Abstract:Reactive sulfur species (RSS) are a family of crucial biosignals for regulating cell processes. Among these, hydrogen polysulfide (H2Sn, n 2) is a hallmark of tumor suppressor activation and regarded as the actual regulator to mediate sulfur-related biology. However, high effective recognition of intracellular H2Sn is insurmountable due to its extremely low concentration and the disturbance of RSS analogues. Here an H2Sn-responsive macromolecule that can distinguish H2Sn from intracellular RSS through polymer degradation in ultrasensitive and highly selective manner is reported. This kind of polymers can further self-assemble into vesicular nanostructure. Upon cell uptake, they can be function as all-in-one H2Sn-nanoplatforms, in order to fulfill multiple ambitious tasks including monitoring the H2Sn biosynthetic pathways, unraveling the puzzles of H2Sn-mediated cellular events, and conducting H2Sn pathological milieu-specific drug delivery.

Abstract:A new diketopyrrolopyrrole amphiphile, namely 2-(2-(2-methoxyethoxy)-ethyl)- 3,6-di(pyridin-2-yl)-2,5-dihydropyrrolo[3,4-c]pyrrole-1,4-dione (PyDPP), is designed and synthesized as a novel ratiometric/fluorescent chemosensor for Cu2+/Hg2+. PyDPP forms soluble loose nanoaggregates by molecular self-assembly in water and maintains intrinsic bright fluorescence and color. The UV–vis absorption spectra data show that PyDPP nanoaggregates exhibits high sensitivity and selectivity toward Cu2+/Hg2+ among other metal cations in PBS (pH 7.4) solution. The reaction mechanism involved in PyDPP-Cu/Hg was investigated by MS, XPS and DFT calculation. The limit of detection of the present nanosensor for Cu2+ is calculated to be 87 nM. This research provides another view in the design and application of FONs nanosensor for heavy metal ions detection.
A Chiral Polymer‐based Turn‐on Fluorescent Sensor for Specific Recognition of Hydrogen Sulfate

Abstract:A new kind of polymeric chemosensor containing chiral naphthaldimine moiety in the side chain was synthesized by the reversible addition-fragmentation chain transfer polymerization of N-{[2-(4-vinylbenzyloxy)-1-naphthyl]-methylene}-(S)-2-phenylglycinol (VNP). The resulting polymers (PVNP) showed high selectivity for hydrogen sulfate relative to other anions including F-, Cl-, Br-, H2PO?4-, CH3CO?2-, and NO?3- in tetrahydrofuran (THF) solution as judged from UV-vis, fluorescence, and circular dichroism spectrophotometric titrations. Compared with its monomer, the polymer has proven to be more attractive for detection of HSO?4- in terms of sensitivity and reproducibility. Upon addition of the anion it gives remarkable spectral responses concomitant with detectable color change from colorless to pale yellow. Furthermore, the HSO?4--induced CD or fluorescence signal can be totally reversed with addition of base and eventually recovered the initial state, leading to a reproducible molecular switch with two distinguished on and off states. Abstract:Hydrogen sulfide (H 2 S) as an endogenous signaling molecule, plays an irreplaceable role in many important physiological activities. It is also closely related to sewage treatment, wine quality evaluation, and food spoilage. Herein, we have successfully synthesized a novel polymer-based probe P1 for fluorescently ratiometric sensing of H 2 S with a high selectivity and sensitivity. By virtue of ring-opening metathesis polymerization (ROMP), P1 was prepared with the disulfide bond linked coumarin-norbornene dyad NB-SS-COU as energy donor, the aggregation-induced emission (AIE) fluorophore anchored norbornene NB-TPE as energy receptor, and the polyethylene glycol (PEG) attached norbornene NB-PEG as a hydrophilic chain. At the 400 nm excitation, P1 displays a bright red emission at 615 nm due to the efficient fluorescence resonance energy transfer (FRET) from energy donor COU to energy acceptor TPE . Upon addition of H 2 S, it shows strong COU -based blue emission at 473 nm for cleavage of the disulfide bond. We also constructed a smartphone sensing platform to conduct visual quantitative detection of H 2 S by calculating the B/R (blue/red) emission ratio values. Moreover, P1 can be successfully employed in evaluating the level fluctuations of endogenous and exogenous H 2 S in living cells, testing water samples/wine samples, and monitoring food freshness.

