Abstract:The four stereoisomers 11,12,15(S)-THETA were prepared by a triply convergent, asymmetric route that exploited the stereospecific, copper mediated cross-coupling of α,β-dialkoxystannanes and the utility of dialkylthionocarbamates as orthogonal alcohol protective groups. Only 11(R),12(S),15(S)-THETA was comparable to natural material by HPLC, GC/MS, and in vitro bioassay.

Abstract:The stereochemistry of the major isomer of 14,15-dihydroxy-5,8,10,12-eicosatetraenoic acid formed from 15-hydroperoxyeicosatetraenoic acid in human leukocytes was determined. The structure (erythro-14(R),15(S]-14,15-dihydroxy-5,8-cis-10,12-trans-eicosatetraenoi c acid) was assigned based on sodium arsenite thin-layer chromatography, NMR spectroscopy, and comparison with material prepared by total synthesis. This compound was found to inhibit leukotriene B4-induced superoxide anion generation in human neutrophils (IC50 = 10(-8)-10(-7) M). Superoxide anion generation induced by either formylmethionyl-leucyl-phenylalanine or arachidonic acid was not affected.

Abstract:(2S,3aR,7aS)-Perhydroindolic acid, the key intermediate in the synthesis of trandolapril, and its trans-isomers, were readily prepared. These proline-like molecules are unique in that they contain a rigid bicyclic structure, with two hydrogen atoms trans to each other at the bridgehead carbon atoms. These molecules were used successfully as chiral organocatalysts in asymmetric domino Michael addition/cyclization reactions of aldehyde esters with beta,gamma-unsaturated a-keto esters. They proved to have excellent catalytic behavior, allowing for the synthesis of multi-substituted, enantiomerically enriched hemiacetal esters. Under optimal conditions (using 10 mol% catalyst loading), a series of beta,gamma-unsaturated a-keto esters was examined with up to 99% de, ee and yield, respectively. Additionally, the enantiomerically enriched hemiacetal esters could be readily transformed into their corresponding bioactive pyrano[2,3-b]pyrans (possessing a multi-substituted bicyclic backbone).

Abstract:The C16-C22 fragment with the acetylene terminus was constructed through the asymmetric dihydroxylation of the corresponding olefin, while the 15-iodo-olefin corresponding to the C11-C15 part was prepared via the asymmetric transfer hydrogenation of the corresponding acetylene ketone followed by hydrozirconation/iodination. Both pieces were joined by a Sonogashira coupling, and the product was further converted into the title compound via a Wittig reaction with the remaining C1-C10 segment and Boland reduction using Zn with TMSCl.

Abstract:A versatile strategy featuring a Colvin rearrangement, hydrozirconation, a Sonogashira cross-coupling reaction and a Z-selective Wittig olefination, was successfully developed for the construction of a conjugated E,E,Z-triene subunit, flanked on both sides by two Z-allylic hydroxyl groups. This chemical pattern is found in many endogenous lipid metabolites such as maresin 1 (MaR1), neuroprotectin D1 (NPD1), and its aspirin triggered-isomer AT-NPD1, which not only counter-regulate inflammation but also actively orchestrate (at nanomolar doses) the resolution and termination program of acute inflammation while promoting wound healing, return to homeostasis and neuroprotection. Unlike previous approaches, the advantages of the present strategy are obvious, as it allows us to modify the nonpolar tail, the carboxylated head or both ends of the molecule without repeating the whole synthetic sequence (about 26-34 steps according to the literature). Thus, the first total syntheses of NPD1 methyl ester epimer (which can also be considered as an enantiomer of AT-NPD1) and its n-3 docosapentaenoic acid derived analogue were achieved from a highly functionalized and late advanced pivotal intermediate. This innovative route may be easily adapted to gain access to other dihydroxylated metabolites and analogues of polyunsaturated fatty acids containing a conjugated E,E,Z-triene subunit. Different epimers/diastereoisomers may be obtained by purchasing the suitable optically pure (S)- and/or (R)-1,2,4-butanetriol(s) as a chiral pool for both stereogenic centers.

Abstract:The catalytic enantioselective desymmetrizing bromoaminocyclization of prochiral cyclohexa-1,4-dienes has been achieved by using chiral anion phase-transfer catalysis, providing a range of enantioenriched cis-3a-arylhydroindoles bearing an all-carbon quaternary stereocenter in good yields (up to 78%) and excellent enantioselectivities (up to 97% ee). Abstract:A new procedure has been developed for synthesizing enantiomerically pure beta-amino acids, alpha-hydroxy-beta-amino acids and certain alkaloids from aspartic acid. By protection, anhydride formation and regioselective reduction, L-aspartic 10 is converted to the N-tosylamino lactone 12. Hydroxylation of 12 by an oxaziridine gives the trans-2-hydroxy-3-N-tosylamino derivative 20. Opening of 12 and 20 by trimethylsily iodide and ethanol affords the iodo-homoserine esters 13 and 21 respectively. Submission of 13 and 21 to Gilman reagents followed by saponification and deprotection gives 4-substituted 3-amino and 3-amino-2-hydroxybutyric acids (15 and 23), exemplified by the syntheses of cyclohexylnorstatine (25), and the components of bestatin (23, R=Ph) and microginin (14). Certain beta-amino acids are transformed into solenopsin A (33) and indolizidine 209D (41).

