acquired clearly. The equivalent wavelength is 0.024 mm. Then, the three dimensional data of the microstructure profile respectively in high frequency and low frequency regions are obtained by frequency filtering. The roughnesses corresponding to different frequency region are 33.2 nm, 19.3 nm and 23.4 nm respectively. The affecting factors for different machining mark are analyzed. The cutting parameters of the FTS are also deduced.

Abstract:Conventional optical refractometry methods are often limited by a narrow measurement range, complex hardware, or relatively high cost. Here, we present a novel refractometry method to measure the bulk refractive index (RI) of materials (including solids and liquids) using lensless holographic on-chip imaging and autofocusing, which is simple, cost-effective, and has a large RI measurement range. As a proof of concept, two compact prototypes were built to measure the RIs of solid materials and liquids, respectively, and they were tested by measuring the RIs of a ZnSe plate and a microscopy immersion oil. Experimental results show that our devices have an average accuracy of similar to 3 x 10(-4) RI unit (RIU) with an estimated precision of similar to 3 x 10(-3) RIU for solids; and an average accuracy of similar to 1 x 10(-4) RIU with an estimated precision of similar to 3 x 10(-4) RIU for liquids. We believe that this cost-effective and portable RI measurement platform holds promise to be used in laboratory and industrial settings. Abstract:By utilizing the internal-reflection effect at air–glass boundary, we have proposed a novel method of nano-displacement measurement. Excellent linearity of the reflectance versus the angle of incidence in internal reflection is achieved by making use of a differential detection scheme. Thereby, the target's displacement which causes the change of incident angle can be determined accurately by measuring reflectance. Using a differential detector such as quadrant detector, a compact system based on the above method has been developed, with advantages of simple configuration and low costs compared with interferometers and traditional confocal methods (CMs). The resolution of this system is about 2nm in a wide measurement range. Factors such as the initial angle of incidence, the number of reflections, and the polarization state of the light determine the resolution and measurement range. It is very suitable for many potential applications, especially in the automatic focus of the state-of-art microscopy, in which the traditional problem of focus drift may be solved. In addition, theoretical analysis and experimental results of a prototype sensor are presented.

Abstract:We design an effective microsystem by integrating ultra thin Ag film patterned with two-dimensional nanodisk array with a microfluidic channel. The refractive index of liquid, which flowing through the channel, is determined by measuring the transmission spectra of this integrated microfluidic device. We systematically simulated the influence of structure parameters to refractive index sensitivity. By adjusting the structure parameters of two-dimensional nanodisk array, such as period, thickness and duty cycle, the sensitivity of refractive index can be improved. The results indicated that the refractive index sensitivity can reach up to 308 nm / refractive index unit (RIU) with proper design. By this way, we can achieve intuitive, convenient and non-polluted refractive index measurement.

Abstract:Abstract Magnetic resonance and electrical resonance can be excited simultaneously by the incident light source in high refractive index all-dielectric metasurfaces. They can manipulate light waves to a certain extent and can be used in many fields. Silicon is widely used in all-dielectric metasurfaces due to its high refractive index and low loss in near infrared range. In this paper, we use silicon nanodisk structure as refractive index sensor and PDMS microfluidic channels as detection channels to realize liquid refractive index sensing. The magnetic resonance peak of the nanodisk structure is selected for sensing. The position of the magnetic resonance is shifted in the transmission spectrum with the change of the refractive index of the surrounding environment. When the refractive index increases, the resonance peak moves towards the long-wave direction, resulting in red shift, and vice versa. In this paper, the shift of resonance peak reaches 230 nm RIU−1.

Abstract:In this chapter, we introduce digital holographic microscopy (DHM) as a marker-free method to determine the refractive index of single, spherical cells in suspension. The refractive index is a conclusive measure in a biological context. Cell conditions, such as differentiation or infection, are known to yield significant changes in the refractive index. Furthermore, the refractive index of biological tissue determines the way it interacts with light. Besides the biological relevance of this interaction in the retina, a lot of methods used in biology, including microscopy, rely on light-tissue or light-cell interactions. Hence, determining the refractive index of cells using DHM is valuable in many biological applications. This chapter covers the main topics which are important for the implementation of DHM: setup, sample preparation and analysis. First, the optical setup is described in detail including notes and suggestions for the implementation. Following that, a protocol for the sample and measurement preparation is explained. In the analysis section, an algorithm for the determination of the quantitative phase map is described. Subsequently, all intermediate steps for the calculation of the refractive index of suspended cells are presented, exploiting their spherical shape. In the last section, a discussion of possible extensions to the setup, further measurement configurations and additional analysis methods are given. Throughout this chapter, we describe a simple, robust, and thus easily reproducible implementation of DHM. The different possibilities for extensions show the diverse fields of application for this technique.

