Abstract:Octopamine (4-(2-amino-1-hydroxy-ethyl)-phenol hydrochloride) acts as a neurotransmitter, neurohormone,or neuromodulator in invertebrate nerve tissue, and is an ideal target for development of highly active, highly selective,and biorationally designed pesticides. In addition, octopamine has the ability to strengthen body metabolism and curediseases such as neurasthenia. Phenol and aminoacetonitrile hydrochloride undergo nitrile acylation in the presence ofanhydrous aluminum tricloride, followed by hydrolysis to yield a carbonylated product, and then a Raney nickelcatalyzed hydrogenation reaction to give the final product with an overall yield of 65%.

Abstract:An improved process was reported to solve the problems such as low yield and high costs in the synthesis of 3-n-propyl-2, 4-dihydroxyacetophenone, which is a key intermediate of Leukotriene receptor antagonist. The target compound was synthesized starting from m-dihydroxybenzene, via acylation, allylation, Claisen rearrangement and hydrogenation reduction. The optimal reaction conditions were as follows. The acylation reaction which used acetic acid as acylation agent and ZnCl 2 as catalyst was carried out under reflux for 5 h, and 77.9% yield was obtained. The allylation reaction using allyl chloride as allylation reagent and NaBr, KI as catalyst was carried out at 40°C for 2 h and continued at 70°C for 14 h, to give 98.6% yield. In Claisen rearrangement carried out at 205°C for 4.5 h, diphenyl ether was used as reaction solvent and NaCl was used as catalyst, and the yield was improved to 67.4%. In the final step, the hydrogenated reduction with Pd/C (w(Pd) is 5%) as catalyst was carried out at 45°C for 6 h, and 88.6% yield was obtained. The process is much of industrial value because of high yield, cheap and available materials, and convenient operations. The structure of the main compounds was confirmed by 1H-NMR, GC-MS and IR.

Abstract:A procedure for synthesis of 2-methoxyphenothiazine (1) has been developed, starting from resorcinol and aniline by condensation, following methylation and cyclization. p-Toluenesulfonic acid and polysubstituted aromatics were employed as the catalyst of condensation and the solvent of cyclization, respectively, to improve the yield. The use of parallel experiments, statistical experimental design, and multivariate modeling made the total yield of the procedure as high as 74.2%. J. Heterocyclic Chem., (2012).

Abstract:ZJ273,propyl 4-[2-[(4,6-dimethoxy-2-pyrimidinyl)oxy]benzylamino]benzoate,is a novel herbicide.The microsynthetic method of the compound was developed from 4,6-dimethoxy-2-pyrimidine,propyl(4-benzylamino) benzoate and phenol,and then purified by preparative-HPLC.The yield of end product was 53%.Its chemical structure was characterized by means of()~1H-NMR,MS,UV,and its chemical purity was determined to be exceed 98% by HPLC.The microsynthetic method of propyl 4[2-[(4,6-dimethoxy-2-pyrimidinyl)oxy] benzylamino]benzoate developed sets up a basis to synthesize the radioisotopic labeled target compound.

Study on the Synthesis of L-4-Nitrophenylalanine

Abstract:L-4-Nitrophenylalanine is an important intermediate in many medicine syntheses. It is commonly synthesized by the nitration of L-phenylalanine with mixed acid nitrate composed of concentrated H2SO4 and concentrated HNO3 in a batch reactor; and the yield of L-4-Nitrophenylalanine can reach 65.2% when the volume ratio of concentrated H2SO4 and concentrated HNO3 of 2:1 is used at reaction temperature of 0℃ with reaction time of 3 h. The structure of by-product was identified with 1H-NMR and 13C-NMR, and the by-product was proved to be a condensation product of L-4-nitrophenylalanine and L-2-nitrophenylalanine. Based on the by-product structure found, the possible nitration mechanism was proposed. In order to depress the side reaction, the tubular reactor was used for the nitration of L-phenylalanine. The orthogonal experiments were adopted to optimize the reaction conditions of nitration with tubular reactor, and the results show that, under the found optimized conditions of the volume ratio of concentrated H2SO4 and concentrated HNO3 equal to 2:1, the reaction temperature of 50℃ and reaction time of 5 min, the L-4-nitrophenylalanine yield could reach 80.9%. Compared with the batch nitration, the nitration of L-phenylalanine with tubular reactor is more adoptable for it can free of dimerization of L-4-nitrophenylalanine and L-2-nitrophenylalanine; moreover, it can also get a higher reaction rate and enhance the reaction efficiency.

Abstract:3, 3-Dimethoxypropanenitrile was synthesized from 2-chloro-3-phenylsulfonylpropanenitrile by elimination and addition reactions. 3-phenylsulfonylpropanenitrile was obtained by eliminating HCl from 2-chloro-3-phenylsulfonylpropanenitrile at 0-20℃ with a yield of 88%. 3-Phenylsulfonylpropanenitrile was reacted with sodium methoxide in methanol, giving 3, 3-dimethoxypropanenitrile with a yield of 98%. Elimination reaction was innovated by using THF as a solvent instead of ethyl ether and addition reaction was innovated by using an excess of sodium methoxide in methanol, so as to have the reaction condition optimized.

These compounds 3a-3p were synthesized in good yield by refluxing alpha-aminoacetophenones and hydrazines in glacial acetic acid. Abstract:A novel and convenient one-step method for the synthesis of 1,3,4,-Triaryl-substituted-3,4,5,6-tetrahydropyrimidin-2(1H)-ones (3) was developed by cyclization of aromatic isocyanates (1) with 3-arylamino-1-phenylpropan-1-ones (2) under refluxing of toluene. Effects of reaction conditions on the converting ratio and yield were investigated. The method offered several advantages including simple experimental procedure, easily available starting materials and excellent yields (79%similar to 96%).

Abstract:The title compound, C9H15NOS, was prepared by asymmetric synthesis from 3-dimethylamino-1-(2-thienyl) propan-1-one hydrochloride. In the molecule, the essentially planar CCCN chain makes a dihedral angle of 16.7 (3)degrees with the thiophene ring. In the crystal structure, molecules are linked by O-H center dot center dot center dot N hydrogen bonds to form a chain along the b axis.

Abstract:A convenient one-pot synthesis of enantiopure (R)-2-amino-3,3,3-trifluoro-2-methyl-N-phenylpropanamide derivatives has been developed. The key step in this synthetic methodology turned out to be the amide formation in which (R)-2-amino-3,3,3-trifluoro-2-methylpropanoic acid hydrochloride was simultaneously protected and activated by Vilsmeier reagent.
