Abstract:As a common gaseous signaling molecule, hydrogen sulfide (H2S) plays a vital role in physiology and pathology. The development of fluorescent probes for detecting H2S has attracted widespread attention. However, most of the reported fluorescent probes with nitrobenzoxadiazole (NBD) as the recognition group have been widely used to simultaneously detect biothiols and H2S, instead of specifically detecting H2S. Herein, a novel NBD-based near-infrared (NIR) fluorescent probe named CX-N for the detection of H2S is synthesized. The selectivity of CX-N for H2S is significantly higher than that for biothiols and other potential interferences. After reacting with H2S, CX-N shows a significant increase in NIR fluorescence (75-fold), large Stokes shift (155 nm) and fast response (4 min). And the possible response mechanism of CX-N to H2S is given and confirmed by HPLC and HRMS. Based on the low cytotoxicity of CX-N, it has been used for H2S imaging in live cells and zebrafish. More importantly, CX-N has also been successfully applied for the real-time imaging of H2S in inflammatory and tumor mice based on its NIR emission, which provides a reliable platform for the specific recognition of H2S in complex biological systems.

Abstract:Hydrogen sulfide (H2S) has emerged as an exciting endogenous gasotransmitter in addition to nitric oxide and carbon monoxide. However, its precise measurement in living cells and animals remains a challenge. In this study, a novel bioluminescence H2S probe was designed and synthesized by modifying the 6'-amino group of d-aminoluciferin into a 6'-azido group, which was highly selective against other reactive sulfur, nitrogen, and oxygen species. Our H2S probe azidoluciferin sensitively reacted with H2S to release d-aminoluciferin with a strong bioluminescence signal. On the basis of its high selectivity and sensitivity, the H2S probe was used to detect H2S production in live cancer cells and nude mice. The bioluminescence signal decreased in mice treated with propargylglycine, an inhibitor of H2S, suggesting that our H2S probe can detect endogenous H2S in real time, in vivo. Overall, the excellent sensing properties of the probe combined with its bioimaging capability make it a useful tool to study H2S biological roles.

Abstract:Hydrogen sulfide (H2S) is an important gaseous mediator in cellular physiology and pathology. For understanding the biological action of H2S to both healthy and disease states, it is important to develop H2S-selective sensor. Herein we report a colorimetric and fluorescent probe, which employs an NBD moiety as fluorophore, and is equipped with azide group as H2S-active unit. The probe exhibits selective and sensitive response to H2S in aqueous solution. By employing 1 mu M sensor, the detection limit was evaluated to be 680 nM with a signal-to-noise ratio of 3. Abstract:Hydrogen sulphide (H2S), the third endogenous gaseous signalling molecule, has attracted attention in biochemical research. The selective detection of H2S in living systems is essential for studying its functions. Fluorescence detection methods have become useful tools to explore the physiological roles of H2S because of their real-time and non-destructive characteristics. Herein we report a near-infrared fluorescent probe, NIR-HS, capable of tracking H2S in living organisms. With high sensitivity, good selectivity and low cytotoxicity, NIR-HS was able to recognize both the exogenous and endogenous H2S in living cells. More importantly, it realized the visualization of endogenous H2S generated in cells overexpressing cystathionine β-synthase (CBS), one of the enzymes responsible for producing endogenous H2S. The probe was also successfully applied to detect both the exogenous and endogenous H2S in living mice. The superior sensing properties of the probe render it a valuable research tool in the H2S-related medical research.

Abstract:Hydrogen sulfide (H2S) is an important endogenous gas signal molecule in living system, which participates in a variety of physiological processes. Very recent evidence has accumulated to show that endogenous H2S is closely associated with various cancers and can be regarded as a biomarker of cancer. Herein, we have constructed a new near-infrared fluorescent probe (DCP-H2S) based on isophorone-xanthene dye for sensing hydrogen sulfide (H2S). The probe shows remarkable NIR turn-on signal at 770 nm with a large Stokes shift of 200 nm, together with high sensitivity (15-fold) and rapid detection ability for H2S (4 min). The probe also possesses excellent selectivity for H2S over various other analytes including biothiols containing sulfhydryl (-SH). Moreover, DCP-H2S has been successfully applied to visualize endogenous and exogenous H2S in living cells (293T, Caco-2 and CT-26 cells). In particular, the excellent ability of DCP-H2S to distinguish normal mice and tumor mice is shown, and it is expected to be a powerful tool for detection of H2S in cancer diagnosis.

