Abstract:Objective To investigate the effects of continuously intrathecal injection rapamycin on neuro pathic pain behaviors in mice.Methods 48 male adult C57/BL6 mice received intrathecal catheter implantation successfully and without motor dysfunction and serious weight loss,were choosed and randomly divided into shamoperation group ( sham,n =24) and chronic constriction injury model group ( CCI,n =24 ).After operation,each group randomly divided into 3 group again.Group I did nothing,group Ⅱ intrathecally injected rapamycin 1 μg/5μlon day 1 to 6 after operation,group Ⅲ intrathecally injected 20％ DMSO 5μl on the same time ( sham,CCI,sham +R,sham + V,CCI + R,CCI + V,n =8 ).Bilateral mechanical paw withdrawal threshold (WMT) and thermal paw withdrawal latency(TWL) were tested on day 1 before CCI and day 1,3,5,7,10,14,17,21,28 after operation.Results Compared with sham group,both WMT and TWL (7d,MWT:( 1.02 ±0.12)g vs (0.42 ±0.12)g,F=51.01,P＜0.05;TWL:(7.03 ±0.71 )s vs (3.26 ±0.66)s,F=38.27,P＜0.05) were significantly decreased after CCI on the ipsilateral side.When intrathecally injected Rapamycin 1 μg/5 μl on day 1 ～6 after CCI,the mechanical allodynia relieved obviously ( 7 d,M WT:( 0.42 ± 0.18 ) g vs ( 0.86 ± 0.25 ) g,F =6.56,P ＜ 0.05 ),and at least continued to 10 d.On the contrary,the effects of rapamycin on thermal hyperalgesia just showed a trend of inhibition,there was no statistic meaning.In addition,the sham group and contralateral pain behaviors did not change (P＞ 0.05 ).Conclusion Rapamycin can relieve the neuropathic pain behaviors in mice after CCI,mainly the mechanical allodynia,but not thermal hyperalgesia.
Effects of Intrathecal Morphine on Excitatory Amino Acid Contents of Spinal Cord in a Rat Model of Incision Pain

Abstract:Objective To observe the analgesic mechanism in the spinal cord of intrathecal administration of morphine and the effects on contents of excitatory amino acid in L 4～L 5 spinal cord of rat model of incision pain.Methods Male SD rats weighing 250±20g were anesthetized with intraperitoneal pentobarbital sodium 40mg/kg.Intrathecal(IT) catheter was inplanted and the tip of the catheter reached the lumbar region,according to the method of Yaksh.Incision method of Brennan.32 male SD rats were divided randomly into four groups.Group Ⅰ received IT artificial cerebrospinal fluid(ACSF) 20μl and 30min later inhaled 1.4% isoflurane for 5min but no incision was made.Group Ⅱ received IT ACSF 20μl before incision.Group Ⅲ received morphine 10μl(10μg) 30min after incision.Group Ⅳ received IT morphine 10μl(10μg) 30min before incision.In groups Ⅲ and Ⅳ ACSF 10μl was flushed through IT morphine administration.A cumulative pain score was utili{ed to assess pain behavior.High performance liquid chromatography(HPLC) combined with fluorescent detection was used to measure the contents of aspartate,glutamate in L 4～L 5 spinal cord of rat model of incision pain.Results The cumulative pain score of rats in operation group was much higher than those of group Ⅰ(P0.01),Asp and Glu contents of the spinal cord were increased significantly in group Ⅱ as compared with those in group Ⅰ(P0.05).Cumulative pain score in group Ⅲ and Ⅳ(group Ⅲ:3±2,gropu Ⅳ:2.5±2) was significantly lower than that in group Ⅱ(17±3)(P0.01).Asp contents of the spinal cord in group Ⅲ and Ⅳ(group Ⅲ 2.00±0.34μmol/g and group Ⅳ 2.02±0.29μmol/g) were significantly lower than that in group Ⅱ(2.36±0.39μmol/g)(P0.05).Glu contents of the spinal cord in group Ⅲ and Ⅳ(group Ⅲ 3.9±0.59μmol/g,group Ⅳ 4.0±0.56μmol/g) were dramatically lower than that in group Ⅱ(4.8±0.89μmol/g)(P0.05).There were no marked differences of the contents of both Glu and Asp among group Ⅰ,group Ⅲ and group Ⅳ(P0.05).Conclusion In a rat model of antinocieptive effect of IT administration morphine may be involved in decline of excitatory amino acid in spinal cord.
Intrathecal rapamycin attenuates the mechanical hyperalgesia of paclitaxel-induced peripheral neuropathy in mice

