Abstract:Adenosine has received great attentions acting as a potential biomarker for monitoring lung cancer. Most of the reported studies for adenosine detection require large instruments and complicated procedures. Herein, a sensitive, rapid and in-situ colorimetric aptasensor was developed for adenosine detection. Moreover, a homemade biomimetic electronic-eye (E-eye) was established and utilized as a portable in-time detection equipment. The entire measurement can be completed within 20 min, including the combination of aptamer with adenosine or AuNPs and the detection of adenosine. Four different kinds of aptamer were compared and the results showed that the AuNPs-aptamer-biotin system was the most stable and with the widest detection range of 5.0 mu M-60.0 mu M and the lowest LOD of 0.17 mu M. Moreover, the artificial urine samples were also tested with a linear range from 5.0 to 50.0 mu M and a LOD of 0.48 mu M. The results validated that the aptasensor together with the E-eye can be a promising platform for adenosine detection. (C) 2020 Elsevier B.V. All rights reserved.

Abstract:Adenosine can be a possible biomarker for monitoring the progress of lung cancer. Here, a sensitive and easily operated colorimetric methods based on aptasensor for the detection of adenosine has been developed. Gold nanoparticles (AuNPs) will aggregate in high-salt solutions, which causes color change, and single-stranded DNA will prevent this from happening. Based on changes in solution color, adenosine can be quantitatively determined. Besides a home-made bionic electronic-eye (E-eye) system was developed as a portable in-situ detection platform. Two different aptamers were used for comparison for adenosine detection. The linear correlation existed between 5.0-60.0μM with a detection limit of 0.17μM in water with modified aptamer. Moreover, the performance of the method was validated by testing the artificial urine samples with the linear range from 5.0 to 50.0μM and a detection limit of 0.48μM in artificial urine. This simple and cost-effective approach indicating its great potential for point-of-care applications.

Abstract:In the present study, we report a novel aptasensor based on silver nanoparticle enhanced fluorescence for the detection of adenosine. First, the distance dependence nature of silver nanoparticle enhanced fluorescence was investigated through fluorescent dyes modified oligonucleotides to control the spacing distance between dyes and AgNP. The results showed that the fluorescence intensity reached the maximum value with the spacing distance of dyes about 8 nm from AgNP surface. The fluorescence intensity decreases when the spacing distance is either above or below this value. Based on this result, a fluorescence switch is constructed. In the "OFF" state, without the target molecules, there is a greater spacing distance between the Cy3 dyes and the AgNP giving comparatively lower fluorescence intensity. While in the "ON" state, in the presence of target molecules, the fluorescence signals increased for the conformation structure change of the aptamer which shorten the spacing distance between the Cy3 dyes and the AgNP to 8 nm. Using adenosine as target, the aptasensor produced a linear range from 200 nM to 200 μM with a correlation coefficient of 0.9949 and the detection limit was 48 nM estimated using 3σ. The aptasensor was also found to be specific in targeting adenosine. The presented method shows a new strategy of combining aptamer recognition and silver nanoparticle for fluorescence signal enhancement and increasing sensitivity.

Abstract:In this work, based on purifying the analysis system by magnetic separation, we constructed a turn-on fluorescent assay for highly sensitive detection of possible biomarkers, employing adenosine as the model target analyte. Aptamer labeled-carbon dots (Apt-CDs) was applied as fluorescence probes as well as selective recognition elements, while Fe3O4@polypyrrole (Fe3O4@PPY) was used as fluorescence quenching and magnetic separating materials for system purification. In absence of adenosine, the Apt-CDs were attached to the surface of PPY through pi-pi stacking and hydrophobic interaction and its fluorescence was quenched by Fe3O4@PPY. In presence of adenosine, the aptamer could specifically bind with adenosine to form a three dimensional Apt-CDs-adenosine complex and detach from PPY, leading to the fluorescence recovery of Apt-CDs. More importantly, with analysis system purification via twice convenient magnetic separations of free CDs and Apt-CDs-adenosine complex respectively, the detection limit could be reduced from 8 nmol L-1 to 0.15 nmol L-1. This concept offers potential application for simple, rapid, cost-effective, and highly sensitive assay of biological samples. (C) 2017 Elsevier B.V. All rights reserved.

