Abstract:Enormous interest in biocatalysis in non-aqueous phase has recently been triggered due to the merits of good enantioselectivity, reverse thermodynamic equilibrium, and no water-dependent side reactions. It has been demonstrated that enzyme has high activity and stability in non-aqueous media, and the variation of enzyme activity is attributed to its conformational modifications. This review comprehensively addresses the stability and activity of the intact enzymes in various non-aqueous systems, such as organic solvents, ionic liquids, sub-/super-critical fluids and their combined mixtures. It has been revealed that critical factors such as Log P, functional groups and the molecular structures of the solvents define the microenvironment surrounding the enzyme molecule and affect enzyme tertiary and secondary structure, influencing enzyme catalytic properties. Therefore, it is of high importance for biocatalysis in non-aqueous media to elucidate the links between the microenvironment surrounding enzyme surface and its stability and activity. In fact, a better understanding of the correlation between different non-aqueous environments and enzyme structure, stability and activity can contribute to identifying the most suitable reaction medium for a given biotransformation.

Abstract:Ionic liquids, with their unique properties, including low volatility, high polarity, good thermal stability over a wide temperature range and selective dissolving capacity by a proper choice of cations or anions, have attracted increasing interest in the organic transformations as reactioin medium as well as catalyst (ligand). Compared with traditional organic solvents, ionic liquids have great advantages in obtaining products with high isolated yields and good to excellent selectivities, accelerationg reaction rate in some cases, ease of work-up, as well as recycling use with or without catalysts after simply recoveration. The paper reviews the advanced progress in the application of ionic liquids as reaction medium as well as catalysts to organic synthesis, mainly including coupling reaction, Michael addition, Baylis-Hillman reaction, Diels-Alder reaction, Aldol reaction, Knoevenagel condensation, cyclorization reaction, alkylation and acylation, oxidation and reduction etc.

Abstract:It is well known that non-aqueous enzymatic catalysis has emerged as an important area of enzyme engineering with the advantages of higher substrate solubility, increased stereoselectivity, modified substrate specificity and suppression of unwanted water-dependent side reactions. As a result, non-aqueous enzymatic catalysis has been applied in the biocatalytic synthesis of important pharmaceuticals and nutriceuticals. With the advancement of non-aqueous enzymatic catalysis in recent years, the efforts have been centered on the discovery and modification of solvent-tolerant biocatalysts for non-aqueous environments. Additionally, with the inevitable trends of green chemistry and sustainable development, green solvents have been utilized for increased number of enzymatic reactions to replace conventional organic solvents. In this review, modification, immobilization and mutagenesis of various enzymes for non-aqueous catalysis are discussed. Recent progress of non-aqueous enzymatic catalysis in solvent-free environments, reverse micelles, supercritical liquid and ionic liquid are also presented. In particular, while direct evolution, high-throughput screening and site-directed mutagenesis are combined as powerful tools for protein engineering, vapor/solid/ice water mixture, sticky solid-state liquid crystal and high density salt suspension are the future directions for solvent engineering in order to broaden the utility and elevate the efficiency of non-aqueous enzymatic catalysis.

Abstract:Water is the universal solvent in nature to catalyze the biological transformation processes. However, owing to the immiscibility of many reagents in water, synthesis chemistry relies heavily on organic solvent. Micellar media is a green alternative to traditional petroleum feedstock derived solvents, which is recently attracting increasing research attention. The present review deals with the recent advances in micellar catalysis with an emphasis on the new "tailor-made" surfactants for various reactions. A brief overview of commercial surfactants, including anionic micelles, cationic micelles, and nonionic micelles is presented. More importantly, an attempt was made to discuss systematically the recent research progress on new surfactants by introducing structures, micellar effects and recycling process, aiming to serve as the basis for future development of surfactants. Abstract:Enzymes have been widely used as efficient, eco-friendly, and biodegradable catalysts in organic chemistry due to their mild reaction conditions and high selectivity and efficiency. In recent years, the catalytic promiscuity of many enzymes in unnatural reactions has been revealed and studied by chemists and biochemists, which has expanded the application potential of enzymes. To enhance the selectivity and activity of enzymes in their natural or promiscuous reactions, many methods have been recommended, such as protein engineering, process engineering, and media engineering. Among them, the additive approach is very attractive because of its simplicity to use and high efficiency. In this paper, we will review the recent developments about the applications of additives to improve the catalytic performances of enzymes in their natural and promiscuous reactions. These additives include water, organic bases, water mimics, cosolvents, crown ethers, salts, surfactants, and some particular molecular additives.

