Abstract:Objective: To assess associations of single and combined exposures to lead and stress during different stages of pregnancy with offspring neurodevelopment.Methods: We measured prenatal lead (maternal blood-lead in early-pregnancy and umbilical-cord-blood-lead) and maternal stress levels in Shanghai-Birth-Cohort from 2013 to 2016. Maternal stress was assessed using Center-for-Epidemiological-Studies-Depression-Scale and Self-Rating-Anxiety-Scale during mid-pregnancy. The Ages-Stages-Questionnaires-3 (at 6/12-months-of-age) and Bayley-III (at 24-months-of-age) were both used to assess neurodevelopment. Results: A total of 2132 mother-child pairs with both prenatal lead and stress measurements were included. The geometric-means of blood-lead in early-pregnancy and cord-blood-lead were 1.46 mu g/dL and 1.33 mu g/dL, respectively. Among the study women, 1.89 % and 0.14 % were screened positive for depression and anxiety. Adjusting for related confounders, the combined exposures had stronger adverse associations with offspring social-emotional skills than single exposures; and the combined exposure in early-pregnancy was associated with greater neurodevelopmental differences than combined exposure around-birth, especially in social-emotion at 24 months-of-age [beta (95 %CI): 10.48(-17.42,-3.54) vs. 5.95(-11.53,-0.36)]. Conclusions: Both single and combined prenatal exposures to lead/stress impaired infant neuro-development, and the effects of combined exposure may be more profound than single exposures. Combined exposure in early pregnancy may be associated with worse neurodevelopmental outcomes than combined exposure around birth, especially in social-emotional development.

Abstract:Background and aim: To assess the current exposure levels of lead and stress during pregnancy, and compared the effects of prenatal combined exposure of lead and stress during different stages of pregnancy on the neurodevelopment of offspring. Methods: A total of 5270 mother-child pairs were assessed in the Shanghai Birth Cohort, maternal blood lead in early pregnancy and umbilical cord blood lead were measured. Assessments of prenatal maternal stress, the Center for Epidemiological Studies-Depression Scale and the Self-Rating Anxiety Scale during mid-pregnancy, postnatal maternal stress using the Edinburgh Postnatal Depression Scale were conducted, and groups of prenatal single exposure to lead/stress and combined exposure were therefore formed. The Ages-Stages-Questionnaires (at 6- and 12-months-of-age) and the Bayley-III (at 24-months-of-age) were performed to assess the offspring neurodevelopment. Results: The geometric means of maternal blood lead in early pregnancy and umbilical cord blood lead were 1.45 μg/dL and 1.53 μg/dL, respectively. A total of 1.86% and 0.18% of the mothers were screened positive for depression and anxiety. Adjusting for related confounders, the prenatal combined exposure in early pregnancy was associated with more deficits in infant communication skills and social-emotional skills, especially in social-emotional at 24-month-old (P=0.016); the prenatal combined exposure around birth was adversely associated with infant neurodevelopment levels in multiple domains. Conclusions: The prenatal combined exposure to lead and stress may affect offspring neurodevelopment more profoundly (especially on social-emotional development), and the combined exposure in early pregnancy may induce more deficits in offspring social-emotional development than the combined exposure around birth. Abstract:Objective To explore the effects of prenatal exposure to stress and lead on spatial learning and memory development in rats. Methods All 32 Sprague-Dawley (SD) pregnant rats were divided randomly into 4 groups, 8 per group in line with the Random Number Table. The four groups were: no maternal stress, no Pb exposure (NS/C) ; non-maternal stress, Pb exposure (NS/L), maternal stress, no Pb exposure (S/C), and maternal stress plus Pb exposure (S/L). The spatial learning and memory ability, the serum corticosterane level both pre and post-testing of 30-day old offsprings, and lead concentration in hippocampns were tested by means of Morris Water Maze, radioimmunoassay and Inductively Coupled Plaswa Mass Spectrometry(ICP-MS). Results The residence time of male and female in NS/L was ( 16. 08±3.41 ) s, ( 15.72 ± 3.33 ) s, which were significantly shorter than NS/L (25.42 ± 4. 76) s, (24. 55± 4. 43 ) s and S/C ( 20. 96±3.45 ) s, ( 20. 65 ±2. 98 ) s, and significant difference was observed in the joint exposure effect( F = 5. 478, P < 0. 05 ). The effect of the joint exposure was significant on pest-testing serum eorticosterone. The hippocampus lead concentrations of NS/L and S/L were (0. 4378±0. 1041 ) μg/g and ( 0. 4679±0. 1243 ) μg/g without significant differences ( F = 0. 298, P 0. 05). Conclusion Prenatal joint exposure to restraint stress and lead might increase the effects of single exposure on the spatial learning and memory ability and serum corticosterone level of offsprings, and the joint influence on cortieosterone level might be one of the reasons of further impairment of learning and memory.
Prenatal Maternal Stress in Relation to the Effects of Prenatal Lead Exposure on Toddler Cognitive Development

