Abstract:The first tertiary amine catalyzed enantioselective Morita-Baylis-Hillman (MBH) reaction of isatins with acrylates has been demonstrated, allowing asymmetric synthesis of biologically significant 3-substituted-3-hydroxy-2-oxindoles in good yields and with excellent enantioselectivities. The C6'-OH group of β-isocupreidine (β-ICD) is believed to facilitate the key proton transfer step in the MBH reaction, via an intramolecular proton relay process.

Abstract:The first highly enantioselective decarboxylative addition of beta-ketoacids to isatins mediated by a bifunctional tertiary amine-thiourea catalyst has been developed, allowing facile synthesis of biologically important 3-hydroxy oxindoles In good yields and excellent enantioselectivities. Abstract:The organocatalytic asymmetric Morita-Baylis-Hillman (MBH) reaction of isatin derivatives with various vinyl sulfones is disclosed. Chiral sulfone-containing 3-hydroxyoxindoles were produced in good to high yields and with good to high ee's. This report displays an unprecedented example to apply activated alkenes with sulfone moiety other than carbonyl groups in asymmetric MBH reactions and provides an efficient strategy to incorporate the sulfone functional group for the synthesis of chiral 3-hydroxyoxindoles.

Abstract:A highly regio- and stereoselective [3 + 2] cycloaddition reaction for constructing novel 3,3'-cyclopentenyldispirooxindoles incorporating two adjacent quaternary spirostereocenters is reported. Under the mild conditions, the asymmetric annulation of isatin-derived MBH carbonates with 3-methyleneoxindoles involving a chiral tertiary amine catalyst provides the corresponding dispirooxindole frameworks with an extraordinary level of enantioselective control. Further synthetic utility of this method was demonstrated by the gram-scale experiment and simple transformation of the obtained product. Moreover, a plausible mechanism for this annulation reaction was also proposed on the basis of the control experiments.

Abstract:Nickel‐catalyzed asymmetric intramolecular addition of aryl halides to α‐ketoamides has been achieved to afford chiral 3‐substituted‐3‐hydroxy‐2‐oxindoles in excellent yields and high enantioselectivities (up to 99% yield and 98% ee), which provides efficient access to valuable molecules containing 3‐hydroxy‐2‐oxindole core. The gram‐scale reaction proved the potential utility of the methodology.

Abstract:The catalytic enantioselective alkynylation of isatins, catalyzed by a Zn(OCOCF3)(2)/chiral bis(sulfonamide)-diamine (BSDA) catalyst under mild condition, has been developed. The desired chiral 3-alkynyl-3-hydroxy-2-oxindoles were obtained in moderate to excellent yields (up to 96%) and good to excellent enantioselectivites (up to 97% ee).
