Abstract:Cancer pain is one of the most severe chronic pains. The mechanisms underlying cancer pain are still unclear. Because of the pain-relieving effects of Cdk5 (Cyclin-dependent kinase 5) antagonist roscovitine in inflammation pain models, we tested whether roscovitine would induce antihyperalgesia in cancer pain. Our previous study showed that the NR2B (N-methyl-D-aspartate receptor 2B) in the spinal cord participates in bone cancer pain in mice. In this study, we used a mouse model of bone cancer pain to investigate whether roscovitine could attenuate bone cancer pain by regulating the expression level of NR2B mRNA in spinal cord. C3H/HeJ mice were inoculated into the intramedullary space of the right femur with Osteosarcoma cells to induce ongoing bone cancer pain behaviors. At day 14 after operation, inoculation of Osteosarcoma cells significantly enhanced mechanical allodynia and thermal hyperalgesia, which was attenuated by intrathecal administration of different doses of roscovitine. Correlated with the pain behaviors changes, RT-PCR experiments in our study revealed that there was a marked increase in the expression of NR2B mRNA in spinal cord after operation, which was attenuated by intrathecal administration of roscovitine. These results suggest that roscovitine may be a useful adjunct therapy for bone cancer pain, and NR2B in spinal cord may participate in this effect.
Influences of Intrathecal Administration of Roscovitine on Bone Cancer Pain in Mouse Models

Abstract:Objective To investigate the influences of intrathecal administration of roscovitine on bone cancer pain in mouse models. Methods Twenty four C3H/Hej mice were divided randomly into group S(sham + vehicle ), group C(tumor + vehicle) or group R (tumor+ roscovitine) (n = 8 ). Osteosarcoma NCTC 2472 cells were implanted into the intramedullary space of the right femurs of mice to make bone cancer pain models. α-MEM (group Sand group C) or roscovitine (group R) was injected in 14 post-implanted day. Pain behaviors such as paw withdrawal mechanical threshold(PMWT) and paw withdrawal thermal latency (PWTL) were observed at 1,6, 24,48 and 72 post-injective hour, respectively. Results The level of PMWT and PTWL among every group had no difference before operation. At day 7 after inoculation, PMWT of group tumor decreased to (1.08±0.24) g, and at day 10 PTWL shortened to (12.7±1.4) s, and then the pain behaviors of group tumor showed aggravation along with the development of cancer pain. There was significant difference in the pain behavior thresholds between group sham and group tumor (P＜0.05). The level of PM WT [ (0.70±0.19 ) g ] and PTWL [ ( 14.16±1.07 ) s] increased compared with group C and base level at 6 h after intrathecal administration of 20 μg Roscovitine, and kept increasing until reaching maximum level after 12 h, then the value decreased gradually, lowering to the level of group C after 72 h. Conclusion Intrathecal administration of roscovitine can relief bone cancer pain in mouse models.
Intrathecal Injection of Metabotropic Glutamate Receptor Subtype 3 and 5 Agonist/Antagonist Attenuates Bone Cancer Pain by Inhibition of Spinal Astrocyte Activation in a Mouse Model

Abstract:Background Astrocytes and metabotropic glutamate receptors play important roles in nociceptive processing. However, their roles in bone cancer pain were not well understood. This study sought to investigate whether selective mGluR3 and mGluR5 agonist or antagonist develop antinociceptive effects on bone cancer pain by inhibition of spinal astrocyte activation. Methods C3H/HeNCrlVr mice were inoculated into the intramedullary space of the femur with sarcoma NCTC 2472 cells to induce bone cancer pain. Quantitative real-time reverse transcription-polymerase chain reaction and Western blot experiments examined messenger RNA and protein expression of spinal glial fibrillary acidic protein, mGluR3, and mGluR5. The authors further investigated effects of intrathecal treatment with the mGluR3 agonist (APDC), the mGluR3 antagonist (LY341495), the mGluR5 agonist (CHPG), or the mGluR5 antagonist (MTEP) on nociceptive behaviors and spinal astrocyte activation associated with bone cancer pain. Results Inoculation of sarcoma cells, but not α-MEM solution, induced progressive bone cancer pain and resulted in up-regulation of glial fibrillary acidic protein, mGluR3, and mGluR5 expression on days 10, 14, and 21 postinoculation. Intrathecal administration of APDC and MTEP attenuated bone cancer-evoked spontaneous pain, mechanical allodynia, thermal hyperalgesia, and reduced spinal glial fibrillary acidic protein expression. However, treatment with LY341495 and CHPG induced thermal hyperalgesia and spinal glial fibrillary acidic protein expression in control mice. Conclusions Spinal mGluR3 activation or mGluR5 inhibition reduced bone cancer pain. Inhibition of spinal astrocyte activation may contribute to the analgesic effects. These findings may lead to novel strategies for the treatment of bone cancer pain.