Abstract:Paclitaxel is an extensively used chemotherapy antitumor drug and paclitaxel-induced peripheral neuropathy (PIPN) is one of the most common side effect. Rapamycin, originally used as an adjuvant drug for chemotherapy, has recently been found to possess potential neuroprotective activities. Our purposes of this study are to verify the effect of rapamycin on PIPN, which contributes to a new target for PIPN treatment. Mice were given paclitaxel or rapamycin with different injection methods. Paw withdrawal threshold was tested at different time points for mechanical sensitivity assessment. Administration of paclitaxel, both 2 mg/kg and 5 mg/kg, could induce mechanical hypersensitivity. 0.01 mg intrathecal injection of rapamycin showed the best effect on attenuate the mechanical hyperalgesia of PIPN. Intrathecal injection of only rapamycin would not induce the mechanical hyperalgesia while when rapamycin and paclitaxel were used together the mechanical hyperalgesia induced by paclitaxel could be attenuated. Paclitaxel could induce mechanical hyperalgesia in mice and rapamycin could attenuate such mechanical hyperalgesia of PIPN.

Abstract:Mammalian target of rapamycin complex 1 (mTORC1) inhibitors increase the incidence of pain in patients, and this finding has been replicated in animal models. However, reports on possible analgesics for this condition are scant. Accumulating evidence finds that nicotinic acetylcholine receptors (nAChRs) are involved in mediating pain. However, whether nicotine, a full agonist of nAChRs, alleviates mTORC1 inhibition-induced pain and its underlying mechanisms remain unknown. In this study, pain was induced in naive male C57BL/6J mice by intraperitoneally injecting rapamycin acutely or repeatedly. Subsequently, pain thresholds, including mechanical and thermal pain, were measured. The involving signaling pathway was tested using western blot analysis and immunofluorescent assay. Changes in neuronal excitability caused by different treatments were also analyzed using whole-cell recording. Microinjection into the anterior cingulate cortex (ACC) was used to test the role of nAChRs containing the alpha 4 beta 2 or alpha 7 subtype in this brain region in pain modulation. Our results showed that nicotine significantly reduced hyperalgesia in mice that received acute or repeated rapamycin injections, and reversed the effects of rapamycin on the phosphorylation of S6K, 4E-BP1, insulin receptor substrate-1 (IRS-1) at Ser636/639, AKT at Ser473, and ERK at Thr202/Tyr204. Whole-cell recording results showed that nicotine reduced the firing rates of pyramidal neurons in the ACC, and a pharmacological blockade of nAChRs containing the alpha 4 beta 2 or alpha 7 subtype in ACC inhibited the antinociceptive effects of nicotine in mice with rapamycin-induced pain. Our findings indicate that analgesics targeting nAChRs can be developed to help patients with rapamycin-induced pain.
Effect of Intrathecal Sufentanil and Protein Kinase C Inhibitor on Pain Threshold and the Expression of NMDA Receptor/cgrp in Spinal Dorsal Horn in Rats with Neuropathic Pain

Abstract:OBJECTIVE:To investigate the effect of intrathecal sufentanil and protein kinase C inhibitor on pain threshold and the expression of N-methyl-D-aspartate receaptors (NMDAR)/calcitonin generelated peptide (CGRP) in spinal dorsal horn in rats with neuropathic pain.METHODS:Fifty-four healthy male Sprague-Dawley rats were randomly divided into 6 groups (9 in each group). The rats in the sham group(Group S) + spared nerve injury (SNI), SP+SNI, and P+SNI were intrathecally injected sufentanil (1 μg), sufentanil (1 μg) and chelerythrine chloride (11 μg), chelerythrine chloride (11 μg) followed by 10 μL normal saline once every day for 14 days postoperatively, respectively. Similarly, rats in the control group (Group C), the sham group (Group S), and SNI model group (Group SNI) were intrathecally injected 20 μL normal saline in the uniform interval. Pain behaviours were measured on Day 1 pre-surgery and on Day 1, 2, 7, and 14 after the intrathecal injection. The expressions of NMDAR and CGRP in the spinal dorsal horn of L5 segment were determined by immunohistochemistry on Day 2, 7, and 14 after the intrathecal injection.RESULTS:Compared with Group C and Group S, mechanical allodynia threshold in group SNI was decreased after the surgery (P<0.01), and expressions of NMDAR and CGRP immunoreactive soma in the spinal dorsal horn was significantly increased (P<0.01). Mechanical stimulation pain threshold was elevated in Group S+SNI, Group P+SNI, and Group SP+SNI compared with Group SNI (P<0.01), while expressions of NMDAR and CGRP immunoreactive soma in Group S+SNI, Group P +SNI, and Group SP+SNI were significantly decreased (P<0.05 or 0.01).CONCLUSION:Intrathecal administration of sulfentanil and protein kinase C inhibitor can provide significant antinociception in rats with neuropathic pain and obviously inhibit the upregulation of NMDAR and CGRP expressions in the spinal dorsal horn of SNI rat models.
Effect of Intrathecal Administration of a Mixture of Butorphanol and Ketamine on Camp-Pka-creb Signal Transduction Pathway in Spinal Dorsal Horn of Rats with Inflammatory Pain

