Abstract:We developed an approach for direct selective hydroxylation of heterobenzylic methylenes to secondary alcohols avoiding overoxidation to ketones by using a KOBu-t/DMSO/air system. Most reactions could reach completion in several minutes to give hydroxylated products in 41–76% yields. Using DMSO-<i>d</i>6, this protocol resulted in difunctionalization of heterobenzylic methylenes to afford α-deuterated secondary alcohols (&gt;93% incorporation). By employing this method, active pharmaceutical ingredients carbinoxamine and doxylamine were synthesized in two steps in moderate yields. The Supporting Information is available free of charge at <a class="ext-link" href="/doi/10.1021/acs.orglett.0c03108?goto=supporting-info">https://pubs.acs.org/doi/10.1021/acs.orglett.0c03108</a>.Experimental details and spectroscopic data (<a class="ext-link" href="/doi/suppl/10.1021/acs.orglett.0c03108/suppl_file/ol0c03108_si_001.pdf">PDF</a>)This article has not yet been cited by other publications.

What problem does this paper attempt to address?
The problem this paper attempts to address is the development of an efficient, selective, and transition-metal-free method for the direct hydroxylation of benzylic (heterocyclic) methyl C-H bonds to generate secondary alcohols. Specifically, the researchers aim to avoid over-oxidation to ketones and achieve this transformation using air as the sole oxidant. Additionally, the researchers explored the use of DMSO-d6 as a solvent to achieve site-selective hydroxylation-deuteration reactions for the preparation of α-deuterated secondary alcohols. ### Main Issues: 1. **Direct Selective Hydroxylation**: Develop a method to directly hydroxylate heterocyclic benzylic methyl C-H bonds to secondary alcohols under mild conditions, avoiding over-oxidation to ketones. 2. **High Chemical Selectivity**: Ensure the reaction has high chemical selectivity, allowing for the selective hydroxylation of target sites in complex molecules. 3. **Transition-Metal-Free Catalyst**: Develop a hydroxylation method that does not rely on transition-metal catalysts, improving the simplicity and cost-effectiveness of the reaction. 4. **Synthesis of α-Deuterated Secondary Alcohols**: Achieve site-selective hydroxylation-deuteration reactions via a "one-pot" method to prepare α-deuterated secondary alcohols, which is significant for medicinal chemistry. ### Background: - Direct selective oxidation of benzylic C-H bonds to alcohols (the lowest oxidation state) remains a challenge because most known methods tend to over-oxidize to aldehydes or ketones. - Transition-metal-catalyzed hydroxylation methods, while effective, are costly and complex to operate. - Deuterated compounds have important applications in medicinal chemistry because deuteration can improve the metabolic stability and bioavailability of drugs. ### Solution: - Using a KOBu-t/DMSO/air system, selective hydroxylation of heterocyclic benzylic methyl C-H bonds to secondary alcohols was achieved. - By optimizing reaction conditions, high chemical selectivity and moderate to good yields were achieved. - Using DMSO-d6 as a solvent, site-selective hydroxylation-deuteration reactions were achieved to prepare α-deuterated secondary alcohols, with a deuteration rate exceeding 93%. - This method was successfully applied to synthesize two active pharmaceutical ingredients (APIs): Carbinoxamine and Doxylamine. ### Conclusion: This study successfully developed an efficient, selective, and transition-metal-free method for the direct hydroxylation and hydroxylation-deuteration of heterocyclic benzylic methyl C-H bonds, providing a new tool for medicinal chemistry and organic synthesis.
