Abstract:We report a one-step hydrothermal method for the preparation of fluorescent molybdenum disulfide quantum dots (MoS2 QDs) and demonstrate the feasibility of fluorescent "turn-off-on" sensing of adenosine triphosphate (ATP) by using the MoS2 QDs. MoS2 QDs having strong blue-green fluorescence at 506 nm and good water-solubility were successfully synthesized by using ammonium tetrathiomolybdate as a single precursor. The fluorescence of MoS2 QDs was first quenched by Fe3+ through the formation of a MoS2 QDs/Fe3+ complex. ATP with the ability to coordinate with Fe3+ caused the dissociation of MoS2 QDs/Fe3+, resulting in the final release of MoS2 QDs and the recovery of fluorescence through a one-step competitive chelating process that took only 10 min to reach equilibrium at room temperature (RT). Facile and rapid sensing of Fe3+ and ATP could thus be achieved through the fluorescent "turn-off-on" strategy. Good linear relationships were obtained over the concentration ranges of 0-200 μM for Fe3+ and 0-140 μM for ATP with a lowest detectable concentration of 5 μM for ATP. Satisfactory results were obtained when the method was applied to a standard addition recovery trial of ATP in human serum samples. No complex surface modification during the preparation or detection process was needed based on the fluorescent "turn-off-on" method of the MoS2 QDs, which suggests its great potential in fluorescent sensing.
Red-emission carbon dots-quercetin systems as ratiometric fluorescent nanoprobes towards Zn2+ and adenosine triphosphate

Abstract:Carbon dots (CDs) emitting red fluorescence (610 nm) were synthesized by solvent thermal treatment of p-phenylenediamine in toluene. Upon 440 nm excitation, quercetin (QCT) alone endowed slight effects on the red fluorescence of CDs. Once Zn2+ was further introduced, the QCT-Zn2+ complex was quickly formed. This complex absorbs excitation light and emits bright green fluorescence at 480 nm. The red fluorescence of CDs was greatly quenched owing to the inner-filter effect. The ratio of fluorescence intensity at 480 nm and 610 nm (I480/I610) gradually increases with increasing concentration (c) of Zn2+. Al3+ exhibits the same phenomen like Zn2+. Fluoride ions form a more stable complex with Al3+ than QCT-Al3+ complex but have a negligible effect on the QCT-Zn2+ complex. The possible interference of Al3+ on Zn2+ can thus be avoided by adding certain amount of F-. The CD-QCT-F- system was constructed as a ratio-metric fluorescent nanoprobe toward Zn2+ with determination range of 0.14-30 μM and limit of detection (LOD) of 0.14 μM. Due to the stronger affinity of adenosine triphosphate (ATP) to Zn2+ than QCT, the I480/I610 value of CD-QCT-F--Zn2+ system gradually decreases with increasing cATP. The ratiometric fluorescent nanoprobe toward ATP was established with detection ranges of 0.55-10 and 10-35 μM and a LOD of 0.55 μM. The above two probes enable the quantitative determination of Zn2+ and ATP in tap and lake water samples with satisfactory recoveries. Abstract:The design and fabrication of high visible-light activated photoelectrode are essential to precisely detect biomolecule in biological system. Herein, an ultrasensitive photoelectrochemical (PEC) aptasensor for specific recognition of adenosine is established based on carbon dots sensitized-amorphous molybdenum sulfide (a-MoSx/CDs) photoanode and dual amplification strategy. The heterostructured photoanode achieved by sequential electrodeposition reveals significantly boosted photocurrent with good stability and repeatability under visible light illumination, giving the credit to highly activated visible light absorption, uniform coverage and good electric contact to the underlying substrate, as well as the energy-band alignment between the two components. By stepwisely immobilizing complementary DNA probe (NH2-DNA) and adenosine aptamer (Apt), followed by methylene blue (MB) binding with the guanine base on Apt, a dual amplified self-powered PEC aptasensor for adenosine detection is constructed. Based on the co-sensitization effect of CDs and MB, ultrasensitive and high-affinitive determination of adenosine is realized over the concentration range of 0.01 nM-1000 nM at 0 V (vs. SCE), with satisfactory stability and reproducibility. The detection limit is as low as 3.3 pM, demonstrating a performance even surpassing most of the sensors reported so far. The prospective application of the co-sensitized a-MoSx photoanode for ultrasensitive aptasensing is highlighted in this work.