Abstract:Objective To investigate the effect of intrathecal administration of a mixture of butorphanol and ketamine on cAMP-PKA-CREB signal transductian pathway in the spinal dorsal ham of the rats with inflammatory pain. Methods Twenty-four male SD rats, weighing 240-280 g,in which intrathecal catheters were successfully placed, were divided into 4 groups randomly (n = 6 each): inflammatory pain group (group IP), butorphanol group (group B), ketamine group (group K), and butorphanol + ketamine group (group BK). The inflammatory pain was induced by injection of 5% formalin 50 μl into the plantar surface of left hind paw. Normal saline 10 μl, butorphannl 12.5 μg, ketamine 50 μg, and a mixture of butorphanol 12.5 μg and ketamine 50 μg was injected intrathecally 30 min before subcutaneous injection of formalin in group IP, B, K and BK respectively.Pain intensity score (PIS) was used to assess pain behavior every 5 min within an hour after subcutaneous injection of formalin. The animals were killed at 2 h after subcutaneous injection of formalin, and the L5 segment of the spinal cord was removed for determination of protein kinase A (PKA) and phosphorylated cAMP response element binding protein (p-CREB) expression using immunohistochemistry. Results Fonnahn administration induced pain behaviour expressed as two phases. PIS scores, PKA and p-CBEB expression, and staining scores were significantly lower during the fast and second phases in group BK than in group IP (P 0.05). Conclusion lntrathecal injection of a mixture of butorphanol and ketamine can reduce inflammatory pain in rats, and the mechanism may be related to the cAMP-PKA-CREB signal transduction pathway. Abstract:The aim of the present study was to determine whether the cAMP response element binding protein (CREB) contributes to neuropathic pain during development stage. Adult (7-8 weeks old) male C57BL/6 mice weighing 20-25 g were used. Intrathecal catheter implantation and chronic constriction of the sciatic nerve of the animals were performed. Western blotting and reverse transcription PCR experiments were carried out. Our study demonstrated that the expression of spinal NMDAR after peripheral nerve injury was modulated by central CREB. Chronic constriction nerve injury (CCI) in mice induced thermal hyperalgesia and mechanical allodynia. The increase of NR1 and NR2B subunits of the NMDAR was significantly diminished by intrathecal administration of the CREB antisense oligonucleotide against CREB and pCREB. Additionally, nociceptive behavior induced by CCI was attenuated by intrathecal administration of the CREB antisense oligonucleotide during the period of injection, and the above effects of relieving pain lasted at least 12 days following the last injection. Our results suggested that central functional pCREB may contribute to the development of neuropathic pain and regulate the expression of the NR1 and NR2B subunits of the NMDAR in the process.
Effects of Intrathecal Injection of Ropivacaine on Spinal Cord in Rats Based on Genomics.