Abstract:Objective: To evaluate the effects of maternal lead exposure during pregnancy on toddler cognitive development and the potential effect modification by maternal stress.Methods: We conducted a prospective birth-cohort study in Shanghai from 2010 to 2012 and investigated 225 mother-infant pairs. The mothers were recruited in mid-to-late pregnancy and children were followed up until 24-36 months old. A self-administered Symptom Checklist-90-Revised Scale (SCL-90-R) was used to assess maternal emotional stress during pregnancy. Maternal whole blood lead levels were measured during gestational weeks 28-36. The toddlers' cognitive levels were assessed using the Gesell Development Scale. Multiple linear regression models were established to explore the main effects of prenatal lead exposure on toddlers' cognitive abilities and the modifying effects of maternal stress. Covariate information was collected through interviews, questionnaires and medical records.Results: The mean maternal blood lead concentration was 3.30 (95%CI: 3.05, 3.57) mu g/dL. After adjusting for relevant confounders, no significant associations of maternal blood lead concentrations with toddlers' cognitive levels were observed in all five domains of the Gesell scale (P>0.05). However, the interaction between prenatal maternal blood lead and stress was significant in the domains of adaptive behavior, language and social behavior. When stratified by maternal stress levels, compared with non-significant associations (P>0.05) among low (P1-P75) prenatal stress group, adverse associations between maternal blood lead concentrations (log10-transformed) and toddlers' cognitive levels were observed among high (P75-P100) prenatal stress group in the domains of language (beta =-33.82, 95%CI:-60.04,-7.59), social behavior (beta = -41.00, 95%CI:-63.11,-18.89) and adaptive behavior (beta =-17.93, 95%CI:-35.83,-0.03).Conclusion: Prenatal maternal stress may exacerbate the deleterious effects of prenatal exposure to lead on toddler cognitive development. Effects of developmental stress and lead (Pb) on corticosterone after chronic and acute stress, brain monoamines, and blood Pb levels in rats

Abstract:Despite restrictions, exposure to lead (Pb) continues. Moreover, exposure varies and is often higher in lower socioeconomic status (SES) families and remains a significant risk to cognitive development. Stress is another risk factor. Lower SES may be a proxy for stress in humans. When stress and Pb co-occur, risk may be increased. A few previous experiments have combined Pb with intermittent or acute stress but not with chronic stress. To determine if chronic developmental stress affects outcome in combination with Pb, we tested such effects on growth, organ weight, brain monoamines, and response to an acute stressor. Sprague Dawley rats were gavaged with Pb acetate (1 or 10 mg/kg) or vehicle every other day from postnatal day (P)4-29 and reared in standard or barren cages. Subsets were analyzed at different ages (P11, 19, 29). Chronic stress did not alter blood Pb levels but altered HPA axis response during early development whereas Pb did not. Pb treatment and rearing each altered organ-to-body weight ratios, most notably of thymus weights. Both Pb and rearing resulted in age- and region-dependent changes in serotonin and norepinephrine levels and in dopamine and serotonin turnover. The model introduced here may be useful for investigating the interaction of Pb and chronic developmental stress.

Abstract:This study used event-related potentials (ERPs) to assess effects of low-level prenatal lead exposure on auditory recognition memory in 2-month-old infants. Infants were divided into four groups according to cord-blood lead concentration: (1) <2.00 μ g/dL, (2) 2.00-2.99 μ g/dL, (3) 3.0-3.7 μ g/dL, and (4) ≥3.7 μ g/dL. The first group showed the normally expected differences in P2, P750, and late slow wave (LSW) amplitudes elicited by mothers' and strangers' voices. These differences were not observed for one or more ERP components in the other groups. Thus, there was electrophysiological evidence of poorer auditory recognition memory at 2 months with cord-blood lead ≥2.00 μ g/dL.