Abstract:Hydrogen polysulfides (H2Sn, n >= 2) is recently regarded as a potential signaling molecule which shows a higher efficiency than hydrogen sulfides (H2S) in regulating enzymes and ion channels. However, the development of specific fluorescent probes for H2Sn with long-wavelength emission (>600 nm) are still rare. In this work, a semi-naphthorhodafluor-based red-emitting fluorescent probe SNARF-H2Sn containing a phenyl 2-(benzoylthio) benzoate responsive unit was constructed. SNARF-H2Sn was capable of selectively detecting H2Sn over other reactive sulfur species. Treatment with H2Sn would result in a > 1000-fold fluorescence enhancement within 10 min. SNARF-H2Sn showed a low limit of detection down to 6.7 nM, and further enabled to visualize exogenous/endogenous H2Sn in living A549 cells and zebrafish. (C) 2020 Elsevier B.V. All rights reserved.

Abstract:Metabolic disorders involving endogenous H 2 S have been linked to a variety of serious human diseases, particularly cancer. In this study, we employed nanofibers with surface-enhanced Raman scattering (SERS) activity for the detection of H 2 S within live cells. These nanofibers were chosen for their minimal invasiveness, high spatial resolution, and enhanced SERS sensitivity. To improve the performance of SERS, highly sensitive core-shell multibranched-Au NPs (MBAuNP)@Ag NPs were decorated on the nanofibers as SERS tags for H 2 S detection. A SERS probe named MBN, embedded between the Au core and Ag shell, was utilized for quantitative detection. These nanofibers exhibited excellent reproducibility (relative standard deviation (RSD) within 5.7 %) and demonstrated a strong linear relationship with sulfide concentrations ranging from 50 nM to 1 μM, with an estimated detection limit of 0.12 nM. As a proof of concept, the aforementioned nanofibers were successfully applied to detect endogenous H 2 S in living cells, offering a potential analytical method in the related research of detection.

Abstract:Hydrogen sulfide (H2S) is an appealing signaling molecule that plays fundamental roles in health and disease. However, H2S-mediated selective chemical transformations for the construction of imaging probes are limited, retarding the interrogation of H2S-related biological processes. Here, we present an alternative approach for engineering a new generation of efficient probes with a nonchemoselective moiety as a building block. To demonstrate our design concept, we developed a sulfoxide-functionalized BODIPY that exhibited a substantial redshift in its absorption and emission spectra upon reduction with H2S. However, such a probe also showed reactivity toward various competing biothiols under aqueous buffer conditions. To achieve high chemoselectivity, we used core-shell silica nanoparticles as an encapsulation matrix to confine the designed molecule probe within their interiors. The inherent molecular-size sieving character of the porous silica shell was capable of impeding competing biothiols from accessing the molecule probe within the core while allowing the specific reaction with the small target H2S. Thus, this strategy avoided disturbance from coexisting biothiols and achieved highly chemoselective detection in ratiometric and near-infrared (NIR) turn-on fluorescence modes. In light of these promising features, together with fast responsiveness and favorable cellular uptake, such a silica nanocomposite was successfully used to detect the endogenous production of H2S in estrogen-induced cardiomyocytes and living mouse model. To our knowledge, the approach reported here is the first to exploit the usefulness of common thiol-sensitive moieties for building chemoselective probes.

Abstract:Probes bearing good aqueous solubility and biocompatibility as well as fast response can serve as ideal tools for evaluating the underlying molecular mechanism of endogenous production of H2S caused by drugs; however, they are still lacking but highly desirable. Here, we demonstrate a novel strategy for constructing highly efficient H2S nanoprobes through locking Förster resonance energy transfer borondipyrromethene (BODIPY) pairs in water-dispersible core-shell silica nanoparticles. Importantly, these nanocomposites can effectively confine complementary guests within the same cores due to the existence of a shield, thus guaranteeing efficient Förster resonance energy transfer. Interestingly, the interior microenvironment of such nanoparticles could be tuned by silylation agents. In this way, an ideal probe for rapid and ratiometric detection of H2S within 15 s is established by optimizing the amount of silylation agent with a polar organic group. Obviously, the silylation agents are explored to serve as a platform not only for establishment of robust structures but also for optimizing the microenvironment of the interior to afford an ideal probe. These silica nanocomposites have also been successfully employed in disclosing the endogenous production of H2S induced by estrogen in cardiomyocytes.