Abstract:OBJECTIVES:There are clinical reports of nerve injury caused by ropivacaine. The mechanism for nerve injury induced by ropivacaine has not been fully clarified. This study aims to investigate the changes of pain threshold and L3 spinal cord genomics at 6 h and 24 h after intrathecal injection of 0.5% and 1.0% ropivacaine, and to explore the underlying mechanisms for nerve injury caused by ropivacaine.METHODS:A total of 30 male Sprague Dawley rats weighing 220-260 g were successfully implanted with microspinal catheter. The rats were randomly divided into 5 groups (each n=6): a control group (given saline), a ropivacaine group 1 and a ropivacaine group 2 (both given 1% ropivacaine), a ropivacaine group 3 and a ropivacaine group 4 (both given 0.5% ropivacaine). The rats received continuous intrathecal injection of corresponding drugs at 8.3 μL/h for 24 h via an implanted intrathecal catheter followed by 24 h-pause of injection for the ropivacaine group 2, the ropivacaine group 4 and the control group, 6 h-pause of injection for the ropivacaine group 1 and the ropivacaine group 3. For each group, the observation of behavioral change and the paw withdrawal mechanical threshold (PWMT) was conducted immediately after the injection and again after the pause of injection. After the PWMT observation, the rats were dissected to acquire L3 spinal cords. Illumina sequencing was applied to construct gene libraries. Then the statistical methods were used to find out differentially expressed genes between the groups. Gene Ontology (GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) signaling pathway analysis were conducted for those genes. Real-time RT-PCR was used to determine different expressions of some of those genes.RESULTS:Compared with control group, the PWMT got higher in the ropivacaine group 1-4 and was positively correlated with concentration, negatively correlated with discontinuation duration. Compared with control group, the ropivacaine group 1 had 488 differentially expressed genes, of which 456 were up-regulated and 32 were down-regulated; the ropivacaine group 2 had 1 194 differentially expressed genes, of which 1 092 were up-regulated and 102 were down-regulated; the ropivacaine group 3 had 518 differentially expressed genes, of which 384 were up-regulated and 134 were down-regulated; and the ropivacaine group 4 had 68 differentially expressed genes, of which 46 were up-regulated and 22 were down-regulated. GO enrichment analysis and KEGG signaling pathway analysis showed that most of these differentially expressed genes were related to signaling pathways of inflammatory response.CONCLUSIONS:After intrathecal injection of 0.5% ropivacaine and 1.0% ropivacaine for 24 h, the differentially expressed genes in L3 spinal cord of rats are mainly related to signaling pathways of inflammatory response.
Effect of intrathecal injection of KN93 on pain behaviors in rats with neuropathic pain

Abstract:Objective To investigate the effect of KN93,an inhibitor of Ca2 +/calmodulin dependent protein kinase Ⅱ (CaMK Ⅱ),on the neuropathic pain of the chronic compression of dorsal root ganglion (CCD) model rats.Methods Forty-eight male SD rats were randomly divided into 3 groups:sham group (group S,n=11),CCD model group (group C,n=11),KN93 group (group K,n=26).Group C and group K were operated with the model of neuropathic pain induced by CCD,group S were treated as sham operated rats.Fourteen days after operation,group S and group C received intrathecal injection of 10％ dimethyl sulfoxide (DMSO) 25 μl,while group K received intrathecal injection of 50 μg KN93 dissolved in 25 μl 10％ DMSO.Pain behavior tests including paw withdrawal thermal latency (PWTL) and paw withdrawal mechanical threshold (PWMT) were detected on eight rats randomly selected from each group before(t1) and 4(t2),7(t3),10(t4),14 d(t5) after operation.Group S,group C and group K also received the same tests 2(t6),4(t7),10(t8),12(t9),24 h(10) after administration of DMSO or KN93.In all three groups before administration and in group K after administration at each time point (t6-t9),3 randomly selected rats were sacrificed and each expansion part of spinal cord was obtained.Western blot was used to detect the expression level of CaMK Ⅱ protein.Results From t6 to t9 after administration,group K PWTL[(14.7±1.6),(18.6±1.8),(21.2±2.5),(15.3±2.0) s] and PWMT[(12.0±1.0),(15.4±1.4),(17.5±1.7),(14.9±1.6) g] were increased when compared with group C PWTL[(11.6±1.8),(10.7±1.7),(11.7±2.4),(9.9±1.7) s] and PWMT[(8.4±0.9),(9.6±1.6),(10.6±1.7),(8.9±1.3) g](P＜0.05).Before and 24 h(t10) after administration,group K PWTL and PWMT had no significant difference when compared with group C (P＞0.05).Compared to the basic value (t5) (1.55±0.12) prior to the administration,the expression of CaMK Ⅱ from t6 to t9[(1.37±0.11),(1.15±0.12),(0.75±0.08),(0.86±0.12)] were significantly decreased (P＜0.05).Conclusions Intrathecal administration of KN93 may play an important role in relieving pain of neuropathic pain rats.

