P3‐133: COMT VAL158MET AND WORKING MEMORY: MAIN EFFECTS AND INTERACTION WITH GENDER

Abstract:The effects of Catechol-O-methyl transferase (COMT) Val158Met (rs4680) polymorphism on cognition and brain function are both markedly sexually dimorphic. In this study, we explore the role of COMT genotype and the interaction between COMT genotype and gender on cortical activity and task-based functional connectivity during working memory task. 121 participants (50Male/71Female, 44 Met carriers/77 Val homozygotes) who aged 52–82 years old, scored 24 or higher on MMSE, had at least 6 years and successfully completed N-Back task during fMRI were included. The participants were divided into 4 groups based on their genotype and gender. Task-related activation and background connectivity were compared across groups to explore the main effects and interaction between genotype and gender during 2-back task. With age, years of education and APOE genotype included as covariates, results were statistically significant at alphasim corrected p < 0.05. No significant differences on education, MMSE score and reaction time between groups. While male were older and performed better than female in accuracy. Compared to Val homozygotes group, the Met carriers group produced significantly greater activation of left inferior temporal gyrus and left inferior frontal gyrus (IFG.L), with activation in IFG.L was significantly positive related to accuracy in Female/Val. Stronger activation in left angular (ANG.L) was also found for male groups compared to female, with higher activation in this region correlated with better accuracy mostly in female. The interaction effects were found in deactivation regions: right precuneus and superior occipital gyrus (SOG.R), with Male/Met exhibited the highest activation. Further analyses revealed background connectivity from group differences regions also influenced by main and interaction effects of genotype and gender. And IFG.L- left calcarine connectivity, SOG.R-right supramarginal, ANG.L-right superior temporal gyrus connectivity were negative correlated with accuracy in Female/Val. Both task-related activation and background connectivity in working memory were influenced by COMT genotype. And this effect was modified by gender, with female were more susceptible.