Abstract:Lipases, crucial catalysts in biochemical synthesis, find extensive applications across industries such as food, medicine, and cosmetics. The efficiency of lipase-catalyzed reactions is significantly influenced by the choice of solvents. Polar organic solvents often result in a decrease, or even loss, of lipase activity. Conversely, nonpolar organic solvents induce excessive rigidity in lipases, thereby affecting their activity. While the advent of new solvents like ionic liquids and deep eutectic solvents has somewhat improved the activity and stability of lipases, it fails to address the fundamental issue of lipases' poor solvent tolerance. Hence, the rational design of lipases for enhanced solvent tolerance can significantly boost their industrial performance. This review provides a comprehensive summary of the structural characteristics and properties of lipases in various solvent systems and emphasizes various strategies of protein engineering for non-aqueous media to improve lipases' solvent tolerance. This study provides a theoretical foundation for further enhancing the solvent tolerance and industrial properties of lipases.

Abstract:Molecular sieves with different acidities and pore-sizes have been applied to catalyze the acetalization and ketalization of carbonyl compounds with glycol. A comparative research on the catalytic activities of the catalysts for certain reactions has been carried out to figure out the relationship between the characters of the catalysts and the activities. The results show that the pore-size of the molecular sieves is one of the most important factors that determine the catalytic activity. Appropriate pore-size is needed for the certain reaction. The acidity is another factor that affects the activities. They also have great effects on the recycled activities of the catalysts.

Abstract:Chemoselectivity of lipoprotein lipase from Pseudomonas specie is strongly influenced by solvation phenomena and can be modulated by changing the reaction medium. Experimental results obtained from enzymatic acylation of L-Ser-β-naphthylamide indicated that chemoselectivity depends on the nature of hydrogen bonds taking place among the molecular species present in the reaction medium. Inverted selectivity was observed upon moving from hydrogen bond donor solvents to hydrogen bond acceptor solvents. Solvent hydrophobicity can also influence chemoselectivity inducing intramolecular electrostatic interactions which alter the reactivity of the nucleophilic group(s). Solvation phenomena were studied by means of 1H NMR. An explanation of interactions between the solvated substrate and the enzyme is also proposed.

Abstract:Optimal enzyme activity depends on a number of factors, including structure and dynamics. The role of enzyme structure is well recognized; however, the linkage between protein dynamics and enzyme activity has given rise to a contentious debate. We have developed an approach that uses an aqueous mixture of organic solvent to control the functionally relevant enzyme dynamics (without changing the structure), which in turn modulates the enzyme activity. Using this approach, we predicted that the hydride transfer reaction catalyzed by the enzyme dihydrofolate reductase (DHFR) from Escherichia coli in aqueous mixtures of isopropanol (IPA) with water will decrease by ∼3 fold at 20% (v/v) IPA concentration. Stopped-flow kinetic measurements find that the pH-independent khydride rate decreases by 2.2 fold. X-ray crystallographic enzyme structures show no noticeable differences, while computational studies indicate that the transition state and electrostatic effects were identical for water and mixed solvent conditions; quasi-elastic neutron scattering studies show that the dynamical enzyme motions are suppressed. Our approach provides a unique avenue to modulating enzyme activity through changes in enzyme dynamics. Further it provides vital insights that show the altered motions of DHFR cause significant changes in the enzyme's ability to access its functionally relevant conformational substates, explaining the decreased khydride rate. This approach has important implications for obtaining fundamental insights into the role of rate-limiting dynamics in catalysis and as well as for enzyme engineering.

