Abstract:We report on an integrated fiber optic design to implement multifiber angular compounding optical coherence tomography, which enables angular compounding for speckle reduction. A multi-facet fiber array delivers three light beams to the sample with different incident angles. Back-reflective/back-scattered signals from these channels were simultaneously detected by a three-channel spectrometer. The axial and lateral resolution was measured to be similar to 3 and similar to 3.5 mu m, respectively, in air with similar to 100 dB sensitivity. We conducted ex vivo experiments on a rat esophagus to demonstrate a contrast to noise improvement of 1.58. (C) 2016 Optical Society of America

Abstract:An improved sparse representation algorithm is proposed for reducing speckle noise in optical coherence tomography (OCT) images. To reduce the dictionary training time, the orthogonal matching pursuit algorithm (OMP) is replaced by a piecewise orthogonal matching pursuit algorithm (ST-OMP), and a threshold standard is set to train multiple atoms simultaneously to improve the tracking efficiency. Normal, diabetic macular edema, and fundus neovascularization membrane retina OCT images were tested using the proposed algorithm and other denoising algorithms. The results show that the proposed algorithm exhibits an excellent denoising ability. The details and edge information in the OCT images were well preserved by the proposed algorithm. Meanwhile, the image processing time was reduced by half with the proposed algorithm compared with the traditional K-SVD algorithm.

Abstract:Multi-channel detection is an effective way to improve data throughput of spectral-domain optical coherence tomography(SDOCT).However,current multi-channel OCT requires multiple detectors,which increases the complexity and cost of the system.We propose a novel multi-channel detection design based on a single spectrometer.Each camera pixel receives interferometric spectral signals from all the channels but with a spectral shift between two channels.This design effec-tively broadens the spectral bandwidth of each pixel,which reduces relative intensity noise(RIN)by√M times with M being the number of channels.We theoretically analyzed the noise of the proposed design under two cases:shot-noise limited and electrical noise or RIN limited.We show both theoretically and experimentally that this design can effectively improve the sensitivity,especially for electrical noise or RIN-dominated systems.

Abstract:We developed spectral domain optical coherence tomography using a dual-channel spectrometer for complex conjugate artifacts (CCA) suppression. We used a three-line charge coupled device to simultaneously detect two interferometric spectra with 2 pi/3 phase difference. The complex interferometric signal was reconstructed by trigonometric manipulation of two real interferometric spectra, and then full-range images were obtained by use of inverse Fourier transform. Artifacts at direct current (DC) and the ghost remnant of the CCA are common issues with the previously reported two-spectrometer method because the slight mismatching between two spectral detection channels had strong negative effects on CCA suppression and appeared to be the limiting factor on system performance. This novel dual-channel spectrometer uses the same spectrometer optics for the two spectral detection channels and, therefore, achieves better matching between two spectral detection channels and consequently better performance in CCA suppression as compared with the dual spectrometer solution. Fullrange imaging with CCA suppression up to similar to 25 dB was demonstrated when imaging an attenuated reflector. The efficacy of both CCA and DC suppressions also was validated by imaging the anterior segment of a rat eye ex vivo. The quality of CCA-suppressed images was significantly improved with regard to those obtained with the dual-spectrometer design. (C) 2017 Optical Society of America

Abstract:Optical coherence tomography (OCT) provides high resolution and cross-sectional images of biological tissue and is widely used for diagnosis of ocular diseases. However, OCT images suffer from speckle noise, which typically considered as multiplicative noise in nature, reducing the image resolution and contrast. In this study, we propose a two-step iteration (TSI) method to suppress those noises. We first utilize augmented Lagrange method to recover a low-rank OCT image and remove additive Gaussian noise, and then employ the simple and efficient split Bregman method to solve the Total-Variation Denoising model. We validated such proposed method using images of swine, rabbit and human retina. Results demonstrate that our TSI method outperforms the other popular methods in achieving higher peak signal-to-noise ratio (PSNR) and structure similarity (SSIM) while preserving important structural details, such as tiny capillaries and thin layers in retinal OCT images. In addition, the results of our TSI method show clearer boundaries and maintains high image contrast, which facilitates better image interpretations and analyses.

