Abstract:In this paper, enantioselectivities of D-amino acid oxidase (DAAO) in ten ionic liquids were investigated in detail. Among the ionic liquids studied, 3-methylimidazolium formate ([MIM][COO]) was able to trigger enzyme activation with L-Ala as substrate. The enzyme activity of DAAO reached a maximum of 4.26 U mg(-1) min(-1) in the presence of 40% [MIM][COO], while no activity toward L-Ala was observed when DAAO was incubated with other ILs. Moreover, DAAO activities towards other L-amino acids such as L-Pro and L-Arg were also improved in [MIM][COO], which indicated that the enzymatic enantioselectivity of amino acids was lowered. This was further confirmed by capillary electrophoresis, circular dichrosim and fluorescence analysis. Molecular optimization supported that the protein-ionic liquid interactions modulated the structure of the enzyme, especially in [MIM][COO], leading to relaxation in the enantioselectivity of DAAO. This observation and exploration might open a new window for modulation enantioselectivity resolution in ionic liquids.

Abstract:D-amino acid oxidases (DAAO) are stereospecific enzymes which are generally almost inactive towards L-enantiomer in neutral solution when L-, D-amino acids are supplied as substrates. In this paper, the D-amino acid oxidase can catalytic oxidize L-amino acids by modulating pH of aqueous solution. With L-Pro as substrate, the catalytic rate ( k cat ) and the affinity ( K m ) of DAAO were 6.71 s −1 and 33 mM at pH 8.0, respectively, suggesting that optimal pH condition enhanced the activity of DAAO towards L-Pro. Similar results were obtained when L-Ala (pH 9.8), L-Arg (pH 6.5), L-Phe (pH 9.0), L-Thr (pH 9.4), and L-Val (pH 8.5) were catalyzed by DAAO at various pH values. The racemization of the L-amino acids was not found by capillary electrophoresis analysis during oxidation, and quantification analysis of L-amino acids before and after catalytic reaction was performed, which confirmed that the modulation of enantioselectivity of DAAO resulted from the oxidation of L-amino acids rather than D-amino acids by changing pH. A mechanistic model was proposed to explain enhanced activity of DAAO towards L-amino acids under optimal pH condition.

Abstract:Ionic liquids, with their unique properties, including low volatility, high polarity, good thermal stability over a wide temperature range and selective dissolving capacity by a proper choice of cations or anions, have attracted increasing interest in the organic transformations as reactioin medium as well as catalyst (ligand). Compared with traditional organic solvents, ionic liquids have great advantages in obtaining products with high isolated yields and good to excellent selectivities, accelerationg reaction rate in some cases, ease of work-up, as well as recycling use with or without catalysts after simply recoveration. The paper reviews the advanced progress in the application of ionic liquids as reaction medium as well as catalysts to organic synthesis, mainly including coupling reaction, Michael addition, Baylis-Hillman reaction, Diels-Alder reaction, Aldol reaction, Knoevenagel condensation, cyclorization reaction, alkylation and acylation, oxidation and reduction etc.

Abstract:The enantio,separation of D,L-alpha-amine acid on(+)-( 18-crown-6) -2,3,11,12-tertcarboxyl acid chiral stationary phase ( CSP), and the influence of the methanol and acid addictives on the separation were investigated. The chiral recognition mechanism was also discussed. It is possible that the solute R-NH2 was protoned to R-NH3+, and the fitness and interaction in the crown ester cavity between the solutes and CSP were the dominant chiral recognition mechanism for the solutes. The concentration of methanol and composition of acid additive influenc ed the equilibrium of R-NH3+ between mobile phase and stationary phase, which resulted in the chiral recognition. In addition, the influence of column temperature on the enantiosepartion of phenylalanine and tyrosine was also investigated. And all the tested temperatures were lowered than the isotemperature T = 467. 3 K. The result showed that the configuratoin of stationary phase didn't have evidence changed, and reservation mechanism and enantiotropy selection action remained changeless. Delta Delta H-0 and Delta Delta S-0 were negative which showed that the enantioseparation'of two solutes was enthalpy control.

Abstract:As a privileged chiral scaffold, cinchona alkaloid and its derivatives have reached remarkable success in catalytic asymmetric organic synthesis. In addition to the wide applications of point chirality control, Smith and co-workers recently discovered a quinidine-derived ammonium cation-catalyzed O-alkylation of tetralones, providing an effective approach for the synthesis of axially chiral biaryls. Using density functional theory (DFT) calculations, we studied the reaction mechanism and origins of enantioselectivity of this novel transformation. A stepwise strategy is adopted to ensure efficient and thorough exploration of the massive conformational space of transition state. Our computations suggested that enolate oxygen forms two hydrogen bonds with the chiral ammonium catalyst, and the non-covalent interactions between the cationic benzylic fragment and the methoxy group of enolate plays a critical role in determining the enantioselectivity.