Abstract:We report herein an exonuclease-assisted aptamer-based target recycling amplification strategy for sensitive and selective chemiluminescence (CL) determination of adenosine. This aptasensor is based on target-induced release of aptamers from capture probes immobilized on the 96-well plate surface, and thus leading to a decreased hybridization with gold nanoparticle-functionalized reporter sequences followed by a CL signal. The introduction of exonuclease III catalyzes the stepwise removal of mononucleotides from 3′-hydroxyl termini of duplex DNAs of aptamers, liberating the adenosine. Therefore, a single copy of target adenosine can lead to the release and digestion of numerous aptamer strands from the 96-well plates and ultimately an enhanced sensitivity is achieved. Experimental results revealed that the exonuclease-assisted recycling strategy enabled the monitoring of adenosine with wide working ranges and low detection limits (LOD: 0.5nM). This new CL strategy might create a novel technology for the detection of various targets and could find wide applications in the environmental and biomedical fields.

Abstract:We have developed a novel sensitive chemiluminescence (CL) aptasensor for the target assay as exemplified by using adenosine as a model target. In this work, we have demonstrated the signaling mechanism to make detection based on magnetic separation and 3,4,5-trimethoxyl-phenylglyoxal (TMPG), a special CL reagent as the signaling molecule, which reacts instantaneously with guanine nucleobases (G) of adenosine-binding aptamer strands. Briefly, amino-functioned capture DNA sequences are immobilized on the surface of carboxyl-modified magnetic beads, and then hybridized with label-free G-rich (including 15 guanine nucleobases) adenosine-binding aptamer strands to form our CL aptasensor. Upon the introduction of adenosine, the aptamer on the surface of magnetic beads is triggered to make structure switching to the formation of the adenosine/aptamer complex. Consequently, G-rich aptamer strands are forced to dissociate from magnetic beads sensing interface, resulting in a decrease of CL signal. The decrement of peak signal is proportional to the amount of adenosine. The effects of the amounts of capture DNA, aptamer, magnetic beads are investigated and optimized. It was found that the CL intensity had a linear dependency on the concentration of adenosine in the range of 4×10−7 to 1×10−5M. With a low detection limit of 8×10−8M and simplicity in CL detection, this novel technique will offer a great promise for future target/aptamer analysis.

Abstract:Adenosine is a vital biological small molecule that regulates various physiological processes in the human body. A high expression of adenosine in cells can facilitate tumor growth. Therefore, detecting adenosine is crucial for early disease diagnosis. In this paper, we designed a fluorescent biosensor for the sensitive detection of adenosine based on the cationic comb-type copolymer PLL-g-Dex for assisted rapid hybridization of nucleic acids at room temperature. In this strategy, adenosine preferentially binds to the aptamer immobilized on the surface of magnetic nanobeads, releasing free aDNA in solution as the primer strand, which rapidly forms DNA nanowires with auxiliary probes of bDNA with the assistance of PLL-g-Dex. SYBR Green I is embedded in DNA duplexes to generate strong fluorescence. The experimental results showed that PLL-g-Dex promotes DNA hybridization reactions at room temperature to form ultra-long DNA nanowires, thus achieving signal amplification and shortening the detection time. In addition, magnetic nanobeads can reduce the background signal during the reaction. Compared with several previous studies on the fluorescence detection of adenosine, this strategy has a lower detection limit of 2.32 nM. Furthermore, this novel system exhibited a good detection performance even under complex environments, such as serum, providing some reference for the quantitative detection of adenosine in early disease diagnosis.

Abstract:Adenosine is an endogenous molecule that plays a vital role in biological processes. Research indicates that abnormal adenosine levels are associated with a range of diseases. The development of sensors capable of detecting adenosine is pivotal for early diagnosis of disease. For example, elevated adenosine levels are closely associated with the onset and progression of cancer. In this study, we designed a novel DNA biosensor utilizing chaperone copolymer-assisted catalytic hairpin assembly for highly sensitive detection of adenosine. The functional probe comprises streptavidin magnetic beads, an aptamer, and a catalytic chain. In the presence of adenosine, it selectively binds to the aptamer, displacing the catalytic chain into the solution. The cyclic portion of H1 hybridizes with the catalytic strand, while H2 hybridizes with the exposed H1 fragment to form an H1/H2 complex containing a G-quadruplex. Thioflavin T binds specifically to the G-quadruplex, generating a fluorescent signal. As a nucleic acid chaperone, PLL-g-Dex expedites the strand exchange reaction, enhancing the efficiency of catalytic hairpin assembly, thus amplifying the signal and reducing detection time. The optimal detection conditions were determined to be a temperature of 25 °C and a reaction time of 10 min. Demonstrating remarkable sensitivity and selectivity, the sensor achieved a lowest limit of detection of 9.82 nM. Furthermore, it exhibited resilience to interference in complex environments such as serum, presenting an effective approach for rapid and sensitive adenosine detection.