Abstract:In this paper, we present a distal-scanning common path probe for optical coherence tomography (OCT) equipped with a hollow ultrasonic motor and a simple and specially designed beam-splitter. This novel probe proves to be able to effectively circumvent polarization and dispersion mismatch caused by fiber motion and is more robust to a variety of interfering factors during the imaging process, experimentally compared to a conventional noncommon path probe. Furthermore, our design counteracts the attenuation of backscattering with depth and the fall-off of the signal, resulting in a more balanced signal range and greater imaging depth. Spectral-domain OCT imaging of phantom and biological tissue is also demonstrated with a sensitivity of similar to 100dB and a lateral resolution of similar to 3 mu m. This low-cost probe offers simplified system configuration and excellent robustness, and is therefore particularly suitable for clinical diagnosis as one-off medical apparatus.

Abstract:High-resolution optical coherence tomography (OCT) is of critical importance to disease diagnosis because it is capable of providing detailed microstructural information of the biological tissues. However, a compromise usually has to be made between its spatial resolutions and sensitivity due to the suboptimal spectral response of the system components, such as the linear camera, the dispersion grating, and the focusing lenses, etc. In this study, we demonstrate an OCT system that achieves both high spatial resolutions and enhanced sensitivity through utilizing a spectrally encoded source. The system achieves a lateral resolution of 3.1 μm and an axial resolution of 2.3 μm in air; when with a simple dispersive prism placed in the infinity space of the sample arm optics, the illumination beam on the sample is transformed into a line source with a visual angle of 10.3 mrad. Such an extended source technique allows a ~4 times larger maximum permissible exposure (MPE) than its point source counterpart, which thus improves the system sensitivity by ~6dB. In addition, the dispersive prism can be conveniently switched to a reflector. Such flexibility helps increase the penetration depth of the system without increasing the complexity of the current point source devices. We conducted experiments to characterize the system’s imaging capability using the human fingertip in vivo and the swine eye optic never disc ex vivo. The higher penetration depth of such a system over the conventional point source OCT system is also demonstrated in these two tissues.

Abstract:As a high-resolution imaging mode of biological tissues and materials, optical coherence tomography (OCT) is widely used in medical diagnosis and analysis. However, OCT images are often degraded by annoying speckle noise inherent in its imaging process. Employing the bilateral sparse representation an adaptive singular value shrinking method is proposed for its highly sparse approximation of image data. Adopting the generalized likelihood ratio as similarity criterion for block matching and an adaptive feature-oriented backward projection strategy, the proposed algorithm can restore better underlying layered structures and details of the OCT image with effective speckle attenuation. The experimental results demonstrate that the proposed algorithm achieves a state-of-the-art despeckling performance in terms of both quantitative measurement and visual interpretation. Abstract:Optical coherence tomography (OCT), as a low-coherence interferometric imaging technique, inevitably suffers from speckle noise, which can reduce image quality and signal-to-noise (SNR). In this paper, we present a dual-beam angular compounding method to reduce speckle noise and improve SNR of OCT image. Two separated parallel light beams are created on the sample arm using a 1x2 optical fiber coupler and are focused into samples at different angles. The epi-detection scheme creates three different light path combinations of these two light beams above. The three combinations produce three images in single B-scan, which are completely separated in depth. The three images show uncorrelated speckle patterns and therefore can be averaged to create a new image with reduced speckle noise. Compared to those reported angular and spatial compounding methods, our method retains their advantages, and moreover has a faster imaging speed and keep the transverse resolution. This method was evaluated on human fingertips in vivo. The results demonstrated a good improvement in speckle contrast.