Abstract:A micro digital in-line holographic particle tracking velocimetry (micro-DHPTV) system has been developed and applied to investigate the three-dimensional flow field in straight and Y-junction microchannels. The micro-DHPTV system comprises a cooled frame-transfer CCD camera and a double-pulsed laser. The processing algorithm introduced to evaluate the three-dimensional velocity is based on the combination of integrated cross-correlation and nearest neighbor matching algorithms, taking advantage of information from both the reconstructed particle field and the original holograms fringes patterns. Tests on simulated pairs of holograms show that the particles can be detected, located, and paired with high probability and accuracy. Results obtained in the straight and Y-junction microchannels show that the superimposed vector field is physically reasonable.

Abstract:High time-resolution flow field measurements in a micro-flow is performed by a micro digital-holographic particle-tracking velocimetry (micro-DHPTV) method. The system consists of an objective lens, a high-speed camera, and a single high-frequency double pulsed laser. Particle positions in a three-dimensional field can be reconstructed by a computer-generated hologram. The error of reconstruction in the z-directions is evaluated by traverse of particles on a glass plate. A velocity error in z-direction is obtained by uncertainty analysis. The time evolution of a three-dimensional water flow in a semicircular micro-channel of 101.9 [μm] width and 32.5[μm] depth and in a circular micro-pipe of 92 [μm] inner diameter are obtained successfully using this micro-DHPTV system. The volume of the system is defined by 409.6 [μm] ×92 [μm] ×92 [μm], and is irradiated by a laser beam with a resolution time of 100 [μs], and a repetition rate of 1 [kHz]. Consequently, approximately 100 instantaneous velocity vectors on each 1 000 frames for 1 [s] in the micro-channel can be obtained.

Abstract:In the present study, droplets of colloidal suspensions or emulsions are characterized using rainbow refractometry according to diameter, relative refractive index and concentration of the dispersed phase. The position and angular spacing of the rainbow are used to retrieve the relative refractive index and droplet size. For the measurement of colloid concentration, a novel method using the intensity ratio of the p = 2 (second-order refraction) and p = 0 (reflection) scattering components present in the rainbow signal is introduced. The experimental system, comprising a monodisperse droplet generation system and standard rainbow refractometry, has been used for validation of the technique. Distilled water mixed with monodisperse polystyrene nanoparticles at different volume concentrations (C-V = 0 - 0.3%) were tested, providing ground truth values of concentration. The measured relative refractive index and droplet diameter agree well with results obtained using the extended effective medium approximation for turbid media and with the known experimental values. The measured highly sensitive relation found to exist between intensity ratio attenuation and colloid concentration is numerically verified using a Monte Carlo ray tracing method. Abstract:A theoretical model of digital inline holography system reveals that the local focus metric curves (FMCs) of different parts of an irregular micro-object present spatial shift in the depth direction which is resulted from the depth shift. Thus, the 3D morphology of an irregular micro-object can be accurately measured using the cross correlation of the local FMCs. This method retrieves the 3D depth information directly, avoiding the uncertainty inherited from the depth position determination. Typical 3D morphology measurements, including the 3D boundary lines of tilted carbon fibers and irregular coal particles, and the 3D swimming gesture of a live Caenorhabdities elegans, are presented.

Abstract:Photonic biosensors based on photonic integrated circuits (PICs) and microfluidic channels (MFCs) have become the subject of intensive research for point-of-care (POC) device applications. In the presented work, we demonstrate the possibility of identifying the complex refractive index (RI) of analyzed liquids through the optimization of the geometry configuration of MFCs under PICs by experimental and numerical approaches. Our results suggest that the real and imaginary parts of the RI for analytes under study can be determined from spectrum of devices with optimized MFCs width. This work paves the way for promising opportunities to identify the presence and concentration of biological markers by using RI sensors for in situ POC applications.

