Abstract:The chlorination of a bioactive compound can change its physiological properties and improve its pharmacokinetic and pharmacological profiles. It therefore has been an important strategy for drug discovery and development. However, the direct aromatic chlorination of complex bioactive molecules is too difficult to be practical. In fact, many functional groups such as hydroxyls, amines, amides or carboxylic acids may strongly restrain the reactivity of Cl+ by forming a halogen bond. Here we report a highly efficient aromatic chlorination of arenes that is catalysed by dimethyl sulfoxide with N-chlorosuccinimide as the chloro source. The mild conditions, easy-availability and stability of the catalyst and reagents, as well as good functional-group tolerance, showed the approach to be a versatile protocol for the late-stage aromatic chlorination of complex natural products, drugs and peptides. The multi-gram experiment and low-cost of N-chlorosuccinimide and dimethyl sulfoxide shows great potential for drug discovery and development in industrial applications. Late-stage aromatic chlorination of active pharmaceutical ingredients has enormous potential in drug discovery yet still features limited applicability due to issues of functional-group tolerance. Now, dimethyl sulfoxide is reported as catalyst for the chlorination of a diverse family of bioactive molecules in combination with N-chlorosuccinimide.

Abstract:An efficient and practical method for the bromination of electron-rich arenes and heteroarenes was developed by using S -methyl methanethiosulfonate as the oxidant. All the bromine atoms were basically transferred to the brominated products, demonstrating the exceptional atom economy and practicality of the proposed protocol. The method reduces the amount of bromine required for this reaction system and obtains products in moderate to good yields. Abstract:A mild and practical method for synthesizing sulfonyl derivatives, which have a wide range of applications in pharmaceuticals, materials, and organic synthesis, was described through the oxidative functionalization of thiols with DMSO/HBr. The simple conditions, low cost and ready availability of DMSO/HBr, as well as the versatility of the transformations, make this strategy very powerful in synthesizing a variety of sulfonyl derivatives including sulfonamides, sulfonyl fluorides, sulfonyl azides, and sulfonates. Mechanistic studies revealed that DMSO served as the terminal oxidant, and HBr acted as both a nucleophile and a redox mediator to transfer the oxygen atom.

Abstract:A mild and general procedure for the difluoromethylation of organochalcogen compounds has been accomplished via sulfoxide and selenoxide-directed [3,3]-sigmatropic rearrangement/Haller-Bauer reaction with difluoroenoxysilanes. The reactions showed good functional group compatibility, wide substrate applicability, and facile scalability. The synthetic utility is highlighted by the iterative use of this protocol to construct multi-CF2H-containing chalcogenides and the late-stage modification of pharmaceuticals.

Abstract:A highly regioselective iodosulfonylation of allenes in the presence of CuI and 1,10-phenanthroline has been developed for the synthesis of various useful ( E)-α-iodomethyl vinylsulfones in moderate to excellent yields. This practical reaction is fast, operationally simple, and in particular, proceeds under very mild conditions to afford the target products with high regio- and stereoselectivity. The selectivity was illustrated by a conceptual DFT analysis.

Abstract:Aromatization-driven deconstruction and functionalization of spiro dihydroquinazolinones via dual photoredox/nickel catalysis is developed. The aromatization effect was introduced to synergistically drive unstrained cyclic C-C bond cleavage, with the aim of overcoming the ring-size limitation of nitrogen-centered radical induced deconstruction of carbocycles. Herein, we demonstrate the synergistic photoredox/nickel catalyzed deconstructive cross-coupling of spiro dihydroquinazolinones with organic halides. Remarkably, structurally diverse organic halides including aryl, alkenyl, alkynyl, and alkyl bromides were compatible for the coupling. In addition, this protocol is also characterized by its mild and redox-neutral conditions, excellent functional group compatibility, high atom economy, and easy scalability. A telescoped procedure involving condensation and ring-opening/coupling was found to be accessible. This work provides a complementary strategy to the existing radical-mediated C-C bond cleavage of unstrained carbocycles.

Abstract:Practical approaches for chemoselective mono-bromination, di-bromination, and tetra-bromination of terminal alkynes to generate 1-bromoalkynes, 1,2-dibromoalkenes, α,α-dibromoketones, and 1,1,2,2-tetrabromoalkanes based on efficient oxidative brominations mediated by a hypervalent iodine reagent have been developed. Chemoselective bromination can be realized under mild conditions by altering the bromine source. The tetrabutylammonium bromide (TBAB)/(diacetoxyiodo)benzene (PIDA) system is specific for mono-bromination to provide 1-bromoalkynes, while the NaBr/PIDA system is selective toward di-bromination to achieve 1,2-dibromoalkenes. When a certain amount of water was added to the NaBr/PIDA system, a different di-bromination product, α,α-dibromo ketones, was generated. Tetra-bromination of terminal alkynes provides an efficient protocol for the synthesis of 1,1,2,2-tetrabromoalkanes in a system with an excess loading of NaBr/PIDA in one pot. This bromination affords good yields (up to 99%) with excellent chemoselectivity (up to 100%). These methods can be applied to the efficient chemoselective synthesis of bromide derivatives, intermediates, and related biologically active compounds.