Intrathecal Injection of the Peptide Myr-NR2B9c Attenuates Bone Cancer Pain Via Perturbing N-Methyl-D-Aspartate Receptor-PSD-95 Protein Interactions in Mice

Abstract:BACKGROUND: N-methyl-D-aspartate receptor (NMDARs)-dependent central sensitization plays an important role in cancer pain. Binding of NMDAR subunit 2B (NR2B) by postsynaptic density protein-95 (PSD-95) can couple NMDAR activity to intracellular enzymes, such as neuronal nitric oxide synthase (nNOS), facilitate downstream signaling pathways, and modulate NMDAR stability, contributing to synaptic plasticity. In this study, we investigated whether perturbing the specific interaction between spinal NR2B-containing NMDAR and PSD-95, using a peptide-mimetic strategy, could attenuate bone cancer-related pain behaviors.METHODS: Osteosarcoma cells were implanted into the intramedullary space of the right femurs of C3H/HeJ mice to induce progressive bone cancer-related pain behaviors. Western blotting was applied to examine the expression of spinal phospho-Tyr1472 NR2B, nNOS, and PSD-95. We further investigated the effects of intrathecal injection of the mimetic peptide Myr-NR2B9c, which competitively disrupts the interaction between PSD-95 and NR2B, on nociceptive behaviors and on the upregulation of phospho-Tyr1472 NR2B, nNOS, and PSD-95 associated with bone cancer pain in the spinal cord.RESULTS: Inoculation of osteosarcoma cells induced progressive bone cancer pain and resulted in a significant upregulation of phospho-Tyr1472 NR2B, nNOS, and PSD-95. Intrathecal administration of Myr-NR2B9c attenuated bone cancer-evoked mechanical allodynia, thermal hyperalgesia, and reduced spinal phospho-Tyr1472 NR2B, nNOS, and PSD-95 expression.CONCLUSIONS: Intrathecal administration of Myr-NR2B9c reduced bone cancer pain. Internalization of spinal NR2B and dissociation NR2B-containing NMDARs activation from downstream nNOS signaling may contribute to the analgesic effects of Myr-NR2B9c. This approach may circumvent the negative consequences associated with blocking NMDARs, and may be a novel strategy for the treatment of bone cancer pain.

Abstract:Although bone cancer pain is a common intractable clinical symptom, its underlying mechanisms are still elusive. Accumulating evidence reveals that the N-methyl-d-aspartate (NMDA) receptor containing a 2B subunit (NR2B) in the spinal cord contributes to bone cancer pain. Our preliminary study demonstrated that intrathecal injection of fusion peptide Myr-RC-13 could disrupt spinal KIF17/mLin10 interaction, which is an essential component of KIF17-mediated NR2B transport. Here we report a means by infusion of the selected peptide Myr-RC-13 intrathecally to attenuate bone cancer pain. The results showed that inoculation of fibrosarcoma NCTC 2472 cells into the femur cavity of C3H/HeJ mice induced progressive bone cancer pain and resulted in up-regulation of KIF17 and NR2B in the spinal cord. In addition, repetitive spinal delivery of Myr-RC-13 relieved bone cancer-related mechanical allodynia and spontaneous pain behaviors, and down-regulated expression of spinal KIF17 and NR2B. Finally, our results demonstrated that selected peptide Myr-RC-13 was able to attenuate bone cancer pain via decreasing spinal KIF17 and NR2B expressions. Therefore, selected peptide Myr-RC-13 might be a potential analgesic strategy for bone cancer pain.
Intrathecal Administration of Roscovitine Prevents Remifentanil-Induced Postoperative Hyperalgesia and Decreases the Phosphorylation of N-methyl-D-aspartate Receptor and Metabotropic Glutamate Receptor 5 in Spinal Cord.

