Abstract:In this study, bioassay-guided fractionation of extracts from the leaves of Morus alba L led to the isolation of 15 bioactive constituents with alpha-glucosidase and tyrosinase inhibitory activities, among which prenylated stilbenes were proved to be a new group of alpha-glucosidase inhibitors apart from iminosugars derived from Morus alba. Their structures were identified on the basis of extensive spectroscopic analysis and chemical evidence, as well as comparing with data from the literature. Among them, compounds (2R)/(2S)-Euchrenone a(7) (6a/6b), Chalcomoracin (7), Moracin C(8), Moracin D (9) and Moracin N (10) exhibited a significant degree of alpha-glucosidase inhibitory activity with IC50 of 6.28, 2.59, 4.04, 2.54 and 2.76 mu M, respectively, while (2R)/(2S)-Euchrenone a(7) (6a/6b), Moracin N (10), Quercetin (13), Norartocarpetin (14), the interconvertible epimeric mixture of (2R)/(2S)-7-methoxyl-8-ethyl-2',4'-dihydroxylflavane-2"-O-beta-D-glucopyranoside (1a/1b) and the interconvertible enantiomers of (2R)/(2S)-7-methoxyl-8-hydroxyethyl-2',4'-dihydroxylflavane (5a/5b) displayed a potent tyrosinase inhibitory effect with IC50 of 0.260, 0.924, 0.523, 0.0824, 0.616 and 0.528 mu M, respectively. Especially, (2R)-7-methoxyl-8-ethyl-2',4'dihydroxylflavane-2"-O-beta-D-glucopyranoside (1a), (2S)-7-methoxyl-8-ethyl-2',4'-dihydroxylflavane-2"-O-beta-D-glucopyranoside (1b), (2S)-8-hydroxyethy1-7,4'-dimethoxylflavane-2'-O-beta-D-glucopyranoside (2), (2R)-7-methoxyl-8-hydroxyethyl-2',4'-dihydroxylflavane (5a) and (2S)-7-methoxyl-8-hydroxyethyl-2',4'-dihydroxylflavane (5b) were identified as new compounds. Abstract:Bioassay-guided fractionation, based on inhibition of -glucosidase, of Euonymus alatus led to the isolation of a new compound, catechin lactone A (7), and seven known compounds (1-6 and 8). Their structures were established on the basis of extensive 1D- and 2D-NMR experiments. The absolute configurations of compound 7 have been determined using vibrational circular dichroism spectroscopy. All these compounds were evaluated for their inhibition activities on -glucosidase and differentiation of 3T3-L1 cells. Compounds 1, 3-5, 7 and 8 showed potent yeast -glucosidase inhibition with IC50 values of 119.1, 39.6, 31.6, 23.0, 71.8 and 102.5 mu M, respectively; compounds 3-5, 7 and 8 can remarkably inhibit the differentiation of 3T3-L1 pre-adipocytes. This is the first report about -glucosidase and 3T3-L1 inhibitory activities of compounds 3, 7 and 8.

Abstract:The twigs of Morus alba L. were found to show strong tyrosinase inhibition activity, and the responsible active components in the extract were further investigated in this study. A flavone, named morusone (1), and sixteen known compounds 2-17 were isolated from M. alba twigs and their structures were identified by interpretation of the corresponding ESI-MS and NMR spectral data. In the tyrosinase inhibitory test, the compounds steppogenin (IC50 0.98 ± 0.01 µM), 2,4,2',4'-tetrahydroxychalcone (IC50 0.07 ± 0.02 µM), morachalcone A (IC50 0.08 ± 0.02 µM), oxyresveratrol (IC50 0.10 ± 0.01 µM), and moracin M (8.00 ± 0.22 µM) exhibited significant tyrosinase inhibition activities, much stronger than that of the positive control kojic acid. These results suggest that M. alba twig extract should served as a good source of natural tyrosinase inhibitors for use in foods as antibrowning agents or in cosmetics as skin-whitening agents.