Abstract:The first total syntheses of four stereoisomers of 13-hydroxy-14-methylhexadecanoic acid have been accomplished. Central to this strategy are asymmetric alkynylation of aldehyde, acid-catalyzed lactonization, the selective protection of primary alcohol and Wittig reaction. The product 1a was obtained in 17 steps in 2% overall yield. Moreover, these synthetic chiral hydroxy fatty acids 1a-1d are valuable for the development of antibacterial agents.

Abstract:Enantioselective total synthesis of (+)-sapinsigin H, (+)-agallochaol C, and (+)-16α, 17-dihydroxy-atisan-3-one have been accomplished starting from enantiopure Wieland–Miescher ketone. Key features of the syntheses include a benzannulation step to construct the tricyclic core, an oxidative dearomatization step to generate the diene, and a Diels–Alder reaction with ethylene gas to establish the bicyclo[2.2.2]octane framework. Efficient late-stage functionalisation of the A-ring by aerobic oxidation and Baeyer−Villiger oxidation completed the atisane target molecules.

Abstract:A series of novel 2-[(4,6-dimethylpyrimidin-2-yl)oxy]-3,3-diphenyl butyric acid derivatives were synthesized and evaluated for their antagonistic activity for endothelin-1-induced contraction in rabbit aorta. Within this series of compounds, 2-[(4,6-dimethylpyrimidin-2-yl)oxy]-3-cyano-3,3-diphenylpropionic acid (4) displays comparable potency with ambrisentan (1), and warrants further investigation.

Abstract:The first total synthesis of both C-11 epimers of 13-hydroxy-α-eudesmol 1a and 1b by the use, as a key reaction, of the Sharpless asymmetric dihydroxylation of alkene 7 is presented. The absolute configuration of natural 13-hydroxy-α-eudesmol is established through comparison of the 1H NMR spectrum of natural diol and synthetic diols. In our synthesis another natural product (+)-α-selinene 2 has also been accomplished.

Abstract:The enantioselective syntheses of (-)-indolizidine 167B and (+)-coniine were described based on the asymmetric hydrogenation of racemic d-hydroxy esters via kinetic resolution. With optically active chiral d-hydroxy ester (S)-4 and chiral 1,5-diol (R)-5 obtained by asymmetric hydrogenation of racemic ethyl 5-hydroxyoctanoate (rac-4) with chiral spiro iridium catalyst Ir-(R)-SpiroPAP as chiral starting materials, the efficient enantioselective syntheses of (-)-indolizidine 167B and (+)-coniine were achieved by using intramolecular reductive amination and N-substitution/cyclization, respectively, as a key step to construct the chiral aza-bicyclic[4.3.0]nonane skeleton and chiral piperidine ring. This provides new efficient methods for enantioselective syntheses of indolizidine and piperidine alkaloids.y

Abstract:The concise total syntheses of the potent HIV inhibitors aetheramides A and B (IC50 values of 15 and 18 nM), as well as three pairs of their stereoisomers, were achieved, which allowed the complete stereochemical assign ment of aetheramides for the first time. With a longest linear sequence of 15 steps, the convergent, fully stereocontrolled route provided aetheramides A and B in 5.3% and 3.6% yields, respectively. The synthetic strategy features efficient Stille coupling for macrocyclization, asymmetric aldol reactions to establish the ambiguous stereochemistries at C-17 and C-26, and implementation of mild conditions to avoid the epimerization of the sensitive polyketide moiety and the migration of the labile lactone.

Abstract:The A/B/C ring intermediate of antithrombotic actinophyllic acid methyl(S)-3-(2-(2-chloroacetyl)-2′,2′-dimethyl-1,2,3,4,5,7-hexahydrospiro[azocino[4,3-b]indole-6,5′-[1,3]diox-an]-5-yl)-3-oxopropanoate(9)was synthesized from(S)-3-(2,2-dimethyl-5-(3-methyl-1H-indol-2-yl)-1,3-dioxan-5-yl)pyrrolidin-2-one via the key steps of reductive ring-opening of pyrrolidone, the tandem hydrogenative deprotection/intramolecular reductive amination, etc.9 was further converted into the A/B/G ring intermediate methyl(S)-7-(2-(2-chloroacetamido)ethyl)-2′,2′-dimethyl-8-oxo-7,8-dihydro-5H-spiro [cyclohepta[b]indole-6,5′-[1,3]dioxane]-9-carboxylate(10)by oxidation. Nine novel compounds were involved in the synthetic route.The structures were characterized by 1 H NMR, 13C NMR,2D NMR and LC-MS(ESI).