Abstract:Long-chain L-α-hydroxy acid oxidase (LCHAO) is a flavin mononucleotide (FMN)-dependent oxidase that dehydrogenates l-α-hydroxy acids to keto acids. There were two different mechanisms, named as hydride transfer (HT) mechanism and carbanion (CA) mechanism, respectively, proposed about the catalytic process for the FMN-dependent L-α-hydroxy acid oxidases on the basis of biochemical data. However, crystallographic and kinetic studies could not provide enough evidence to prove one of the mechanisms or eliminate the alternative. In the present studies, theoretical computations were carried out to study the molecular mechanism for LCHAO-catalyzed dehydrogenation of L-lactate. Our molecular dynamics (MD) simulations indicated that L-lactate prefers to bind with LCHAO in a hydride transfer mode rather than a carbanion mode. Quantum mechanics/molecular mechanics (QM/MM) calculations were further carried out to obtain the optimized structures of reactants, transition states, and products at the level of ONIOM-EE (B3LYP/6-311++G(d,p)//B3LYP/6-31G(d,p):AMBER). Quantum chemical studies indicated that LCHAO-catalyzed dehydrogenation of L-lactate would be a stepwise catalytic reaction in a hydride transfer mechanism but not a carbanion mechanism. MD simulations, binding free energy calculations, and QM/MM computations were also implemented on the complex between L-lactate and Y129F mutant LCHAO. By comparing the Y129F mutant system with the wild-type system, it was further confirmed that the key residue Tyr129 in the active site of LCHAO would not affect L-lactate's binding to LCHAO but play an important role on the catalytic reaction process through an H-bond interaction.
Enantioselective Toxicities of Chiral Ionic Liquids 1-Alkyl-3-methylimidazolium Lactate to Aquatic Algae.

Abstract:With the wide application of chiral ionic liquids (CILs) as green solvents, their threats to the aquatic environment cannot be ignored. Thus, risk assessment and the prospective design of inherently safe CILs have become more urgent. However, whether enantioselectivity is a feature of the aquatic toxicity of CILs is poorly understood. Herein, we describe the first investigation into the ecotoxicities of CILs toward green algae Scenedesmus obliquus and Euglena gracilis. A series of methylimidazolium lactic ionic liquids, which cation parts with different alkyl chains and anion part is enantiomers of lactate, are used as representative CILs. The results of S. obliquus showed that the EC50 value of L-(+)-1-ethyl-3-methylimidazolium lactate (L-(+)-EMIM L) was more than 5000 mu M, while the EC50 value of D-(-)-1-ethyl-3-methylimidazolium lactate (D-(-)-EMIM L) was 2255.21 mu M. Such a distinct difference indicates the enantioselective toxicity of CILs to algae. This enantioselectivity initially persisted with increasing carbon chain length, but no longer exhibited when with greater carbon chain lengths, due to changes in the toxicity weightings of the cation parts. Further research showed that the enantioselective effects of CILs resulted from the differences in the production of reactive oxygen species, the damage to cell membrane integrity and cell wall after exposure to CILs. Results from this study showed that monitoring for the racemate CILs will give an inadequate or misleading environmental risk assessment. Thus, we should improve our ability to predict their effects in natural environments. In the meantime, non-selective use of CILs will do harm to aquatic organisms. Therefore, to minimize their potential for environmental impact, the enantioselective toxicities of CILs with short alkyl chains should be taken into consideration. (C) 2014 Elsevier B.V. Controlling Enantioselectivity of Esterase in Asymmetric Hydrolysis of Aryl Prochiral Diesters by Introducing Aromatic Interactions.

Abstract:Aromatic interactions specific to aryl radicals were introduced into two esterases, BioH from Escherichia coli and RspE from Rhodobacter sphaeroides to control their enantioselectivity in the asymmetric hydrolysis of prochiral aryl glutaric acid diesters. As a result, the enantiomeric excess (ee) of the S-product of dimethyl 3-phenylglutarate was increased from 25% (BioH wild type) to 96% (B_L83F/L86F) and from 13% (RspE wild type) to >99% (R_Y27R), respectively, while another variant of RspE R_M121F gave a reversed ee of 50% (R-product). Similar enhancement or reversion of enantioselectivity were also observed in the hydrolysis of three other prochiral aryl diesters (dimethyl 3-(4-flouro)-phenylglutarate, dimethyl 3-(4-cholo)-phenylglutarate and dimethyl 3-(3,4-dicholo)-phenylglutarate). Especially, the mutant R_Y27R was shown to be an excellent S-selective hydrolase for prochiral aryl diesters, with ee of all S-products >99%. In the mutants with altered enantioselectivity, the successful introduction of designed aromatic interactions was confirmed by molecular dynamics simulations and binding free energy analysis. These results demonstrate that aromatic interaction is one of the origins of enzyme enantioselectivity, the tuning of which leads to dramatic change in enantioselectivity. Besides, the successful engineering of the enantioselectivity in two different proteins toward four different substrates suggests that the introduction of aromatic interactions is a generally applicable strategy in the control of enantioselectivity toward aryl substrates.

