Abstract:Indole-2-carboxylic acid and indoline-2-carboxylic acid are common building block in natural products with bioactivities and anti-hypertension pharmaceuticals. The syntheses and transformation from each other were reviewed. Indoline-2-carboxylic acid can be got from the reduction of indole-2-carboxylic acid and the S enantiomer can be achieved from the resolution of the racemic mixture with chemical reagents or enzymes. The chiral pool synthesis, emerged recently, will play an important role in the future. Indole-2-carboxylic acid can be formed from Fischer way or substituted anilines and benzaldehydes.

Abstract:AIM:To design and synthesize novel AchE inhibitors.METHODS:The condensation of 2-bromo-5, 6-dimethoxy-indan-1-one with various aminoalkyl phenols in the presence of K2CO3 and acetonitrile gave the corresponding title compounds, and the in vitro AchE and BchE inhibitory activities were evaluated by the modified Ellman method.RESULTS:Sixteen novel target compounds 8a - p were synthesized, their structures were confirmed by 1H NMR, MS, IR and elemental analysis. Preliminary pharmacological test demonstrated that most of these compounds displayed high AchE inhibitory activities, the IC50 of the most potent inhibitor 8h was 50.0 nmol x L(-1), similar to that of Huperzine A (IC50 = 53.0 nmol x L(-1)), while all the compounds were almost inactive against BchE.CONCLUSION:2-Phenoxy-indan-1-one derivatives exhibit high activities of AchE inhibition and are worthy of further investigation.

Abstract:A series of novel 3-substituted-indole derivatives with a benzyl tertiary amino moiety were designed, synthesized and evaluated as H3 receptor antagonists and free radical scavengers for Alzheimer's disease therapy. Most of these synthesized compounds exhibited moderate to potent antagonistic activities in CREs driven luciferase assay. In particular, compound 2d demonstrated the most favorable H3 receptor antagonistic activity with the IC50 value of 0.049μM. Besides, it also displayed high binding affinity to H3 receptor (Ki=4.26±2.55nM) and high selectivity over other three histamine receptors. Moreover, 2d and other two 3-substituted indole derivatives 1d and 3d exerted potent ABTS radical cation scavenging capacities similar to melatonin. Above results illustrate that 2d is an interesting lead for extensive optimization to explore new drug candidate for AD therapy.

Abstract:A series of aromatic acetic acid compounds were designed and synthesized on the basis of Non-steroidal anti-inflammatory drugs indomethacin and diclofenac. Compounds 5a, 7a, 5h, 7h and 17 could strongly promote insulin-regulated differentiation of 3T3-L1 cells in vitro. They acted as full or partial PPARγ agonist, or improved insulin resistance through non-PPARγ pathway.

Abstract:A series of novel indene-derived retinoic acid receptor α (RARα) agonists have been designed and synthesized. The use of receptor binding, cell proliferation and cell differentiation assays demonstrated that most of these compounds exhibited moderate RARα binding activity and potent antiproliferative activity. In particular, 4-((3-isopropoxy-2,3-dihydro-1H-inden-5-yl)-carbamoyl)benzoic acid (36d), which showed a moderate binding affinity, exhibited a great potential to induce the differentiation of NB4 cells (68.88% at 5 μM). Importantly, our work established indene as a promising skeleton for the development of novel RARα agonists.

Abstract:The title compound, C(19)H(19)NO(10), was obtained from the reaction of α-d-1-bromo-2,3,4-tri-O-acetylxylose with N-hy-droxy-phthalimide in the presence of potassium carbonate. The asymmetric unit contains two independent mol-ecules, in which the O-CH-O-N torsion angles are 73.0 (4) and 65.0 (4)°. The hexa-pyranosyl rings adopt chair conformations and the substituent groups are in equatorial positions. In the crystal, mol-ecules are linked by nonclassical C-H⋯O hydrogen bonds.

Abstract:Objective: In this paper we have reported the synthesis of novel chromeno[2,3- d ]pyrimidine-2,4(3 H )dione derivatives ( IVa–IVh ) by three component reaction between barbituric acid, indole and aromatic aldehydes useing L-Proline as a catalyst. Methods: The structures of synthesized compounds were confirmed by spectroscopic techniques. The synthesized compounds were examined for Acetyl CoA inhibitory activity. Results and Discussion: Compound ( IVb ) showed good activity as compared to other compounds. Also, the synthesized compounds were examined for Anti-inflammatory activity against MMP-2 and MMP-9 using gelatin zymography; in which compound ( IVd ) exhibited very good efficacy viz inhibited MMP-2 and MMP-9 to 80 and 50%, respectively. Furthermore, the molecular docking studies were carried out using a 1PG4 receptor. Conclusions: From the molecular docking study it was found that compound IVa exhibits a best dock confirmation with higher binding affinity (−11.0 kcal/mol) by establishing 3 hydrogen bonds.

Abstract:Thiazolin-4-ones and their derivatives represent important heterocyclic scaffolds with various applications in medicinal chemistry. For that reason, the synthesis of two 5-substituted thiazolidin-4-one derivatives was performed. Their structure assignment was conducted by NMR experiments (2D-COSY, 2D-NOESY, 2D-HSQC and 2D-HMBC) and conformational analysis was conducted through Density Functional Theory calculations and 2D-NOESY. Conformational analysis showed that these two molecules adopt exo conformation. Their global minimum structures have two double bonds (C=N, C=C) in Z conformation and the third double (C=N) in E. Our DFT results are in agreement with the 2D-NMR measurements. Furthermore, the reaction isomerization paths were studied via DFT to check the stability of the conformers. Finally, some potential targets were found through the SwissADME platform and docking experiments were performed. Both compounds bind strongly to five macromolecules (triazoloquinazolines, mglur3, Jak3, Danio rerio HDAC6 CD2, acetylcholinesterase) and via SwissADME it was found that these two molecules obey Lipinski's Rule of Five.

Abstract:Aim To synthesize 5'(S)-1,1-dioxolane-5-oxo-(5'-ethyl-5'-hydroxyl-2'H,5'H,6'H-6-oxopyrano)-[3',4'-f]-Δ6(8)-tetrahydroindolizine,a key intermediate for the total synthesis of anticancer drugs 20(S)-camptothecin and its deriveatives.Methods Target compound was synthesized from 1,1-dioxolane-5-oxo-(5'-ethyl-2'H,5'H,6'H-6-oxopyrano)-[3',4'-f]-Δ6(8)-tetrahydroindolizine,via a three-step process including iodine catalyzed oxidation,kinetic resolution and base-hydroxylation.Results and conclusion The synthetic procedure was simple to manipulate and the target compound is obtained with ee 96% optical purity in 15.7% overall yield.