Abstract:Building biomimetic polymer vesicles that can sense a biological signaling molecule is a tremendous challenge at the cross-frontier of chemistry and biology. We develop a new class of o-azidomethylbenzoate (AzMB)-containing block copolymer that can respond to an endogenous signaling molecule, hydrogen sulfide (H2S). Such a gasotransmitter can trigger cascade chemical reactions to sever the AzMB side functionalities, which alters the polymer amphiphilicity and further leads to a controllable disassembly of their self-assembly vesicular nanostructure. Moreover, if we introduce cystathionine γ-lyase (CSE), a specific enzyme converting cysteine into H2S, onto the vesicle membrane, the polymersomes can extend their responsive scope from H2S to a specific amino acid bioactivator. We anticipate that this polymer model could open up a new avenue for constructing biosignal-triggered nanocapsules for intracellular applications.

Abstract:Hydrogen polysulfide (H2Sn, n≥2), as a kind of active sulfur species (RSS), has become a hot topic in RSS. It can regulate the biological activity of many proteins through S-sulfhydrylation of cysteine residues (protein Cys-SSH), and has a protective effect on cells. Although there have been some studies on hydrogen polysulfide, its production, degradation pathway and regulation mechanism still need further be researched. In presented study, an original lysosome-localized fluorescent probe for determining H2Sn was developed utilizing rhodamine as the fluorogen. The probe used morpholine as the locating unit of lysosomes and chose 2-fluoro-5-nitrobenzoate as the recognizing group. Before adding H2Sn, the proposed probe displayed a spironolactone structure and emitted very weak fluorescence. After adding H2Sn, a conjugated xanthene was formed and the probe demonstrated green fluorescence. When the H2Sn concentration was varied from 6.0×10-7 mol·L-1 to 10.0×10-5 mol·L-1, the fluorescence intensity of the probe was linearly dependent on the H2Sn concentration. And the detection limit was 1.5×10-7 mol·L-1. The presented probe owned a fast response speed, good selectivity, excellent sensitivity and broad pH work scope. In addition, the probe had been well utilized to sense endogenic and exogenic H2Sn in lysosomes.
A polymeric film probe with a turn-on fluorescence response to hydrogen sulfate ions in aqueous media

Abstract:A new kind of hydrophilic copolymer poly(HEMA-co-VNP) was designed and synthesized by the radical copolymerization of 2-hydroxyethylmethacrylate (HEMA) and 2-{[2-(4-vinylbenzyloxy) naphthalen-1-yl] methyleneamino}-(S)-2-phenylethanol (VNP). The desired copolymer showed a highly selective red-shifted emission and a unique chiroptical response upon HSO4- binding in organic solution. UV-vis and H-1 NMR spectroscopic studies revealed that the hydrogen bonding between the imine moiety in the VNP repeating units and HSO4- is crucial for the high selectivity of the receptor to this anion. As expected, the incorporation of HEMA into the polymer matrix endowed the copolymer excellent hydrophilicity, flexibility and good film-forming properties. Thus, high-quality film sensors could be easily fabricated on quartz plates through spin-casting techniques. The resultant polymeric films can recognize HSO4- ions among a series of common anions in aqueous solution with high selectivity and sensitivity. The promising new film probe for HSO4- has distinct characteristics such as a rapid response, enough stability in an aqueous media and practicality.

Abstract:As a signal transduction molecule in organism, H2S plays an important role in many physiological processes. In this paper, a novel H2S fluorescent probe SBD was designed based on 1,8-naphthalimide and dinitrobenzenesulfonyl group. In DMSO-PBS solution, probe SBD could recognize H2S and change the color of solution from yellow to pink, realizing the naked eye recognition for H2S. In addition, probe SBD displayed high fluorescence enhancement rate (45-fold), good selectivity, strong anti-interference ability and the detection limit was as low as 152 nM. Most importantly, probe SBD showed very low cytotoxicity and had been successfully applied to image exogenous H2S in living cells.

Abstract:In order to quantitatively detect hydrogen polysulfides (H2Sn, n > 1) in biosystems, we propose a new two-photon turnon fluorescent probe, named Np–Sn, based on the D–π–A skeleton two-photon fluorophore of 4 -hydroxy-1,8-naphthalimide derivative and deprotection of 2-fluoro-5-nitrobenzoic ester by H2Sn–Np–Sn displayed a more than 80-fold enhancement towards H2Sn in 550 nm and high sensitivity with a detection limit as low as 33 nM. Additionally, the probe Np–Sn was further used for fluorescence imaging of H2Sn in living cells under two-photon excitation (820 nm), which showed a high-resolution imaging, thus demonstrating its practical application in biological systems for the study of physiological and pathological functions of H2Sn.