Abstract:Introduction: Neuro-developmental effects of in utero exposure to lead were documented in previous studies, however, limited studies focused on the association between lead exposure and maternal stress during pregnancy among pregnant women with non-occupational lead exposure. Methods: A total of 1931 pregnant women were investigated using stratified-cluster-sampling in Shanghai in 2010, and women were recruited during late pregnancy (28-36 gestational weeks). Maternal life event stress and emotional stress were assessed using “Event-Stress-Scale-for-Pregnant-Women” and “Symptom-Checklist-90-R-Scale” (SCL-90) respectively. Maternal whole blood lead concentration was determined. Other data on covariates were collected from maternal interviews and from medical records. Results: Mean maternal blood lead level was 4.33μg/dL (SD 1.71, range 0.8–14.8 μg/dL). After adjusting for potential confounders, a nonlinear relationship was observed between blood lead and total scores of SCL-90. When blood lead levels below 2.6μg/dL, the total scores of SCL-90 increased with increasing blood lead levels (β=4.19, p=0.027), however, when above 2.6μg/dL, the total scores of SCL-90 did not significantly increase with increasing blood lead levels (β=0.44 ,p=0.233), the trend difference was statistically significant (p=0.024).The similar patterns can be observed for the subscales of SCL-90 such as somatization, depression, anxiety and psychoticism. Conclusions: Our findings suggested a positive association between lead and emotional stress under low-lead-exposure conditions during pregnancy, but a non-significant association under high-lead-exposure conditions.

Abstract:Numerous observations in clinical and preclinical studies indicate that the developing brain is particular sensitive to lead (Pb)'s pernicious effects. However, the effect of gestation-only Pb exposure on cognitive functions at maturation has not been studied. We investigated the potential effects of three levels of Pb exposure (low, middle, and high Pb: 0.03%, 0.09%, and 0.27% of lead acetate-containing diets) at the gestational period on the spatial memory of young adult offspring by Morris water maze spatial learning and fixed location/visible platform tasks. Our results revealed that three levels of Pb exposure significantly impaired memory retrieval in male offspring, but only female offspring at low levels of Pb exposure showed impairment of memory retrieval. These impairments were not due to the gross disturbances in motor performance and in vision because these animals performed the fixed location/visible platform task as well as controls, indicating that the specific aspects of spatial learning/memory were impaired. These results suggest that exposure to Pb during the gestational period is sufficient to cause long-term learning/memory deficits in young adult offspring.

Abstract:Lead (Pb) exposure and prenatal stress (PS) during development are co-occurring risk factors with shared biological substrates. PS has been associated with transgenerational passage of altered behavioral phenotypes, whereas the transgenerational behavioral or biochemical consequences of Pb exposure, and modification of any such effects by PS, is unknown. The present study sought to determine whether Pb, PS, or combined Pb and PS exposures produced adverse transgenerational consequences on brain and behavior. Maternal Pb and PS exposures were carried out in F0 mice. Outside breeders were used at each subsequent breeding, producing four F1-F2 lineages: [F1 female-F2 female (FF), FM (male), MF, and MM]. F3 offspring were generated from each of these lineages and examined for outcomes previously found to be altered by Pb, PS, or combined Pb and PS in F1 offspring: behavioral performance [fixed-interval (FI) schedule of food reward, locomotor activity, and anxiety-like behavior], dopamine function [striatal expression of tyrosine hydroxylase (Th)], glucocorticoid receptor (GR) and plasma corticosterone, as well as brain-derived neurotrophic factor (BDNF) and total percent DNA methylation of and genes in the frontal cortex and hippocampus. Maternal F0 Pb exposure produced runting in F3 offspring. Considered across lineages, F3 females exhibited Pb-related alterations in behavior, striatal BDNF levels, frontal cortical total percentage DNA methylation levels and serum corticosterone levels, whereas F3 males showed Pb- and PS-related alterations in behavior and total percent DNA methylation of hippocampal . However, numerous lineage-specific effects were observed, most of greater magnitude than those observed across lineages, with outcomes differing by F3 sex. These findings support the possibility that exposures of previous generations to Pb or PS may influence the brain and behavior of future generations. Observed changes were sex-dependent, with F3 females showing multiple changes through Pb-exposed lineages. Lineage effects may occur through maternal responses to pregnancy, altered maternal behavior, epigenetic modifications, or a combination of mechanisms, but they have significant public health ramifications regardless of mechanism. https://doi.org/10.1289/EHP4977.