Abstract:Hydrogen polysulfide (H2Sn), derived from hydrogen sulfide (H2S), has attracted increasing attention, which is suggested to be the actual signal molecule instead of H2S in physiological and pathological processes. Reversible detection of H2Sn through a NIR-excitable fluorescence probe is an effective means to understand its functions but is quite challenging. Herein, we reported a NIR-excitable ratiometric nanoprobe for the reversible detection of H2Sn based on luminescence resonance energy transfer principle with upconversion nanoparticles as the energy donor and an organic molecule, SiR1, as the energy acceptor and reversible recognition unit of H2Sn. The as-prepared nanoprobe exhibited high selectivity and fast response for the reversible detection of H2Sn, which can monitor the formation and consumption of endogenous H2Sn in living cells. Because of the reduced autofluorescence by NIR excitation, it was successfully applied for tracking the fluctuation of H2Sn concentration of mice in physiological and pathological processes including inflammation and liver injury.

Abstract:Two fluorescent chemosenors, Mito-HS and Lyso-HS, were rationally designed and synthesized with positive charge at physiological conditions. The positive charge showed triple functions as target moieties for subcellular mitochondria and lysosome of living cells, soluble moieties for chemosenors, as well as effective sequesters of HS(-) (H2S at physiological conditions). These two probes showed faster and more efficient fluorescence H2S detection than the similar literature-reported probe without positive charge. In addition, visualizing of hydrogen sulfide in tumors of living mice was successfully achieved for the first time using the probe Mito-HS.

Abstract:Hydrogen sulfide (H2S) is an important signaling molecule that exists in a variety of tissues and has important effects on various physiological functions of the human body. Studies have shown that chronic kidney disease, liver cirrhosis and cardiovascular disease are closely related to abnormal levels of H2S in the body. In addition, endogenous H2S is closely associated with a variety of cancers and can be regarded as a biomarker for specific cancers. In this study, we have successfully designed a rhodamine hybrid polymethine-based small molecule dye (FL-NO2), whose fluorescence in the second near-infrared window (NIR-II, 900-1700 nm) can be dramatically enhanced after exposure to H2S. After being encapsulated by amphiphilic polymer F-127, the obtained water-soluble nanoparticles (n-FL-NO2) can be used for the sensitive detection of H2S in the NIR-II region, which provides large penetration depth and high signal-to-noise ratio. The probe also exhibited high selectivity towards H2S, avoiding interference from multiple biothiols. The detection of endogenous H2S in vivo was demonstrated with a colon cancer model, which expressed a high level of H2S. In summary, this study provides a promising NIR-II fluorescent probe for the rapid detection of H2S, which may become an effective tool for disease diagnosis.

Abstract:A dicyanoisophorone-based near-infrared fluorescent probe (NIR-NP) was successfully designed for hydrogen sulfide (H2S) detection. 7-Nitro-1,2,3-benzoxadiazole (NBD) amine, an electron-withdrawing group, was applied to quench the fluorescence of dicyanoisophorone-based fluorescent dyes and could be removed by H2S-specific thiolysis of an NBD amine bond, leading to a significant fluorescence turn-on response. The probe could quantitatively detect H2S with a lower detection limit (0.03 mu M) in vitro and in living cells. The probe with a large Stokes shift (186 nm) showed a high selectivity to H2S in the presence of other biothiols, including glutathione, homocysteine, and cysteine. Moreover, the probe was successfully applied to image endogenous H2S in different tumor cells and in liver tissues. The excellent properties of the probe and its applications in cell and tissue imaging indicated its potential for further exploration and application of H2S in the pathophysiology of cancers and even in the diagnosis of H2S-related diseases.

The probe employs cyanine dye as a fluorophore, and is equipped with a nitro group as a fluorescent modulator. Abstract:Taking apart in numerous physiological and pathological activities, hydrogen sulfide (H 2 S) has been selected as an excellent target spot for the early diagnosis of cancer. So far, there are many mature probes that apply single optical imaging to monitor endogenous H 2 S. Nevertheless, a single modality is not an ideal method to afford accurate diagnostic information in comprehensive biological organisms. Herein, we developed a dual-modal imaging probe BWS. This designed probe showed a specific response to H 2 S with both chemiluminescence and NIR fluorescence light-up. The concurrence of fluorescence and chemiluminescence signal provided "double insurances" for highly accurate monitoring of H 2 S. Satisfactorily, this dual-modal imaging probe performed precise visualization of endogenous H 2 S in living cells and in vivo . We envisaged that this chemiluminescence/fluorescence real-time dual-modality strategy for H 2 S detection will expand and optimize the multimodal imaging methods for efficient diagnosis and treatment of cancer.