Abstract:Here we report a novel approach for the detection of adenosine in cancer cells by electrochemiluminescence (ECL) on a wireless indium tin oxide bipolar electrode (BPE). In this approach, ferrocene (Fc) which is labeled on adenosine aptamer is enriched on one pole of the BPE by hybridization with its complementary DNA (ssDNA) and oxidized to Fc(+) under an external voltage of 5.0V at the two ends of BPE. Then, a reversed external voltage was added on the BPE, making Fc(+) enriched pole as cathode. The presence of Fc(+) promotes the oxidation reaction on the anodic pole of the BPE, resulting in a significant increase of ECL intensity using Ru(bpy)3(2+)/tripropylamine (TPA) system as test solution. The presence of target adenosine was reflected by the ECL signal decrease on the anodic pole caused by the target-induced removal of ferrocene-aptamer on the cathodic pole. The decrease of ECL signal was logarithmically linear with the concentration of ATP in a wide range from 1.0 fM to 0.10 μM. This ECL biosensing system could accurately detect the level of adenosine released from cancer cells.

Abstract:Three simple fluorescence turn-on sensors have been designed for the selective detection of adenosine (AD) based on a boronic acid receptor and a coumarin fluorophore (NLC, NJC, NDC). In the absence of AD, the fluorescence emission intensity of the sensors was relatively weak due to the effective electronic transfer from the to the coumarin to the boronic acid moiety. When the AD was introduced into the system, an obvious fluorescence enhancement was observed. It was attributed to the boronic acid covalently binding to the cis-diol unit of the AD resulting in the electronic transfer process being inhibited. Under the optimal conditions, these fluorescent sensors not only could perform good selectivity and fast response (< 30 s), but also show good water solubility. Finally, the proposed sensor NLC has been utilized for the AD determination in urine without any complicated pretreatment and the recovery was in the range of 95–103%. As a result, it is promising for application on the AD detection in complex biological samples. Moreover, there are no small molecular sensors detecting AD have been reported, yet, it is promising for providing a new platform to detect AD by small molecular sensor.

Abstract:Coupling nano-probe technology,aptamer recognition with resonance light scattering analysis,through recognizing reaction between adenosine and its specific aptamer in homogeneous matrix,causing the disaggregation of the aggregated network structure of adenosine specific aptamer functionalized nanogold structure,a sharply change on the resonance light scattering signal can be obtained.Based on it,a novel homogeneous aptamer recognizing based method for adenosine detection was established.The linear concentration range was 0.1～10μmol/L for adenosine with a detection limit of 0.04μmol/L(3S/N).The proposed method also shows good selectivity for adenosine detection and no interference from guanosine,thymidine,cytidine and uridine.

Abstract:We demonstrate a simple force-based label-free strategy for the highly sensitive sensing of adenosine. An adenosine ssDNA aptamer was bound onto an atomic force microscopy (AFM) probe by covalent modification, and the molecular-interface adsorption force between the aptamer and a flat graphite surface was measured by single-molecule force spectroscopy (SMFS). In the presence of adenosine, the molecular recognition between adenosine and the aptamer resulted in the formation of a folded, hairpin-like DNA structure and hence caused a variation of the adsorption force at the graphite/water interface. The sensitive force response to molecular recognition provided an adenosine detection limit in the range of 0.1 to 1 nM. The addition of guanosine, cytidine, and uridine had no significant interference with the sensing of adenosine, indicating a strong selectivity of this sensor architecture. In addition, operational parameters that may affect the sensor, such as loading rate and solution ionic strength, were investigated.

Abstract:A simple and sensitive colorimetric aptasensor for rapid and facile detection of adenosine triphosphate (ATP) has been demonstrated here based on aptamer-based hairpin probes and unmodified gold nanoparticles (AuNPs). The hairpin probe is constructed by adding another five nucleotides to the 5′-end of an anti-ATP aptamer which can hybridize to nucleotides at the 3′-end of the aptamer, forming a hairpin-shaped structure. In the absence of ATP, the hairpin probes are rigid, and the AuNPs are susceptible to salt-induced aggregation. Conversely, upon binding with target ATP, the hairpin probes undergo conformational changes, forming aptamer-ATP complexes and exposing flexible ends which coat the surface of AuNPs to inhibit their aggregation in the high salt solution. Subsequently, a blue-to-red color change can be recognized by the naked eye. The aptasensor achieved selective responses toward ATP with a detection limit of 0.1μM, and exhibited high-quality detection performance in biological samples. In addition, this detection method is simple, rapid and cost-effective, holding great potential for further applications in point-of-care research.