Their structures were elucidated by spectroscopic analysis. Moracins X (3) and Y (4) represent unusual substituted group compared with 2-arylbenzofuran derivatives, which were isolated from the genus Morus. Compounds 2-5 were evaluated for cytotoxicity against several human cancer cell lines.

Abstract:Three new compounds (1, 11, and 12), together with 32 known ones, were isolated from the root bark of Morus alba L. using various chromatographic methods. The structures of the undescribed compounds were elucidated based on 1D, 2D NMR, and HRESIMS dataanalysis, while the known ones were identified by comparison of their spectroscopic data with those reported in the literature. All the isolates were evaluated for their cytotoxic activities against human gastric cancer HGC27 cells by CCK-8 assay. Among them, compounds 5, 8, 10, and 30 exhibited cytotoxic activities on HGC27 cells with IC50 values of 33.76 ± 2.64 μM, 28.94 ± 0.72 μM, 6.08 ± 0.34 μM, and 10.24 ± 0.89 μM, respectively. Furthermore, compound 10 was confirmed to reduce proliferation ability, increase apoptosis rate, and inhibit cell migration pathway by annexin V/PI double staining experiment, transwell experiment, and Western blot analysis.

Abstract:A new 2-arylbenzofuran derivative, mornigrol D (1), along with two new flavones, mornigrol G (2) and mornigrol H (3), and six known compounds, norartocarpetin (4), dihydrokaempferol (5), albanin A (6), albanin E (7), moracin M (8), and albafuran C (9), were isolated from the barks of Morus nigra. Their structures were elucidated by spectroscopic analysis. Compounds 1 and 9 showed antioxidative activities in vitro with inhibition ratios of 98 and 99% at the concentration of 10-4mol/l, and of 74 and 75% at the concentration of 10-5mol/l. In addition, compounds 1 and 4 showed potent anti-inflammatory activities (inhibition of release of -glucuronidase from rat polymorphonuclear leucocytes induced by platelet activating factor) with inhibitory ratios of 65.9% (P0.01) and 67.7% (P0.01) at a concentration of 10-5mol/l.

Abstract:Abstract Objective To study the chemical constituents from root bark of Morus alba and their α-glucosidase inhibition and DPPH radical scavenging activities. Methods The chemical constituents were isolated and purified by repeated column chromatographies on silica gel, Sephadex LH-20, and preparative HPLC. Their structures were elucidated by 1D and 2D NMR spectra and HR-ESI-MS. Results Thirteen compounds 1 – 13 were isolated and identified. The bioactive assays revealed that compounds 1, 3 and 8 displayed strong α-glucosidase inhibitory activity with IC 50 values of (147.1 ± 1.1), (314.1 ± 0.8), and (207.6 ± 0.1) µmol/L, respectively, which were stronger than the positive control of acarbose (418.6 ± 0.1 µmol/L). Compounds 10 and 11 displayed potent DPPH scavenging activity with EC 50 values of (2.9 ± 0.1) and (5.0 ± 0.1) µmol/L [EC 50 of positive control Vitamin C was (54.8 ± 0.1) µmol/L], respectively. Conclusion To the best of our knowledge, this is the first report about the compounds 1, 3 , and 8 of M. alba with α-glucosidase inhibitory effects.

Abstract:Twenty-two flavonoids and stilbenes (1-22) were obtained from the leaf of Morus alba var. multicaulis. Among them, morusalbanosides A (1), B1 (2), and B2 (3) were new compounds. Moreover, compounds 1, 3, 4-11, 15-18, and 22 displayed inhibitory effects on triglyceride (TG) accumulation in HepG2 cells in a concentration dependent manner. Furthermore, compounds 1, 3, 11, and 22 could activate the phosphorylation of AMP-activated protein kinase α (AMPKα), reduce the synthesis of TG by inhibiting the expression of fatty acid synthase (FAS) and stearoyl-CoA desaturase 1 (SCD1). While, only compounds 1 and 11 could promote the phosphorylation of acetyl-CoA carboxylase 1 (ACC1) and accelerate the oxidation of fatty acids by up-regulating carnitine palmitoyltransferase 1A (CPT1A). In brief, this study found that most of the researched flavonoids and stilbenes could regulate TG metabolism in vitro. They might play the role by up-regulating phosphorylation of AMPKα, inhibiting TG biosynthesis, and promoting the oxidation of fatty acids.