Abstract:Digital holographic microscopy is an important measurement method for micro-nano structures. However, when the structured features are of high-slopes, the interference fringes can become too dense to be recognized. Due to the Nyquist's sampling limit, reliable wavefront restoration and phase unwrapping are not feasible. To address this problem, the interference fringes are proposed to be sparsified by tilting the reference wavefronts. A data fusion strategy including region extraction and tilt correction is developed for reconstructing the full-area surface topographies. Experimental results of high-slope elements demonstrate the validity and reliability of the proposed method.
A Novel Approach for Measurement of Free-Form Optical Elements with Digital Holographic Microscopy

Abstract:Free-form optical elements face significant challenges in high-precision measurement due to their high complexity and non-rotational symmetry. Digital holographic microscopy (DHM), as one of the methods for the measurement of free-form optical elements, has promising applications due to its ultra-high precision and non-destructive and fast characteristics. Therefore, we have designed a novel measurement method that combines transmission DHM and reflection DHM to obtain thickness information and surface information of elements to deduce the 3D structure. With this method, we completed the measurement of a free-form optical element. The DHM system we built has recorded holograms under 4× and 20× objectives and successfully recovered the 3D surface shape of the element. The measurements are consistent with the designed and manufactured parameters, demonstrating the unique advantages of DHM for measuring special types of optical elements.

Abstract:A refractive index (RI) sensor based on micro-nano fiber (MN-fiber) with chirped fiber Bragg grating (CFBG) Fabry–Perot cavity (FP-cavity) for a microfluidic chip has been proposed. A single-mode fiber is drawn by hydrogen flame heading come into MN-fiber. Two CFBGs are written into this MN-fiber by the ultraviolet (UV) laser mask exposure method. One is at the tapered region, another is at the micro-nano region. Then a micro-nano fiber with chirped fiber Bragg grating (MN-CFBGs) FP-cavity sensor is formed. The Bragg reflection wavelengths of two CFBGs are 1620 nm, 3-dB bandwidth are above 50 nm. The reflectance of two CFBGs are 70% and 99%, respectively. The effects of reflectivity and bandwidth of the CFBGs FP-cavity, diameter and length of MN-fiber with this sensor’s optical properties are analysed is and discussed. This sensor is embedded in a microfluidic chip and the MN-fiber region is immersion microfluid in different channels. The experimental results show that refractive index sensitivity of the sensor is –986 nm/refractive index unit (RIU), and the signal of the sensor has little noise. The CFBG-FP sensor not only has high sensitivity and lager measurement range, but also high contrast resonance signal and stability.

Abstract:This paper presents a simple and rapid method to measure the depth of the channel of microfluidic chip. With the depth of focus (DOF) low to few micro-meters, general microscopes show the strong axial (or longitudinal) resolving power. With the assistant of fine-adjustment, the objective was controlled accurately to focus on the channel side and channel bed respectively. Then the depth was calculated from the scales that fine adjustments turned. Experiments results demonstrated the possibilities of depth measurement with normal microscope. Compared with other methods, this method showed high degree of precision with a deviation less than 0.6µm while the magnification was 100X. This method is rapid, simple and reliable, and it is very suitable for monitoring the etching process to optimize the function of microfluidic chip.

Abstract:Digital holography (DH) has been widely used for imaging and characterization of micro and nanostructures in materials science and biology and has the potential to provide high-resolution, non-destructive measurements of fluid surfaces as well. Digital holographic setups capture the complex wavefronts of light scattered by an object or reflected from a surface, allowing for quantitative measurements of their shape and deformation. However, their use in fluid profilometry is scarce and has not been explored in much depth. We present an alternative usage for a DH setup that can measure and monitor the surface of fluid samples. Based on DH reflectometry, our modelling shows that multiple reflections from the sample and the reference interfere and generate multiple holograms of the sample, resulting in a multiplexed image of the wavefront. The individual interferograms can be isolated in the spatial-frequency domain, and the fluid surface can be digitally reconstructed from them. We further show that this setup can be used to track changes in the surface of a fluid over time, such as during the formation and propagation of waves or evaporation of surface layers.