Abstract:A colorimetric method is presented for ultrasensitive determination of adenosine. The assay is based on side-by-side self-assembly of aptamer-functionalized gold nanorods (Au NRs). It relies on the fact that the conjugation of the helper DNA predominantly occurs at the terminal ends of the Au NRs rather than at their sides. The adenosine aptamers consist of two pieces of ssDNA (termed C1 and C2) that were individually attached to the sides of Au NRs. In the presence of adenosine, it will be captured by C1 and C2 to form a stable sandwich structure. As a result, a side-to-side assembly of the Au NRs occurs. If the adenosine concentration is increased, the absorbance of the Au NRs at 742 nm gradually decreases, and the color changes from brick red to dark brown. Response is linear range in the 10 pM to 5 nM adenosine concentration range, and the detection limit is as low as 3.3 pM. Adenosine analogues such as uridine and cytidine do not interfere. The method was used to quantify adenosine in serum samples at concentrations as low as 10 pM.Schematic representation of an effective colorimetric method for adenosine detection based on target adenosine-induced side-by-side self-assembly of gold nanorods (Au NRs).

Abstract:An abasic site called dSpacer has been introduced into duplex regions of the 8-17 DNAzyme and adenosine aptamer for label-free fluorescent detection of Pb(2+) and adenosine, respectively. The dSpacer can bind an extrinsic fluorescent compound, 2-amino-5,6,7-trimethyl-1,8-naphthyridine (ATMND), and quench its fluorescence. Addition of Pb(2+) enables the DNAzyme to cleave its substrate and release ATMND from DNA duplex, recovering the fluorescence of ATMND. Similarly, the presence of adenosine induces structural switching of the aptamer, resulting in the release of ATMND from the DNA duplex and a subsequent fluorescence enhancement. Under optimized conditions, this label-free method exhibits detection limits of 4 nM for Pb(2+) and 3.4 muM for adenosine, which are even lower than those of the corresponding labeled-DNAzyme and aptamer sensors. These low detection limits have been obtained without compromising any of the selectivity of the sensors. Finally, the dynamic range of the adenosine sensor has been tuned by varying the number of hybridized base-pairs in the aptamer duplex. The method demonstrated here can be applied for label-free detection and quantification of a broad range of analytes using other DNAzymes and aptamers.

Abstract:We present a simple and low-cost approach for adenosine deaminase (ADA) determination by assembling a purpose-designed carboxyfluorescein (FAM)-labeled aptamer with oxidized single-walled carbon nanohorns (oxSWCNHs). A carboxyfluorescein (FAM) labeled aptamer that had a strong affinity for adenosine was synthesized. The mixing of FAM-labeled aptamer with oxSWCNHs formed by a self-assembly of two components via their electrostatic interactions. As a result, the fluorescence of the FAM-labeled aptamer was quenched by the efficient electron transfer for the determination of adenosine deaminase. In the presence of adenosine, the FAM-labeled aptamer in the self-assembly was released from the oxSWCNHs surface, thereby inducing the fluorescence recovery. Upon reaction with ADA, the fluorescence of the complex was quenched as a consequence of the removal of adenosine converted into inosine which has no affinity for this aptamer. Thus, a simple and rapid strategy for assaying ADA is reported with a detection limit of 0.10 U/mL and linear range from 0.25 to 3.125 U/mL. The method was employed ADA in human and mouse serum.

Abstract:Nucleic acid aptamers, which are short single-stranded RNA or DNA molecules, undergo conformational changes upon recognizing small molecules. However, their structural characterization is challenging due to the flexibility of single-stranded oligonucleotides. Nanopores provide a sensitive method for single-molecule analysis of molecular conformational changes as they transport through the pore. In this study, we employed α-hemolysin (α-HL) nanopore with high spatiotemporal resolution to investigate the conformational changes of DNA aptamer (ADO-23) upon binding to adenosine (Ado) molecules. Unlike the events generated by the aptamer alone, the DNA-Ado complex produced two distinct new characteristic events, indicating the formation of two unique secondary structures. By analyzing current events, the structures formed by the DNA-Ado complex were speculated to be hairpin DNA and G-quadruplex (G4) DNA. Furthermore, we demonstrated the potential of the aptamer as a chiral selector within the nanopore. The results showed that the aptamer specifically bound to D-Ado and not to L-Ado. Finally, we realized specific and sensitive detection of small molecule Ado by α-HL pore, with a detection limit as low as 10.5 nM, and successfully applied it to the detection of real human serum samples. The nanopore platform provides a powerful tool for studying small molecule-biomolecule conformational changes, and we constructed sensors for detecting adenosine through the aptamer. This work also offers a promising new approach to the recognition of enantiomers at the single-molecule level.