Racemates of amines, amino alcohols, and amino acids could generate enantioselective precipitate with multicomponent self-assemblies under mild conditions. Affinity-binding immobilization of D-amino acid oxidase on mesoporous silica by a silica-specific peptide

Abstract:Enzyme immobilization is widely used for large-scale industrial applications. However, the weak absorption through physical methods limits the recovery ability. Here, affinity-binding immobilization of enzymes was explored using a silica-specific affinity peptide (SAP) as a fusion tag to intensify the binding force between the enzyme and mesoporous silica (MPS) carrier. D-amino acid oxidase (DAAO) of Rhodosporidium toruloides was used as a model enzyme. The optimal screened SAP (LPHWHPHSHLQP) was selected from a M13 phage display peptide library and fused to the C-terminal of DAAO to obtain fused DAAOs with one, two and three SAP tags, respectively. The activity of DAAO-SAP-MPS was superior comparing with DAAO-2SAP-MPS and DAAO-3SAP-MPS; meanwhile DAAO-SAP-MPS shows 36% higher activity than that of DAAO-MPS. Fusion with one SAP improved the thermal stability with a 10% activity increase for immobilized DAAO-SAP-MPS compared to that of DAAO-MPS at 50 °C for 3 h. Moreover, the activity recovery of immobilized DAAO-SAP-MPS was 25% higher in operation stability assessment after six-batch conversions of cephalosporin to glutaryl-7-amino cephalosporanic acid than that of DAAO-MPS.

Abstract:A novel chiral ionic liquid, compound 1, based on camphorsulfonic acid, was prepared. A catalytic amount of 1 readily promotes L-proline-catalyzed aldol reactions, with good chemoselectivity, both in H2O and in organic solvents. Further, the aldol reaction of cyclohexanone with 4-nitrobenzaidehyde afforded 2-[hydroxy(4-nitrophenyl)methyl]cyclohexanone (6) in 98% yield with high enantioselectivity (94% ee) when large amounts of 1 (5 equiv.) were used as promoter.

Abstract:[AAE]X composed of amino acid ester cations is a sort of typically “bio-based” protic ionic liquids (PILs). They possess potential Brønsted acidity due to the active hydrogens on their cations. The Brønsted acidity of [AAE]X PILs in green solvents (water and ethanol) at room temperature was systematically studied. Various frameworks of amino acid ester cations and four anions were investigated in this work from the viewpoint of structure–property relationship. Four different ways were used to study the acidity. Acid dissociation constants (pKa) of [AAE]X determined by the OIM (overlapping indicator method) were from 7.10 to 7.73 in water and from 8.54 to 9.05 in ethanol. The pKa values determined by the PTM (potential titration method) were from 7.12 to 7.82 in water. Their Hammett acidity function (H0) values (0.05 mol·L−1) were about 4.6 in water. In addition, the pKa values obtained by the DFT (proton-transfer reactions) were from 7.11 to 7.83 in water and from 8.54 to 9.34 in ethanol, respectively. The data revealed that the cationic structures of [AAE]X had little effect and the anions had no effect on the acidity of [AAE]X. At the same time, the OIM, PTM, Hammett method and DFT method were reliable for determining the acidic strength of [AAE]X in this study.

Abstract:A family of novel ionic liquids with amino acids and their derivatives as cations and environmentally benign materials as anions have been synthesized using easy preparation techniques. The ionic liquids obtained have the same characteristics as conventional imidazolium ionic liquids and the same chiralities as natural amino acids. Thermal stabilities, phase behaviour, viscosities and miscibilities of the representative family members have been investigated, generally showing no difference from conventional ionic liquids. These amino acid ionic liquids may be used as catalysts and "fully green'' solvents in the cycloaddition of cyclopentadiene to methyl acrylate, which is a typical Diels-Alder reaction. This approach to treating amino acids and their derivatives can serve as an alternative to traditional ionic liquids having synthetic chemical components.