Abstract:The relationship between prenatal stress (PS) exposure and neurodevelopmental deficits remains inconclusive, especially when assessing the role of PS duration and timing and sex-dependent effects. This study explored a sex-specific association between the duration and timing of exposure and the outcomes of PS-induced neurotoxicity in hippocampal microstructure, synaptophysin expression, and neurobehavioral performance in rats. Pregnant rats were randomly assigned to control, PS-ML (exposed to prenatal restraint stress in the mid-to-late period of pregnancy), or PS-L (exposed in the late period of pregnancy) groups, and offspring in each group were divided into two subgroups by sex. Surface-righting reflex test, cliff avoidance test and Morris water maze test showed that neurodevelopmental levels were reduced in PS-treated pups but without significant sex differences. On postnatal day 22, hippocampal microstructure was examined by electron microscopy, and the expression of hippocampal synaptophysin was assessed by western blot. Abnormal ultrastructural appearance of hippocampal neurons and myelin sheaths, more degenerating neurons and higher G-ratios were found in young PS-ML and PS-L rats as well as reduced expression of hippocampal synaptophysin, although PS-ML pups were more greatly affected than PS-L, with males showing slightly greater impairments than females. These findings suggest that hippocampal hypo-myelination and decreased synaptophysin expression in neurodevelopment may be a duration and time-dependent effect of prenatal stress exposure, modified slightly by sex.
Low-Level Prenatal Lead Exposure and Neurobehavioral Development of Children in the First Year of Life: A Prospective Study in Shanghai

Abstract:We used a prospective study design to assess the effects of prenatal low-level lead exposure on the development of urban, inner-city children in Shanghai. Umbilical cord blood samples were consecutively collected from 605 live newborns. Two hundred and fifty-seven samples were excluded from the study due to clotting. Lead levels were determined on 348 cord blood samples. The geometric mean was 9.2 μg/dl. Based on their cord blood lead levels, infants were classified into two exposure groups: 104 in a relatively low lead group (lead levels ≤30 percentile), and 104 in a relatively high lead group (lead levels ≥70 percentile). Seventy-five subjects failed to complete the study, and 133 babies were included in the final cohort: 69 babies in the high lead group and 64 in the low lead group. At 3, 6, and 12 months, the Bayley Scales of Infant Development were administered and capillary blood lead levels were measured. Detailed information was obtained on a wide range of variables relevant to infant development. At all three ages, the Mental Development Index (MDI) scores, adjusted for confounders, were inversely related to the infants' cord blood lead levels. The difference of the mean adjusted MDI scores between low and high lead groups was 3.4 at 3 months, 6.3 at 6 months, and 5.2 at 12 months of age. These differences were statistically significant at all time points. No significant association between cord blood lead levels and the Psychomotor Development Index (PDI) scores was detected at all three visits after adjustment for confounders. Postnatal lead levels were unrelated to concurrent developmental status. We conclude that prenatal low-level lead exposure, which is relatively common in Shanghai, is associated with an adverse developmental impact on children through the first year of life.

Abstract:Objective To study the effects of prenatal exposure to low level lead on the physical and neuro-behavioral development of mice and the interaction between lead and zinc. Methods Kunming pregnant mice were provided with drinking water containing lead, zinc and lead-zinc jointly and respectively. The body weight of maternal mice, offspring's survival rate and body weight were observed. Neuro-behavioral development of offspring was assessed by observation of offspring's behavior performance. The levels of dopamine (DA), 5-hydroxytryptamine (5-HT) and malonaldehyde (MDA) in offspring's brains were measured. Results At the doses of lead and zinc which did not effect the increase of maternal body weight during pregnancy, the lead exposure group showed low survival rate, growth retardation and delay of physiological development and neonatal reflexes in the offspring, the levels of DA and MDA in offspring's brains increased compared with the control group. Offspring's low survival rate and growth retardation couldn't be antagonized by zinc, but equal mole as lead of zinc could inhibit the harmful effects on neuro-reflex development. Zinc could also inhibit the increase of DA and MDA levels in offspring's brains induced by lead . However joint exposure to lead and half dosage as much as lead of zinc could deteriorate the effects on delay of physical, neuro-behavioral development and accelerate the DA and 5-HT decompose compared with that in the lead exposure group. Conclusion The harmful effects on offspring's growth and neuro-behavioral development could be induced by prenatal exposure to low level of lead. The types of joint-effect of lead and zinc were different according to the concentration of zinc, which indicated that the zinc-lead interaction was very complicated and it should be very prudent to antagonize lead toxicity using zinc.