Abstract:N-methyl-D-aspartate receptor (NMDAR) and metabotropic glutamate receptor 5 (mGluR5) play an important role in nociceptive processing and central sensitization. Our previous study showed that tyrosine phosphorylation of NMDAR subunit 2B (NR2B) at Tyr1472 in spinal dorsal horn contributes to the postoperative hyperalgesia induced by remifentanil. Cyclin-dependent kinase 5 (Cdk5) has been implicated in synaptic plasticity, learning, memory and pain signaling via regulating the phosphorylation of NMDAR and mGluR5. In the present study, a rat model of postoperative pain was used to investigate the role of Cdk5 in spinal dorsal horn in remifentanil-induced hyperalgesia and the intervention of pretreatment with Cdk5 inhibitor roscovitine. Intraoperative infusion of remifentanil (0.04 mg/kg, subcutaneous) significantly enhanced mechanical allodynia and thermal hyperalgesia induced by plantar incision during the postoperative period (each lasting between 2 h and 48 h), which were attenuated by pretreatment with roscovitine. Correlated with the pain behavior changes, Western blotting revealed that there was a significant increase in the expression of Cdk5 and its activator p35/p25, and further the kinase activity of Cdk5 in spinal dorsal horn after intraoperative infusion of remifentanil. The phosphorylation of NR2A at Ser1232, the phosphorylation of NR2B at Tyr1472 and the phosphorylation of mGluR5 at Ser1167 were also significantly up-regulated. Furthermore, these increases were attenuated by pretreatment with roscovitine. These results suggested that Cdk5 may contribute to remifentanil-induced postoperative hyperalgesia via regulating the phosphorylation of NMDAR and mGluR5 in spinal dorsal horn. These findings provide experimental evidence for the further application of Cdk5 inhibitor in preventing remifentanil-induced hyperalgesia.
Effect of Intrathecal Injection of Ifenprodil on the Expression of 2B Subunit Mrna of NMDA Receptor in Spinal Cord in a Mouse Model of Bone Cancer Pain

Abstract:Objective To investigate the effect of intrathecal injection of ifenprodil on the expression of 2B subunit mRNA of NMDA receptor (NR2B) in spinal cord in a mouse model of bone cancer pain. Methods One hundred and forty male C3H/HeJ mice weighing 20-25 g were randomly divided into 5 groups (n=28 each):group S sham operation; group B bone cancer pain; group I1- ifenprodil 2.5, 5 and 10 μg. Bone cancer pain was performed by intra-right-femur inoculations of osteolytic NCTC 2472 cells in group I1- and B. On the 14 d after inoculations, group I1-3 received intrathecal injection of ifenprodil 2.5, 5 and 10 μg dissolved in 5 μl solvent respectively, while group B and S received intrathecal injection of solvent. Seven mice was selected randomly from each group at 1 d before inoculations, at 1 h before, intrathecal injection of ifenprodil or solvent and at 2, 12 and 24 h after the injection (T1-5) tor determination of mechanical and thermal pain threshold. The mice were decapitated at T2.5 after determination of pain threshold and the L3-5 part of spinal cord was removed for determination of the expression of NR2B mRNA by RT-PCR. Results The mechanical and thermal pain threshold were significantly lower in group B and I1-3(P<0.05), except for the thermal pain threshold at T3 in group I3(P>0.05), and the expression of NR2B mRNA was significantly higher in group B and I3 while lower in group I2.3 than in group S (P<0.05). The mechanical and thermal pain threshold were significantly higher and the expression of NR2B mRNA was lower in group I2.3 than in group B (P<0.05), but there were no significant differences in the indexes mentioned above between group I1 and B (P>0.05). The mechanical and thermal pain threshold were significantly higher and the expression of NR2B mRNA was lower in group I3 than in group I2 (P <0.05). Conclusion lntratheeal injection of ifenprodil can attenuate bone cancer pain through blocking NR2B and inhibit hyperalgesia through down-regulating the NR2B mRNA expression in spinal cord.

