Abstract:It has been reported that the catechol-O-methyltransferase(COMT) Val158Met genetic polymorphism,a functional polymorphism that may affect dopamine metabolism,is associated with the prefrontal cognitive function.To explore the change of working memory ability in healthy young adults,a continuous 3-back numerical working memory task for an hour was performed for 12 blocks among different COMT genotypes of subjects.RFLP-PCR was used for the genetyping of COMT Val158Met in 115 healthy young men.Twenty-one individuals were chosen as subjects according to different COMT genotypes,and P3 visual event-related potentials(ERP) evoked from the 3-back task were utilized to observe the relationship between this COMT polymorphism and cortical physiology in the task.In the 3-back task,the participants responded to the present stimulus if it was identical to the stimulus three trials previously.The ERP was recorded with the 3-back task for 5 min,for a total of 12 blocks.The duration of the experimental task was 1 h.The visual stimuli were generated and presented with the NeuroScan STIM software used.The results showed that the frequency of Met/Met in this study group of participants was similar to that of the Japanese population(i.e.about 4%) but much lower than the frequency of the British population or the American population.Clear differences in the structure of genotypic frequency between this experimental group and British[x~2(1) = 34.2,p<.005]or American[x~2(1) = 48.6,p<.005]populations were observed,but no significant difference in structure was found between our experimental group and the Japanese population[x~2(1) = 2.8,p>.05].The distribution of the COMT allele frequency followed the same trend in the structure of COMT gene polymorphisms (p<.01).Val/Met heterozygote subjects had a significantly lower P3 amplitude than bearing the Val homozygote(p<.01).Met/ Met homozygote had intermediate levels of P3 amplitude.Val/Met Heterozygote subjects were associated with the poorest performance of working memory.Relatively low COMT activity,as a result of the Met allele,improved the local cortical stabilization.The high COMT activity encoded by the Val158 allele resulted in an enhanced ability to update the contents of working memory with new information.P3 amplitude was believed to be more closely related to the intensity of processing.There was the appearance of decreased P3 amplitude in the parietal cortex when the task became more complex.Val/Met heterozygote subjects had a significantly lower P3 amplitude than other subjects because Val/Met heterozygote subjects needed more resource engagement in the 3-back task.The stability and flexibility of working memory were regulated by genetic variation.Our findings further support the notion that the COMT Val158Met polymorphism may play a role in cognitive function.It is clear that there is an association between the COMT polymorphism and visual ERP.This association,however,might be affected by ethnicity,testing methods,and different types of stimuli to some different extents.