Abstract:A simple and sensitive colorimetric aptasensor for rapid and facile detection of adenosine triphosphate (ATP) has been demonstrated here based on aptamer-based hairpin probes and unmodified gold nanoparticles (AuNPs). The hairpin probe is constructed by adding another five nucleotides to the 5′-end of an anti-ATP aptamer which can hybridize to nucleotides at the 3′-end of the aptamer, forming a hairpin-shaped structure. In the absence of ATP, the hairpin probes are rigid, and the AuNPs are susceptible to salt-induced aggregation. Conversely, upon binding with target ATP, the hairpin probes undergo conformational changes, forming aptamer-ATP complexes and exposing flexible ends which coat the surface of AuNPs to inhibit their aggregation in the high salt solution. Subsequently, a blue-to-red color change can be recognized by the naked eye. The aptasensor achieved selective responses toward ATP with a detection limit of 0.1μM, and exhibited high-quality detection performance in biological samples. In addition, this detection method is simple, rapid and cost-effective, holding great potential for further applications in point-of-care research.

Abstract:A novel nanoprobe was prepared by encapsulating carbon dots (CDs) and gold nanoclusters (AuNCs) into zeolitic imidazolate framework-8 (ZIF-8) for sensitive detecting adenosine triphosphate (ATP). Under excitation at 360 nm, the obtained CDs/ nanoprobe exhibits dual-emissions at 469 nm and 660 nm, respectively, corresponding to the fluorescence emission of CDs and the aggregation-induced emission enhancement (AIEE) of AuNCs. The framework of ZIF-8 in this probe can be degraded by ATP due to the coordination competition of ATP and 2-Methylimidazole towards zinc ion (Zn 2+ ), resulting in the release of CDs and AuNCs. The following dispersion of CDs would improve efficiencies of the fluorescence excitation and the consequent emission of CDs. On the contrary, the AIEE of AuNCs would be decreased spontaneously after the AuNCs originally restricted in ZIF-8 were allowed to escape. The intensity ratio of fluorescence at 469 nm to that at 660 nm ( I 469 / I 660 ) was conveniently employed as the response signal for representing the amount of ATP. This nanoprobe exhibits excellent sensitivity and selectivity toward ATP, with a limit of detection (LOD) of 0.061 μM. Besides, low cytotoxicity of this nanoprobe facilitates its application as a fluorescent indicator in fluorescence imaging of living cells. Encapsulating two types of fluorescent nanomaterials by a degradable ZIF-8 structure makes the ratiometric fluorescence response of the nanocomposite probe towards the target analyte that destroys the ZIF-8 structure possible, and simplifies the application of the probe.

Abstract:An "off-on" electrochemiluminescence (ECL) strategy was constructed for highly sensitive and selective detection of adenosine 5'-triphosphate (ATP) with a quantum dots (QDs) modified electrode and a DNAzyme signal probe. The immobilized QDs were functionalized with a DNA sequence (DNA1) and then aptamer for recognition of target analyte. The signal probe was prepared by assembling another DNA sequence (DNA2) and G-quadruplex on gold nanoparticle via Au-S chemistry, which was used to bind the probe to electrode surface through a hybridization reaction with aptamer and hemin for forming G-quadruplex/hemin DNAzyme, respectively. Upon the sandwich hybridization of DNA1-aptamer-DNA2, the signal probe could be captured on the aptasensor to catalyze the reduction of dissolved oxygen, the coreactant for cathodic ECL emission of QDs, leading to a decrease of ECL intensity and thus the "off" state. In the presence of target, its recognition by aptamer led to the release of aptamer from electrode surface and decreased the amount of captured signal probe, thus the ECL emission was in its "on" state. The "off-on" strategy resulted from the target-induced structure switching could be used for specific detection of ATP with a linear range of 8-2000 nM and a detection limit of 7.6 nM. The proposed aptasensor could be successfully applied in the ECL detection of ATP in human serum. This method could resist environmental interfering agents and be extended for sensitive and reliable detection of a wide range of analytes in complex sample.
Cu-Deficient CuInSe Quantum Dots for “Turn-On” Detection of Adenosine Triphosphate in Living Cells

