Abstract:Chemical investigation of the root bark of Morus alba led to the isolation of a new flavone, dioxycudraflavone A (1) and a new 2-arylbenzofuran, 5-hydroxyethyl moracin M (2), together with seven known compounds namely sanggenon V (3), morusin (4), morusignin L (5), licoflavone C (6), moracin C (7), alfafuran (8) and mulberrofuran G (9). The structure elucidation of these compounds was based on analyses of spectroscopic data including 1D, 2D NMR and HR-ESIMS. All compounds were evaluated for the alpha-glucosidase inhibitory and cytotoxic activities. Compounds 2-4, 8 and 9 exhibited strong alpha-glucosidase inhibitory activities with IC50 less than 10 mu M, while only 4 and 9 showed moderate cytotoxic effects against lung cancer cells. Abstract:Four new 2-arylbenzofuran derivatives, moracins V-Y (1-4), together with two known compounds, moracin N (5) and moracin P (6), were isolated from the leaves of Morus alba L. Their structures were elucidated by spectroscopic analysis. Moracins X (3) and Y (4) represent unusual substituted group compared with 2-arylbenzofuran derivatives, which were isolated from the genus Morus. Compounds 2-5 were evaluated for cytotoxicity against several human cancer cell lines.

Abstract:Objective To investigate the chemical constituents from the berries of Morus alba L.Methods The compounds were isolated by chromatographies on silica gel column,ODS column,Sephadex LH-20 column,preparative HPLC and their chemical structures were elucidated on the basis of physicochemical properties and spectroscopic data.Results Seven compounds were isolated and their structures were identified to be aurantiamide (1),cytochalasin B (2),(22E)-24-methyl-5α-cholesta-7,22-diene-3β,5α,6β-triol(3),(22E)-5α,8α-epidioxyergosta-6,22-dien-3β-ol (4),7β-hydroxy-sitosterol(5),β-sitosterol(6),and daucosterine(7).Conclusion Compounds 1-5 were isolated from the berries of Morus alba L.for the first time.

Abstract:An undescribed coumarin–limonene hybrid (morusalin A, 1 ), an unreported steppogenin–limonene hybrid (morusalin B, 2 ), and three undescribed isoprenylated flavonoids (morusalins C–E, 3 – 5 ), along with forty-one known ones ( 6 – 46 ), were isolated from the dried roots bark of Morus alba . Their structures with absolute configurations were elucidated by detailed interpretation of NMR spectroscopy, mass spectrometry, single-crystal X-ray diffraction, and ECD calculations. In addition, all these isolated compounds were assayed for their inhibitory activity against lipopolysaccharide (LPS)-induced nitric oxide (NO) production in murine macro-phage RAW 264.7 cells and for their cytotoxic activities against human colon cancer (HCT 8) and hepatoma (HepG 2) cells. Moreover, a biosynthetic pathway for the formation of compounds 1 – 6 is also proposed.

Abstract:A new 2-arylbenzofuran derivative, mornigrol D (1), along with two new flavones, mornigrol G (2) and mornigrol H (3), and six known compounds, norartocarpetin (4), dihydrokaempferol (5), albanin A (6), albanin E (7), moracin M (8), and albafuran C (9), were isolated from the barks of Morus nigra. Their structures were elucidated by spectroscopic analysis. Compounds 1 and 9 showed antioxidative activities in vitro with inhibition ratios of 98 and 99% at the concentration of 10-4mol/l, and of 74 and 75% at the concentration of 10-5mol/l. In addition, compounds 1 and 4 showed potent anti-inflammatory activities (inhibition of release of -glucuronidase from rat polymorphonuclear leucocytes induced by platelet activating factor) with inhibitory ratios of 65.9% (P0.01) and 67.7% (P0.01) at a concentration of 10-5mol/l.

Abstract:Two new 2-arylbenzofuran derivatives, moracinfurol A and B (1–2), and ten known compounds (3–12) were isolated from the leaves of Morus alba. Their structures were determined on the basis of spectroscopic analysis including 1D, 2D NMR and HR-ESI-MS. All of the 2-arylbenzofuran derivatives were evaluated for cytotoxicity against A549 cells. Some cytotoxic 2-arylbenzofuran derivatives might induce autophagy characterized by the accumulation of LC-3 Ⅱ.

Abstract:Four new flavonoids, mortatarins A–D (1–4, resp.), along with eight known flavonoids (5–12) were isolated from the root bark of Morus alba var. tatarica. Their structures were established on the basis of spectroscopic data analysis, and the absolute configuration of 4 was determined by analysis of its CD spectrum. All isolates were tested for inhibitory activities against α‐glucosidase. Compounds 4, 7, and 8 exhibited a significant degree of inhibition with IC50 values of 5.0±0.3, 7.5±0.5, and 5.9±0.2 μM, respectively.