This study estab-lished rat models with prenatal single and combined exposure to Ps and Pb. We investigated the effects of such prenatal single and combined exposure on hippocampal structures using morphological analyses, on learning/ memory using the Morris-water-maze test, and on fecal microbiota using 16S rRNA sequencing. The mediating roles of gut microbiota were analyzed using the bootstrap method. The study found both single and combined exposure affected hippocampal ultra-structures and spatial learning/memory, and the most significant impair-ments were observed in the Pb+Ps group. Prenatal Pb+Ps co-exposure decreased fecal microbial alpha/beta-diversity. Significantly lower levels of B/F-ratio, class-Bacteroidia, order-Bacteroidales, and family-S24-7, and significantly higher levels of class-Bacilli, order-Lactobacillales, family-Lactobacillaceae, and genus -Lactobacillus were observed in the co-exposure group, compared with the controls. Increased relative abundances of genus-Helicobacter mediated the detrimental effect of prenatal Ps+Pb co-exposure on learning/memory [beta (95%CI) for the total and indirect effects: -10.70 (-19.19,-2.21) and -4.65(-11.07,-1.85)], accounting for 43.47% of the total effect. As a result, increased relative abundances of genus -Lactobacillus alleviated the adverse effects of the co-exposure on learning/memory, and the alleviation effect accounted for 44.55% of the direct effect [beta (95% CI) for the direct and indirect effects: -0.28(-0.48,-0.08) and 0.13(0.01, 0.41)]. This study suggested that prenatal combined exposure to Pb and Ps induced more impairments in offspring gut microbiota and neuro-development than single exposure, and alterations in fecal microbiome may mediate the developmental neurotoxicity induced by such prenatal co-exposure.
The Effect of Lead Exposure During Early Stage of Life on Expression of Metabotropic Glutamate Receptor Subtype 5

Abstract:Objective The effect of lead exposure on children is consistently associated with intellectual and other neurologic deficits. However the exact mechanism by which Pb2+ exerts toxic effects on developmental central nervous system remains unknown. Our group has found by gene-chip test that the expression of metabotropic glutamate receptor subtype 5 (mGluR5) mRNA was changed by lead exposure. The present study aimed to examine the effects of different level of lead exposure on the expression of mGluR5 in gestation and lactation. Methods Sprague-Dawley rats were exposed to lead acetate during gestation and lactation. Three concentrations of 0.05% ,0.2% ,and 0.5% lead acetate were applied and considered as low,middle and high exposure group respectively. The Pb levels of blood and hippocampus of pups were analyzed at weaning to evaluate the actual lead content at the end of the exposure. The impact of lead exposure on the expression of mGluR5 mRNA and protein in hippocampal tissue of pups was investigated by quantitative real-time polymerase chain reaction (PCR) and Western blot. The potential role of the expression of mGluR5 mRNA and protein in lead neurotoxicity were discussed. Results The levels of lead in blood and hippocampus from lead-exposed rats were significantly higher than those in the controls and positively related to the degree of lead exposure. The results of real-time PCR and Western blot showed that exposure to lead acetate decreased the expression of mGluR5 mRNA and protein with a dose-dependent manner. Conclusions Hippocampal mGluR5 might be involved in lead-induced neurotoxicity.
Effects of soil lead exposure and land use characteristics on neurodevelopment among children under 3 years of age in northern Taiwan

Abstract:<p>Lead (Pb) exposure increases the risks of neurodevelopmental disorders in children. Child-specific activities and land use scenarios may lead to elevated opportunities for Pb exposure through the soil. Therefore, we investigated hair and fingernail Pb concentrations among young children in northern Taiwan, in relation to soil Pb pollution and land use characteristics. We also explored the effect of the Pb exposure burden and land use scenarios on neurobehavioral development. In total, 139 healthy children under 3 years of age were recruited in October 2011 to April 2014. Pb levels in hair and fingernail samples were determined using an inductively coupled plasma/mass spectrometer. Pb concentrations in soils and land use types surrounding the children's homes were accessed by a geographic information system to identify any associations with hair Pb levels. The Bayley Scales of Infant and Toddler Development (Bayley-III) were used to evaluate the cognitive, language, and motor development of the children. A multivariable regression model was performed to assess the effects of soil Pb levels and land-use status on Pb exposure in children, as well as associations of Pb exposure and land-use scenarios with neurodevelopmental abilities. Geometric mean Pb concentrations in hair, fingernails, and soil were 2.9±4.8 μg/g, 0.8±5.1 μg/g, and 20.8±4.3 mg/kg, respectively. The multivariable analysis indicated that soil Pb concentrations and green areas around residences had potential links with Pb exposure among children in northern Taiwan. Hair Pb concentrations were negatively associated with expressive language scores. Soil Pb exposure was positively associated with hair Pb concentrations. Land use types around the children's homes in northern Taiwan were associated with their neurodevelopment. Increased green areas were negatively associated with hair Pb concentrations. Living near a highway may have had negative impacts on gross motor scores. A healthy residence can avoid potential health risks for children during their early life.</p>