Abstract:Copper indium selenide (CISe) quantum dots (QDs) are a class of extremely appealing candidates for various biological applications due to their composition- and size-dependent photoluminescence and low toxicity. However, the synthesis of high-quality water-soluble CISe QDs with controlled composition in the aqueous phase is still a great challenge, which limits their biological applications. Moreover, the effects of the copper content in CISe QDs on the fluorescence and the corresponding mechanism are still subject to debate. Herein, a composition-tunable aqueous synthesis of high-quality Cu-deficient CISe QDs was presented, and the effects of the copper content on the fluorescence of CISe QDs was studied by the time-resolved fluorescence. The composition of CISe QDs could be flexibly controlled by manipulating the reaction temperature, the reaction time, and the Cu/In precursor ratio. The time-resolved fluorescence of CISe QDs with different Cu/In ratios suggested that the donor–acceptor pair (DAP) recombination was enhanced gradually with the increase of copper vacancies, resulting in the enhancement of fluorescence. However, if copper vacancies achieved an excessive concentration, the fluorescence would decrease possibly due to Auger recombination. The optimized CISe QDs with the maximum quantum yield of 10.3% were obtained with the Cu/In ratio, reaction temperature, and reaction time of 1/5, 170 °C, and 1 h, respectively. Based on the intense emission of the Cu-deficient CISe QDs, a highly sensitive "turn-on" fluorescent probe was developed for the detection of adenosine triphosphate (ATP) in living cells with a limit of detection of 56 nM. Abstract:We report a simple, fast and selective colorimetric assay of adenosine triphosphate (ATP) using unmodified gold nanoparticles (AuNPs) as probes and metal ions as cross-linkers. ATP can be assembled onto the surface of AuNPs through interaction between the electron-rich nitrogen atoms and the electron-deficient surface of AuNPs. Accordingly, Cu2+ ions induce a change in the color and UV/Vis absorbance of AuNPs by coordinating to the triphosphate groups and a ring nitrogen of ATP. A detection limit of 50 nM was achieved, which is comparable to or lower than that achievable by the currently used electrochemical, spectroscopic or chromatographic methods. The theoretical simplicity and high selectivity reported herein demonstrated that AuNPs-based colorimetric assay could be applied in a wide variety of fields by rationally designing the surface chemistry of AuNPs. In addition, our results indicate that ATP-modified AuNPs are less stable in Cu2+, Cd2(+) or Zn2+-containing solutions due to the formation of the corresponding dimeric metal-ATP complexes.

Abstract:In this work, based on purifying the analysis system by magnetic separation, we constructed a turn-on fluorescent assay for highly sensitive detection of possible biomarkers, employing adenosine as the model target analyte. Aptamer labeled-carbon dots (Apt-CDs) was applied as fluorescence probes as well as selective recognition elements, while Fe3O4@polypyrrole (Fe3O4@PPY) was used as fluorescence quenching and magnetic separating materials for system purification. In absence of adenosine, the Apt-CDs were attached to the surface of PPY through pi-pi stacking and hydrophobic interaction and its fluorescence was quenched by Fe3O4@PPY. In presence of adenosine, the aptamer could specifically bind with adenosine to form a three dimensional Apt-CDs-adenosine complex and detach from PPY, leading to the fluorescence recovery of Apt-CDs. More importantly, with analysis system purification via twice convenient magnetic separations of free CDs and Apt-CDs-adenosine complex respectively, the detection limit could be reduced from 8 nmol L-1 to 0.15 nmol L-1. Two-Dimension (2D) Cu-MOFs/aptamer Nanoprobe for In Situ ATP Imaging in Living Cells