Abstract:Objective To investigate the analgesic efficacy of intrathecal injection of RC-13,a competitive kinesin superfamily protein 17 antagonist,in a mouse model of bone cancer pain.Methods Forty male C3H/HeJ mice,aged 6-8 weeks,weighing 20-25 g,were randomly divided into 5 groups (n=8 each): sham operation group (group S); bone cancer pain + 5 μl dimethyl sulfoxide (DMSO) group (group R0); bone cancer pain + 2.5 μg RC-13 group (group R1); bone cancer pain + 5 μg RC-13 group (group R2) and bone cancer pain + 10 μg RC-13 group (group R3).In groups R0-3,bone cancer pain was induced by implantation of α-min-imal essence medium (α-MEM) containing osteosarcomaNCTC 2472 cells into the intramedullary space of right femur.In group S,culture medium α-MEM containing no cancer cell was injected instead.10％ DMSO 5 μl and RC-13 2.5 μg/5 μl,5μg/5μ1 and 10 μg/5 μ1 dissolved in 10％ DMSO were injected intrathecally in groups R0-3,respectively,once a day for 3 consecutive days starting from 14th day after inoculation of the tumor cells.Pain behavior was assessed by the paw withdrawal mechanical threshold (PWMT) and spontaneous lifting times (SLTs) measured at 1 day before inoculation and at 3,5,7,10,14 days after inoculation.The same tests were also performed at 1,3,5 and 7 days after administration in groups R0-3.Results Compared with group S,PWMT was significantly decreased and SLTs were increased at 7-14 days after inoculation in the other groups (P ＜ 0.05).Compared with group R0,PWMT was significantly increased and SLTs were reduced at 1 day after administration in group R1,at 1and 3 days after administration in group R2,and at 1,3 and 5 days after administration in group R3 (P ＜ 0.05).Compared with group R1,PWMT was significantly increased and SLTs were reduced at 3 days after administration in group R2,and at 1,3 and 5 days after administration in group R3 (P ＜ 0.05).Compared with group R2,PWMT was significantly increased and SLTs were reduced at 1 and 3 days after administration in group Rs (P ＜ 0.05).Conclusion Intrathecal RC-13,a competitive kinesin superfamily protein 17 antagonist,has a good analgesic efficacy in a mouse model of bone cancer pain and the efficacy is dose-dependent.
Effects of Repeated Intrathecally KIF17 Antisense Oligodeoxynucleotide on the Expression of Mlin10 and NR2B in Spinal Cord in a Mouse Model of Bone Cancer Pain

Abstract:Objective To investigate the effects of repeated intrathecally kinesin superfamily protein 17 (KIF17) antisense oligodeoxynucleotide (ODN) on the expression of mLin10 and NR2B in spinal cord in a mouse model of bone cancer pain.Methods Fifty-six male C3H/HeJ mice,aged 4 ～ 6 weeks,weighting 20 ～ 25 g,were randomly divided into two groups:sham operation group (group S,n=20) and bone cancer pain group (group T,n=36).20μl α-minimal essence medium (α-MEM) which containing 2× 105 NCTC2472 osteosarcoma cells was injected into the intramedullary space of the right femur in group T.In group S,no cancer cell was instead.The number of spontaneous flinches (NSF) and the paw withdrawal mechanical threshold (PWMT) were measured at the day before (base) and the days 4,7,10 and 14 after inoculation.According to the corresponding time points,twenty-four mice were sacrificed for determination the expression of KIF17,mLin10 and NR2B using Western blot.Then,the mice of group T were randomly divided into three groups (n=8,T1,T2,T3,group).In group S and group T1,Saline 5 μl was injected intrathecally.KIF17 sense ODN and antisense ODN,5 μg/5μl were respectively injected in group T2 and T3 for 6 consecutive days.Pain behaviors were assessed at the days 2-6 after the first injection.And determinated the KIF17,mLin10 and NR2B expression,again.Results Compared with group S,the NSF was increased and the PWMT was decreased at the days 7,10 and 14 after inoculation in group T (P＜0.05).Compared with the base ((0.65±0.15),(1.06±0.06),(1.01±0.14)),the expression of KIF17,mLin10 and NR2B (14d:(1.13 ±0.06),(2.17 ± 0.37),(1.85 ± 0.32)) were increased at the days 7,10 and 14 after inoculation in group T(P＜0.05).During the course of the injection,compared with group T1 and T2,the NSF was decreased and the PWMT was increased significantly in the group T3(P＜0.05),the expression of KIF17,mLin10 and NR2B((0.88±0.08),(0.96±0.11),(1.03±0.08)) were reduced in group T3 (P＜0.05).Conclusion Intrathecal KIF 17 antisense ODN in the mice of bone cancer pain improves the pain behaviors,and inhibits the up-regulated of KIF17,mLin10 and NR2B during the course of the injection.
The Role of N‐methyl‐d‐aspartate Receptor Subunit NR2B in Spinal Cord in Cancer Pain