Abstract:Image quality is an important parameter characterizing the performances of an optical coherence tomography (OCT) system. Low image quality not only deteriorates the image analysis and interpretations, but also impacts on the clinical applications of OCT systems, leading to misdiagnosis. Speckle noise is always present in OCT signals, and thus inevitably affects the OCT image quality. This paper studies the speckle noise reduction problem in OCT systems, and tries to compare a variety of the wavelet transform based methods. Specifically, we give the logical flow diagram of the dual-density dual-tree complex wavelet method first, and then combine it with the local variance estimation based bivariate contraction model for speckle noise reduction. By performing experiments on OCT images of human retina, swine eye and human dental, we compare the speckle noise reduction effects of the dual-density method, dual-density dual-tree real wavelet method (R2D) and dual-density dual-tree complex wavelet (C2D) method. Results show that the C2D method can effectively eliminate the speckle noise while retaining the important edge detail information of the OCT images.

Abstract:In this study, we propose a generic speckle simulation for optical coherence tomography (OCT) signal, by convolving the point-spread function (PSF) of the OCT system with the numerically synthesized random sample field. We validate our model and use the simulation method to study the statistical properties of cross-correlation coefficients between A-scans, which have been recently applied in transverse motion analysis by our group. The results of simulation show that oversampling is essential for accurate motion tracking; exponential decay of OCT signal leads to an underestimate of motion that can be corrected; lateral heterogeneity of sample leads to an overestimate of motion for a few pixels corresponding to the structural boundary. Abstract:Optical coherence tomography (OCT) is a micrometer-resolution, cross-sectional imaging modality for biological tissue. It has been widely applied for retinal imaging in ophthalmology. However, the large speckle noise affects the analysis of OCT retinal images and their diagnostic utility. In this article, we present a new speckle reduction algorithm for 3D OCT images. The OCT speckle noise is approximated as Poission distribution, which is difficult to be removed for its signal-dependent characteristic. Thus our algorithm is consisted by two steps: first, a variance-stabilizing transformation, named Anscombe transformation, is applied to redistribute the multiplicative speckle noise into an additive Gaussian noise; then the transformed data is decomposed and filtered in 3D Shearlet domain, which provides better representation of the edge information of the retinal layers than wavelet and curvelet. The proposed method is evaluated through the three parameters using high-definition B-scans as the ground truth. Quantitative experimental results show that our method gives out the best evaluation parameters, and highest edge contrast, compared with state-of-the-art OCT denoising algorithms.

Abstract:Optical coherence tomography (OCT) has been an important and powerful tool for biological research and clinical applications. However, speckle noise significantly degrades the image quality of OCT and has a negative impact on the clinical diagnosis accuracy. In this paper, we propose a novel speckle noise suppression technique which changes the spatial distribution of sample beam using a special optical chopper. Then a series of OCT images with uncorrelated speckle patterns could be captured and compounded to improve the image quality without degradation of resolution. Typical signal-to-noise ratio improvement of ∼6.4 dB is experimentally achieved in tissue phantom imaging with average number n = 100. Furthermore, compared with conventional OCT, the proposed technique is demonstrated to view finer and clearer biological structures in human skin in vivo, such as sweat glands and blood vessels. The advantages of low cost, simple structure and compact integration will benefit the future design of handheld or endoscopic probe for biomedical imaging in research and clinical applications.