Abstract:The dependence of the enantioselectivity and enantiomer migration order (EMO) on the chirality and stereo-conformation of ligands used for the chiral selectors of Cu(II) complexes was investigated. It was interestingly observed that EMOs were reversed not only by a change of the ligand chirality of selector, but also by the trans- or cis-conformation of a 4-hydroxy substituent in 4-hydroxyproline. In the presence of the sodium dodecyl sulfate (SDS) micellar phase, the EMOs were reversed when both the bidentate and tridentate ligands of the proline family were used. The resolution (Rs) of enantiomers decreases when the cis-hydroxy substituent exists in D- or L-hydroxyproline. SDS micellar phase improves the resolution of amino acid enantiomers. The relationships among the EMO, ligand chirality, complex stability and stereo-selectivity are discussed.
Preparation of amino acid chiral ionic liquid and visual chiral recognition of glutamine and phenylalanine enantiomers

Abstract:In this paper, the amino acid chiral ionic liquid (AACIL) was prepared with L-phenylalanine and imidazole. It was characterized by CD, FT-IR, 1H NMR, and 13C NMR spectrum. The chiral recognition sensor was constructed with AACIL and Cu(II), which exhibited different chiral visual responses (solubility or color difference) to the enantiomers of glutamine (Gln) and phenylalanine (Phe). The effects of solvent, pH, time, temperature, metal ions, and other amino acids on visual chiral recognition were optimized. The minimum concentrations of Gln and Phe for visual chiral recognition were 0.20 mg/ml and 0.28 mg/ml, respectively. The mechanism of chiral recognition was investigated by FT-IR, TEM, SEM, TG, XPS, and CD. The location of the host-guest inclusion or molecular placement has been conformationally searched based on Gaussian 09 software.
Enhancement of the substrate specificity of D-amino acid oxidase based on tunnel-pocket engineering

Abstract:D-Amino acid oxidase (DAAO) selectively catalyzes the oxidative deamination of D-amino acids, making it one of the most promising routes for synthesizing optically pure L-amino acids, including L-phosphinothricin ( L-PPT), a chiral herbicide with significant market potential. However, the native DAAOs that have been reported have low activity against unnatural acid substrate D-PPT. Herein, we designed and screened a DAAO from Rhodotorula taiwanensis (RtwDAAO), and improved its catalytic potential toward D-PPT through protein engineering. A semirational design approach was employed to create a mutation library based on the tunnel-pocket engineering. After three rounds of iterative saturation mutagenesis, the optimal variant M3rd -SHVG was obtained, exhibiting a >2000-fold increase in relative activity. The kinetic parameters showed that M3rd -SHVG improved the substrate binding affinity and turnover number. This is the optimal parameter reported so far. Further, molecular dynamics simulation revealed that the M3rd -SHVG reshapes the tunnel-pocket and corrects the direction of enzyme-substrate binding, allowing efficiently catalyze unnatural substrates. Our strategy demonstrates that the redesign of tunnel-pockets is effective in improving the activity and kinetic efficiency of DAAO, which provides a valuable reference for enzymatic catalysis. With the M3rd -SHVG as biocatalyst, 500 mM D, L-PPT was completely converted and the yield reached 98%. The results laid the foundation for further industrial production.
Investigation of laccase activity in cholinium-based ionic liquids using experimental and molecular dynamics techniques

Abstract:Laccases hold great potential for biotechnological applications, particularly in environmental pollutant remediation. Laccase activity is governed by the solvent environment, and ionic liquids (ILs) emerge as a versatile solvent for activation or stabilization of enzymes. Herein, effects of cholinium-based ILs formulated with carboxylic acids, inorganic acid, and amino acids as anionic species, on the catalytic activity of laccase from Trametes versicolor were investigated by experimental and computational approaches. Experimental results showed that laccase activity was enhanced by 21.39 % in 0.5 M cholinium dihydrogen citrate ([Cho][DHC]), in relation to the laccase activity in phosphate buffer medium. However, cholinium aminoate ILs negatively affected laccase activity, as evidenced by the partial deactivation of laccase in both cholinium glycinate and cholinium phenylalaninate, at concentrations of 0.1 M and 0.5 M, respectively. Molecular dynamics studies revealed that the enhancement of laccase activity in [Cho][DHC] might be attributed to the highly stabilized and compact structure of laccase, facilitating a better internal electron transfer during the laccase-substrate interactions. Enhanced catalytic performance of laccase in [Cho][DHC] was postulated to be driven by the high accumulation level of dihydrogen citrate anions around laccase's surface. [Cho][DHC] holds great promise as a cosolvent in laccase-catalyzed biochemical reactions.

Enantioselective Alkoxylation with Electrogenerated Chiral Carbocation Intermediates

Abstract:Electrochemical reactions via carbocation intermediates remain fundamental transformations that build up molecular functionality and complexity in a sustainable manner. Enantioselective control of such processes is a great challenge in a highly ionic electrolyte solution. Here, we report an anodic generation of chiral α-imino carbocation intermediates by enamine catalysis. The chiral carbocation intermediates can be intercepted by simple alcohols to deliver chiral tertiary ethers with high stereoselectivity. The key SN1 step proceeds via a tertiary amine-mediated proton shuttle that facilitates facial selection in reacting with carbocation. This study presents a viable electrochemical strategy for externally tuning enamine catalysis.