Abstract:Objective: To investigate the potential mechanisms and the analgesic effects of intrathecal administration of Ro 25-6981, a selective antagonist of the NR2B subunit of NMDA receptors (NR2B), on neuropathic pain in rats. Methods: (1) L5 spinal nerve ligations (SNL) were performed after intrathecal catheterizations were succeeded in male Sprague-Dawley rats. The rats with neuropathic pain evaluated by von Frey filaments-induced mechanical allodynia were divided into three groups randomly. Ro 25-6981 at the dose of 10 μg (20 μl) or 100 μg (20 μl), or saline solution at the same volume, was administrated to rats in each group by intrathecal injection (i.t.). The 50% paw withdrawal thresholds (PWT) of the rats were tested with von Frey filaments at 30 min, 60 min, 90 min, and 120 min after Ro 25-6981 or saline solution administration. And the locomotor functions were also examined by the slope board test at the same time points. (2) The effects of Ro 25-6981 on the discharges of spinal wide dynamic range (WDR) neurons were examined through in vivo extracellular electrophysiological recording techniques in normal rats. Results: (1) Intrathecal injection of Ro 25-6981 at the dose of 10 μg did not show significant effects on 50% PWT and % MPE in SNL rats as compared with that i.t. the same volume of saline solution (p 0.05, n = 8); while at the dose of 100 μg showed significant increases in 50% PWT and %MPE (P0.05, n =8). Moreover, no obvious impairment on locomotor function was observed after i.t. Ro 25-6981 at both of the above doses. (2) Spinal administration of Ro 25-6981 at the dose of 100 μg had no significant effect on Aβ-fiber evoked discharges of WDR neurons (P0.05, n = 6), but showed significant enhance in C-fiber evoked discharges (P0.05, n = 6). Conclusions: Intrathecal injection of Ro 25-6981 at the dose of 100 μg had significant analgesic effects on rats with neuropathic pain, but didn't affect their locomotor functions; it is likely that such analgesic effects were achieved via antagonizing the spinal NR2B subunit of NMDA receptors, and then inhibiting the C-fiber evoked discharges of spinal WDR neurons.

Abstract:BACKGROUND: The fact that transient receptor potential ankyrin 1 (TRPA1) on the peripheral terminals could attenuate hyperalgesia is widely accepted, but the effect of spinal TRPA1 in the modulation of hyperalgesia has not been fully demonstrated. In the present study, we investigated the effect of intrathecal (i.t.) administration TRPA1 antagonists on chronic pain and expression of TRPA1 and phosphorylation N-methyl-D-aspartate receptor 2B subunit (p-NR2B) in the spinal cord with chronic compression of the dorsal root ganglia (CCD) in rats. METHODS: The study was conducted in 2 parts. Part 1: Sixteen rats were divided into 2 groups (n = 8 each): a sham group and CCD group. Paw withdrawal mechanical thresholds (PWMT) were measured at baseline and 1, 3, 7, 10, 14, and 21 days after CCD. Sixteen other rats were used to evaluate expression of TRPA1 and p-NR2B in spinal cord on the seventh and 14th days after CCD; Western blotting was used to evaluate expression levels (n = 4 each). Part 2: 40 rats were divided into 5 groups (n = 8 each): CCD group, CCD + Vehicle group, CCD + HC-030031(10 mu g, i.t.) group, CCD + HC-030031(25 mu g, i.t.) group, and CCD + HC-030031(50 mu g, i.t.), group. PWMTs were measured at baseline and 0.5, 1, 2, 4, and 6 hours after i.t. HC-030031on the third, seventh, 10th, and 14th days after CCD. Another 48 rats were used to evaluate expression of TRPA1 and p-NR2B in spinal cord 2 hours after injection on the seventh and 14th days after CCD in groups CCD, CCD + Vehicle, and CCD + HC-030031(50 mu g, i.t.) using Western blotting (n = 4 each). RESULTS: Compared with the sham group, PWMT was significantly decreased, and protein expression of TRPA1 and p-NR2B were upregulated, in spinal cord on the seventh and 14th days after CCD operation. TRPA1 antagonists (HC-030031, 50 mu g, i.t.) increased the PWMT after CCD and downregulated the protein level of TRPA1 and p-NR2B in spinal cord at 2 hours after the injection on the seventh and 14th days after CCD. CONCLUSIONS: These data demonstrated that the i.t. administration of TRPA1 antagonists could attenuate neuropathic pain in CCD rats, and this effect could be partially reduced by p-NR2B receptor expression in spinal cord.

Abstract:Aim:To study the analgesic effect of intrathecal APA microencapsulated NIH3T3/rPENK on neuropathic pain in rats.Methods:50 SD rats were randomly divided into five groups:CCI group,sham-operated group,APA group,NIH3T3/rPENK and APA-NIH3T3/rPENK groups.The thermal threshold of the operated hindpaws was examinated before and after transplantation in APA,NIH3T3/rPENK and APA-NIN3TE/rPENK groups.The analgesisc effect of neuropathic pain was observed after infecting of naloxone in rats,The concentrations of enkephalin in cerebrospinal fluid perfusion fluid were measured with radioimmunoassay.Results:The thermal threshold of the operated hindpaws of CCI was significantly lower than that of the control side. The thermal threshold of the rats in NIH3T3/rPENK group and APA-NIH3T3/rPENK group were mainly higher than APA group(P0.05).When compared to APA-NIH3T3/rPENK group,the thermal threshold of the rats in NIH3T3/rPENK group was lower(P0.05).The concentration of the leucine-enkephalin in NIH3T3/rPENK group was lower than that of APA-NIH3T3/rPENK group(P0.05).Conclusion:Intrathecal implantation of NIH3T3/rPENK or APA-NIH3T3/rPENK can significantly reduce the pain-sensitive behavior in neuropathic pain rats,especially APA-NIH3T3/rPENK,and the APA microcapsule can give good effects on immune isolation.