Thermodynamic predictions for biocatalysis in nonconventional media: theory, tests, and recommendations for experimental design and analysis

Abstract:This article discusses the application of thermodynamic and related analysis to reaction systems for enzymic or whole cell catalysis, in which there are high proportions of organic liquid, gas, or supercritical fluid. A variety of predictions may be made, especially based on the partitioning of components between the different phases normally present. In many cases, observed behavior can be explained without invoking any direct molecular effects on the biocatalyst. The predictable changes should always be allowed for before seeking explanations for the residual effects, which are often very different from the crude observations. A summary of the general thermodynamics of multiphase systems is presented, and then the main classes of component that distribute between the phases are discussed in turn. Thermodynamic water activity (aw) determines the mass action effects of water on hydrolytic equilibria. It also describes the distribution of water between the various phases that can compete in binding water. Because catalytic activity is very sensitive to the hydration of the enzyme molecules, aw often predicts an unchanging optimum as other aspects of the system are changed. Hence the aw should be measured and/or controlled in these systems, whether the primary aim is to study the effects of water or of other changes. The methods available for measurement and control of aw are discussed. Adverse effects of organic solvents or similar nonpolar species partly reflect their tendency to partition into the relatively polar phase around the biocatalyst, especially when this is dilute aqueous. The well-established log P parameter is a measure of this. But other mechanisms of inactivation can occur: directly through contact of the biocatalyst with the phase interface, or indirectly via hydration changes. In these cases the molecular property log P is probably not the best solvent parameter. In low-water systems the biocatalyst remains in a separate phase even when water-miscible solvents are used. Hence, the categorization of solvents in terms of miscibility becomes less relevant. This accounts for the "two peak" dependence of catalytic activity on water content in some miscible systems. Differential solvation of reactants and products, as the bulk phase is altered, causes changes in concentration-based equilibrium constants and yields. These changes in solvation may be monitored through partition coefficient or solubility measurements. Reactant solvation can also account for differences in biocatalyst kinetics, whether or not partitioning into a dilute aqueous phase is involved. These predictable effects should be allowed for when studying effects of solvent or similar changes on activity or specificity.(ABSTRACT TRUNCATED AT 400 WORDS)

The uses of ionic liquids in biocatalysis include improved separations and phase behaviour, reduction in toxicity, and stabilization of protein structures. As the ionic liquid state of the art has progressed, concepts of what can be achieved in biocatalysis using ionic liquids have evolved and more beneficial effects have been discovered. Abstract:Re-engineering enzymes with high activities in the given environments different from the physiological one has been constantly pursued for application of enzymatic catalysis in industrial biocatalytic processes, pharmaceutical industry, biosensing etc. Re-engineering enzyme catalysts by chemical approaches, including immobilization and chemical modification, represents a simple but effective route. The unusual phenomenon that immobilized or chemically modified enzymes display higher activities than native enzymes has been observed in both single- and multiple-enzyme systems. Recent achievements in enhancing enzymatic activities in both single-and multiple-enzyme systems by chemical approaches are summarized in this review. We propose that these enhanced enzymatic activities can be attributed to the well-designed specific interactions between immobilization carriers (or chemical modifiers) and enzymes, substrates, or reaction media. In addition to this mechanism, which is applicable for both single- and multiple-enzyme systems, other important factors responsible for enhanced activities of multiple-enzyme systems, including substrate channeling, kinetic matching, and an ordered spatial distribution of enzymes, are also discussed. Understanding the origin of enhanced activity in enzymatic catalysis may provide design efficient enzyme catalysts for practical applications.