Abstract:Cancer pain is one kind of the most common and severe kinds of chronic pain. No breakthrough regarding the mechanisms and therapeutics of cancer pains has yet been achieved. Based on the well established involvement of the NMDA (N-methyl-D-aspartate) receptor containing NR2B in inflammatory pain and neuropathic pain and the effective pain relief obtained with ketamine in cancer patients with intractable pain, we supposed that NR2B in the spinal cord was an important factor for cancer pain. In this study, we investigated the possible role of NR2B in the spinal cord using a murine model of bone cancer pain. C3H/HeJ mice were inoculated into the intramedullary space of the right femur with Osteosarcoma NCTC 2472 cells to induce ongoing bone cancer-related pain behaviors. At day 14 after operation, the expression of NR2B mRNA and NR2B protein in the spinal cord were higher in tumor-bearing mice compared to the sham mice. Intrathecal administration of 5 and 10 mu g of NR2B subunit-specific NMDA receptor antagonist ifenprodil attenuated cancer-evoked spontaneous pain, thermal hyperalgesia and mechanical allodynia. These results suggest that NR2B in the spinal cord may participate in bone cancer pain in mice, and ifenprodil may be a useful alternative or adjunct therapy for bone cancer pain. Abstract:Objective To investigate the effect of ifenprodil in the mice of bone cancer pain.Methods 96 male C3H/HeJ mice were divided randomly into tumor group( Group T),control group( Group C) and sham group( Group S).The α-minimal essence media(ct-MEM) with 2 × l05 osteosarcoma NCTC 2472 cells were implanted into the intramedullary space of the right femurs of mice to induce ongoing bone cancer related pain behaviors.The sham group was inoculated by α-MEM without any cells.On the 14th d after inoculation,pain ethology indexes such as the spontaneous lifting behaviors,the paw withdrawal mechanical threshold(PWMT) and the paw withdrawal thermal latency (PWTL)were observed on 1 d before inoculation and on 3 d,5 d,7 d,10 d,14 d,17 d,19 d,23 d after inoculation.Lumbar intumescentia of mice in each group were taken out to investigate the expression level of NR2B western blot after pain behaviors tests at the same time point after intrathecal injection.Results ( 1 ) At day14 after the operation,the obvious increasing of spontaneous lifting behaviors ( ( 12.88 ±1.64) ) and the expression of NR2B (2.12 ±0.13),the significant decreasing of PWMT( (0.39 ±0.17)g) and PWTL( ( 11.59 ± 1.67 ) s ) were observed in group T compared with group S and preoperative base level (P ＜ 0.05 ).(2) At day 17,day 19 and day 23 after the operation,compared with the basal level of dayl4 before administration and group C,the spontaneous lifting behaviors ( (5.13 ± 1.38),(4.70 ± 1.58),(5.64 ± 1.17) ) of group T were obviously decreased,PWMT ( ( 1.10 ± 0.65 ) g,( 0.95 ± 0.56 ) g,( 1.05 ± 0.26 ) g) and PWTL ( ( 15.17 ± 1.27) s,( 15.93 ± 2.18 ) s,( 16.28 ± 1.48 ) s ) were increased,the expression of NR2B ( ( 1.42 ± 0.17),(1.67 ±0.53),(1.14 ±0.79) ) were significantly decreased(P＜0.05).Conclusion Repeated intratheal injection of ifenprodil can efficiently relieve spontaneous lifting behaviors,mechanical hyperalgia and thermal hyperalgia and decrease the expression of lumbar intumescentia NR2B in the mouse model of bone cancer pain.

Abstract:The aim of the present study was to determine whether the cAMP response element binding protein (CREB) contributes to neuropathic pain during development stage. Adult (7-8 weeks old) male C57BL/6 mice weighing 20-25 g were used. Intrathecal catheter implantation and chronic constriction of the sciatic nerve of the animals were performed. Western blotting and reverse transcription PCR experiments were carried out. Our study demonstrated that the expression of spinal NMDAR after peripheral nerve injury was modulated by central CREB. Chronic constriction nerve injury (CCI) in mice induced thermal hyperalgesia and mechanical allodynia. The increase of NR1 and NR2B subunits of the NMDAR was significantly diminished by intrathecal administration of the CREB antisense oligonucleotide against CREB and pCREB. Additionally, nociceptive behavior induced by CCI was attenuated by intrathecal administration of the CREB antisense oligonucleotide during the period of injection, and the above effects of relieving pain lasted at least 12 days following the last injection. Our results suggested that central functional pCREB may contribute to the development of neuropathic pain and regulate the expression of the NR1 and NR2B subunits of the NMDAR in the process.