Abstract:Objective To evaluate the effect of intrathecal ropivacaine on spinal histone deacetylase 1 (HDAC1) and HDAC2 expression in the rats with neuropathic pain (NP).Methods Thirty adult male Sprague-Dawley rats, weighing 220-250 g, in which intrathecal catheters were successfully placed, were randomly divided into 3 groups (n=10 each) using a random number table: sham operation group (group S), group NP, and NP + ropivacaine group (group R).NP was induced by chronic constriction injury (CCI) in anesthetized rats.Sciatic nerve was exposed and 4 loose ligatures were placed on the left sciatic nerve at 1 mm intervals with 4-0 chromic catgut.Starting from 7th day after CCI, 0.25％ ropivacaine 20 μl was injected intrathecally in group R, while the equal volume of normal saline was given instead of ropivacaine in S and CCI groups once a day for 7 consecutive days.At 1 day before CCI (T0) ,and 3, 7, 10, 14, 17 and 21 days after CCI (T1-6) , the mechanical paw withdrawal threshold (MWT) and thermal paw withdrawal latency (TWL) were measured.After the pain threshold was measured at T4,3 rats from each group were sacrificed, and their lumbar enlargements were harvested for determination of the expression of HDAC1 and HDAC2 by Western blot.Results Compared with group S, the MWT was significantly decreased, and the TWL was shortened at T2-6, and the expression of HDAC 1 and HDAC2 was up-regulated at T4 in NP and R groups (P＜0.05).Compared with group NP, the MWT was significantly increased at T4, and the TWL was prolonged at T3.4, and the expression of HDAC1 and HDAC2 was downregulated at T4 in group R (P＜0.05).Conclusion The mechanism by which intrathecal ropivacaine alleviates NP may be related to inhibited up-regulation of spinal HDACI and HDAC2 expression in rats.
Effects of Intrathecal Butorphanol on Rat's Pain Threshold and Changes of Neuronal NMDA Receptor Expression in Rat Spinal Dorsal Horn Following Spared Nerve Injury of Sciatic Nerve

Abstract:[Objective] To observe the effect of intrathecal Butorphanol on rat's pain threshold and changes of nervronal NMDA receptor expression in rat spinal dorsal horn following spared nerve injury of sciatic nerve by using immunohistochemistry method. [Methods] Twenty healthy male Sprague-Dawley rats (200~270 g) were randomly devided into four groups (n =5,in each group): control group (group C), normal saline group (group NS), Butorphanol 12 μg group (group B1), Butorphanol 24 μg group (group B2). A microapinal catheter was insterted intrathecally into the lumbar region (about 8 cm) according to the method of Yaksh. After 5 days, group C involved exposure of the sciatic nerve and its branches without any lesion. the other groups were made to the spared nerve injury model (SNI).group C, group NS, groupB1 and group B2 were intrathecally injected 20 μL normal saline or 10 μl Butorphanol (12 μg and 24 μg) followed by 10 μL normal saline after 2 days post-surgery, respectively. 2 hours after intrathecally injection, all rats were killed, and the L5 spinal cord were dissected for determination of the expression of NMDA receptor using immunohistochemical techniques. [Result] The number of immunoreactive soma in the superficial laminae of the L5 spinal cord was significantly incressed significantly in group NS than that in group C (P 0.05). The number of immunoreactive soma in group B1 and B2 increased significantly compared with that in group NS, and there was also difference between group B1 and B2 (P 0.05). The immunoreactive soma quantity density of NMDAR was higher in group NS than those in group C (P0.05), but the quantity density in group B1and B2 were lower than that in the group NS, and between group B1 and B2 the quantity density was also different (P 0.05). The immunoreactive soma optical density of NMDAR was higher in SNI model group than that in control group (P 0.05). [Conclusions] Intrathecal Butorphanol (24 μg) can lessen hyperalgia of neuropathic pain and the dosage was correlation with the effect. The expression of NMDA receptor in spinal cord is up regulated following spared nerve injury of sciatic nerve in rats and intrathecal Butorphanol can restrain this change.
Effect of Intrathecal Injection of Neuropeptide S and Gabapentin on Pain and Anxiety Behavior in Rats with Neuropathic Pain