Abstract:Background A complete explanation of the mechanisms by which Pb 2+ exerts toxic effects on developmental central nervous system remains unknown. Glutamate is critical to the developing brain through various subtypes of ionotropic or metabotropic glutamate receptors (mGluRs). Ionotropic N-methyl-D-aspartate receptors have been considered as a principal target in lead-induced neurotoxicity. The relationship between mGluR3 / mGluR7 and synaptic plasticity had been verified by many recent studies. The present study aimed to examine the role of mGluR3 / mGluR7 in lead-induced neurotoxicity. Methods Twenty-four adult and female rats were randomly selected and placed on control or 0.2% lead acetate during gestation and lactation. Blood lead and hippocampal lead levels of pups were analyzed at weaning to evaluate the actual lead content at the end of the exposure. Impairments of short -term memory and long-term memory of pups were assessed by tests using Morris water maze and by detection of hippocampal ultrastructural alterations on electron microscopy. The impact of lead exposure on mGluR3 and mGluR7 mRNA expression in hippocampal tissue of pups were investigated by quantitative real-time polymerase chain reaction and its potential role in lead neurotoxicity were discussed. Results Lead levels of blood and hippocampi in the lead-exposed rats were significantly higher than those in the controls ( P < 0.001). In tests using Morris Water Maze, the overall decrease in goal latency and swimming distance was taken to indicate that controls had shorter latencies and distance than lead-exposed rats ( P = 0.001 and P < 0.001 by repeated-measures analysis of variance). On transmission electron microscopy neuronal ultrastructural alterations were observed and the results of real-time polymerase chain reaction showed that exposure to 0.2% lead acetate did not substantially change gene expression of mGluR3 and mGluR7 mRNA compared with controls. Conclusion Exposure to lead before and after birth can damage short-term and long-term memory ability of young rats and hippocampal ultrastructure. However, the current study does not provide evidence that the expression of rat hippocampal mGluR3 and mGluR7 can be altered by systemic administration of lead during gestation and lactation, which are informative for the field of lead-induced developmental neurotoxicity noting that it seems not to be worthwhile to include mGluR3 and mGluR7 in future studies.
Effects of Prenatal Exposure to Lead on Learning and Memory of Rats' Offspring

Abstract:Objective To explore the effects of prenatal exposure to lead on learning and memory of rats' offspring. Methods The pregnant rats were randomly divided into 4 groups provided with double evaporated water, 50, 100 and 200 mg/L lead acetate solution via drinking water respectively. The lead-exposure period for exposure groups was limited from the 1st day after pregnancy to the 20th day when the offspring began to be weaned. For learning ability 20-day old offspring were tested by water maze. For active learning and memory ability, the 20-day old, 40-day old and 60-day old offspring were tested by Y maze. Results The frequency of the mistakes in water maze made by offspring increased with the increase of the prenatal lead-exposure doses of their mothers and showed significantly higher levels in 100 and 200 mg/L groups compared with that of control group (P0.05 or P0.01). Only the 200 mg/L group showed significantly longer incubation period of escape compared with that of control group (P0.05). The qualified rates of 0-min, 10-min and 24-h escape of Y maze test for each dose group revealed the lowest levels when the offsping were 20-day old, and increased gradually after the offspring were suspended for exposure to lead and reached the peaks when they were 60-day old. The qualified rates of 0-min escape for each dose group showed no significant differences compared with that of control group when they were 60-day old (P0.05). But the qualified rates of 0-min and 24-h escape for 100 and 200 mg/L groups showed sigificantly lower levels compared with that of control group when they were 60-day old (P0.05 or P0.01). Conclusion Prenatal exposure to lead impaired the learning and memory ability first, then the locomotion ability of offspring. After the offspring were suspended to be exposed to lead, the working memory ability could be recovered better, but the reference memory ability and learning ability couldn't be recovered quite well.
