Abstract:An efficient synthesis of 2,5-disubstituted oxazoles via Co(III) catalysis is described herein. The synthesis is achieved under mild conditions through [3+ 2] cycloaddition of N-pivaloyloxyamides and alkynes. The reaction operates through an internal oxidation pathway and features a very broad substrate scope. The one-step synthesis of natural products such as texamine and balsoxin has been demonstrated via this protocol.

Abstract:A gold-catalyzed regioselective [3 + 2] cycloaddition of alkynyl triazenes with 1,2,4-dioxazoles was developed. The triazene group in the products could be replaced to obtain iodo-oxazoles, providing potential transformations to diverse oxazole structures. This protocol features readily available starting materials, mild reaction conditions and scalability. A plausible mechanism involving a nitrene transfer process was proposed.

Abstract:Herein, we report the first intramolecular C-N bond formation of phenylpropanamide derivatives via organocatalytic oxidative reactions, affording 3,3-disubstituted oxindole derivatives with up to 99% yield. The high efficiency of this reaction is exemplified by the transition metal-free mild conditions and the ability to perform the reaction on a gram scale. Meanwhile, the DFT calculation of the catalytic oxidative transformation pathway has also been studied.

Abstract:An efficient synthesis toward 2-alkyl substituted oxazoles has been achieved through Suzuki cross-coupling reaction. Treatment of various alkyl iodides with 9-MeO-BBN, followed by in situ palladium-catalyzed carbon–carbon formation with 2-iodooxazoles obtained the 2-alkyl substituted oxazoles in good to excellent yields. Regioselective C2-alkylation on 2,4-di-iodooxazole under the same reaction conditions was firstly disclosed.

Abstract:A general and stereoselective synthesis of (Z)-ketoeneynes 9 and 11a-f was established by using a Pd(O)-Cu(I)-catalyzed cross-coupling of the labile (Z)-ketoenol triflate 3 with 1-alkynes under carefully controlled reaction conditions. Isomerization of the coupling products into the more stable (E)-ketoeneynes 8 and 12a-f was observed and could be minimized by earring out the coupling reaction in CH3CN at low temperature using Et3N as the base. Abstract:Oxazoles are among the most important heterocyclic scaffolds in the fields of natural products and medicinal chemistry. Herein is developed a tandem reaction for the synthesis of a diverse array of 4,5-difunctionalized oxazoles utilizing easily-accessible ethyl 2-isocyanoacetate and aldehydes (26 examples, 31-83% yields). This cascade reaction is facilitated by catalytic CuBr and molecular oxygen as the oxidant. The process involves a catalytic cycloaddition oxidative dehydroaromatization mechanism. The broad aldehyde substrate scope, mild reaction conditions, and atom economy make this protocol an attractive alternative to access functionalized oxazoles.

Abstract:Aromatic heterocycles have been identified as effective directing groups (DGs) in C–H functionalization but can be retained as undesired bulky substituents in the final products. Herein, we report a Co(III)-catalyzed 1-aminoisoquinoline synthesis strategy based on oxadiazole-directed aromatic C–H coupling with alkynes and a subsequent redox-neutral C–N cyclization reaction. This labile N–O bond-based protocol has allowed the toleration of a broad range of functional groups.

Abstract:The cobalt-catalyzed cross-dehydrogenative coupling of (benz)oxazoles and ethers is described. Access to some important bioactive heteroaryl ether derivatives was achieved using CoCO3 as an inexpensive catalyst at levels as low as 1.0 mol %. Abstract:A mild and efficient methodology involving Pd(PPh3)4-catalyzed intramolecular cyclization of N-alkynyl alkyloxycarbamates with CuCl2 or CuBr2 for the synthesis of 4-halo-oxazolones was developed. This reaction exhibiting good functional tolerance provided a new, efficient, and rapid synthetic process to 4-halo-oxazolones. The resulting 4-halo-oxazolones can serve as great potential precursors for the 3,4,5-trisubstituted oxazolones via a Pd-catalyzed cross-coupling reaction.

Abstract:A cobalt‐catalyzed annulation of o‐phenylenediamines and internal alkynes for the synthesis of quinoxalines is developed. This method provides a variety of quinoxalines in good to high yields under a simple and mild reaction conditions with molecular O2 as a terminal oxidant. Preliminary mechanistic studies show that a novel reaction mechanism is involved in the current reaction, which is different from the previous reported dicarbonyl compound mechanism via the oxidation of the alkyne.