Abstract:Objective To investigate the effects of intracerebroventricular injection of neuropeptide S(NPS) on pain and anxiety behaviors in rats with neuropathic pain, and explore the possible mechanism. Methods 40 male Sprague-Dawley rats were randomly divided into Sham-Vehicle group, Sham-NPS group, chronic constriction injury(CCI) group, Neuropeptide S low-dose group(NPSl group, 0.1 nmol/L)and Neuropeptide S high-dose group(NPSh group, 1 nmol/L), with 8 rats in each group. Pain-related behaviors, anxiety-related behaviours,and expression of NPS receptor(NPSR) in the left amygdaloid nucleus were measured on the 14 th day via intracerebroventricular injection.Results Compared with the Sham-Vehicle group, the CCI group demonstrated significant pain and anxiety behaviors 14 days after operation(P0.01), the NPSh group significantly relieved(P0.01). And there was no significant difference between the NPSl group and the CCI group(P0.05). Compared with the CCI group, NPSR expression in amygdaloid nucleus increased in the NPSh group. Conclusion NPS can dose-dependently relieve the pain and anxiety behaviors in CCI rats, which may be related with the increase of NPSR in amygdaloid nucleus.

Abstract:Objecfive To investigate the analgesic effect of intrathecal(IT)DREAM.short hairpin RNA (shRNA)in rats with neuropathic pain.Methods Healthy male SD rats weighing 280-320 g were used in this study.Neuropathic pain was induced by chronic constrictive iniury(CCI)to sciatic nerve.A microspinal catheter W&q inserted into subaraehnoid space according to Yak8h method at day 3 after CCI.Twenty.four rats in which IT catheter was successfully implanted were randomly divided into 4 groups(n=6 each):group Ⅰsham operation (S).group Ⅱ neuropathic pain(NP),groupⅢRNA interference(BNAi)and groupⅣblank vector(BV).Lentivims with DREAM-shRNA 5μl was injected IT at day 8 after CCI in group RNAi(Ⅲ).Blank vector 5 μl was injected IT once a day for 7 days,starting from the day 8 after CCI in group BV(Ⅳ).The thermal plan threshold and mechanical pian threshold were measured at day 1 before CCI(To,baseline)and at day 7-14 after CCI(T1-8).The animals were killed at day 15 after CCI.The lumbar segment of the spinal cord(L4-6)was removed for determination of expression of green fluorescent protein(GFP)in the dorsal hom of the spihal cord by immune-fluorescent method.Results The thermal threshold was signillcanfly decreased at T1-4 as compared with the baseline at To in all 4 groups and returned to the baseline levels at T5-8 in group S and RNAi ( group Ⅰ and Ⅱ ) but remained low in group NP (Ⅱ ) and BV (Ⅳ ). The mechanical pian threshold was significantly decreased after CC1/sham operation and was significanty higher at T8 in group BNAi than in other 3 groups. The green fluorescence was observed in group BNAi but not in other 3 groups. Conclusion IT DBEAN-shRNA can ameliorate hyperalgesia in rats with neuropathic pain.

Abstract:Objective To evaluate the effect of intrathecai (IT) sufentanil on NMDAR and CGRP expression in dorsal horn of the spinal cord in a rat model of neuropathic pain. Methods Thirty-six healthy male SD rats weighing 220-280 g were randomly divided into 4 groups (n=9 each): group Ⅰ normal control (group C);group Ⅱ sham operation (group S);group Ⅲ spared sciatic nerve injury (group SNI) and group Ⅳ sufentanil + SNI (group S + SNI). A polyethylene micrespinal catheter was inserted through atlanto-occipital membrane into the subarachnoid space and advanced caudally until the tip reached lumbar segment according to Yaksh. In group Ⅲ and Ⅳ left sciatic nerve and its three terminal branches--tibial, common peroneal and sural nerves were exposed. Tibial and common pereneal nerves were ligated and severed at a point distal to the ligature, while sural nerve was kept intact. In group Ⅱ the sciatic nerves and its branches were exposed but not ligated and severed. In group Ⅳ sufentanil 1 μg in normal saline (NS) 10 μl was injected IT once a day for 14 consecutive days after operation. In group Ⅰ , Ⅱ and Ⅲ NS 10 μl was injected IT instead of sufentanil. Paw withdrawal threshold to von Frey filament stimulation and paw withdrawal latency to noxious thermal stimuli were measured 2 day before (baseline) and 1,2,7, 14 d of IT sufentanil/NS injection. Three animals were killed at 2, 7 and 14 d of IT injection respectively. The lumbar segment of the spinal cord was removed for determination of NMDAR and CGRP expression in the dorsal horn of the spinal cord (L5) using immuno-histochemistry. Results Compared with group C and S (Ⅰ, Ⅱ), mechanical pain threshold was significantly decreased and the expression of NMDAR and CGRP was significantly increased in group SNI (Ⅲ), but there was no significant difference in thermal paw withdrawal latency. Compared with group SNI (Ⅲ), mechanical pain threshold was increased and thermal paw withdrawal latency was prolonged in group S + SNI (Ⅳ) and the expression of NMDAR and CGRP was decreased. Conclusion Intrathecal sufentanil can reduce neuropathic pain by inhibiting the up-regulation of NMDAR and CGRP expression in the dorsal horn of the spinal cord.
Effects of Intrathecal Injection of Spironolactone on a Model of Radicular Pain in Rats