Abstract:Three new compounds (1, 11, and 12), together with 32 known ones, were isolated from the root bark of Morus alba L. using various chromatographic methods. The structures of the undescribed compounds were elucidated based on 1D, 2D NMR, and HRESIMS dataanalysis, while the known ones were identified by comparison of their spectroscopic data with those reported in the literature. All the isolates were evaluated for their cytotoxic activities against human gastric cancer HGC27 cells by CCK-8 assay. Among them, compounds 5, 8, 10, and 30 exhibited cytotoxic activities on HGC27 cells with IC50 values of 33.76 ± 2.64 μM, 28.94 ± 0.72 μM, 6.08 ± 0.34 μM, and 10.24 ± 0.89 μM, respectively. Furthermore, compound 10 was confirmed to reduce proliferation ability, increase apoptosis rate, and inhibit cell migration pathway by annexin V/PI double staining experiment, transwell experiment, and Western blot analysis.

Abstract:The secondary metabolites of Morus alba leaves were analysed with chromatographic and spectral methods.Ten compounds were isolated from the leaves.Their structures were identified as 5,7-dihydroxycoumarin-7-O-β-D-glucopyranoside(1),5,2′,4′-trihydroxyflavone-7-O-β-D-glucopyranoside(2),scopolin(3),butanedioic(4),3,4-dihydroxylbenzoic acid(5),kaempferol-3-O-(6″-O-2-butenoyl)-β-D-glucopyranoside(6),kaempferol-3,7-di-O-β-D-glucopyranoside(7),uridine(8),thymine(9),D-galactitol(10).Compounds 5～10 were isolated from the genus Morus for the first time.Compound 2 is a novel natural product,and compound 1 is a new compound.

Abstract:The column chromatography on silica gel, Sephadex LH-20 and semi-preparative HPLC were used to separate and purify the compounds from the EtOAc extract of medium and MeOH extract of cell cultures of Morus alba. Eight compounds were isolated. Based on physico-chemical properties and spectroscopic data, their structures were identified as isobavachalcone (1), genistein (2), norartocarpetin (3), albanin A (4), guangsangon E (5), mulberrofuran F (6), chalcomoracin (7), kuwanon J (8). Compounds 3-6 were isolated from the cell cultures of M. alba for the first time.

Abstract:Ultraviolet-B (UV-B) radiation is harmful to plants and human beings. Many secondary metabolites, like flavonoids, alkaloids, and lignin, are UV-B absorbing compounds, which can protect the genetic material of plants. Furthermore, they are active components of herbal drugs. UV-B radiation can activate the self-protective secondary metabolism system. The results of this paper provide a method to induce bioactive secondary metabolites from mulberry leaves (Morus alba L.) by UV-B irradiation in vitro. Five significantly different chromatographic peaks were found by HPLC fingerprint after induction, from which two active compounds were identified: One was chalcomoracin, a natural Diels-Alder type adduct with antibacterial activity; the other one was moracin N, which is a precursor of chalcomoracin. Their contents were 0.818 mg/g and 0.352 mg/g by dry weight, respectively.

They might play the role by up-regulating phosphorylation of AMPKα, inhibiting TG biosynthesis, and promoting the oxidation of fatty acids.

Abstract:OBJECTIVE:To study the chemical constituents of the twigs of Morus atropurpurea.METHOD:The compounds of the EtOAc fraction were isolated and purified by column chromatography on silica gel, polyamide, Sephadex LH -20, and their structures were elucidated on the basis of spectroscopic evidence (MS, NMR).RESULT:Eleven compounds were identified as mulberrin (1), cyclomulberrin (2), morusin (3), cyclomorusin (4), 2', 4',4, 2"-tetrahydroxy-3'-{3"-methylbut-3"-enyl-}-chalcone (5), mulberrofran G (6), scopoletin (7), moruchalcone A (8), kaempferol (9), ursolic acid (10), beta-daucosterol (11).CONCLUSION:Except compounds 9 and 11, all the other compounds were obtained from M. atropurpurea for the first time.

Abstract:The twigs of Morus alba L. were found to show strong tyrosinase inhibition activity, and the responsible active components in the extract were further investigated in this study. A flavone, named morusone (1), and sixteen known compounds 2-17 were isolated from M. alba twigs and their structures were identified by interpretation of the corresponding ESI-MS and NMR spectral data. In the tyrosinase inhibitory test, the compounds steppogenin (IC50 0.98 ± 0.01 µM), 2,4,2',4'-tetrahydroxychalcone (IC50 0.07 ± 0.02 µM), morachalcone A (IC50 0.08 ± 0.02 µM), oxyresveratrol (IC50 0.10 ± 0.01 µM), and moracin M (8.00 ± 0.22 µM) exhibited significant tyrosinase inhibition activities, much stronger than that of the positive control kojic acid. These results suggest that M. alba twig extract should served as a good source of natural tyrosinase inhibitors for use in foods as antibrowning agents or in cosmetics as skin-whitening agents.