Abstract:A substrate-induced strategy for the bromochlorination and bromoazidation of diverse 1,3-dienes has been reported. This protocol offers straightforward and highly selective access to vicinal or allyl chlorobromides and azidobromides, which are ubiquitous moieties in many natural products and valuable synthetic intermediates. Additionally, a series of transformations of the products were performed to demonstrate their synthetic potential. Furthermore, control experiments were performed to elucidate the mutual conversion pathway between vicinal and allyl chlorobromides. Bromochlorination and bromoazidation of diverse 1,3-dienes to prepare various chlorobromides and azidobromides with high efficiency and selectivity using a general protocol with mild conditions and simple operation.

Abstract:Organic thioethers play an important role in the discovery of drugs and natural products. However, the green synthesis of organic sulfide compounds remains a challenging task. The convenient and efficient synthesis of 5-alkoxy-3-halo-4-methylthio-2(5H)-furanones from DMSO is performed via the mediation of 1,3-dibromo-5,5-dimethylhydantoin (DBDMH), affording a facile route for the sulfur-functionalization of 3,4-dihalo-2(5H)-furanones under transition metal-free conditions. This new approach has demonstrated the functionalization of non-aromatic Csp2-X-type halides with unique structures containing C-X, C-O, C=O and C=C bonds. Compared with traditional synthesis methods using transition metal catalysts with ligands, this reaction has many advantages, such as the lower temperature, the shorter reaction time, the wide substrate range and good functional group tolerance. Notably, DMSO plays multiple roles, and is simultaneously used as an odorless methylthiolating reagent and safe solvent.

Abstract:DMSO‐TsCl has been developed for direct methylthiolation of various electron‐rich heteroarenes (more than 40 examples) with high regioselectivity in moderate to excellent yields (up to 96%). Especially, pyrroles, furans and thiophenes can be efficiently mono‐methylthiolated. This practical method features scalable, metal‐free, mild conditions which are compatible with air and moisture. Several applications of methylthiolated product were demonstrated for the first time. Based on controlled experimental results, a plausible mechanism was proposed with two key intermediates involved in mechanism being detected by HRMS.

Abstract:We developed an approach for direct selective hydroxylation of heterobenzylic methylenes to secondary alcohols avoiding overoxidation to ketones by using a KOBu-t/DMSO/air system. Most reactions could reach completion in several minutes to give hydroxylated products in 41–76% yields. Using DMSO-<i>d</i><sub>6</sub>, this protocol resulted in difunctionalization of heterobenzylic methylenes to afford α-deuterated secondary alcohols (>93% incorporation). Abstract:We report, for the first time, manganese-mediated direct functionalization of the Hantzsch esters with readily accessible alkyl iodides through an aromatization–dearomatization strategy. Applying this protocol, a library of valuable 4-alkyl-1,4-dihydropyridines were facilely afforded in good yields. This simple and practical reaction proceeds under visible-light irradiation at room temperature and displays high functional-group compatibility. Abstract:The textbook alkene halogenation reaction establishes straightforward access to vicinal dihaloalkanes. However, a robust catalytic method for dihalogenizing electron-deficient olefins in an enantioselective manner is still under development, and its mechanism remains controversial. Herein, we disclose efficient regio-, anti-diastereo-, and enantioselective dibromination, bromochlorination, and dichlorination reactions of enones catalyzed by a chiral N,N'-dioxide/Yb(OTf)3 complex. With the combination of electrophilic halogen and halide salts as halogenating agents, an array of homo- and heterodihalogenated derivatives is achieved in moderate to good enantioselectivities. Moreover, DFT calculations reveal that a novel triplet halo-radical pylon intermediate is probable in accounting for the exclusive regio- and anti-diastereoselectivity.

Abstract:The introduction of methyl group into aromatic compounds is a valuable transformation. A large number of known methods use organohalides as the starting materials. However, those methods require pre-synthesized methyl metal reagents or toxic methyl electrophiles. Herein, a palladium-catalyzed reductive coupling reaction between aryl bromides and trimethylsilyl- diazomethane is developed, and the following desilicification process can afford the methylated products. This transformation has broad functional group tolerance and allows methylation of (hetero)aryl halides in moderate to good yields. Thus, it has the potential to be an attractive approach for methylation of organic. In addition, this reductive coupling can also serve as an efficient way for the introduction of silylmethyl group.
