Abstract:A regio-, diastereo- and enantioselective 1,3-dipolar cycloaddition reaction between nitrones and electron-deficient alkenes has been developed employing C 1 -symmetrical Ti(OTos) 2 - and TiCl 2 -TADDOLate as catalysts. In the presence of powdered 4 Å molecular sieves and 100 mol% of Ti(OTos) 2 -TADDOLate as the catalyst, the nitrones smoothly react with alkenes at room temperature to give isoxazolidines in high conversion, giving the trans-diastereomer of the isoxazolidines with de's up to >90% and ee up to 58%. Furthermore, C 1 -symmetrical TiCl 2 -TADDOLate has a better chiral induction, giving the trans-diastereomer of the isoxazolidines with high enantioselectivity up to 70% ee. The influence of catalyst amount, temperature and solvents on the reaction course has been investigated

Abstract:1,3-Dipolar cycloaddition is a commonly exploited method to access 5-membered chemical entities with a variety of peripheral functionalities and their stereochemical arrangements. Nitrones are isolable 1,3-dipoles that exhibit sufficient reactivity toward electron-deficient olefins in the presence of Lewis acids to deliver highly substituted isoxazolidines. Herein we document that α,β-unsaturated amides, generally regarded as barely reactive in a 1,3-dipolar reaction manifold, were effectively activated using the designed 7-azaindoline auxiliary in an In(OTf)3 /bishydroxamic acid catalytic system. The broad substrate scope and clean removal of the 7-azaindoline auxiliary from the product highlight the synthetic utility of the present catalysis.

Abstract:The first nitro-group-initiated redox-neutral [3+2] cycloaddition of nitrocyclopropanes with alkenes by using visible-light-absorbing transition-metal complexes was reported. High diastereoselectivities were observed for two quaternary carbon centers on the ring and validated by DFT calculations. Spiro- or polycyclic structures can be constructed smoothly. Cyclic γ-amino acid derivatives and polysubstituted cyclic amino alcohols can be obtained easily through reduction of the nitro group.

Abstract:A selective synthesis of either 2H-pyran-2-ones (4) or chiral dihydropyranones (6) from the same substrates of 3-bromoenals and 1,3-dicarbonyl compounds upon oxidative N-heterocyclic carbene catalysis is presented. It is shown that the oxidative transformation of 3-bromoenals under NHC catalyst can be well controlled to proceed through two pathways, i.e., elimination of reducible beta-bromide or by an external oxidant 3, allowing the selective generation two sorts of unsaturated acyl azoliums, respectively.

Abstract:Efficient strategies to assemble enantioenriched pyridine derivatives are highly important, given their significance in both synthetic and medicinal chemistry. Here, we report an enantioselective nickel-catalyzed intermolecular [2 + 2 + 2] cycloaddition of alkyne-tethered malononitriles with alkynes for the synthesis of densely substituted pyridines. The α-all-carbon quaternary center adjacent to pyridine is introduced by desymmetrizing the two cyano groups of disubstituted malononitriles. Notably, terminal alkynes are also tolerated with good regioselectivity to afford tetrasubstituted pyridines. Zinc halide is essential to enable the occurrence of the transformation by promoting the hetero-cyclometallation step. The reaction uses bio-renewable 2-MeTHF as the solvent and features mild reaction conditions, good functional group compatibilities, and enantioselectivities. This study offers a straightforward approach to the valuable enantioenriched heteroarenes from feedstock chemicals by forming three bonds and one quaternary stereocenter simultaneously in a single reaction step.

Abstract:The first catalytic asymmetric decarboxylative [3 + 2] cycloaddition reaction of ethynylethylene carbonates with malononitrile has been developed successfully by an organo/copper cooperative system. This strategy led to a series of optically active polysubstituted dihydrofurans in good yields with high levels of enantioselectivities (up to 99% yield, 97% ee). The presence of the terminal alkynyl and the cyano group in the dihydrofuran products provides a wide scope for further structural transformations. More importantly, this organo/metal cooperative catalytic system will broaden the substrate scope and enable fundamentally new reactions.

Abstract:Alkynyl Fischer carbene complexes were found to undergo chemoselective, regioselective, and rate-enhanced 1,3-dipolar cycloaddition with nitrones to give 2,3-dihydroisoxazole carbene complexes in excellent yields. These alkynyl complexes can serve as synthons for substituted propiolate esters since the metal pentacarbonyl group of the cycloadducts can be easily oxidatively removed with DMSO. The tungsten carbene complex 4b reacted with 3 different series of N-alkyl nitrones: N-tert-butyl 2a-f, N-methyl 1a-h and N-benzyl 3 to give cycloadducts 6a-f, 7a-h, and 8-9, respectively, while the chromium carbene complex 4a reacted only with N-tert-butyl nitrones 2a-g to give isolable cycloadducts 5a-g. The structure of N-methyl cycloadduct 7d has been established by a single-crystal X-ray analysis. The reactivity of N-alkyl nitrones toward carbene complexes was in the order: N-Me > N-Bn > N-(t)Bu. In addition, our results showed that the rate of cycloaddition reaction increased as the electron donating ability of para-substituent in N-tert-butyl nitrone 2a-f increased.

Abstract:The high nitrogen content and diverse reactivity of malononitrile are widely harnessed to access nitrogen-rich fine chemicals. Although the facile substitutions of malononitrile can give structurally diverse quaternary carbons, their access to enantioenriched molecules, particularly chiral amines that are prevalent in bioactive compounds, remains rare. Here we report a cobalt-catalysed desymmetric reduction of disubstituted malononitriles to give highly functionalized β-quaternary amines. The pair of cobalt salt and sodium borohydride is proposed to generate a cobalt-hydride intermediate and initiate the reduction. Meanwhile, the enantiocontrol of the dinitrile is achieved through a tailored bisoxazoline ligand with two large flanks that create a narrow gap to host the bystanding nitrile and thus restrict the C( ipso )−C(α) bond rotation of the complexed one. Combined with the extensive derivatization possibilities of all substituents on the quaternary carbon, this asymmetric reduction unlocks pathways from malononitrile as a bulk chemical feedstock to intricate, chiral nitrogen-containing molecules.

Abstract:A diastereo- and enantioselective formal [3+2] cycloaddition of N-tosylaziridines and aldehydes catalyzed by a Ni(II)-bisoxazoline complex has been accomplished. The 1,3-oxazolidine products are obtained with high diastereoselectivity, good yields (up to 99%) and high ee values (up to 96% ee). The challenging long distant stereocenter control is achieved by a chirality transfer approach.

Abstract:Highly enantioselective Michael addition of malonates to enones catalyzed by dipeptide-derived multifunctional phosphonium salts has been developed. The newly established catalytic system was characterized with its wide substrate scope featured with aliphatic aldehyde-derived enones and substituted malonates. The gram scale-up synthesis of adducts can also be successfully achieved under optimal conditions with both excellent yield and enantioselectivity.