Abstract:Developing a ratiometric H2S fluorescent probe with fast response is of great importance for studying the H2S physiology. Herein, two hemicyanine-based H2S probes were constructed; the one with a propanoic acid group (CouPa) showed poor sensitivity while the other one with theN,N-diethylpropionamide moiety (CouDE) exhibited distinctly improved performance. CouDE showed the ability to detect mitochondrial H2S level fluctuation, which was triggered by alteration of CBS enzyme activity. Moreover, endogenous H2S change in solid tumours was monitored using CouDE.

Abstract:A coumarin-based fluorescence chemoprobe was developed and evaluated for the selective and sensitive detection of hydrogen sulfide in degassed PBS buffers and fetal bovine serum. Fluorescence detection of hydrogen sulfide in living cells was also successfully achieved using two-photon confocal fluorescence imaging. Abstract:Hydrogen sulfide (H 2 S) performs essential functions in several physiological and pathological procedures, such as liver cirrhosis, diabetes mellitus, and cancer. To optimize the H 2 S detection in practical applications, herein, a new probe YG-H 2 S for the fluorescent imaging of H 2 S was developed. Compared with the current H 2 S probes, the main advantages of YG-H 2 S were a wide pH range (5.0–9.0), a relatively fast response (within 10 min), and high selectivity from the competitive species (including biological mercaptan). Owing to the low cytotoxicity of YG-H 2 S , it was good for the biological depth imaging in biological events. YG-H 2 S could detect endogenous H 2 S in HeLa cells. In addition, YG-H 2 S could monitor the level of H 2 S in 4 T1-xenograft and acute liver injury model mice as well as in separate tissues. Consequently, YG-H 2 S might be a potential method for detecting H 2 S in preliminary diagnosis.

Abstract:As a signal transduction molecule in organism, H2S plays an important role in many physiological processes. In this paper, a novel H2S fluorescent probe SBD was designed based on 1,8-naphthalimide and dinitrobenzenesulfonyl group. In DMSO-PBS solution, probe SBD could recognize H2S and change the color of solution from yellow to pink, realizing the naked eye recognition for H2S. In addition, probe SBD displayed high fluorescence enhancement rate (45-fold), good selectivity, strong anti-interference ability and the detection limit was as low as 152 nM. Most importantly, probe SBD showed very low cytotoxicity and had been successfully applied to image exogenous H2S in living cells.

Abstract:H2S has been reported to play essential roles in a variety of physiological and pathological procedures. In this work, a novel fluorescent probe, Rho-HS, for detecting H2S was developed by introducing the ortho-halogen to activate the least reactive recognition group 2,4-dinitrophenyl moiety. In combination of the structures from both Rhodamine B and fluorescein, Rho-HS could generate both the colorimetric and fluorescent responses. This feature was not frequently achieved and could lead to the quantitative and convenient for the end-user. In comparison with recent probes for H2S, the major advantages of Rho-HS included suiting wide pH range (6.0-10.0), relatively rapid response (within 15 min) and the high selectivity among the competing species including the biothiols. With low cytoxicity, Rho-HS was further applied in the biological imaging in living MCF7 cells and Caenorhabditis elegans. We hope that the designing strategy in this work might provide useful information for more preferable implements in this field.

Abstract:Hydrogen sulfide (H 2 S) is traditionally considered as an ecologically toxic gas, has been proven to be the third endogenous signaling gasotransmitter and plays important roles in physiological and pathophysiological conditions. Herein, we designed and synthesized a novel "turn off" fluorescent probe for the detection of hydrogen sulfide(H 2 S) in vitro. Probe A could detect H 2 S in CH 3 CN/PBS system with significant fluorescence quenching. Probe A also showed obviously aggregation- induced emission (AIE) effects. Fluorescence titration experiments revealed probe A combine with H 2 S in a 1: 1 ratio with a detection limit of 6.54 μM. Cell imaging results demonstrated the potential applicability of probe A for H 2 S detection in living cells. The supramolecular assembly of probe A and β-cyclodextrin further revealed the interaction with H 2 S.