Abstract:Monitoring the intracellular adenosine triphosphate (ATP) level is vital for elaborating its function in physiological states. However, the intracellular fluorescence sensing of ATP using ATP aptamer remains difficult owing to non-target displacement and susceptibility toward enzymatic degradation. Herein, by combining 2D Cu-MOF nanosheets and FAM labelled ATP aptamer, we developed a fluorescent Cu-MOFs/aptamer nanoprobe to image and sense intracellular ATP. This nanoprobe reveals a very low fluorescence intensity because of the excellent quenching efficiency of 2D Cu-MOF nanosheets. The presence of ATP was capable to dissociate the FAM-aptamer from Cu-MOF nanosheets and resulted in an intense fluorescence signal. The Cu-MOFs/aptamer nanoprobe enables highly sensitive and selective measurement of ATP level ranging from 10 μM to 800 μM with a detection limit of 4.24 μM. This nanosystem also further realized in situ detection of the undulation of ATP trigged by drug stimulation, depending on the selective delivery of the nanoprobe and attractive capability of resisting nonspecific displacement. Signal-On Photoelectrochemical Aptasensor For Adenosine Triphosphate Detection Based On Sensitization Effect Of Cds:Mn@Ru(Bpy)(2)(Dcbpy) Nanocomposites

Abstract:A novel photoelectrochemical aptasensor for adenosine triphosphate (ATP) detection was developed by introducing inorganic organic photoactive nanocomposites as sensitization agents to achieve significant signal amplification. Specifically, a TiO2/Au hybrid structure was first fabricated by decorating Au nanoparticles on the surface of TiO2 film, which then was used as the photoelectrochemical matrix for the immobilization of ATP aptamer probes; amino-functionalized CdS:Mn (CdS:Mn-NH2) nanocrystals covalently bound with Ru(bpy)(2)(dcbpy)(2+) (bpy = 2,2'-bipyridine; dcbpy = 2,2'-bipyridine-4,4'-dicarboxylic acid) to form CdS:Mn@Ru(bpy)(2)(dcbpy) photoactive nanocomposites, which were employed as signal amplification element labeling on the terminal of ATP aptamer probes. The ATP detection was performed via the photocurrent variation produced by the conformation change of ATP aptamer probes after specifically binding with ATP molecules. Before incubation with ATP, the ATP aptamer probe hybridized with its partly complementary DNA to form a rodlike double helix, which made the labeled sensitization agents of CdS:Mn@Ru(bpy)(2)(dcbpy) far from the TiO2/Au electrode surface, resulting in depressed sensitization effect. In the presence of ATP, the ATP aptamer probe specifically bound with ATP molecule to form a G-quadruplex structure, which made the CdS:Mn@Ru(bpy)(2)(dcbpy) very close to the TiO2/Au electrode surface, resulting in noticeably enhanced photocurrent intensity due to full activation of the sensitization effect. Accordingly, a signal-on photoelectrochemical aptasensor was constructed. The designed aptasensor exhibited a wide linear range from 0.5 pM to 5 nM with a low detection limit of 0.18 pM for ATP detection.
COLORIMETRIC DETERMINATION OF ADENOSINE IN URINE USING BIOTIN MODIFIED APTAMER AND GOLD NANOPARTICLES

Abstract:Adenosine can be a possible biomarker for monitoring the progress of lung cancer. Here, a sensitive and easily operated colorimetric methods based on aptasensor for the detection of adenosine has been developed. Gold nanoparticles (AuNPs) will aggregate in high-salt solutions, which causes color change, and single-stranded DNA will prevent this from happening. Based on changes in solution color, adenosine can be quantitatively determined. Besides a home-made bionic electronic-eye (E-eye) system was developed as a portable in-situ detection platform. Two different aptamers were used for comparison for adenosine detection. The linear correlation existed between 5.0-60.0μM with a detection limit of 0.17μM in water with modified aptamer. Moreover, the performance of the method was validated by testing the artificial urine samples with the linear range from 5.0 to 50.0μM and a detection limit of 0.48μM in artificial urine. This simple and cost-effective approach indicating its great potential for point-of-care applications.