Abstract:Objective To investigate effects of metabotropic glutamate receptor subtype 5 (mGluR5) antagonist MTEP on the nociceptive behavior and the expression of glial fibrillary acidic protein (GFAP) in spinal cord associated with bone cancer pain. Methods C3H/HeNCrlVr 60 male mice were randomly divided into 5 groups: ( 1 ) normal control group: the mice were given food and water ad libitum; ( 2 ) MTEP + Tumor group: the mice were treated by intrathecal gdministration ( once daily on the days 14 ～20 after inoculation of tumor cells)with MTEP (150 nmol); (3) physiological saline + Tumor group:the tumor mice were treated with the same volume of physiological saline; (4) MTEP + Sham group: the sham mice were treated with the same dose of MTEP;(5) physiological saline + Sham group: the sham mice were treated with the same volume of physiological saline.the mice pain behaviors were assessed with the paw withdrawal thermal latency (PWTL) at the corresponding time points, then the mice were killed and the samples of spinal cord were used to real-time PCR and western blot detection of GFAP mRNA and protein expression. Results The basic values of PWTL had no significant differences among all groups (P＜0.05). At day 14 after operation,no significant difference was found in the PWTL value between normal control group and the sham operation group. But in tumor group, the PWTL value was significantly lower than in the normal control group (P＜ 0.05 ). At day 21 after operation,the PWTL and the level of GFAP expression in the spinal cord had no significant differences among normal control group, MTEP + Sham group and physiological saline + Sham group (P ＞ 0.05 ); the PWTL ( (6. 18 ± 1.29 ) s) in physiological saline + Tumor group was significantly lower than in normal control group ( ( 15.91 ± 1.65 )s), physiological saline + Sham group ( ( 16.57 ± 1.86) s) and MTEP + Sham group ( ( 17.05 ± 2.43 ) s) (P ＜ 0.05 ), but the level of GFAP expression was higher than in the above three groups. In MTEP +Tumor group ,the PWTL (9.39 ± 1.94s) was higher than in physiological saline + Tumor group, and the level of GFAP expression was lower than in physiological saline +Tumor group (P ＜ 0.05 ). Conclusion Inhibiting spinal activation of astrocytes may be one of the MTEP anticancer pain mechanisms.

Abstract:Objective To study the effects of intrathecal injeetion of cyclin-dependent kinase 5 inhibitor Roseovitine on the pain behavior in chronic compression of dosal root ganglia(CCD)rats.Methods 24 male SD rats with intrathecal catheter properly inserted,were randomly divided into four groups(n=6):sham operation group(S group),50%DMSO was used as the vehicle control for the intrathecal injection;CCD group treated with 50%DMSO(operated for chronic compression of right dorsal root ganglion,C group);Roscovitine group(Roscovitine 50μg,R group)and CCD0 group was not injected any drugs(C0 group),just as that in control.To evaluate the behaviorsl changes,paw withdrawal mechanical threshold(PWMT)and paw withdrawal thermal latency (PWTL)were measured 0,7 days and 1,4,8 h after injection on the 7th day of CCD.Results Compared with the value-based,with the exception of S group,all of the rats after CCD PWMT and PWTL were significantly lower(P＜0.05).PWMT of R group was no significant change at all times point.Compared to value-based PWTL of R group was a marked increase.and remained at a relatively high level(P＜0.05).Conclusion Cdk5 involves in spinal dorsal horn of neuropathic pain signal transduction process.
Effect of Intrathecal Fluorocitrate on the Mechanical and Heat Hyperalgesia in Mice Bearing Bone Cancer

