Abstract:An enantioselective synthesis of pharmaceutically important spirobarbiturates has been achieved via spirocyclic chiral phosphine-catalyzed asymmetric [4 + 2] annulation of barbiturate-derived alkenes with allenoates. With the use of this tool, various spirobarbiturate-cyclohexenes are obtained in good to excellent yields with excellent diastereo- and enantioselectivities. A wide range of α-substituted allenoates and barbiturate-derived alkenes were tolerated.

Abstract:A palladium-catalyzed asymmetric [4+2] cycloaddition of 2-methylidenetrimethylene carbonate with alkenes derived from pyrazolones, indandione, or barbiturate has been successfully developed, affording pharmacologically interesting chiral tetrahydropyran-fused spirocyclic scaffolds. The target compounds were generated in good to excellent yields and with high enantioselectivity (up to 99 %ee). Furthermore, this cycloaddition reaction could be efficiently scaled up, and several synthetic transformations were accomplished for the construction of other useful chiral spiropyrazolone and spiroindandione derivatives.

Abstract:Herein, we report the first enantioselective annulation of α-arylidene pyrazolones through a formal C(sp3 )-H activation under mild conditions enabled by highly variable RhIII -Cpx catalysts. The method has a wide substrate scope and proceeds with good to excellent yields and enantioselectivities. Its synthetic utility was demonstrated by the late-stage functionalization of drugs and natural products as well as the preparation of enantioenriched [3]dendralenes. Preliminary biological investigations also identified the spiropyrazolones as a novel class of Hedgehog pathway inhibitors.

Abstract:An efficient route to chiral carbocyclic nucleoside analogues containing a quaternary stereocenter and a C═C double bond has been established via a highly enantioselective [3 + 2] annulation of Morita-Baylis-Hillman (MBH) carbonates with α-purine-substituted acrylates. With 20 mol % (S)-SITCP as the catalyst, various chiral carbocyclic nucleoside analogues with a quaternary stereocenter and a C═C double bond were obtained in high yields (up to 92%) with good diastereoselectivities (up to 10:1 dr) and excellent enantioselectivities (up to 96% ee). Furthermore, the corresponding products were subjected to diverse transformations to afford interesting and potentially useful chiral carbocyclic nucleosides.

Abstract:A highly enantioselective formal [4 + 2] annulation involving electron-deficient allenes as C2-synthons has been developed under bifunctional phosphonium salt catalysis. With this catalytic protocol, a wide range of synthetically interesting and highly functionalized chiral 4H-pyran derivatives were readily prepared in good yields (up to 99%) with outstanding diastereo- and enantioselectivities (up to >20:1 dr, up to >99.9% ee). The utility and the practicality of this method were demonstrated by the gram-scale reaction and facile elaboration. Of note, this is the first example of an asymmetric annulation reaction involving allenic reactants under a phase-transfer catalysis system.

Abstract:A chiral phosphoric acid-catalyzed enantioselective synthesis of spiro[indoline-3,6'-indolo[1,2-c]quinazolin]-2-ones has been developed by condensation/N-alkylation cascade from 2-(1H-indolyl)anilines and isatins, affording the spiro aminals with quaternary stereogenic centers with up to 93% ee. The protocol could be expanded to 2-(3,5-dimethyl-1H-pyrrol-2-yl)aniline and a variety of pyrrole-derived spiro chiral aminals were also obtained in good yields (71-91%) with excellent enantioselectivities (89%-94% ee). Abstract:We herein present the construction of a chiral 1,4-enyne featuring tertiary or quaternary stereogenic center via Pd-catalyzed branch-, enantio-, and diastereoselective allylic C-H alkylation. Alkynyl carbon bearing bulky substituents appeared to exhibit competitive reaction performance, and the desired chiral 1,4-enynes were obtained in up to 93% yield and with up to >20:1 b/l, >20:1 dr, and 98% ee. A gram-scale experiment, the feasible operation of benzothiazole ring, and the preparation of the key intermediate to access (+)-Breynolide and prostaglandin are represented as a demonstration of multifarious synthetic utility in chemical synthesis.

Abstract:Chiral phosphines were found to catalyze the enantioselective asymmetric gamma-addition of heteroaromatic compounds to allenoates in good yields with high enantiomeric ratios and regioselectivity in the presence of (S)-BINOL. Both pyrazole and imidazole could be employed in this process. The synthetic value of these gamma-addition products was demonstrated by the preparation of biologically relevant molecules and structural scaffolds. Remarkably, the synthetic utility of this strategy was demonstrated through a two-step synthesis of a Janus kinase (JAK) inhibitor.