Abstract:Adenosine triphosphate (ATP) is a major chemical energy carrier in organisms and is involved in numerous biological processes. ATP levels are associated with many diseases, cell viability, and food freshness. Thus, it has become an important biomarker. Many strategies have been used to detect ATP. However, the problems of difficult-to-prepare materials, too much dependence on instruments, and complicated processes restrict the application of these methods. In this study, we proposed a novel ATP detection sensor. The method is based on the fluorescence enhancement effect of dimeric G-quadruplex (Di-G4) on thioflavin T (ThT). First, the cleavage of Di-G4 by S1 nuclease decreases system fluorescence. However, it can be recovered by increases in ATP concentrations, which act as an inhibitor of S1 nuclease. Under the optimized conditions, a good linear relationship was observed between fluorescence intensity and ATP concentrations within the range of 0.5-120 µM. The detection limit was 245 nM. The method was utilized to measure the ATP content in apples and compared with ATP assay kits, resulting in satisfactory results.
Aptamer-based flares hybridized with single-stranded DNA-conjugated MoS2 nanosheets for ratiometric fluorescence sensing and imaging of potassium ions and adenosine triphosphate in human fluids and living cells

Abstract:Addressing the limitations observed in previous studies, where the quantitative range of nanoprobes for detecting K+ and adenosine triphosphate (ATP) did not cover concentrations found within living cells, the present study aimed to develop ratiometric nanoprobes that can accurately sense changes in K+ and ATP levels in living cells and quantify them in human fluids. The proposed nanoprobes consisted of recognition flares modified with 6-carboxyfluorescein (FAM) and 5-carboxytetramethylrhodamine (TAMRA), along with thiolate single-stranded DNA (ssDNA) and molybdenum disulfide nanosheets (MoS2 NSs). The thiolate ssDNA acts as a linker between the flares and the MoS2 NSs, directly forming a functional nanostructure at room temperature. The direct conjugation of labeled flares to the MoS2 NSs simplifies the fabrication process. In the absence of K+ and ATP, the hybridization of flares and thiolate ssDNA caused FAM to move away from TAMRA, suppressing the fluorescence resonance energy transfer (FRET) process. However, upon the introduction of K+ and ATP, the flares undergo a structural transformation via the formation of G-quadruplex formation and the generation of hairpin-shaped structures, respectively. This structural change leads to the release of the flares from the ssDNA-conjugated nanosheet surface. The release of the flares brings FAM and TAMRA into close proximity, allowing FRET to occur, leading to FRET and static quenching. By monitoring the ratio between the fluorescence intensities of FAM and TAMRA, the concentration of K+ (5-100 mM) and ATP (0.3-5 mM) can be accurately determined by the proposed nanoprobes. The advantages of these nanoprobes lie in their ability to provide ratiometric measurements, which enhance the accuracy and reliability of the quantification process. The proposed nanoprobes offer potential applications as ratiometric imaging probes for monitoring K+ and ATP-related reactions in living cells, providing valuable insights into cellular processes. Additionally, they can be employed for determining the levels of K+ and ATP in human fluids, offering potential diagnostic applications in various clinical settings.

Abstract:Abstract Background Ex vivo and in vivo detection and imaging of adenosine triphosphate (ATP) is critically important for the diagnosis and treatment of diseases, which still remains challenges up to present. Results We herein demonstrate that ATP could be fluorescently detected and imaged ex vivo and in vivo. In particular, we fabricate a kind of fluorescent ATP probes, which are made of titanium carbide (TC) nanosheets modified with the ROX-tagged ATP-aptamer (TC/Apt). In the constructed TC/Apt, TC shows superior quenching efficiency against ROX (e.g., ~ 97%). While in the presence of ATP, ROX-tagged aptamer is released from TC surface, leading to the recovery of fluorescence of ROX under the 545-nm excitation. Consequently, a wide dynamic range from 1 μM to 1.5 mM ATP and a high sensitivity with a limit of detection (LOD) down to 0.2 μM ATP can be readily achieved by the prepared TC/Apt. We further demonstrate that the as-prepared TC/Apt probe is feasible for accurate discrimination of ATP in different samples including living cells, body fluids (e.g., mouse serum, mouse urine and human serum) and mouse tumor models. Conclusions Fluorescence detection and imaging of ATP could be readily achieved in living cells, body fluids (e.g., urine and serum), as well as mouse tumor model through a new kind of fluorescent ATP nanoprobes, offering new powerful tools for the treatment of diseases related to abnormal fluctuation of ATP concentration.