Abstract:Aim To investigate the effects of intrathecal delivery of spironolactone on radicular pain behaviors and the underlying mechanisms.Methods Using rat model of radicular pain induced by chronic compression of dorsal root ganglion(CCD),77 male SD rats successfully received intrathecal catheter implantation and without motor dysfunction were randomly divided into 3 groups:sham-operation group(Sham group,n=27),chronic compression of dorsal root gan-glion group(CCD group,n=27) and spironolactone group(Spir group,n=23).Rats in Spir group were intrathecally treated with spironolactone 3 μg/10 μl twice daily on day 2～4 after CCD,while rats in CCD and Sham group received 10 μl 10% alcohol.Paw withdrawal mechanical threshold(PWMT)and paw withdrawal thermal latency(PWTL)were tested on day 1 before CCD and day 1,4,7,10,14 after CCD(n=12 in CCD and Sham group,n=8 in Spir group).The expression of phosphorylated p65(p-p65)among 3 groups on day 4,7,10 after CCD was revealed by Western blot(n=5 in each group).Results Compared with Sham group,both PWMT(P0.01)and PWTL(P0.01)were significantly decreased after CCD on the ipsilateral side,so were the expression of p-p65(P0.01).No significant difference was found in sham group(P0.05)and on the contralateral side(P0.05).Intrathecal spironolactone significantly reduced mechanical allodynia(11.58±1.38 vs 7.71±0.82 on Day 4,P0.01;10.89±1.46 vs 7.49±1.11 on Day 7,P0.01)and thermal hyperalgesia(14.14±1.52 vs 8.19±1.32 on Day 4,P0.01;13.56±1.63 vs 8.56±1.56 on Day 7,P0.01)in comparison to vehicle-treated CCD rats.Consistently,the expression of p-p65 also significantly decreased(P0.01).Conclusions Intrathecal spironolactone had therapeutic effects on radicular pain induced by chronic compression of dorsal root ganglion in rats.The inhibition of MR-NF-κB pathway was one of the main underlying mechanisms.

Abstract:Background and Objectives: Recent studies have suggested that nuclear factor kappa B (NE-kappa B) may play a role in mediating nerve injury-induced neuropathic pain. Here, we examined the effects of intrathecal pyrrolidine dithiocarbamate (PDTC), a NE-kappa B inhibitor, on the development of neuropathic pain, spinal microglial activation, and CX3CR1 expression induced by sciatic chronic constriction injury (CCI) model in rats.Methods: Under chloral hydrate anesthesia, male Sprague-Dawley rats (300-350 g) fitted with intrathecal catheters underwent either sciatic CCI or sham surgery. Intrathecal saline or PDTC (100 or 1000 pmol/d) was infused I day before or 3 days after CCI (n 8). The rat hind-paw withdrawal threshold to mechanical stimuli and withdrawal latency to radiant heat were determined before surgery and from days I to 7 after CCI. Spinal microglial activation was evaluated with OX-42 immunoreactivity, and spinal CX3CR1 expression was assessed by Western blotting.Results: Chronic constriction injury induced mechanical allodynia and thermal hyperalgesia and microglial activation as demonstrated by OX-42 expression. Whereas it had no apparent effect on spinal cord histology, intrathecal administration of PDTC prevented the development of the mechanical and thermal hyperalgesia and inhibited nerve injury induced microglial activation and spinal CX3CR1 expression.Conclusions: In this study, we have shown the protective effect of intrathecal PDTC on the development of nociceptive behaviors induced by CCI in rats. The activation of NF-kappa B pathway may contribute to spinal microglial activation and CX3CR1 up-regulation. (Reg Anesth Pain Med 2010;35: 231-237)