Abstract:Objective By investigating the effect of intrathecal fluorocitrate (FC) on the mechanical and heat hyperalgesia on bone cancer mice, to evaluate the role of astrocytes in nociceptive transmission at the spinal level. Methods Fourty-eight C3H/HeJ mice weighed 20-25 g were randomly divided into 6 groups (n=8 each): sham operation, bone cancer + NS, bone cancer + FC 0.25 nmol/μl, bone cancer + FC 0.50 nmol/μl, and bone cancer + FC 0.75 nmol/μl groups. The mechanical withdrawal threashold (MWT) and thermal withdrawfal latency (TWL) were measured before operation and 7, 12, 17 and 21 days after operation. NS and FC were intrathecally administered 17 days after the operation, and the nociceptive thresholds were tested before drug administration and 15 min, 30 min, 1 h and 2 h afterwards. Results In the bone cancer groups, steady mechanical and heat hyperalgesia were noticed from the 12th day after operation, and FC increased the MWT and TWL in a dose-dependent pattern. Two hours after the drug, the nociceptive thresholds returned to the original resting level. The effect of FC 0.75 nmol/μl was the strongest at 30 min and lasted for 2 h. Conclusion Intrathecal FC can reduce the mechanical and heat hyperalgesia in mice with bone cancer pain, and the astrocytes may play a role in the nociceptive transmission at the spinal level.
Intrathecal Injection of NR2B Antagonist Ifenprodil Alleviate the Bone Cancer Pain in Mouse

Abstract:Objective To investigate the analgesic effect of intrathecal injection of NR2B antagonist ifenprodil in mouse of bone caner pain. Methods The mouse model of bone cancer pain was developed by intra-femur inoculations of α-minimal essence media (α-MEM) with osteolytic NCTC 2472 cells in C3H/HeJ mice(group T). The sham group (group S,n =10) were inoculated by α-MEM without any cells. Pain ethology indexes such as the spontaneous lifting behaviors, the paw withdrawal mechanical threshold(PWMT) and the paw withdrawal thermal latency (PWTL) were observed on 1 d before inoculation and on 3 d,5 d,7 d,10 d,14 d after inoculation to evaluate the mouse model of bone cancer. On the 14th d after inoculation, the succeed mouse model of bone cancer pain was divided randomly into 2.5 μg, 5 μg, 10 μg ifenprodil group(group I1, I2, I3,n =10) and solventia group (20%DMSO group, group C,n =10). Group I1～I3 were treated by intrathecal injection of corresponding dose of ifenprodil. Group C and group S were treated by intrathecal injection of solventia. All the volume of physic liquor of intrathecal injection was 5 μl. Pain ethology indexes mentioned above was observed at 2 h, 12 h, 24 h after administration. Results (1) The significant increasing of spontaneous lifting behaviors(13.16±1.78), the obvious decreasing of PWMT (0.47±0.19)g and PWTL (10.01±1.42)s were observed on 14th d after inoculation in group T compared with group S and preoperative base level( P ＜0.05). (2) The spontaneous lifting behaviors [I2:(9.00±1.80),(10.90±1.17);I3:(6.80± 2.51),(8.80±1.64)] was decreased, the PWMT [I2:(1.12±0.27)g,(0.82±0.30)g;I3:(1.44±0.34)g,(1.16±0.37)g] and PWTL [I2:(13.86±1.51)s,(12.12±1.27)s;I3:(15.95±1.70)s,(13.74±1.98)s] was increased in group I2 and I3 compared with the basal level on 14th d before administration and group C at 2 h and 12 h after administration ( P ＜0.05) The analgesic effect of group I3 was more conspicuous compared with group I2( P ＜0.05). No obvious change of pain ethology indexes was observed in group I1 ( P ＞0.05). Conclusion Intrathecal injection of ifenprodil can efficiently relieve mechanical hyperalgia and thermal hyperalgia in the mouse model of bone cancer pain.
Effect of Intrathecal Injection of Bovineadrenalmedulla 8-22 on Pain Behavior in a Mouse Model of Bone Cancer Pain