Abstract:Optical coherence tomography (OCT) images are usually degraded by significant speckle noise, which will strongly hamper their quantitative analysis. However, speckle noise reduction in OCT images is particularly challenging because of the difficulty in differentiating between noise and the information components of the speckle pattern. To address this problem, the spiking cortical model (SCM)-based nonlocal means method is presented. The proposed method explores self-similarities of OCT images based on rotation-invariant features of image patches extracted by SCM and then restores the speckled images by averaging the similar patches. This method can provide sufficient speckle reduction while preserving image details very well due to its effectiveness in finding reliable similar patches under high speckle noise contamination. When applied to the retinal OCT image, this method provides signal-to-noise ratio improvements of >16 dB with a small 5.4% loss of similarity. Abstract:Optical coherence tomography (OCT) is an important imaging technique extensively applied in medical sciences. However, OCT images often suffer from speckle noise, which is a kind of multiplicative noise inherited in coherent imaging systems. A speckle noise reduction algorithm with total variation (TV) regularization is proposed to restore speckled OCT images. It constructs the regularization parameter and utilizes the tuning function. The proposed algorithm realizes sufficient speckle noise reduction and delicate edge preservation. Simulations demonstrate the performance of the proposed algorithm with respect to visual effects, processing time and image quality metrics of signal-to-noise ratio (SNR), equivalent number of looks (ENL), contrast-to-noise ratio (CNR), relative mean square error (RMSE) and edge preservation factor. Compared with some classical and typical despeckling algorithms, the proposed algorithm exhibits good results in edge preservation, recovery error and time efficiency, and presents better SNR, ENL and CNR. The applicability of the proposed algorithm with regard to OCT in-device preprocessing is discussed in details. Abstract:Optical Coherence Tomography (OCT) has been developed for more than one decade. With the optimum of system configuration such as light source, the imaging elements, the imaging quality has been improved to a higher level. However, many ideal assumptions including dispersion cancellation in the study of OCT system have become inapplicable. Actually, dispersion, can lead to a wavelength dependent phase distortion in sample arm, and finally result in a degrading in image resolution. Therefore, many dispersion compensation methods have been presented by researchers to correct the distorted image. In this paper, the principle of dispersion in OCT imaging system is discussed, and we demonstrate how it affects image quality Then, with respect to the compensation methods as our knowledge, we classify them into hardware compensation and software compensation and present the detailed procedures and their characteristics, respectively At last, a detailed discussion has been made to conclude that novel algorithms which can perform higher order compensation with depth variant are necessary and uniform evaluating criteria as well.

Abstract:We describe an angular compounding method by full-channel B-scan modulation encoding for speckle reduction in optical coherence tomography. The complex-valued spectral interferogram (SI) is reconstructed by removing one of the conjugate terms in the depth space. Fourier transform of the complex SI along the lateral direction enables a full-channel (with both negative and positive Fourier space) for B-scan modulation in the spatial frequency (ν) domain. A full-size probe beam, determined by the scanning mirror size, is centered on the mirror pivot, which allows the negative and positive half-channels working in parallel. Compared with the existing method, where only a half-channel (negative or positive) works alone, the proposed full-channel method offers more than twice the performance in speckle reduction. The feasibility of the proposed full-space approach is validated through both the phantom and in vivo human thumbnail experiments.

Abstract:This paper presents a common-path (CP) swept-source optical coherence tomography (SSOCT) system based on a special endoscopic probe design with an in-line internal reflection as the reference and a two-axis electrothermal microelectromechanical system mirror for image scanning. The rear surface of a gradient reflective index (GRIN) lens inside the probe is set as the reference reflection plane. The length of the GRIN lens is optimized to eliminate the artifacts in SSOCT images successfully. Doppler OCT is also demonstrated based on the CP endoscopic probe. The diameter of the probe is only 2.5 mm, so it can be easily inserted into the biopsy channel of traditional endoscopes to access human internal organs for in vivo diagnoses.
Interchangeable Side-Viewing Endoscopic Probes for Optical Coherence Tomography (conference Presentation)

Abstract:We present a proof-of-concept for side-viewing endoscopic optical coherence tomography probes designed for real-time imaging. The design employs a commercial rotating micro-engine (Kinetron), with a 1 mm outer diameter and maximum speed of 10,000 rpm, to steer the beam from a GRIN lens-terminated optical fiber (Agiltron), 360 degrees around the probe body. The engine is encapsulated inside a PET tube with an outer diameter of 1.6 mm, and coupled to a swept-source based optical coherence tomography (SS-OCT) system operating at 1300 nm with an A-scan rate of 100 kHz. Large material dispersion mismatch between the reference and sample arms, which would otherwise degrade the axial resolution, is compensated for by using the Master-Slave OCT technique to process the interferograms. This allows for more convenient interchange of probes without exactly matching the fibre length. The probe can be configured using GRIN lenses at the end of the fibers with different focal lengths, and the fiber lengths can differ by several cm. When the probe is changed, the reference path is adjusted, but no dispersion compensation is needed, due to use of the Master Slave method developed by our group. We will demonstrate the performance of such systems with full 360 degree tomographic images of scattering phantoms taken at different probe driving speeds, with different configurations of the probe heads.