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Morus alba L. (mulberry) is a well-known medicinal species that has been used by herbalist doctors for the treatment of diabetes for a long history, and modern ethnopharmacological studies have demonstrated the ameliorating effects of different mulberry extracts toward diabetes-related symptoms and identified a number of α-glucosidase inhibitors as hypoglycemic ingredients.AIM OF THE STUDY: The present study aims to explore new potent α-glucosidase inhibitors from the root bark of M. alba (known as Sang-Bai-Pi in traditional medicine) based on an in vivo antidiabetic evaluation of its extract fractions and further characterize the preliminary mechanism of the new active constituents.MATERIALS AND METHODS: α-Glucosidase inhibitory assay and diabetic mice model were used to locate and evaluate the active fractions from the extract. Diverse separation techniques (e.g. Sephadex LH-20 column chromatograph (CC) and HPLC) and spectroscopic methods (e.g. MS, NMR and ECD) were employed to isolate and structurally characterize the obtained constituents, respectively. Fluorescence quenching, kinetics and molecular docking experiments were conducted to investigate the enzyme inhibitory mechanism of the active compounds.RESULTS: The 80% ethanol eluate from the macroporous resin CC exerted good antidiabetic effects in the tested mice. Fifty-two flavonoids including 22 new ones were then separated and identified, and most of them showed strong inhibition against α-glucosidase with their structure-activity relationship being also discussed. The four new most active ingredients were further characterized to be mixed type of α-glucosidase inhibitors, and their binding modes with the enzyme were also explored.CONCLUSIONS: Our current work has demonstrated that the root bark of M. alba is an extremely rich source of flavonoids as potent α-glucosidase inhibitors and potential antidiabetic agents, which makes it a promising candidate species to develop new natural remedies for the prevention and treatment of diabetes.

Abstract:Two novel rearranged Diels–Alder adducts, morunigrines A (1) and B (2), and four new prenylated flavonoids, morunigrols A–D (3–6), were isolated from the twigs of Morus nigra, together with four known prenylated phenolic compounds, including two flavonoids (7 and 8) and two 2-arylbenzofurans (9 and 10). Morunigrines A (1) and B (2) are a novel class of Diels–Alder adducts with unprecedented carbon skeletons featuring a rearranged chalcone–stilbene/2-arylbenzofuran core decorated with a unique methylbiphenyl moiety. The structures of the new compounds were assigned by analysis of spectroscopic data. The absolute configuration of compound 6 was determined by the measurement of specific rotation. A plausible biogenetic pathway for 1 and 2 is also proposed. Abstract:Ultraviolet-B (UV-B) radiation is harmful to plants and human beings. Many secondary metabolites, like flavonoids, alkaloids, and lignin, are UV-B absorbing compounds, which can protect the genetic material of plants. Furthermore, they are active components of herbal drugs. UV-B radiation can activate the self-protective secondary metabolism system. The results of this paper provide a method to induce bioactive secondary metabolites from mulberry leaves (Morus alba L.) by UV-B irradiation in vitro. Five significantly different chromatographic peaks were found by HPLC fingerprint after induction, from which two active compounds were identified: One was chalcomoracin, a natural Diels-Alder type adduct with antibacterial activity; the other one was moracin N, which is a precursor of chalcomoracin. Their contents were 0.818 mg/g and 0.352 mg/g by dry weight, respectively.