Abstract:Objective To investigate the effect of intrathecal injection bovineadrenalmedulla 8-22 (BAM8-22) on pain behavior in a mouse model of bone cancer pain.Methods 50 male 8-10 week old C3H/HeJ mice weighing 18-22 g were divided randomly into 5 groups (n =8):sham group (S group),BCP group:artificial cerebrospinal fluid treat group (B0 group) and BAM8-22 treat groups (B1-B3 group).Group C and B were induced mouse models of bone cancer pain by intra-right-femur inoculations of osteolytic NCTC2472 cells while group S were injected only α-MEM.On the 14 d after inoculations,group S and BCP received intrathecal injection of artificial cerebrospinal fluid 5 μl,while group B1-B3 received intrathecal injection of BAM8-22 0.8,2.4,8.0 nmol,which dissolved in 5 μl artificial cerebrospinal fluid.They were intrathecally injected,respectively,once a day for 7 consecutive days starting from day 14 after inoculation.Mice received pain behavior tests including quantification of spontaneous flinches (NSF),paw withdrawal mechanical threshold (PWMT) 0.5 h before and at 2 h,after administration.Results Compared with S group [(1.80±0.20) times,(1.87 ±0.22) g],NSF [(14.67±2.65) times] was significantly increased (P =0.000) and MWT [(0.42 ± 0.22) g] was significantly decreased (P =0.002) on 7-21 days after inoculation in BCP group.Compared with B0 group [(12.38 ±1.12) times,(0.48 ± 0.22) g] and the basal level on 14th day before administration [(12.65 ±1.10) times,(0.46 ±0.19) g,P=0.012,0.011,0.001,0.001],PWMT was significantly decreased,and NSF was increased at 2 h after administration in B2 group [(7.28 ±0.82) times,(1.01 ±0.13) g] and B3 groups [(4.18 ±1.02) times,(1.38 ±0.12) g,P=0.027,0.025,0.013,0.009].The analgesic effect of group B3 was more conspicuous compared with group B2 (P =0.007,0.031).No obvious change of pain ethology indexes wag observed in group B1 [(12.03 ± 1.22) times,(0.51 ± 0.16) g] (P =0.096,0.074,0.527,0.474).Conclusion Intrathecal BAM8-22 injection can effectively attenuated bone cancer pain with increased doses in the mouse model of bone cancer pain.

Abstract:Objective: To investigate the expression of NR2B mRNA in the anterior cingulate cortex(ACC) and L4-6spinal cord in the bone cancer pain mice.Methods: 75 adult male C57BL/6 mice were randomly and assigned into three groups(n = 5): bone cancer pain group(group C);sham-operated group(group O);normal control group(group N).On postoperative 6 d,10 d,14 d,18 d and 22 d after operation,the mechanical allodynia and the expression of NR2B mRNA in the L4-6 spinal cord and ACC were evaluated with the mechanical pain threshold and fluorescence quantitative RT-PCR respectively.Results: Compared with group O and group N,mechanical pain threshold in group C was decreased at 10 d,14 d,18 d and 22 d after operation(p < 0.05).Compared with group O and group N,the expression of NR2B mRNA in ACC was increased at 14 d and 18 d after operation(p < 0.05).Compared with group O and group N,the expression of NR2B mRNA in L4 ~ 6 spinal cord was increased at 10 d,14 d,18 d and 22 d after operation(p < 0.05).Conclusion: The expressions of NR2B mRNA in ACC and L4-6 spinal cord are increased in bone cancer pain mice.These suggest that the generation and maintenance of bone cancer pain is related to both the spinal cord and the ACC.

Abstract:Background: Neuropathic pain induced by spinal or peripheral nerve injury is very resistant to common pain killers, nerve block, and other pain management approaches. Recently, several studies using stem cells suggested a new way to control the neuropatic pain. In this study, we used the spinal nerve L5 ligation (SNL) model to investigate whether intrathecal rat mesenchymal stem cells (rMSCs) were able to decrease pain behavior, as well as the relationship between rMSCs and reactive oxygen species (ROS).Methods: Neuropathic pain of the left hind paw was induced by unilateral SNL in Sprague-Dawley rats (n = 10 in each group). Mechanical sensitivity was assessed using Von Frey filaments at 3, 7, 10, 12, 14, 17, and 24 days post-ligation. rMSCs (10 mu l, 1 x 10(5)) or phosphate buffer saline (PBS, 10 mu l) was injected intrathecally at 7 days post-ligation. Dihydroethidium (DHE), an oxidative fluorescent dye, was used to detect ROS at 24 days post-ligation.Results: Tight ligation of the L5 spinal nerve induced allodynia in the left hind paw after 3 days post-ligation. ROS expression was increased significantly (P < 0.05) in spinal dorsal horn of L5. Intrathecal rMSCs significantly (P < 0.01) alleviated the allodynia at 10 days after intrathecal injection (17 days post-ligation). Intrathecal rMSCs administration significantly (P < 0.05) reduced ROS expression in the spinal dorsal horn.Conclusions: These results suggest that rMSCs may modulate neuropathic pain generation through ROS expression after spinal nerve ligation.
