Abstract:The Winterfeldt oxidation (NaOH, DMF, air, rt) of substituted 1,2,3,4-tetrahydro-gamma-carbolines has been developed, which provides a convenient and efficient method for the synthesis of the corresponding dihydropyrrolo[3,2-b]quinolones in moderate to excellent yields (38-94%). The generality and substrate scope of this reaction are explored and a possible mechanism is proposed. The results imply that electron-withdrawing groups on N-2 of tetrahydro-gamma-carbolines and N-5-H are necessary. The synthesis of 5 or 7-substituted pyrrolo[3,2-b]quinolones in near quantitative yields was also achieved through deprotection and aromatization of N-1-Boc-dihydropyrrolo[3,2-b]quinolones.

Abstract:The synthesis of diversely substituted quinazoline-2,4(1H, 3H)-diones by cyclization of tert-butyl (2-cyanoaryl) carbamates using readily accessible Boc protected o-amino nitriles is reported. The reaction proceeds smoothly at room temperature using just 1 equiv. of H2O2 as an oxidant under basic conditions. This reaction is compatible with a variety of aromatic/heteroaromatic substrates with different functional groups. This strategy can be utilized for simplified synthesis of Goshuyuamide II and an alkaloid isolated from Zanthoxylum arborescens in good yield. This method was also applied to the synthesis of quinazoline-2,4(1H, 3H)-diones that are precursors to medicinally important compounds: Alfuzosin, Terazosin, Prazosin, Terazosin, IAAP, Doxazosin, FK 366 (Zenarestat) and KF31327.

Abstract:The resin-bound cyclic malonic acid ester 2 reacted with aryl isothiocyanate and alkyl halides to afford the key intermediate arylthioaminomethylene cyclic malonic ester resin 3. Subsequently, resin 3 proceeded thermal cyclization giving the 2-alkylthio-4(1H)-quinolones in good yields and excellent purities.

Abstract:A rhodium-catalyzed highly stereoselective formal [2 + 4]-cycloaddition reaction of α-diazo pyrazoleamides and 2-aminophenyl ketones that produces 4-hydroxy-2-quinolinones in good yields with excellent diastereoselectivities has been developed. A pyrazolium ylide species that is generated from α-diazo pyrazoleamides is used as a C2 synthon for this cycloaddition. This protocol offers an efficient approach to a variety of 4-hydroxy-2-quinolinones featuring sequential quaternary centers.

Abstract:A highly efficient method for the synthesis of 1,2,3,4-tetrahydroquinoline derivatives via a molecular iodine catalyzed domino reaction of anilines with cyclic enol ethers, such as 2,3-dihydrofuran and 3,4-dihydro-2H-pyran, is described. The reaction may proceed through an aza-Diels–Alder process between an in situ generated 2-azadiene and another equivalent of cyclic enol.

Abstract:A very mild protocol for fixation of carbon dioxide with 2-alkynamides in DMSO at 30 °C using a CO(2) balloon in the presence of K(2)CO(3) has been developed, which leads to an efficient assembly of oxazolidine-2,4-diones. It is observed that the regioselectivity was controlled by the aryl group.

Abstract:In AgSbF6/Rh-2(OAc)(4)/DCM system, two-component reaction of 2-diazo-3,5-dioxo-6-ynoates (sulfones) and two equivalent of aldehydes provided easy access to 2-(1,3-dioxolanes-4-yl)-4H-pyran-4-ones. This represents the first method for generating the novel heterocyclic compounds. A possible mechanism is proposed to explain this result. The most significant features of the tandem cyclization are simple procedure, mild conditions and high diastereoselectivity.

Abstract:A facile and efficient method for the synthesis of 3-aryl-4-hydroxy-1,3-thiazolidin-2-ones by the reaction of 1,4-dithiane-2,5-diol with acyl azides is reported.
