Abstract:An efficient and practical approach to trifluoromethylthiolation of α-haloketones/α-haloarylmethanes was developed. The transformation employs only AgSCF3 and KI in situ generated active nucleophilic trifluoromethylthio species and cleanly occurs in up to quantitative yield at room temperature, thereby providing an unprecedentedly easy entry to various α-SCF3-substituted ketones/arylmethanes.

Abstract:The introduction of the (trifluoromethyl) thio moiety into organic molecules has aroused comprehensive interest recently. We describe herein an efficient and chemoselective synthesis of trifluoromethanesulfanylbenzofulvenes through BiCl3- or TsOH-promoted cascade electrophilic cyclization reactions. The presence of the trifluoromethanesulfanyl cation (CF3S+) would facilitate the electrophilic ring closure and nucleophilic attack of chloride or TsOH. This reaction provides a concise method for the synthesis of chlorine-or tosyloxy-substituted trifluoromethanesulfanylbenzofulvenes in good to excellent yields.

Abstract:Triphenylphosphine-mediated metal-free trifluoromethylthiolation and difluoromethylthiolation of thiols by CF3SO2Cl and CHF2SO2Cl to synthesize trifluoromethyl disulfides and difluoromethyl disulfides, respectively, was achieved at room temperature. Iodine generated in situ from iodide facilitated this reaction via the formation of iodotriphenylphosphonium iodide which could serve as a reducing agent in this transformation. Readily available reagents and mild reaction conditions without transition-metals allow this protocol to be more practical than traditional methods.

Abstract:A new method has been developed for the copper(I)-mediated trifluoromethylthiolation of α-bromoketone using commercial anhydrous potassium fluoride, elemental sulfur, and (trifluoromethyl)-trimethylsilane in anhydrous N,N-dimethylformamide. This protocol provides facile access to a variety of α-trifluoromethylthiolated carbonyl compounds in moderate to excellent yields under mild and ligand free conditions.

Abstract:Using (trifluoromethyl)trimethylsilane (TMSCF3 ) as the trifluoromethylating agent, MgCl2-catalyzed trifluoromethylation of carbonyl compounds proceeded readily at room temperature. In the presence of 10 mol% of MgCl2, a variety of carbonyl substrates such as aliphatic/aromatic aldehydes, acyclic/cydic ketones and esters could be trifluoromethylated in DMF, giving the corresponding trimethylsilyl ethers (ketals) in up to 93% isolated yields. Trifluoromethylketones could be readily obtained after hydrolysis of the trimethylsilyl ketals. The reactions could tolerate air and moisture, and the use of oxygen and moisture-free conditions was not required. (C) 2017 Published by Elsevier Ltd.

Abstract:Abstract We report a simple protocol for (ethoxycarbonyl)difluoromethylthiolation of nucleophilic compounds using a difluoroalkyl sulfonium salt which can be prepared in situ via Tf2O-triggered electrophilic activation of a benzyl difluoroalkyl sulfoxide. With the protocol, difluoroalkylthiolated arenes, heteroarenes, α-difluoroalkylthiolated carbonyl compounds, etc. were obtained smoothly with good to excellent yields. Merits of the reaction include the readily available difluoroalkylthiolation reagent and substrates, mild conditions, and excellent regioselectivity.

Abstract:AbstractS‐((Phenylsulfonyl)difluoromethyl)thiophenium salts were designed and prepared by a triflic acid catalyzed intramolecular cyclization of ortho‐ethynyl aryldifluoromethyl sulfanes. The thiophenium salts were found to be efficient as electrophilic difluoromehtylating reagents for introduction of a CF2H group to sp3‐hybridized carbon nucleophiles such as of β‐ketoesters and dicyanoalkylidenes. The (phenylsulfonyl)difluoromethyl group can be readily transformed into CF2H under mild reaction conditions. Enantioselective electrophilic difluoromethylation was also achieved in the presence of bis(cinchona) alkaloids.

Abstract:A metal‐free direct trifluoromethylthiolation of sulfonium ylides with an electrophilic trifluoromethylthiolating reagent has been established, in which sulfonium salt or α‐bromoacetic ester is employed as sulfonium ylide precursors. This trifluoromethylthiolation enables the straightforward construction of SCF3‐substituted sulfonium ylides from a wide range of substrates, including ketones, esters, and even PEGylated substrates. Moreover, the application of this approach in large‐scale preparation and the fluorescence and fluorine‐19 magnetic resonance imaging capabilities of the product are also explored.

Abstract:We report the development of bifunctional trifluoromethylsilyl reagents for selective trifluoromethylation. The newly developed reagent, Me2 (CH2 Cl)SiCF3 , allows highly enantioselective trifluoromethylations of ketones with broad scope. Notably, by taking advantage of the chloromethyl group, a tandem synthesis of chiral trifluoromethylated oxasilacyclopentanes is developed, paving way to α-CF3 tertiary alcohols with vicinal tertiary or quaternary stereocenters. Theoretical studies revealed the important role of nonclassical C-H⋅⋅⋅F-C interactions in stabilizing the transition state, and that the presence of the chlorine atom enhances such interactions for better enantiofacial control.

Abstract:Comprehensive Summary This article described an unprecedented synthesis of trifluoromethylthiophosphonium salts from allyl trifluoromethyl sulfoxide and phosphines in the presence of trimethylsilyl trifluoromethanesulfonate (TMSOTf) through Mislow‐Evans‐type rearrangement. The resulting trifluoromethylthiophosphonium salts were firstly isolated and fully characterized. These fluoroalkylphosphonium salts, as exemplified by trifluoromethylthio triphenylphosphonium salt (6a), exhibited unique and versatile reaction with nucleophiles and deoxygenative trifluoromethylthiolation reaction of carboxylic acids. Furthermore, the photoredox catalytic tunable radical hydrotrifluoromethylation and hydrotrifluoromethythiolation of unactivated alkenes with 6a was successfully developed. Abstract:We described a trifluoromethylation of alkenes using PhICF3Cl as bifunctional reagent. Chlorotrifluoromethylated products were obtained when non-conjugated alkenes were treated with PhICF3Cl in 1,4-dioxane at 60 °C, while vinyl C-H trifluoromethylated products were yielded by further elimination of hydrochloride in the case of those conjugated alkene substrates in DMF. Broad substrate scope, especially including complex alkenes bearing biological active motifs, suggests that this mild and catalyst-free reaction is feasible for late-stage modification in drug discovery.

Abstract:A novel and efficient tandem reaction of sulfonyl chloride and trifluoromethylsulfanylamide is described here for the synthesis of various trifluoromethyl thiolsulphonates with a broad functional group tolerance. In the process, it is believed that sulfinate generated from sulfonyl chloride is a critical intermediate and the additive 4-methylbenzenesulfonic acid (p-TsOH) facilitates the formation of "CF3S"'.