Abstract:A DNAzyme-based colorimetric aptasensor has been fabricated for fast readout and facile detection of adenosine triphosphate (ATP). The sensor consists of an ATP-specific aptamer flanked with cytosine-rich arms at both ends (50 & 30) and a peroxidase mimicking DNAzyme which were complexed together to form a functional triple-helix molecular switch (THMS). The DNAzyme strand serves as a signal transduction probe (STP) in the molecular switch. In the presence of ATP, the aptamer binds to it and the THMS complex disassembles. Subsequently the released DNAzyme sequence can form a catalytic G-quadruplex DNAzyme leading to obvious color change. This sensor provides an efficient and stable diagnostic system whilst conserving the inherent catalytic activity of the DNAzyme and the selectivity of the aptamer. The aptasensor response was linear for ATP concentration between 5 and 230 nM (R-2 = 0.9854) with a detection limit of 2.4 nM (S/N = 3). Besides, ATP can be detected in human serum samples with satisfactory results, which demonstrates the potential applications for real analysis.
Terbium ion-coordinated carbon dots for fluorescent aptasensing of adenosine 5'-triphosphate with unmodified gold nanoparticles

Abstract:This work reports on a novel time-resolved fluorescent aptasensing platform for the quantitative monitoring of adenosine 5'-triphosphate (ATP) by interaction of dispersive/agglomerate gold nanoparticles (AuNPs) with terbium ion-coordinated carbon dots (Tb-CDs). To construct such a fluorescent nanoprobe, Tb-CDs with high-efficient fluorescent intensity are first synthesized by the microwave method with terbium ions (Tb(3+)). The aptasensing system consists of ATP aptamer, AuNP and Tb-CD. The dispersive/agglomerate gold nanoparticles are acquired through the reaction of the aptamer with target ATP. Upon target ATP introduction, the aptamers bind with the analytes to form new aptamer-ATP complexes and coat on the surface of AuNPs to inhibit their aggregation in the high salt solution. In this case, the fluorescent signal of Tb-CDs is quenched by the dispersive AuNPs on the basis of the fluorescence resonance energy transfer (FRET). At the absence of target analyte, gold nanoparticles tend to aggregate in the high salt state even if the aptamers are present. Thus, the added Tb-CDs maintain their intrinsic fluorescent intensity. Experimental results indicated that the aptasensing system exhibited good fluorescent responses toward ATP in the dynamic range from 40nM to 4.0μM with a detection limit of 8.5nM at 3sblank criterion. The repeatability and intermediate precision is less than 9.5% at three concentrations including 0.04, 0.4 and 2.0μM ATP. The selectivity was acceptable toward guanosine 5'-triphosphate, uridine 5'-triphosphate and cytidine 5'-triphosphate. The methodology was applied to evaluate the blank human serum spiked with target ATP, and the recoveries (at 3 concentration levels) ranged between 97.0% and 103.7%. Importantly, this detection scheme is rapid, simple, cost-effective, and does not require extensive sample preparation or separation.

Abstract:In this work, polymethacrylic acid (PMAA)-templated silver nanoclusters (Ag NCs) were developed as the fluorescent probe for the efficient and sensitive detection of adenosine triphosphate (ATP) in a wide range of pH values. The fluorescence intensity of the Ag NCs could keep stable with pH values ranging from 2.5 to 9.3. The detection of ATP was based on the quenching of the fluorescent Ag NCs in the presence of ATP. The fluorescence quenching of the Ag NCs with increasing ATP concentration was studied at pH 2.5, 4.5, 7.0 and 8.5 which involved a wide pH environment in body fluids. The limit of detection (LOD) for ATP was as low as 0.1 mmol/L in an acidic environment with pH of 2.5 and all the linear correlation coefficients were satisfied under wide-span pH values from 2.5 to 8.5. In addition, the sensitive determination of ATP was also achieved by adding copper ions (Cu2+). The high selectivity and rapid detection process proved that the fluorescent probe had great potential to detect ATP in biological samples under different pH conditions.