Abstract:Hydrogen polysulfide (H2Sn), derived from hydrogen sulfide (H2S), has attracted increasing attention, which is suggested to be the actual signal molecule instead of H2S in physiological and pathological processes. Reversible detection of H2Sn through a NIR-excitable fluorescence probe is an effective means to understand its functions but is quite challenging. Herein, we reported a NIR-excitable ratiometric nanoprobe for the reversible detection of H2Sn based on luminescence resonance energy transfer principle with upconversion nanoparticles as the energy donor and an organic molecule, SiR1, as the energy acceptor and reversible recognition unit of H2Sn. The as-prepared nanoprobe exhibited high selectivity and fast response for the reversible detection of H2Sn, which can monitor the formation and consumption of endogenous H2Sn in living cells. Because of the reduced autofluorescence by NIR excitation, it was successfully applied for tracking the fluctuation of H2Sn concentration of mice in physiological and pathological processes including inflammation and liver injury.

Abstract:We present an approach for constructing a H2S-specific nanoprobe by the entrapment of a small molecule probe within the hydrophobic interior of surface cross-linked micelles (SCMs), endowing the designed nanoprobes with good water solubility and biocompatibility. Importantly, the obtained nanoprobes displayed good responsiveness to H2S in both ratiometric fluorescence and light-up NIR emission modes, thus enabling accurate identification of H2S-rich colorectal cancer cells.

Abstract:The present study introduces a novel fluorescent sensor with an overtone peak reference designed for the detection of mercury (II) ions (Hg 2+ ) and hydrogen sulfide (H 2 S). The study proposes two novel response mechanisms that hinges on the synergistic effect of cation exchange dissociation (CED) and photo-induced electron transfer (PET). This sensor exhibits a remarkable detection limit of 2.9 nM for Hg 2+ . Additionally, the sensor reacts with H 2 S to generate nickel sulfide (NiS) semiconductor nanoparticles, which amplify the fluorescence signal and enable a detection limit of 3.1 nM for H 2 S. The detection limit for H 2 S is further improved to 29.1 pM through the surface functionalization of the nanomaterial with pyridine groups (increasing reactivity) and chelation of gold nanoparticles (AuNPs), which enhances the sensor's specificity. This improvement is primarily due to the surface plasmon resonance (SPR) of AuNPs and their affinity for H 2 S. The single-emission strategy can yield skewed results due to environmental changes, whereas the overtone peak reference strategy enhances result accuracy and reliability by detecting environmental interference through reference emission peaks. In another observation, the low-toxicity dihydropyrene-bipyridine nanorods (TPP-BPY) has been successfully utilized for both endogenous and exogenous H 2 S detection in vivo using a mouse model. The successful development of TPP-BPY is expected to provide an effective tool for studying the role of H 2 S in biomedical systems.

Abstract:One challenge of the structural design of a fluorescent probe is how to improve the detection performance on trace target analytes in complex samples. Herein a new polymer fluorescent nanoprobe (2DSP-C28) has been synthesized, by adopting a two-dimensional (2D), spiropyran (SP)-based nanosheet structure with hydrophobic long-chain alkanes (C28). Unlike a traditional spiropyran-based small molecule probe, the 2DSP-C28 probe can exhibit quantitative-fluorescent and photochromic properties. Under the detection of metal-ions, the nanoprobe in DMSO aqueous solution is selectively fluorescent-quenched-responsive for Fe-ions (~100 μM), with a characteristic stoichiometric ratio (SR) of <10, a high sensitivity (LOD: ~ 0.2 μM). When the nanoprobe is incorporated into electrospun polyethylene oxide (PEO), it can be used for gas detection, and display a color-change with acid-base gas and identify the HF gas. It is expected that this new polymer fluorescent nanoprobe can be promisingly applied for rapidly environmental monitoring on the ion or gas pollution.

A new lysosome-targetable fluorescent probe for detection of endogenous hydrogen polysulfides in living cells and inflamed mouse model.

Abstract:Hydrogen polysulfides (H2Sn, n > 1) belong to sulfane sulfur in the reactive sulfur species (RSS) family and play significant roles in maintaining biological homeostasis in organisms. The detection of H2Sn in living systems is essential, but further understanding of its "intact" function in living cells has been limited, owing to the lack of appropriate analytical tools. In this work, a new fluorescent probe PP-PS was designed for the detection of endogenous H2Sn. The probe has a large Stokes shift (178 nm), low detection limit (1 nM), and short response time (1 minute). Besides, PP-PS was successfully applied in the imaging of endogenous H2Sn in lysosomes of living cancer cells, xenograft mouse tumor tissues, and LPS-induced mouse inflammation tissues. These results revealed that the probe PP-PS could act as a new fluorescence imaging tool to study the function of intracellular hydropolysulfides.
